    Anticoagulantes com diferentes formas de ação têm sido utilizados na hematologia aviária. Entretanto, poucos estudos foram publicados sobre seu efeito e sobre os parâmetros hematológicos dos avestruzes. Visando a preencher essa lacuna, realizou-se um estudo em que foi comparado o efeito de dois anticoagulantes de rotina (EDTA - 2 mg/mL sangue e da Heparina -10U/mL sangue) sobre os eritrócitos de avestruzes. Para tanto, foram avaliados a integridade, o volume globular e a morfometria dos eritrócitos de amostras sanguíneas de 20 aves, tratadas com ambos os anticoagulantes. A integridade eritrocitária foi estimada pelo grau de hemoglobina livre no plasma, sendo o volume globular obtido pelo método de micro-hematócrito e os dados morfométricos (área, diâmetro maior e menor) calculados automaticamente em sistema computadorizado de imagem digital de alta resolução. A hemólise estimada pela dosagem de hemoglobina plasmática, o volume globular, a área e o diâmetro máximo dos eritrócitos foram significativamente maiores nas amostras tratadas com EDTA. Para melhor preservar a integridade e a morfometria dos eritrócitos, concluiuse que a heparina é um anticoagulante mais adequado que o EDTA para avaliar as discrasias eritrocitárias dos avestruzes.Anticoagulants with different modes of action have been used in avian hematology, however, few studies on their effects on the hematological parameters of ostriches were published. Seeking to fill this gap, the effect of two routine anticoagulants (EDTA - 2 mg/mL blood and Heparin - 10 U/mL blood) on erythrocytes of ostriches was compared. Blood samples of 20 ostriches, treated with both anticoagulants were evaluated for integrity, globular volume and morphometry of the erythrocytes. The erythrocyte integrity was estimated by the concentration of free hemoglobin in plasma. The globular volume was obtained by the method of microhematocrit and morphometric data (area, bigger and smaller diameters) were automatically calculated by a computerized system for high resolution digital image. Hemolysis estimated by plasma hemoglobin, packed cell volume, area and maximum diameter of erythrocytes were significantly higher in the samples treated with EDTA. To better preserve the integrity and morphometry of erythrocytes, it was concluded that the heparin rather than EDTA is a more appropriated anticoagulant to evaluate erythrocyte dyscrasia in ostriches

    Primary lung neoplasms are rare in small animals and present variable clinical signs. Here we report a case of a 4-year-old male cat which presented with apathy, dyspnea and pleural effusion. The cytological analysis of the pleural effusion revealed massive presence of atypical cells and the histopathological examination of the lung demonstrated tissue infiltration by atypical cells arranged in solid nests. The cytological and histopathological findings were compatible with squamous cell lung carcinoma

    Foi avaliada a hipótese que, à semelhança dos neutrófilos de mamíferos, a produção de superóxido pelos heterófilos de avestruzes está associada com a maturidade funcional dessas células. Foram utilizados 20 avestruzes de ambos os sexos, divididos em dois grupos: 1-|30 dias (n=10) e 180-|240 dias (n=10) de idade. Para avaliação do metabolismo oxidativo dos heterófilos, estimou-se a produção de superóxido, utilizando o teste citoquímico não estimulado de redução do tetrazólio de nitroazul (NBT). A porcentagem média de redução heterofílica do NBT de avestruzes com até 30 dias de idade (0,7±1,3) foi menor (P<0,001) que a observada naqueles com idade entre seis e oito meses (6,2±2,7). Conclui-se que o metabolismo oxidativo dos heterófilos de avestruzes aumenta com a idade, sugerindo que essa menor produção de superóxido reduz a capacidade bactericida e pode contribuir para a alta mortalidade nos três primeiros meses de vida.It was valuated the hypothesis that, like the neutrophils of mammals, the superoxide production by avian heterophils is associated with functional maturity of these cells. Were used twenty ostriches of both sexes, divided into two groups: 1-|30 days (n=10) and 180-|240 days (n=10) of age. The oxidative metabolism by heterophils was estimated by superoxide production in not stimulated cytochemic test of nitroblue terazolium (NBT). The average percentage of heterophilic NBT reduction in ostriches with up to 30 days of age (0.7±1,3) was lower (P<0,001) than that observed in those aged between six and eight months (6,2±2,7).     This experiment was conducted at an aquicolous enterprise with the objective of evaluating the use of MOS and β-GLU as dietary supplements in an experimental lot in order to follow the zootechnical performance, establishing a relationship with the hematological parameters, the morphological alterations of the intestine, and the enzymatic activity (protease, lipase and amylase), and the water quality of 3,000 tilapia juveniles kept in cages (Wt = 24 ± 0.26g). Nine cages (6.0 m3) were used, with three treatments and three replications. 1: commercial feed without supplementation (control); 2: 0.1% per ton of MOS; 3: 0.03% per ton of purified β-GLU. The feed contained 36% of crude protein (CP) incorporated into the premix MOS and purified β-GLU (BIORIGIN®). The tilapia that had received the diet supplemented with β-GLU in a period of 90 days showed a favorable condition of the immune system, increase in the absorption surface of the front part of the intestine and consequently, growth in the activity of the digestive enzymes, denoting higher efficiency in the use of the nutrients in juveniles, providing satisfactory zootechnical performance in comparison with the other diets. This product may be used as a dietary supplement for this species when kept in cages

    Oxidative metabolism of the neutrophils in sheep treated with sodium monensin and experimentally submitted to ruminal acidosis

    Ruminal acidosis is due to excessive ingestion of carbohydrates of rapid fermentation without previous adaptation of the microorganisms, causing severe metabolic disturbances to the animals. The objective of the present study was to assess the neutrophilic oxidative metabolism in sheep treated with sodium monensin in experimentally induced ruminal lactic acidosis. A total of 18 male sheep, half-bred (ideal x Merino), fistulated in the rumen, were used; nine of them received 33 mg/kg of the ionophore diet per day, for 30 days; the others were controls. The acidosis was induced by supplying 15g of sucrose/kg of body weight. The clinical evaluation and the rumen and blood samples were obtained before (0h) and at 6, 12, 24 and 48 hours post-induction. In both groups, all the animals presented clinical manifestations of ruminal lactic acidosis 6 hours after the induction. From this period on, a significant pH decrease (P<0.05) was observed in the ruminal fluid, which reached levels below 5. There were relevant differences (P<0.05) between the groups 12 hours after the induction, when the sheep treated with monensin had higher values than those of the control group. During this period, the oxidative metabolism of the neutrophils remained inhibited, and the reestablishment of this function only occurred in the sheep which received monensin. Blood pH, plasmatic glucose and the ionizable calcium suffered alterations within its levels. The seric cortisol concentration rose significantly (P<0.05) in both groups, although differences (P<0.05) between them were found at the end of the observation period. The treatment with monensin did not influence the oxidative metabolism of the neutrophils inhibited by the lactic acidosis; however, a faster recovery of this metabolism was verified in the animals treated with the ionophore

    Erythrogram and erythrocytes measurement of ostriches (Struthio camelus) in São José do Rio Preto-SP, Brazil

    Ostriches' erythrocytic parameters help on the diagnosis of specific pathologies and serve as basic knowledge for studies in comparative avian pathology. To obtain reference values of erythrocyte indices for ostriches (Struthio camelus) raised in a commercial system in Brazil and verify if there are differences between gender and age groups, 240 healthy from both sexes animals were bled. Heparinized blood samples were analyzed using standard techniques to determine the red blood cell (RBC) count, hemoglobin concentration, packed cell volume (PCV), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) values, besides the red cell morphometry analysis using a computer software that calculates the greater and the smaller diameters of the erythrocytes. Prior to data analysis, ostriches were divided into three different age groups: from four to 13 months, from 13 to 23 months and from 23 to 30 months the. Younger ostriches presented lower erythrocyte indices than the older ones. The age group effect was only significant in females for the erythrocyte, hemoglobin, MCV, MCH, and MCHC variables. The female ostriches presented PCV, MCV, MCH, and MCHC values significantly higher than males in some age groups. It was observed that the erythrocytes of the female ostriches are more elongated and larger than males. It was concluded that erythrocytic parameters of ostriches in São José do Rio Preto-SP, Brazil are influenced by gender and age, highlighting the importance of consider besides these factors also the geoclimatic conditions to an adequate interpretation of the erythrogram

    Oxidative stress, superoxide production, and apoptosis of neutrophils in dogs with chronic kidney disease

    Oxidative stress is a key component in the immunosuppression of chronic kidney disease (CKD), and neutrophil function may be impaired by oxidative stress. To test the hypothesis that in uremic dogs with CKD, oxidative stress is increased and neutrophils become less viable and functional, 18 adult dogs with CKD were compared with 15 healthy adult dogs. Blood count and urinalysis were done, and the serum biochemical profile and plasma lipid peroxidation (measurement of thiobarbituric acid reactive substances) were determined with the use of commercial reagents. Plasma total antioxidant capacity (TAC) was measured with a spectrophotometer and commercial reagents, superoxide production with a hydroethidine probe, and the viability and apoptosis of neutrophils with capillary flow cytometry and the annexin V-PE system. The plasma concentrations of cholesterol (P = 0.0415), creatinine (P < 0.0001), and urea (P < 0.0001) were significantly greater in the uremic dogs than in the control dogs. The hematocrit (P = 0.0004), urine specific gravity (P = 0.015), and plasma lipid peroxidation (P < 0.0001) were significantly lower in the dogs that were in late stages of CKD than in the control group. Compared with those isolated from the control group, neutrophils isolated from the CKD group showed a higher rate of spontaneous (0.10 ± 0.05 versus 0.49 ± 0.09; P = 0.0033; median ± standard error of mean) and camptothecin-induced (18.53 ± 4.06 versus 44.67 ± 4.85; P = 0.0066) apoptosis and lower levels of superoxide production in the presence (1278.8 ± 372.8 versus 75.65 ± 86.6; P = 0.0022) and absence (135.29 ± 51.74 versus 41.29 ± 8.38; P = 0.0138) of phorbol-12-myristate-13-acetate stimulation. Thus, oxidative stress and acceleration of apoptosis occurs in dogs with CKD, the apoptosis diminishing the number of viable neutrophils and neutrophil superoxide production

    Leishmaniasis causes oxidative stress and alteration of oxidative metabolism and viability of neutrophils in dogs

    The aim of the present study was to test the hypothesis that oxidative stress and alteration of oxidative metabolism and apoptosis of neutrophils in dogs vary with the stage of leishmaniasis and to determine the contribution of uremia to such alterations. Dogs with leishmaniasis were classified into two stages: moderate (Leish II, n = 20) or very severe (i.e. with concurrent uremia; Leish IV, n = 20) according to the LeishVet Consensus. The two leishmaniasis groups were compared with uremic dogs without leishmaniasis (Uremic, n = 10) and to healthy dogs (Control, n = 30). To determine oxidative stress, total antioxidant/oxidant capacity, lipid peroxidation, total glutathione and the plasma antioxidants albumin, uric acid and bilirubin were quantified. Superoxide production was determined using the hydroethidine probe and viability and apoptosis were measured using annexin V-PE by capillary flow cytometry. Oxidative stress was present in both uremia and leishmaniasis with reduced total antioxidant capacity and was associated with increased induced production of superoxide and apoptosis. The greatest amount of oxidants was observed in animals with moderate disease only. Neutrophils from uremic dogs with and without leishmaniasis had decreased viability and an increased apoptosis rate in addition to increased lipid peroxidation. In conclusion, oxidative stress occurs in both stages of leishmaniasis with differences in intensity and levels of plasma markers; however, uremia does contribute to the decreased spontaneous viability of neutrophils in dogs in the final stage of the disease. © 2013 Elsevier Ltd.
