    A high resolution wind&wave forecast model chain for the Mediterranean and Adriatic Sea

    DHI (Danish Hydraulic Institute) and HyMOLab (Hydrodynamics and Met-Ocean Laboratory of the Dept. of Engineering and Architecture of the University of Trieste) have undertaken a joint applied research project with the aim to develop a state-of-art wind-wave forecast service at mid resolution for the Mediterranean Sea and at very high resolution for the Adriatic Sea. Weather routing, civil protection, coastal engineering, oil&gas and renewable energy fields, the planning of operations at sea, ... are just few among the multiple potential applications of this service. The meteorological model used in this study is WRF-ARW, one of the most widely used state-of-the-art open-source non-hydrostatic model. Global Forecast System (GFS) dataset provides the boundary and initial conditions. MIKE21-Spectral Waves is used as wave model with resolution ranging from 0.1 to 0.03 approximately. The use of a local area meteorological model guarantees higher levels of resolution and accuracy in an area such as the Mediterranean Sea where the complex orography and coastline induce short-time/small-space weather scales. The model chain runs daily (or twice a day on demand) on the High Performance Computing (HPC) infrastructure of HyMOLab. The validation of the entire model chain and specifically the forecast data obtained for the sea state is continuously updated according to new available data from satellites and buoys. Anyway, a major verification of the performance of the model chain against historic data (hindcast) is almost mandatory. For this aim, we performed a multi-decade test obtaining very good statistical parameters for the entire model chain performance. In this context the hindcast dataset developed by DHI and HyMOLab consists of 35 years of hourly data for the period 1979-2013, with the same model chain. The CFSR d093.0 hourly dataset with a spatial resolution of 0.5 provides the boundary and initial conditions. The atmospheric and wave models performance is checked against six satellite datasets, missions Envisat, ERS-2, Geosat FO, Jason-1, Jason-2, Topex-Poseidon, using a moving window technique procedure. Wave data close to coast are compared with available data from more than 20 buoys. The paper describes the validation procedure adopted for the hindcasted data. Furthermore the forecast service is described too, with specific emphasis to the very high resolution adopted in the Adriatic Sea

    Infectious Keratitis: Characterization of Microbial Diversity through Species Richness and Shannon Diversity Index

    Purpose: To characterize microbial keratitis diversity utilizing species richness and Shannon Diversity Index. Methods: Corneal impression membrane was used to collect samples. All swabs were processed and analyzed by Biolab Laboratory (level V—SSN Excellence: ISO 9001:2015), Biolab Srl (Ascoli Piceno, Italy). DNA extraction, library preparation, and sequencing were performed in all samples. After sequencing, low-quality and polyclonal sequences were filtered out by the Ion software. At this point, we employed Kraken2 for microbial community analysis in keratitis samples. Nuclease-free water and all the reagents included in the experiment were used as a negative control. The primary outcome was the reduction in bacterial DNA (microbial load) at T1, expressed as a percentage of the baseline value (T0). Richness and Shannon alpha diversity metrics, along with Bray–Curtis beta diversity values, were calculated using the phyloseq package in R. Principal coordinate analysis was also conducted to interpret these metrics. Results: 19 samples were included in the study. The results exhibited a motley species richness, with the highest recorded value surpassing 800 species. Most of the samples displayed richness values ranging broadly from under 200 to around 600, indicating considerable variability in species count among the keratitis samples. Conclusions: A significant presence of both typical and atypical bacterial phyla in keratitis infections, underlining the complexity of the disease’s microbial etiology

    Social marketing and healthy eating : Findings from young people in Greece

This research provides lessons for social marketing to tackle the severe nutrition-related problems in this country by obtaining insight into the eating behaviour of young adults aged 18–23. Also, the main behavioural theories used to inform the research are critically discussed. The research was conducted in Athens. Nine focus groups with young adults from eight educational institutions were conducted and fifty-nine participants’ views towards eating habits, healthy eating and the factors that affect their food choices were explored. The study found that the participants adopted unhealthier nutritional habits after enrolment. Motivations for healthy eating were good health, appearance and psychological consequences, while barriers included lack of time, fast-food availability and taste, peer pressure, lack of knowledge and lack of family support. Participants reported lack of supportive environments when deciding on food choices.     Cytomolecular identification of individual wheat-wheat chromosome arm associations in wheat-rye hybrids

    Chromosome pairing in the meiotic metaphase I of wheatrye hybrids has been characterized by sequential genomic and fluorescent in situ hybridization allowing not only the discrimination of wheat and rye chromosomes, but also the identification of the individual wheat and rye chromosome arms involved in the chromosome associations. The majority of associations (93.8%) were observed between the wheat chromosomes. The largest number of wheat-wheat chromosome associations (53%) was detected between the A and D genomes, while the frequency of B-D and A-B associations was significantly lower (32 and 8%, respectively). Among the A-D chromosome associations, pairing between the 3AL and 3DL arms was observed with the highest frequency, while the most frequent of all the chromosome associations (0.113/ cell) was found to be the 3DS-3BS. Differences in the pairing frequency of the individual chromosome arms of wheat-rye hybrids have been discussed in relation to the homoeologous relationships between the constituent genomes of hexaploid wheat

    Numerical simulation of wind waves on the RĂ­o de la Plata: evaluation of four global atmospheric databases

    The performance of NCEP/NCAR I, NCEP/DOE II, JRA-25 and ERA-Interim global databases, implemented as atmospheric forcings of the SWAN model in the Río de la Plata region, was quantitatively tested by calculating the bias, the mean square root error, the determination coefficient and the slope of the line fitted between observed and simulated wave parameters (significant wave height, mean period and direction). Even though statistical estimators showed no evident differences for wave periods and directions some noticeable differences were observed for simulated significant wave heights depending on the forcing used. The lowest bias (0.22 m) was obtained when the SWAN model was forced by ERA-Interim. With regard to the mean square root errors, the lowest values were obtained when NCEP/NCAR I (0.16 m) and NCEP/DOE II (0.19 m) were used as forcing. In addition, the best slope for simulated heights (0.79) was obtained using NCEP/DOE II. Computed determination coefficients for heights, periods and directions were very similar (0.89-0.93) for all the simulations carried out in this study. Energetic and severe wave events were given special consideration. The most energetic wave episode recorded in the Río de la Plata mouth (24 August, 2005) was analyzed and discussed in particular. It was concluded that during energetic atmospheric conditions the best agreement is achieved by implementing NCEP/DOE II as forcing. In the light of these results it is concluded that NCEP/DOE II is the most suitable atmospheric forcing to simulate wave heights with the SWAN model in the Río de la Plata region.Na região do Rio de la Plata, o desempenho das reanålises globais do NCEP/NCAR I, NCEP/DOE II, JRA-25 e ERAInterim implementadas como forçantes atmosféricas do modelo SWAN foram quantitativamente acessados através do viés, erro quadråtico médio, coeficiente de determinação e inclinação da reta. Estes índices foram obtidos dos parùmetros de ondas observados e simulados (alturas significativas de ondas, período principal e direção). Embora as estimativas estatísticas não mostrem diferenças evidentes para períodos e direçÔes, algumas diferenças notåveis foram obtidas para altura de ondas simuladas, dependendo do vento utilizado. O menor viés para altura significativa (0.22 m) foi obtido quando o SWAM foi forçado com a ERAInterim, enquanto o NCEP/NCAR I (0.16 m) e NCEP/DOE II (0.19 m) forneceram menor erro quadråtico médio. A melhor inclinação da reta entre simulação e observação de altura significativa (0.79) foi obtida usando NCEP/DOE II. No período de estudo, o maior episódio de onda registrado na boca do Río de la Plata foi analisado e discutido. Neste evento de condiçÔes atmosféricas energéticas o melhor ajuste foi alcançado utilizando os ventos do NCEP/DOE II como forçante. Conclui-se que a base de dados NCEP/DOE II é forçante atmosférica mais adequada para simular alturas significativas de ondas com o modelo SWAN na região estudada

    Evaluation of the nutrition knowledge of sports department students of universities

    <p>Abstract</p> <p>Background</p> <p>Individuals who have knowledge on the importance of adequate and balanced diet and reflect this knowledge to their behaviors are considered to be more successful in sports life. The present study aims to evaluate the nutrition knowledge of students receiving sports education in universities.</p> <p>Methods</p> <p>The study sample consists of 343 voluntary students from the Sports Departments of Hacettepe, Gazi and Ankara Universities in Ankara. The questionnaire used in the study included a demographic section, and 30 questions on true-false nutrition knowledge. For the reliability of the questionnaire, the internal consistency coefficient was calculated and the Kuder Richardson (KR-20) value was found to be 0.71. For higher reliability, 9 dysfunctional questions were excluded from the questionnaire. The research data were collected through a questionnaire form and face-to-face interviews. For the statistical analyses of the data, tables were prepared to show mean, standard deviation (<inline-formula><m:math xmlns:m="http://www.w3.org/1998/Math/MathML" name="1550-2783-8-11-i1"><m:mover accent="true"><m:mrow><m:mi>X</m:mi></m:mrow><m:mo class="MathClass-op">̄</m:mo></m:mover><m:mo class="MathClass-bin">±</m:mo><m:mi>S</m:mi><m:mi>D</m:mi></m:math></inline-formula>) and percentage (%) values. In order to determine the nutrition knowledge of students, the "independent t test" was used for nutrition lesson and gender.</p> <p>Results</p> <p>University students receiving sports education and expected to continue their professional lives on sport-related fields were determined to have the lack of knowledge on nutrition. The mean value about the nutrition knowledge of the first year students was found 11.150 ± 2.962, while the mean value of the fourth year students was 13.460 ± 3.703, and the difference is statistically significant (p = .000).</p> <p>Conclusion</p> <p>Students, coaches and teachers in physical education were found not to give the necessary importance to their diets, and they were still not aware of the importance of nutrition on performance.</p

    mTORC1 amplifies the ATF4-dependent de novo serine-glycine pathway to supply glycine during TGF-ÎČ1-induced collagen biosynthesis

    The differentiation of fibroblasts into a transient population of highly activated, extracellular matrix (ECM)-producing myofibroblasts at sites of tissue injury is critical for normal tissue repair. Excessive myofibroblast accumulation and persistence, often as a result of a failure to undergo apoptosis when tissue repair is complete, lead to pathological fibrosis and are also features of the stromal response in cancer. Myofibroblast differentiation is accompanied by changes in cellular metabolism, including increased glycolysis, to meet the biosynthetic demands of enhanced ECM production. Here, we showed that transforming growth factor-ÎČ1 (TGF-ÎČ1), the key pro-fibrotic cytokine implicated in multiple fibrotic conditions, increased the production of activating transcription factor 4 (ATF4), the transcriptional master regulator of amino acid metabolism, to supply glucose-derived glycine to meet the amino acid requirements associated with enhanced collagen production in response to myofibroblast differentiation. We further delineated the signaling pathways involved and showed that TGF-ÎČ1-induced ATF4 production depended on cooperation between canonical TGF-ÎČ1 signaling through Smad3 and activation of mechanistic target of rapamycin complex 1 (mTORC1) and its downstream target eukaryotic translation initiation factor 4E-binding protein 1 (4E-BP1). ATF4, in turn, promoted the transcription of genes encoding enzymes of the de novo serine-glycine biosynthetic pathway and glucose transporter 1 (GLUT1). Our findings suggest that targeting the TGF-ÎČ1-mTORC1-ATF4 axis may represent a novel therapeutic strategy for interfering with myofibroblast function in fibrosis and potentially in other conditions, including cancer

    Maternal Perception of Child Weight Among Mexicans in California and Mexico

    The prevalence of childhood overweight is high in Mexican immigrant communities in the United States. Understanding mother’s perceptions of child weight in immigrants’ country of origin may help to understand this high prevalence. The goal of this study was to examine and compare mothers’ perception of weight in Mexico (MX) and in an immigrant community in California (CA). We assessed perceptions of child weight using a pictorial scale with 314 mothers of 5-year-old children in MX and 60 mothers of 5 year-old-children in CA. We compared maternal reports with children’s objectively measured weight. Using chi-square and Analysis of Variance, we investigated associations of maternal perception of and satisfaction with weight according to socio-demographic characteristics. Mothers were more likely to underestimate their children’s weight in CA than in MX. On average, CA mothers wanted their children to be smaller than they currently were and mothers in MX wanted their children to be bigger than they currently were. This differed by weight status in CA with mothers of normal weight and at-risk-for-overweight children wanting them to be bigger and mothers of overweight children wanting them to be smaller. In order for programs to be effective, mothers must be able to recognize their children as overweight and want to address it. Because underestimation of weight and a desire for a larger size is common in this population, programs to address overweight may be more effective if they focus on alternative benefits of weight control strategies, such as healthy child development

    PRALIMAP: study protocol for a high school-based, factorial cluster randomised interventional trial of three overweight and obesity prevention strategies

    <p>Abstract</p> <p>Background</p> <p>Given the increase in overweight and obesity prevalence in adolescents in the last decade, effective prevention strategies for these conditions in adolescents are urgently needed. The PRALIMAP (Promotion de l'ALImentation et de l'ActivitĂ© Physique) trial aims to evaluate the effectiveness for these conditions of 3 health promotion strategies -- educational, screening and environmental -- applied singly or in combination in high schools over a 2-year intervention period.</p> <p>Methods</p> <p>PRALIMAP is a stratified 2 × 2 × 2 factorial cluster randomised controlled trial including 24 state high schools in Lorraine, northeastern France, in 2 waves: 8 schools in 2006 (wave 1) and 16 in 2007 (wave 2). Students entering the selected high schools in the 4 academic years from 2006 to 2009 are eligible for data collection. Interventional strategies are organized over 2 academic years. The follow-up consists of 3 visits: at the entry of grade 10 (T0), grade 11 (T1) and grade 12 (T2). At T0, 5,458 (85.7%) adolescents participated. The educational strategy consists of nutritional lessons, working groups and a final party. The screening strategy consists in detecting overweight/obesity and eating disorders in adolescents and proposing, if necessary, an adapted care management program of 7 group educational sessions. The environmental strategy consists in improving dietary and physical activity offerings in high schools and facilities, especially catering. The main outcomes are body size evolution over time, nutritional behaviour and knowledge, health and quality of life. An evaluation process documents how each intervention strategy is implemented in the schools and estimates the dose of the intervention, allowing for a per protocol analysis after the main intention-to-treat analysis.</p> <p>Discussion</p> <p>PRALIMAP aims at improving the prevention and management of overweight and obesity in adolescents by translating current evidence into public health practice. Particular attention is paid to clustering, multiple factorials and long-term duration to address common pitfalls in health promotion trials. The results should inform how best to implement, in a school environment, effective nutrition prevention programs targeting adolescents who are at a point their lives when they develop responsibilities and empowerment for health attitude behaviours.</p> <p>Trial registration</p> <p>This trial is registered at ClinicalTrials.gov under <a href="http://clinicaltrials.gov/ct2/show/NCT00814554">NCT00814554</a>.</p

    Filamin A Binds to CCR2B and Regulates Its Internalization

    The chemokine (C-C motif) receptor 2B (CCR2B) is one of the two isoforms of the receptor for monocyte chemoattractant protein-1 (CCL2), the major chemoattractant for monocytes, involved in an array of chronic inflammatory diseases. Employing the yeast two-hybrid system, we identified the actin-binding protein filamin A (FLNa) as a protein that associates with the carboxyl-terminal tail of CCR2B. Co-immunoprecipitation experiments and in vitro pull down assays demonstrated that FLNa binds constitutively to CCR2B. The colocalization of endogenous CCR2B and filamin A was detected at the surface and in internalized vesicles of THP-1 cells. In addition, CCR2B and FLNa were colocalized in lamellipodia structures of CCR2B-expressing A7 cells. Expression of the receptor in filamin-deficient M2 cells together with siRNA experiments knocking down FLNa in HEK293 cells, demonstrated that lack of FLNa delays the internalization of the receptor. Furthermore, depletion of FLNa in THP-1 monocytes by RNA interference reduced the migration of cells in response to MCP-1. Therefore, FLNa emerges as an important protein for controlling the internalization and spatial localization of the CCR2B receptor in different dynamic membrane structures
