A new structural class of serine protease inhibitors revealed by the structure of the hirustasin–kallikrein complex
AbstractBackground: Hirustasin belongs to a class of serine protease inhibitors characterized by a well conserved pattern of cysteine residues. Unlike the closely related inhibitors, antistasin/ghilanten and guamerin, which are selective for coagulation factor Xa or neutrophil elastase, hirustasin binds specifically to tissue kallikrein. The conservation of the pattern of cysteine residues and the significant sequence homology suggest that these related inhibitors possess a similar three-dimensional structure to hirustasin.Results: The crystal structure of the complex between tissue kallikrein and hirustasin was analyzed at 2.4 Å resolution. Hirustasin folds into a brick-like structure that is dominated by five disulfide bridges and is sparse in secondary structural elements. The cysteine residues are connected in an abab cdecde pattern that causes the polypeptide chain to fold into two similar motifs. As a hydrophobic core is absent from hirustasin the disulfide bridges maintain the tertiary structure and present the primary binding loop to the active site of the protease. The general structural topography and disulfide connectivity of hirustasin has not previously been described.Conclusions: The crystal structure of the kallikrein–hirustasin complex reveals that hirustasin differs from other serine protease inhibitors in its conformation and its disulfide bond connectivity, making it the prototype for a new class of inhibitor. The disulfide pattern shows that the structure consists of two domains, but only the C-terminal domain interacts with the protease. The disulfide pattern of the N-terminal domain is related to the pattern found in other proteins. Kallikrein recognizes hirustasin by the formation of an antiparallel β sheet between the protease and the inhibitor. The P1 arginine binds in a deep negatively charged pocket of the enzyme. An additional pocket at the periphery of the active site accommodates the sidechain of the P4 valine
Fluorophore Labeled Kinase Detects Ligands That Bind within the MAPK Insert of p38α Kinase
The vast majority of small molecules known to modulate kinase activity, target the highly conserved ATP-pocket. Consequently, such ligands are often less specific and in case of inhibitors, this leads to the inhibition of multiple kinases. Thus, selective modulation of kinase function remains a major hurdle. One of the next great challenges in kinase research is the identification of ligands which bind to less conserved sites and target the non-catalytic functions of protein kinases. However, approaches that allow for the unambiguous identification of molecules that bind to these less conserved sites are few in number. We have previously reported the use of fluorescent labels in kinases (FLiK) to develop direct kinase binding assays that exclusively detect ligands which stabilize inactive (DFG-out) kinase conformations. Here, we present the successful application of the FLiK approach to develop a high-throughput binding assay capable of directly monitoring ligand binding to a remote site within the MAPK insert of p38α mitogen-activated protein kinase (MAPK). Guided by the crystal structure of an initially identified hit molecule in complex with p38α, we developed a tight binding ligand which may serve as an ideal starting point for further investigations of the biological function of the MAPK insert in regulating the p38α signaling pathway
Diastereoselektive Synthese von [alpha]-Tocopherol durch eine neue, enzym-ähnliche Chromanol-Zyklisierung
Die Tocopherol-Cyclase katalysiert die Bildung von g-Tocopherol 12 aus dem
Phytylhydrochinon 13 und spielt eine SchlĂĽsselrolle in der Biosynthese von Vitamin E.[33, 34]
Durch Inkubationsversuche mit radioaktiv markiertem Substrat konnte der Mechanismus dieser
Zyklisierungsreaktion aufgeklärt werden.[37] Diese verläuft über eine si-Protonierung der
Doppelbindung von 2 gefolgt von einer re-Attacke des phenolischen O-Atoms.
Das Ziel dieser Arbeit war es, eine Modellverbindung fĂĽr die Nachahmung dieser
enzymatischen Reaktion zu finden, als eine komplett neue Synthesestrategie fĂĽr die Herstellung
von enantiomerenreinem a-Tocopherol (1) (Vitamin E).
Das Schema solcher Modellverbindungen (84), die diese Eigenschaften erfüllen könnten, ist
unten dargestellt. FĂĽr die Beeinflussung der Zyklisierung wurde eine chirale Hilfsgruppe
basierend auf Prolin gewählt, die eine saure Gruppe (R2) trägt. Die Hilfsgruppe wurde so
angebracht, dass diese nach erfolgter Zyklisierung mittels Hydrogenolyse wieder abgespalten,
und so das gewĂĽnschte a-Tocopherol (1) in wenigen Reaktionsschritten erhalten werden kann.
FĂĽr die Beeinflussung der Zyklisierung sollte eine geeignete chirale Hilfsgruppe gefunden
werden, mit der ein möglichst hoher Diastereoisomerenüberschuss erzielt werden kann.
Modellverbindungen mit Prolin als chirale Hilfsgruppe:
Als erstes wurden die Modellverbindungen 85 und 105 mit Prolin als chirale Hilfsgruppe,
ausgehend von Phytylbromid 90, synthetisiert. Der SchlĂĽsselschritt dieser Synthese war die
Entwicklung und DurchfĂĽhrung der Mannich-Reaktion von 88 mit Prolin. Als Schutzgruppe
(R) wurde fĂĽr 85 ein Pivaloyl-Ester und fĂĽr 105 ein Camphanoyl-Ester verwendet.
Die so erhaltenen Modellverbindungen 85 und 105 wurden anschliessend auf deren
Zyklisierungseigenschaften unter Verwendung von verschiedenen Lewis-Säuren (LS) getestet.
Die durchgefĂĽhrten Zyklisierungen zeigten, dass mit den entsprechenden Hydrochloridsalzen
von 85 bzw. 105 höhere Diastereoisomerenüberschüsse für die gebildeten Zyklisierungsprodukte
102 bzw. 109 erzielt werden konnten als mit den zwitterionischen Formen. Als bestes
Resultat konnte mit 105·HCl ein Diastereoisomerenüberschuss (de) von 38% bei einer
Ausbeute von 45% erreicht werden, unter Verwendung von AlCl3 als Lewis-Säure.
Modellverbindungen mit Prolinsulphonsäure als chirale Hilfsgruppe:
Die oben durchgefĂĽhrten Zyklisierungsresultate zeigten, dass Prolin einen Effekt auf die
Diastereoselektivität der Zyklisierungsreaktion ausübt, jedoch konnten durch weitere
Veränderungen der Reaktionsbedingungen keine besseren Resultate erzielt werden. Daher
wurde für die nächsten Modellverbindungen das (S)-Prolin durch Prolinsulphonsäure ersetzt,
wodurch eine Möglichkeit geschaffen werden sollte, die Zyklisierung intramolekular
durchzufĂĽhren. Diese Modellverbindungen (112 und 125) wurden dann auf ihre Zyklisierungseigenschaften
hin ĂĽberprĂĽft, wobei ein besonderes Augenmerk auf die Zyklisierung mit
Brönsted-Säuren (BS) gelegt wurde.
Die Zyklisierungen der Modellverbindung 112 mit p-TsOH in CH3CN verliefen nur schlecht;
bessere Resultate wurden mit SnCl4 erreicht, wobei als bestes Resultat ein DiastereoisomerenĂĽberschuss
von 28% bei einer Ausbeute von 95% erhalten werden konnte. Mit der Nmethylierten
Modellverbindung 125 konnte mit p-TsOH, unter Verwendung von
Propylencarbonat (PC) als Lösungsmittel, wesentlich höhere Diastereoisomerenüberschüsse
erreicht werden. Dabei konnte für gute Ausbeute (ca. 70%) de’s von 45% erzielt werden. Für
diese Zyklisierungen wurde immer das nicht natĂĽrliche (SRR)-Diastereoisomer im Ăśberschuss
gebildete. Weitere Untersuchungen mit einem unsubstituierten Tocopherol-Vorläufer zeigten,
dass die Prolinsulphonsäure alleine nicht azide genug ist, die Zyklisierung zu Katalysieren. Die
Zyklisierung verläuft daher mit grosser Wahrscheinlichkeit nicht über eine intramolekulare
Protonierung der Doppelbindung.
Modellverbindungen mit Dipeptiden als chirale Hilfsgruppe:
Des weiteren wurden andere Prolin-Derivate als chirale Hilfsverbindung fĂĽr den diastereoselektiven
Chromanolringschluss getestet. Insbesondere erschienen aber Dipeptide, basierend
auf einer Sequenz aus Prolin und einer beliebig anderen, bevorzugt aber sauren, Aminosäure als
chirale Hilfsverbindung besonders attraktiv zu sein. Zu diesem Zweck wurde die Modellverbindung
(S,S)-Asp-140 durch Kopplung der Asparaginsäure an den Vorläufer (S)-105
hergestellt, und dann unter Verwendung einer Standard-Reaktionssequenz auf deren
Zyklisierungseigenschaften hin untersucht. Diese beinhaltet die Zyklisierung mit SnCl4 und p-
TsOH in CH2Cl2, gefolgt von der Reduktion des Zyklisierungsproduktes zum CamphanoylgeschĂĽtzten
a-Tocopherol (104). Die Resultate sind nachfolgend zusammengefasst.
Durch die Verwendung von Pro-Asp als chirale Hilfsgruppe konnte die Selektivität der
Zyklisierungsreaktion erheblich gesteigert werden. Vor Allem die Zyklisierungen mit p-TsOH
lieferten hohe de’s von bis zu 78%. Mit SnCl4 konnte als bestes Resultat ein de von ca. 60%
erhalten werden.
Da sich erneut das nicht natĂĽrliche (SRR)-Diastereoisomer im Ăśberschuss bildete, wurde die
Stereozentren der chiralen Hilfsgruppe geändert. Die so erhaltene Modellverbindung (R,R)-
Asp-140 wurde ebenfalls auf deren Zyklisierungseigenschaften unter den oben verwendeten
Standardbedingungen getestet. Mit dieser Modellverbindung konnte eine Umkehrung der
Selektivität der Zyklisierungsreaktion erreicht werden. Im Fall von p-TsOH wurden
DiastereoisomerenĂĽberschĂĽsse von 65% zu Gunsten des natĂĽrlichen (RRR)-Diastereoisomers
erhalten. Somit war eine chirale Hilfsgruppe gefunden mit der die Zyklisierung effektiv
kontrolliert werden konnte. Mit der gewählten Synthesestrategie war zudem eine Möglichkeit
geschaffen worden, sowohl das (SRR)- als auch das (RRR)-Diastereoisomer von a-Tocopherol
(1) in guter Isomerenreinheit zu synthetisieren.
Zusätzliche Untersuchungen mit den Monomethylestern von (S,S)-Asp-140 zeigten, dass bei
dieser Zyklisierung für eine gute Reaktivität wie auch für eine hohe Selektivität beide
Säurefunktionen benötigt werden. Darüber hinaus spielt die räumliche Distanz zwischen den
beiden Carbonsäuren ebenfalls eine wichtige Rolle, wie das Beispiel einer Modellverbindung
mit Glutaminsäure zeigte, bei der die Carbonsäuren um eine CH2-Gruppe weiter voneinander
entfernt sind. Dieser kooperative Effekt könnte durch das Vorhandensein einer
Wasserstoffbrücke zwischen den beiden Säurefunktionen von (S,S)-Asp-140 erklärt werden,
wodurch die Azidität der an der Zyklisierungsreaktion beteiligten Carbonsäure wesentlich
erhöht wird. Als Zyklisierungs-Mechanismus wäre eine Kaskadenreaktion denkbar bei der das
Proton der p-TsOH über eine oder beiden Säurefunktionen an die Doppelbindung
weitergegeben wird.
Diese Hypothese könnte die gemachten Beobachtungen erklären. Der Zyklisierungsvorläufer
Asp-140 kann daher als Modellverbindung fĂĽr die Nachahmung des enzymatischen
Chromanolringschlusses angesehen werden
Structures of designed armadillo-repeat proteins show propagation of inter-repeat interface effects
The armadillo repeat serves as a scaffold for the development of modular peptide-recognition modules. In order to develop such a system, three crystal structures of designed armadillo-repeat proteins with third-generation N-caps (YIII-type), four or five internal repeats (M-type) and second-generation C-caps (AII-type) were determined at 1.8 Ă… (His-YIIIM4AII), 2.0 Ă… (His-YIIIM5AII) and 1.95 Ă… (YIIIM5AII) resolution and compared with those of variants with third-generation C-caps. All constructs are full consensus designs in which the internal repeats have exactly the same sequence, and hence identical conformations of the internal repeats are expected. The N-cap and internal repeats M1 to M3 are indeed extremely similar, but the comparison reveals structural differences in internal repeats M4 and M5 and the C-cap. These differences are caused by long-range effects of the C-cap, contacting molecules in the crystal, and the intrinsic design of the repeat. Unfortunately, the rigid-body movement of the C-terminal part impairs the regular arrangement of internal repeats that forms the putative peptide-binding site. The second-generation C-cap improves the packing of buried residues and thereby the stability of the protein. These considerations are useful for future improvements of an armadillo-repeat-based peptide-recognition system
An Invariant Surface Patch on the TRIM5α PRYSPRY Domain Is Required for Retroviral Restriction but Dispensable for Capsid Binding▿
TRIM5α is a retrovirus restriction factor in the host cell cytoplasm that blocks infection before provirus establishment. Restriction activity requires capsid (CA)-specific recognition by the PRYSPRY domain of TRIM5α. To better understand the restriction mechanism, nine charge-cluster-to-triple-alanine mutants in the TRIM5α PRYSPRY domain were assessed for CA-specific restriction activity. Five mutants distributed along the TRIM5α PRYSPRY primary sequence disrupted restriction activity against N-tropic murine leukemia virus and equine infectious anemia virus. Modeling of the TRIM5α PRYSPRY domain based on the crystal structures of PRYSPRY-19q13.4.1, GUSTAVUS, and TRIM21 identified a surface patch where disruptive mutants clustered. All mutants in this patch retained CA-binding activity, a reticular distribution in the cytoplasm, and steady-state protein levels comparable to those of the wild type. Residues in the essential patch are conserved in TRIM5α orthologues and in closely related paralogues. The same surface patch in the TRIM18 and TRIM20 PRYSPRY domains is the site of mutants causing Opitz syndrome and familial Mediterranean fever. These results indicate that, in addition to CA-specific binding, the PRYSPRY domain possesses a second function, possibly binding of a cofactor, that is essential for retroviral restriction activity by TRIM5α
