Multiple moonlighting functions of mycobacterial molecular chaperones
Molecular chaperones and protein folding catalysts are normally thought of as intracellular proteins involved in protein folding quality control. However, in the mycobacteria there is increasing evidence to support the hypothesis that molecular chaperones are also secreted intercellular signalling molecules or can control actions at the cell wall or indeed control the composition of the cell wall. The most recent evidence for protein moonlighting in the mycobacteria is the report that chaperonin 60.2 of Mycobacterium tuberculosis is important in the key event in tuberculosis - the entry of the bacterium into the macrophage. This brief overview highlights the potential importance of the moonlighting functions of molecular chaperones in the biology and pathobiology of the mycobacteria. © 2010 Elsevier Ltd. Evaluating Common Raven Take for Greater Sage-Grouse in Oregon’s Baker County Priority Conservation Area and Great Basin Region
The common raven (Corvus corax; raven) is a nest predator of species of conservation concern, such as the greater sage-grouse (Centrocercus urophasianus). Reducing raven abundance by take requires authorization under the Migratory Bird Treaty Act. To support U.S. Fish and Wildlife Service’s take decisions (e.g., those that authorize killing a specified proportion or number of individuals annually in a defined area), including the most recent one for Oregon’s Baker County Priority Area for Conservation (PAC), we modeled raven population dynamics under hypothetical scenarios with take rates ranging from below to above the maximum sustained yield (MSY; i.e., trmsy= 0.01-0.60). We fit a Bayesian state-space logistic model to estimate abundance based on the Breeding Bird Survey route-level count data for the PAC during 1997-2019 and Great Basin Region (GBR) during 1968-2019. We predicted abundance for 2019-2030 and evaluated potential take levels (PTL) for the PAC and GBR. Abundance averaged 682 (SE = 93) for the PAC during 1997-2019 and 333,027 (SE = 20,504) for the GBR during 1968-2019. With take rates between 0.41 and 0.60, predicted abundance averaged 308 (SD = 405) for the PAC and 142,258 (SD = 53,474) for the GBR during 2019-2030. With management factor F = 0.75-2 for takes ranging from below to above the MSY, the PTL 50th percentiles were 150-401 yr-1 for the PAC and 60,457-161,219 yr-1 for the GBR. Our modeling framework is flexible and can be part of a comprehensive management strategy for ravens in the western United States
Estimating Trends of Common Raven Populations in North America, 1966–2018
Over the last half century, common raven (Corvus corax; raven) populations have increased in abundance across much of North America. Ravens are generalist predators known to depredate the eggs and young of several sensitive species. Quantifying raven population increases at multiple spatial scales across North America will help wildlife resource managers identify areas where population increases present the greatest risk to species conservation. We used a hierarchical Bayesian modeling approach to analyze trends of standardized raven counts from 1966 to 2018 using Breeding Bird Survey data within each Level I and II ecoregion of the United States and Canada. We also compared raven abundance within and outside the distributions of 9 sensitive or endangered species. Although we found substantial evidence that raven populations have increased across North America, populations varied in growth rates and relative abundances among regions. We found 73% of Level I (11/15) and II (25/34) ecoregions demonstrated positive annual population growth rates ranging from 0.2–9.4%. We found higher raven abundance inside versus outside the distributions of 7 of the 9 sensitive species included in our analysis. Gunnison sage-grouse (Centrocercus minimus) had the highest discrepancy, with 293% more ravens within compared to outside of their range, followed by greater sandhill crane (Antigone canadensis tabida; 280%), and greater sage-grouse (C. urophasianus; 204%). Only 2 species, least tern (Sternula antillarum) and piping plover (Charadrius melodus), indicated lower raven abundance within relative to outside their distributions. Our findings will help wildlife resource managers identify regional trends in abundance of ravens and anticipate which sensitive species are at greatest risk from elevated raven populations. Future research directed at identifying the underlying regional drivers of these trends could help elucidate the most appropriate and responsive management actions and, thereby, guide the development of raven population management plans to mitigate impacts to sensitive species
A Rapid Assessment Function to Estimate Common Raven Population Densities: Implications for Targeted Management
Common raven (Corvus corax; raven) populations have increased over the past 5 decades within the western United States. Raven population increases have been largely attributed to growing resource subsidies from expansion of human enterprise. Concomitantly, managers are becoming increasingly concerned about elevated adverse effects on multiple sensitive prey species, damage to livestock and agriculture, and human safety. Managers could benefit from a rapid but reliable method to estimate raven densities across spatiotemporal scales to monitor raven populations more efficiently and inform targeted and adaptive management frameworks. However, obtaining estimates of raven density is data- and resource-intensive, which renders monitoring within an adaptive framework unrealistic. To address this need, we developed a rapid survey protocol for resource managers to estimate site-level density based on the average number of ravens per survey. Specifically, we first estimated raven densities at numerous field sites with robust distance sampling procedures and then used regression to investigate the relationship between those density estimates and the number of ravens per survey, which revealed a strong correlation (R2 = 0.86). For management application, we provide access to R function software through a web-based interface to estimate density using number of ravens per survey, which we refer to as a Rapid Assessment Function (RAF). Then, using a simulation analysis of data from sites with abundant surveys and the RAF, we estimated raven density based on different numbers of surveys to help inform how many surveys are needed to achieve reliable estimates within this rapid assessment. While more robust procedures of distance sampling are the preferred methods for estimating raven densities from count surveys, the RAF tool presented herein provides a reliable approximation for informing management decisions when managers are faced with resource and small sample size constraints
Does gender matter? A cross-national investigation of primary class-room discipline.

With the on-going international debate about educational performance highlighting the widening gender achievement gap between girl and boy pupils, the demand for more male teachers has become prevalent in educational discourse. Concerns have frequently been raised about the underachievement of boys, with claims that the lack of male ‘role models’ in schools has an adverse effect on boys’ academic motivation and engagement. Although previous research has examined ‘teaching’ as institutional talk, men’s linguistic behaviour in the classroom remains largely ignored, especially in regard to enacting discipline. The difference that tenure makes
This paper argues that housing tenures cannot be reduced to either production relations or consumption relations. Instead, they need to be understood as modes of housing distribution, and as having complex and dynamic relations with social classes. Building on a critique of both the productionist and the consumptionist literature, as well as of formalist accounts of the relations between tenure and class, the paper attempts to lay the foundations for a new theory of housing tenure. In order to do this, a new theory of class is articulated, which is then used to throw new light on the nature of class-tenure relations
Clinically relevant patient clusters identified by machine learning from the clinical development programme of secukinumab in psoriatic arthritis
Objectives: Identify distinct clusters of psoriatic arthritis (PsA) patients based on their baseline articular, entheseal and cutaneous disease manifestations and explore their clinical and therapeutic value. Methods: Pooled baseline data in PsA patients (n=1894) treated with secukinumab across four phase 3 studies (FUTURE 2–5) were analysed to determine phenotypes based on clusters of clinical indicators. Finite mixture models methodology was applied to generate clinical clusters and mean longitudinal responses were compared between secukinumab doses (300 vs 150 mg) across identified clusters and clinical indicators through week 52 using machine learning (ML) techniques. Results: Seven distinct patient clusters were identified. Cluster 1 (very-high (VH) – SWO/TEN (swollen/tender); n=187) was characterised by VH polyarticular burden for both tenderness and swelling of joints, while cluster 2 (H (high) – TEN; n=251) was marked by high polyarticular burden in tender joints and cluster 3 (H – Feet – Dactylitis; n=175) by high burden in joints of feet and dactylitis. For cluster 4 (L (Low) – Nails – Skin; n=209), cluster 5 (L – skin; n=283), cluster 6 (L – Nails; n=294) and cluster 7 (L; n=495) articular burden was low but nail and skin involvement was variable, with cluster 7 marked by mild disease activity across all domains. Greater improvements in the longitudinal responses for enthesitis in cluster 2, enthesitis and Psoriasis Area and Severity Index (PASI) in cluster 4 and PASI in cluster 6 were shown for secukinumab 300 mg compared with 150 mg. Conclusions: PsA clusters identified by ML follow variable response trajectories indicating their potential to predict precise impact on patients’ outcomes. Axial Involvement in Psoriatic Arthritis cohort (AXIS): the protocol of a joint project of the Assessment of SpondyloArthritis international Society (ASAS) and the Group for Research and Assessment of Psoriasis and Psoriatic Arthritis (GRAPPA).
BACKGROUND: Involvement of the axial skeleton (sacroiliac joints and spine) is a relatively frequent manifestation associated with psoriatic skin disease, mostly along with involvement of peripheral musculoskeletal structures (peripheral arthritis, enthesitis, dactylitis), which are referred to as psoriatic arthritis (PsA). Data suggest that up to 30% of patients with psoriasis have PsA. Depending on the definition used, the prevalence of axial involvement varies from 25% to 70% of patients with PsA. However, there are currently no widely accepted criteria for axial involvement in PsA.Objective: The overarching aim of the Axial Involvement in Psoriatic Arthritis (AXIS) study is to systematically evaluate clinical and imaging manifestations indicative of axial involvement in patients with PsA and to develop classification criteria and a unified nomenclature for axial involvement in PsA that would allow defining a homogeneous subgroup of patients for research. DESIGN: Prospective, multicenter, multinational, cross-sectional study. METHODS AND ANALYSES: In this multicenter, multinational, cross-sectional study, eligible patients [adult patients diagnosed with PsA and fulfilling Classification Criteria for Psoriatic Arthritis (CASPAR) with musculoskeletal symptom duration of ⩽10 years not treated with biological or targeted synthetic disease-modifying anti-rheumatic drugs] will be recruited prospectively. They will undergo study-related clinical and imaging examinations. Imaging will include radiography and magnetic resonance imaging examinations of sacroiliac joints and spine. Local investigators will evaluate for the presence of axial involvement based on clinical and imaging information which will represent the primary outcome of the study. In addition, imaging will undergo evaluation by central review. Finally, the central clinical committee will determine the presence of axial involvement based on all available information. ETHICS: The study will be performed according to the ethical principles of the Declaration of Helsinki and International Council for Harmonisation Good Clinical Practice guidelines. The study protocol will be approved by the individual Independent Ethics Committee / Institutional Review Board of participating centers.
