    Nothing is Permanent but Change: The Journey to Consolidation and Back

    Responding to a staffing shortage and assessment of usage data, eight of Hale Library's nine service desks were consolidated into one service point. Fourteen months later, the Libraries conducted an assessment of the consolidation using reference transaction data, responses to a survey administered to Librariesā€™ staff, and responses from a student focus group. The assessment revealed many logistical and staffing challenges with the new desk. As a result, a separate service point for reference was created directly outside the consolidated desk. Our statistics indicate that this was a successful change

    Effects of corn supplementation on serum and muscle microRNA profiles in horses

    Laminitis associated with equine metabolic syndrome causes significant economic losses in the equine industry. Diets high in non-structural carbohydrates (NSC) have been linked to insulin resistance and laminitis in horses. Nutrigenomic studies analyzing the interaction of diets high in NSCs and gene expression regulating endogenous microRNAs (miRNA) are rare. This study\u27s objectives were to determine whether miRNAs from dietary corn can be detected in equine serum and muscle and its impacts on endogenous miRNA. Twelve mares were blocked by age, body condition score, and weight and assigned to a control (mixed legume grass hay diet) and a mixed legume hay diet supplemented with corn. Muscle biopsies and serum were collected on Days 0 and 28. Transcript abundances were analyzed using qRT-PCR for three plant-specific and 277 endogenous equine miRNAs. Plant miRNAs were found in serum and skeletal muscle samples with a treatment effect (p \u3c .05) with corn-specific miRNA being higher than control in serum after feeding. Endogenous miRNAs showed 12 different (p \u3c .05) miRNAs in equine serum after corn supplementation, six (eca-mir16, -4863p, -4865p, -126- 3p, -296, and -192) previously linked to obesity or metabolic disease. The results of our study indicate that dietary plant miRNAs can appear in circulation and tissues and may regulate endogenous genes

    The Nature of the Density Clump in the Fornax Dwarf Spheroidal Galaxy

    We have imaged the recently discovered stellar overdensity located approximately one core radius from the center of the Fornax dwarf spheroidal galaxy using the Magellan Clay 6.5m telescope with the Magellan Instant Camera (MagIC). Superb seeing conditions allowed us to probe the stellar populations of this overdensity and of a control field within Fornax to a limiting magnitude of R=26. The color-magnitude diagram of the overdensity field is virtually identical to that of the control field with the exception of the presence of a population arising from a very short (less than 300 Myr in duration) burst of star formation 1.4 Gyr ago. Coleman et al. have argued that this overdensity might be related to a shell structure in Fornax that was created when Fornax captured a smaller galaxy. Our results are consistent with this model, but we argue that the metallicity of this young component favors a scenario in which the gas was part of Fornax itself.Comment: 24 pages including 8 figures and 3 tables.     Tailored for Real-World: A Whole Slide Image Classification System Validated on Uncurated Multi-Site Data Emulating the Prospective Pathology Workload.

    Standard of care diagnostic procedure for suspected skin cancer is microscopic examination of hematoxylin & eosin stained tissue by a pathologist. Areas of high inter-pathologist discordance and rising biopsy rates necessitate higher efficiency and diagnostic reproducibility. We present and validate a deep learning system which classifies digitized dermatopathology slides into 4 categories. The system is developed using 5,070 images from a single lab, and tested on an uncurated set of 13,537 images from 3 test labs, using whole slide scanners manufactured by 3 different vendors. The system\u27s use of deep-learning-based confidence scoring as a criterion to consider the result as accurate yields an accuracy of up to 98%, and makes it adoptable in a real-world setting. Without confidence scoring, the system achieved an accuracy of 78%. We anticipate that our deep learning system will serve as a foundation enabling faster diagnosis of skin cancer, identification of cases for specialist review, and targeted diagnostic classifications

    Lentiviral Expression of Retinal Guanylate Cyclase-1 (RetGC1) Restores Vision in an Avian Model of Childhood Blindness

    BACKGROUND: Leber congenital amaurosis (LCA) is a genetically heterogeneous group of retinal diseases that cause congenital blindness in infants and children. Mutations in the GUCY2D gene that encodes retinal guanylate cyclaseā€“1 (retGC1) were the first to be linked to this disease group (LCA type 1 [LCA1]) and account for 10%ā€“20% of LCA cases. These mutations disrupt synthesis of cGMP in photoreceptor cells, a key second messenger required for function of these cells. The GUCY1*B chicken, which carries a null mutation in the retGC1 gene, is blind at hatching and serves as an animal model for the study of LCA1 pathology and potential treatments in humans. METHODS AND FINDINGS: A lentivirus-based gene transfer vector carrying the GUCY2D gene was developed and injected into early-stage GUCY1*B embryos to determine if photoreceptor function and sight could be restored to these animals. Like human LCA1, the avian disease shows early-onset blindness, but there is a window of opportunity for intervention. In both diseases there is a period of photoreceptor cell dysfunction that precedes retinal degeneration. Of seven treated animals, six exhibited sight as evidenced by robust optokinetic and volitional visual behaviors. Electroretinographic responses, absent in untreated animals, were partially restored in treated animals. Morphological analyses indicated there was slowing of the retinal degeneration. CONCLUSIONS: Blindness associated with loss of function of retGC1 in the GUCY1*B avian model of LCA1 can be reversed using viral vector-mediated gene transfer. Furthermore, this reversal can be achieved by restoring function to a relatively low percentage of retinal photoreceptors. These results represent a first step toward development of gene therapies for one of the more common forms of childhood blindness
