    Spatial and temporal EEG dynamics of dual-task driving performance

    <p>Abstract</p> <p>Background</p> <p>Driver distraction is a significant cause of traffic accidents. The aim of this study is to investigate Electroencephalography (EEG) dynamics in relation to distraction during driving. To study human cognition under a specific driving task, simulated real driving using virtual reality (VR)-based simulation and designed dual-task events are built, which include unexpected car deviations and mathematics questions.</p> <p>Methods</p> <p>We designed five cases with different stimulus onset asynchrony (SOA) to investigate the distraction effects between the deviations and equations. The EEG channel signals are first converted into separated brain sources by independent component analysis (ICA). Then, event-related spectral perturbation (ERSP) changes of the EEG power spectrum are used to evaluate brain dynamics in time-frequency domains.</p> <p>Results</p> <p>Power increases in the theta and beta bands are observed in relation with distraction effects in the frontal cortex. In the motor area, alpha and beta power suppressions are also observed. All of the above results are consistently observed across 15 subjects. Additionally, further analysis demonstrates that response time and multiple cortical EEG power both changed significantly with different SOA.</p> <p>Conclusions</p> <p>This study suggests that theta power increases in the frontal area is related to driver distraction and represents the strength of distraction in real-life situations.</p

    TNFAIP3, a negative regulator of the TLR signaling pathway, is a potential predictive biomarker of response to antidepressant treatment in major depressive disorder

    AbstractInflammation and abnormalities in Toll-like receptor (TLR) expression and activation have been linked to major depressive disorder (MDD). However, negative regulators of TLR pathways have not been previously investigated in this context. Here, we sought to investigate the association of depression severity, measured by the 17-item Hamilton Depression Rating Scale (HAMD-17), with mRNA expression levels of negative regulators of the TLR pathway, including SOCS1, TOLLIP, SIGIRR, MyD88s, NOD2 and TNFAIP3, in peripheral blood mononuclear cells (PBMCs) from 100 patients with MDD and 53 healthy controls, before and after treatment with antidepressants. Positive regulators of the TLR4 pathway, including Pellino 1, TRAF6 and IRAK1, were also investigated. Among all patients, MyD88s, and TNFAIP3 mRNAs were expressed at lower levels in PBMCs from patients with MDD. Multiple linear regression analyses revealed that TNFAIP3 mRNA expression before treatment was inversely correlated with severity of depression and effectively predicted improvement in HAMD-17 scores. Among 79 treatment-completers, only TNFAIP3 mRNA was significantly increased by treatment with antidepressants for 4 weeks. Treatment of human monocytes (THP-1) and mouse microglia (SIM-A9) cell lines with fluoxetine significantly increased TNFAIP3 mRNA expression and suppressed IL-6 levels. The suppressive effect of fluoxetine on IL-6 was attenuated by knockdown of TNFAIP3 expression. These findings suggest that both dysfunction of the negative regulatory system in patients with MDD and antidepressant treatment exert anti-inflammatory effects, at least in part through increased expression of the TNFAIP3 gene. They also indicate that modulating expression of the TNFAIP3 gene to rebalance TLR-mediated inflammatory signaling may be potential therapeutic strategy for treating MDD

    Face recognition using nonparametric-weighted Fisherfaces

    This study presents an appearance-based face recognition scheme called the nonparametric-weighted Fisherfaces (NW-Fisherfaces). Pixels in a facial image are considered as coordinates in a high-dimensional space and are transformed into a face subspace for analysis by using nonparametric-weighted feature extraction (NWFE). According to previous studies of hyperspectral image classification, NWFE is a powerful tool for extracting hyperspectral image features. The Fisherfaces method maximizes the ratio of between-class scatter to that of within-class scatter. In this study, the proposed NW-Fisherfaces weighted the between-class scatter to emphasize the boundary structure of the transformed face subspace and, therefore, enhances the separability for different persons' face. The proposed NW-Fisherfaces was compared with Orthogonal Laplacianfaces, Eigenfaces, Fisherfaces, direct linear discriminant analysis, and null space linear discriminant analysis methods for tests on five facial databases.     Association between interleukin-6 receptor gene variations and atherosclerotic lipid profiles among young adolescents in Taiwan

    <p>Abstract</p> <p>Background</p> <p>To analyze the potential genetic associations between four polymorphisms of interleukin-6 receptor (IL-6R) gene and atherosclerotic lipid profiles among young adolescents in Taiwan.</p> <p>Methods</p> <p>Using data from the Taipei Children Heart Study-II - a cross-sectional survey in 2003. After multi-stage sampling, we selected 418 boys and 441 girls with an average age of 13.1 years. We genotyped the subjects for four IL-6R gene polymorphisms (rs4845617 G/A, rs4845623 A/G, rs8192284 A/C, and rs2229238 C/T) using a TaqMan 5' nuclease assay. Lipid profiles, including total cholesterol (CHOL), triglycerides (TG), high density lipoprotein-cholesterol (HDL-C), low density lipoprotein-cholesterol (LDL-C) were measured using standard methods. We also calculated CHOL/HDL-C ratio, LDL-C/HDL-C ratio, and TG/HDL-C ratio as atherosclerotic indexes.</p> <p>Results</p> <p>IL-6R rs8192284 A/C and rs2229238 C/T variants showed strong associations with high TG (additive model, OR = 1.58, 95%CI: 1.05-2.37; OR = 1.55, 95%CI: 1.04-2.29, respectively), low HDL-C (additive model, OR = 1.57, 95%CI: 1.03-2.39; OR = 1.68, 95%CI: 1.12-2.52, respectively), and high CHOL/HDL-C (additive model, OR = 1.68, 95%CI: 1.08-2.61, OR = 1.82, 95%CI: 1.18-2.79, respectively) in girls. We inferred five common haplotypes using rs4845617 G/A, rs4845623 A/G, and rs2229238 C/T (GAC, GAT, GGC, AAC, and AAT). In girls, the AAT haplotype was associated with a significant risk of high TG, low HDL-C, high CHOL/HDL-C, and abnormal lipid levels (high TG or low HDL-C) when compared with the GAC haplotype (OR range = 3.08-4.40, all p < 0.05).</p> <p>Conclusion</p> <p>The IL-6R rs8192284 A/C and rs2229238 C/T variants are associated with dyslipidemia in girls, but not in boys. The AAT haplotype of the IL-6R gene (rs4845617 G/A, rs4845623 A/G, and rs2229238 C/T) may play an important role in the pathogenesis of dyslipidemia and atherosclerosis in girls.</p

    5-ALA mediated photodynamic therapy induces autophagic cell death via AMP-activated protein kinase

    Photodynamic therapy (PDT) has been developed as an anticancer treatment, which is based on the tumor-specific accumulation of a photosensitizer that induces cell death after irradiation of light with a specific wavelength. Depending on the subcellular localization of the photosensitizer, PDT could trigger various signal transduction cascades and induce cell death such as apoptosis, autophagy, and necrosis. In this study, we report that both AMP-activated protein kinase (AMPK) and mitogen-activated protein kinase (MAPK) signaling cascades are activated following 5-aminolevulinic acid (ALA)-mediated PDT in both PC12 and CL1-0 cells. Although the activities of caspase-9 and -3 are elevated, the caspase inhibitor zVAD-fmk did not protect cells against ALA-PDT-induced cell death. Instead, autophagic cell death was found in PC12 and CL1-0 cells treated with ALA-PDT. Most importantly, we report here for the first time that it is the activation of AMPK, but not MAPKs that plays a crucial role in mediating autophagic cell death induced by ALA-PDT. This novel observation indicates that the AMPK pathway play an important role in ALA-PDT-induced autophagy

    Identifying Ketamine Responses in Treatment-Resistant Depression Using a Wearable Forehead EEG

    This study explores the responses to ketamine in patients with treatment-resistant depression (TRD) using a wearable forehead electroencephalography (EEG) device. We recruited fifty-five outpatients with TRD who were randomised into three approximately equal-sized groups (A: 0.5 mg/kg ketamine; B: 0.2 mg/kg ketamine; and C: normal saline) under double-blind conditions. The ketamine responses were measured by EEG signals and Hamilton Depression Rating Scale (HDRS) scores. At baseline, responders showed a significantly weaker EEG theta power than did non- responders (p < 0.05). Responders exhibited a higher EEG alpha power but lower EEG alpha asymmetry and theta cordance at post-treatment than at baseline (p < 0.05). Furthermore, our baseline EEG predictor classified responders and non-responders with 81.3 +- 9.5% accuracy, 82.1 +- 8.6% sensitivity and 91.9 +- 7.4% specificity. In conclusion, the rapid antidepressant effects of mixed doses of ketamine are associated with prefrontal EEG power, asymmetry and cordance at baseline and early post-treatment changes. The prefrontal EEG patterns at baseline may account for recognising ketamine effects in advance. Our randomised, double- blind, placebo-controlled study provides information regarding clinical impacts on the potential targets underlying baseline identification and early changes from the effects of ketamine in patients with TRD.Comment: This revised article is submitting to IEEE TBM

    One of the most ancient therapeutic maneuvers, acupuncture is part of Traditional Chinese Medicine. Based in ancient philosophic thinking and observation of naturally occurred phenomena, Traditional Chinese Medicine possesses peculiar pathophysiology,complicated diagnosis system based on examination of pulse, tongue and observation of patients´ behavior and attitudes. During long decades, acupuncture has been considered as an exotic alternative to Orthodox Western Medicine, however, recent studies and the panel studies promoted by NIH (National Institutes of Health) in 1988 have elected acupuncture asa validated therapy for a long listed health disorders, thus transforming acupuncture as a serious therapeutic modality. More discussions and studies should be evaluated in order to obtaining more evidence-based indication for acupuncture and to carry out the medical education in teaching acupuncture.Uma das mais antigas modalidades de terapêutica, acupuntura faz parte da MedicinaTradicional Chinesa. Baseada principalmente em um antigo pensamento filosófico chinês e na observação da ocorrência de fenômenos da natureza. Medicina Tradicional Chinesa possui uma fisiopatologia peculiar, um sistema de diagnóstico complicado baseado em exame de língua e pulso e na observação de comportamento e atitudes de pacientes. Durante décadas,a acupuntura foi considerada uma alternativa exótica à Medicina Ocidental Ortodoxa, entretanto, estudos recentes e estudos de painel do NIH norte americano em 1988 elegeram a acupuntura como uma terapia validada para uma extensa lista de afecções, transformando a em uma modalidade terapêutica séria. Necessitamos de mais estudos e discussões para aumentar as indicações baseadas em evidência para outras afecções, além de ajudar a implementar a educação médica em acupuntura

    Acupuncture has been put in evidence more recently. It represents an ascending practice among under-graduated medical students, as well as among graduated physicians in search for their specialization. Recently, acupuncture won BMA approval, which not only recommended NHS to accept acupuncture to treat chronic headache, but suggested that more primary care doctors should be trained to practice acupuncture. In a review of some recent studies, acupuncture has been studied as a validated practice to treat conditions like systemic arterial hypertension and in vitro fertilization, conditions which differ from usual indication of acupuncture. This new situation put a new perspective to acupuncture, perhaps it is on due time that acupuncture should be considered as part of orthodox medicine.A acupuntura está cada vez mais em evidência, além de ser uma especialidade em ascensão tanto entre os estudantes de medicina, como entre os médicos que buscam a sua especialização, ganhou destaque da Associação Médica Britânica que advogou a sua aprovação perante a comunidade médica britânica, além de recomendar o Sistema de Saúde Pública Britânica a aceitar o tratamento de cefaléia crônica por acupuntura, aconselha que o mesmo encoraje os médicos de atenção primária a passar por treinamento em acupuntura. Na revisão de trabalhos recentes, a acupuntura está sendo pesquisada para o tratamento de diversas moléstias ou condições clínicas diferentes daquelas a que eram indicadas. Situações como hipertensão arterial sistêmica e fertilização in vitro, a acupuntura tem sido estudada como opções de tratamento válidas.
