Human Primordial Germ Cells Are Specified from Lineage-Primed Progenitors.
In vitro gametogenesis is the process of making germline cells from human pluripotent stem cells. The foundation of this model is the quality of the first progenitors called primordial germ cells (PGCs), which in vivo are specified during the peri-implantation window of human development. Here, we show that human PGC (hPGC) specification begins at day 12 post-fertilization. Using single-cell RNA sequencing of hPGC-like cells (hPGCLCs) differentiated from pluripotent stem cells, we discovered that hPGCLC specification involves resetting pluripotency toward a transitional state with shared characteristics between naive and primed pluripotency, followed by differentiation into lineage-primed TFAP2A+ progenitors. Applying the germline trajectory to TFAP2C mutants reveals that TFAP2C functions in the TFAP2A+ progenitors upstream of PRDM1 to regulate the expression of SOX17. This serves to protect hPGCLCs from crossing the Weismann's barrier to adopt somatic cell fates and, therefore, is an essential mechanism for successfully initiating in vitro gametogenesis

A high proportion of the world population of the spoon-billed sandpiper occurs at Tiaozini, China, during the post-breeding Moult
The Critically Endangered Spoon-billed Sandpiper Calidris pygmaea breeds in arctic and subarctic Russia and migrates to winter on coastal mudflats in south-east Asia. Its world population is probably about 700 individuals. We report Lincoln-Petersen closed-population estimates of the number of Spoon-billed Sandpipers at Tiaozini, a coastal site in Jiangsu Province, China, based upon resightings and scan surveys of individually-marked leg-flagged birds. Surveys were conducted in September-October, when adult Spoon-billed Sandpipers are moulting their primary feathers and long-distance movements are unlikely. We estimated that 220 birds were present at Tiaozini in 2017 and 224 in 2018. Nearly all of them were adults (one-year old or older), so about 40% of the world population of this age class was estimated to be present. Hence, protection of the mudflats and roost sites at Tiaozini is of global importance for the conservation of the Spoon-billed Sandpiper.Non
Semantic dementia: a complex and culturally influenced presentation.
The variants of frontotemporal dementia (FTD) require careful differentiation from primary psychiatric disorders as the neuropsychiatric manifestations can overshadow the unique cognitive deficits. The language variants of FTD are less readily recognised by trainees despite making up around 43% of cases. This educational article presents an anonymised case of one of the language variants: semantic dementia. The cognitive deficits and neuropsychiatric manifestations (delusions and hyperreligiosity) are explored in terms of aetiology and management. By the end of the article, readers should be able to differentiate FTD from Alzheimer's disease, understand the principles of management and associated risks, and develop a multifaceted approach to hyperreligiosity in dementia
The time-dependent rearrangement of the epithelial basement membrane in human skin wounds
In 62 human skin wounds (surgical wounds, stab wounds and lacerations after surgical treatment) we analyzed the immunohistochemical localization of collagen IV in the epithelial basement membrane. In 27 of these wounds the distribution of collagen VII, which represents a specific component of the basement membrane of stratified epithelia, was also analyzed. We were able to demonstrate a virtually identical co-distribution of both collagen IV and VII in the wound area with no significant time-dependent differences in the appearance of both collagen types. Fragments of the epithelial basement membrane could be detected in the wound area from as early as 4 days after wounding and after 8 days a complete restitution of the epithelial basement membrane was observed. In all cases with a wound age of more than 21 days the basement membrane was completely reformed over the former lesional area. The period between 8 and 21 days after wounding was characterized by a wide variability ranging from complete restitution to deposition of basement membrane fragments or total lack of the epidermal basement membrane
Evaluation of Rapid Diagnostics for Plasmodium falciparum and P. vivax in Mae Sot Malaria Endemic Area, Thailand
Prompt and accurate diagnosis of malaria is the key to prevent disease morbidity and mortality. This study was carried out to evaluate diagnostic performance of 3 commercial rapid detection tests (RDTs), i.e., Malaria Antigen Pf/Pan™, Malaria Ag-Pf™, and Malaria Ag-Pv™ tests, in comparison with the microscopic and PCR methods. A total of 460 blood samples microscopically positive for Plasmodium falciparum (211 samples), P. vivax (218), mixed with P. falciparum and P. vivax (30), or P. ovale (1), and 124 samples of healthy subjects or patients with other fever-related infections, were collected. The sensitivities of Malaria Ag-Pf™ and Malaria Antigen Pf/Pan™ compared with the microscopic method for P. falciparum or P. vivax detection were 97.6% and 99.0%, or 98.6% and 99.0%, respectively. The specificities of Malaria Ag-Pf™, Malaria Ag-Pv™, and Malaria Antigen Pf/Pan™ were 93.3%, 98.8%, and 94.4%, respectively. The sensitivities of Malaria Ag-Pf™, Malaria Antigen Pf/Pan™, and microscopic method, when PCR was used as a reference method for P. falciparum or P. vivax detection were 91.8%, 100%, and 96.7%, or 91.9%, 92.6%, and 97.3%, respectively. The specificities of Malaria Ag-Pf™, Malaria Ag-Pv™, Malaria Antigen Pf/Pan™, and microscopic method were 66.2%, 92.7%, 73.9%, and 78.2%, respectively. Results indicated that the diagnostic performances of all the commercial RDTs are satisfactory for application to malaria diagnosis
Opportunities for topical antimicrobial therapy: permeation of canine skin by fusidic acid
BACKGROUND: Staphylococcal infection of the canine epidermis and hair follicle is amongst the commonest reasons for antimicrobial prescribing in small animal veterinary practice. Topical therapy with fusidic acid (FA) is an attractive alternative to systemic therapy based on low minimum inhibitory concentrations (MICs, commonly <0.03 mg/l) documented in canine pathogenic staphylococci, including strains of MRSA and MRSP (methicillin-resistant Staphylococcus aureus and S. pseudintermedius). However, permeation of canine skin by FA has not been evaluated in detail. This study aimed to define the degree and extent of FA permeation in canine skin in vitro from two sites with different hair follicle density following application of a licensed ophthalmic formulation that shares the same vehicle as an FA-betamethasone combination product approved for dermal application in dogs. Topical FA application was modelled using skin held in Franz-type diffusion cells. Concentrations of FA in surface swabs, receptor fluid, and transverse skin sections of defined anatomical depth were determined using high-performance liquid chromatography and ultraviolet (HPLC-UV) analysis. RESULTS: The majority of FA was recovered by surface swabs after 24 h, as expected (mean ± SEM: 76.0 ± 17.0%). FA was detected within 424/470 (90%) groups of serial sections of transversely cryotomed skin containing follicular infundibula, but never in 48/48 (100%) groups of sections containing only deeper follicular structures, nor in receptor fluid, suggesting that FA does not permeate beyond the infundibulum. The FA concentration (mean ± SEM) in the most superficial 240 μm of skin was 2000 ± 815 μg/g. CONCLUSIONS: Topically applied FA can greatly exceed MICs for canine pathogenic staphylococci at the most common sites of infection. Topical FA therapy should now be evaluated using available formulations in vivo as an alternative to systemic therapy for canine superficial bacterial folliculitis.Peer reviewedFinal Published versio
A new parametric equation of state and quark stars
It is still a matter of debate to understand the equation of state of cold
supra-nuclear matter in compact stars because of unknown on-perturbative strong
interaction between quarks. Nevertheless, it is speculated from an
astrophysical view point that quark clusters could form in cold quark matter
due to strong coupling at realistic baryon densities. Although it is hard to
calculate this conjectured matter from first principles, one can expect the
inter-cluster interaction to share some general features to nucleon-nucleon
interaction. We adopt a two-Gaussian component soft-core potential with these
general features and show that quark clusters can form stable simple cubic
crystal structure if we assume Gaussian form wave function. With this
parameterizing, Tolman-Oppenheimer-Volkoff equation is solved with reasonable
constrained parameter space to give mass-radius relation of crystalline solid
quark star. With baryon densities truncated at 2 times nuclear density at
surface and range of interaction fixed at 2fm we can reproduce similar
mass-radius relation to that obtained with bag model equations of state. The
maximum mass ranges from about 0.5 to 3 solar mass. Observed maximum pulsar
mass (about 2 solar mass) is then used to constrain parameters of this simple

The Virtue of Defects in 4D Gauge Theories and 2D CFTs
We advance a correspondence between the topological defect operators in
Liouville and Toda conformal field theories - which we construct - and loop
operators and domain wall operators in four dimensional N=2 supersymmetric
gauge theories on S^4. Our computation of the correlation functions in
Liouville/Toda theory in the presence of topological defect operators, which
are supported on curves on the Riemann surface, yields the exact answer for the
partition function of four dimensional gauge theories in the presence of
various walls and loop operators; results which we can quantitatively
substantiate with an independent gauge theory analysis. As an interesting
outcome of this work for two dimensional conformal field theories, we prove
that topological defect operators and the Verlinde loop operators are different
