    Resistance to antimicrobial agents in bifidobacteria

    Includes bibliographical references (leaves 95-106).For bifidobacteria to survive and achieve colonisation, they have to interact with inhibitory host-produced substances such as bile salts. Another aspect which should be studied is the safety of the probiotic bacterium and risks of acquisition of genes for resistance to antimicrobial agents. Although bifidobacteria exhibit resistance to a wide range of antibiotics, little is known about the molecular basis for this resistance. The aim of this project was, therefore, to investigate the molecular mechanisms responsible for the resistance to antibiotics and bile salts observed in bifidobacteria, and more specifically, to determine whether efflux systems are involved in this resistance.     From meadows to milk to mucosa – adaptation of Streptococcus and Lactococcus species to their nutritional environments

    Lactic acid bacteria (LAB) are indigenous to food-related habitats as well as associated with the mucosal surfaces of animals. The LAB family Streptococcaceae consists of the genera Lactococcus and Streptococcus. Members of the family include the industrially important species Lactococcus lactis, which has a long history safe use in the fermentative food industry, and the disease-causing streptococci Streptococcus pneumoniae and Streptococcus pyogenes. The central metabolic pathways of the Streptococcaceae family have been extensively studied because of their relevance in the industrial use of some species, as well as their influence on virulence of others. Recent developments in high-throughput proteomic and DNA-microarray techniques, in in vivo NMR studies, and importantly in whole-genome sequencing have resulted in new insights into the metabolism of the Streptococcaceae family. The development of cost-effective high-throughput sequencing has resulted in the publication of numerous whole-genome sequences of lactococcal and streptococcal species. Comparative genomic analysis of these closely related but environmentally diverse species provides insight into the evolution of this family of LAB and shows that the relatively small genomes of members of the Streptococcaceae family have been largely shaped by the nutritionally rich environments they inhabit.

    An evaluation of the falls prevention practice of community-based allied health professionals working in primary care

    Objectives: This study aimed to explore allied health professional’s (AHP) current practice and understanding of falls prevention and to investigate self-reported change following a professional development workshop. Methods: Four professional development workshops were designed targeting exercise, home safety interventions, the LiFE program and medication reviews. Self-reported surveys were collected pre, post and at three-month follow-up. Results: Seven workshops were run in Northern Sydney with 161 participants. AHP identified practice barriers were regularly assessing falls risk and collaborating with other service providers. At three months, change in practice was reported by participants in the exercise (88.1%), home safety (48%), LiFE (78.6%), and medication review (62.5%) workshops. Discussion: These findings provide good support for the effectiveness of professional development workshops in knowledge translation for medication review and exercise based interventions. Further studies and initiatives into promoting screening for falls risk and greater collaboration between community service providers is recommended

    Co-production of 11α-hydroxyprogesterone and ethanol using recombinant yeast expressing fungal steroid hydroxylases

    Background: Bioethanol production from sustainable sources of biomass that limit effect on food production are needed and in a biorefinery approach co-products are desirable, obtained from both the plant material and from the microbial biomass. Fungal biotransformation of steroids was among the first industrial biotransformations allowing corticosteroid production. In this work, the potential of yeast to produce intermediates needed in corticosteroid production is demonstrated at laboratory scale following bioethanol production from perennial ryegrass juice. Results: Genes encoding the 11α-steroid hydroxylase enzymes from Aspergillus ochraceus (11α-SHAoch) and Rhizopus oryzae (CYP509C12) transformed into Saccharomyces cerevisiae for heterologous constitutive expression in p425TEF. Both recombinant yeasts (AH22:p11α-SHAoch and AH22:p509C12) exhibited efficient progesterone bioconversion (on glucose minimal medial containing 300 µM progesterone) producing either 11α-hydroxyprogesterone as the sole metabolite (AH22:p11α-SHAoch) or a 7:1 mixture of 11α-hydroxyprogesterone and 6β-hydroxyprogesterone (AH22:p509C12). Ethanol yields for AH22:p11α-SHAoch and AH22:p509C12 were comparable resulting in ≥75% conversion of glucose to alcohol. Co-production of bioethanol together with efficient production of the 11-OH intermediate for corticosteroid manufacture was then demonstrated using perennial ryegrass juice. Integration of the 11α-SHAoch gene into the yeast genome (AH22:11α-SHAoch+K) resulted in a 36% reduction in yield of 11α-hydroxyprogesterone to 174 µmol/L using 300 µM progesterone. However, increasing progesterone concentration to 955 µM and optimizing growth conditions increased 11α-hydroxyprogesterone production to 592 µmol/L product formed. Conclusions: The progesterone 11α-steroid hydroxylases from A. ochraceus and R. oryzae, both monooxygenase enzymes of the cytochrome P450 superfamily, have been functionally expressed in S. cerevisiae. It appears that these activities in fungi are not associated with a conserved family of cytochromes P450. The activity of the A. ochraceous enzyme was important as the specificity of the biotransformation yielded just the 11-OH product needed for corticosteroid production. The data presented demonstrate how recombinant yeast could find application in rural biorefinery processes where co-production of value-added products (11α-hydroxyprogesterone and ethanol) from novel feedstocks is an emergent and attractive possibility

    A multi-centre study of adults with learning disabilities referred to services for antisocial or offending behaviour: demographic, individual, offending and service characteristics

    This study was carried out as part of a larger study commissioned by the UK Department of Health to investigate the service pathways for offenders with learning disabilities (LD). The study covered three health regions in the UK and included 477 people with LD referred to services because of antisocial or offending behaviour during a 12-month period. Data were collected concerning demographic, individual, offending behaviour and service characteristics. The findings of the study are broadly consistent with contemporary research concerning this population, particularly in relation to the nature and frequency of offending, history of offending, psychopathology, age and gender distribution. However, very few of those referred had any form of structured care plan, despite having significant offending histories, and this may have compromised early identification of their needs and communication between the health, social and other services involved
