We discuss SU(5) Grand Unified Theories in the context of orientifold compactifications. Specifically, we investigate two and three D-brane stack realizations of the Georgi-Glashow and the flipped SU(5) model and analyze them with respect to their Yukawa couplings. As pointed out in arXiv:0909.0271 the most economical Georgi-Glashow realization based on two stacks generically suffers from a disastrous large proton decay rate. We show that allowing for an additional U(1) D-brane stack this as well as other phenomenological problems can be resolved. We exemplify with globally consistent Georgi-Glashow models based on RCFT that these D-brane quivers can be indeed embedded in a global setting. These globally consistent realizations admit rigid O(1) instantons inducing the perturbatively missing coupling 10105^H. Finally we show that flipped SU(5) D-brane realizations even with multiple U(1) D-brane stacks are plagued by severe phenomenological drawbacks which generically cannot be overcome.Comment: 34 pages v2 minor correction

This is the final published version. Available from PLoS via the DOI in this record. All relevant data are available from Figshare, under the DOI http://dx.doi. org/10.6084/m9.figshare.1292883.AIMS: Weight-loss after bariatric surgery improves insulin sensitivity, but the underlying molecular mechanism is not clear. To ascertain the effect of bariatric surgery on insulin signalling, we examined glucose disposal and Akt activation in morbidly obese volunteers before and after Roux-en-Y gastric bypass surgery (RYGB), and compared this to lean volunteers. MATERIALS AND METHODS: The hyperinsulinaemic euglycaemic clamp, at five infusion rates, was used to determine glucose disposal rates (GDR) in eight morbidly obese (body mass index, BMI=47.3 ± 2.2 kg/m(2)) patients, before and after RYGB, and in eight lean volunteers (BMI=20.7 ± 0.7 kg/m2). Biopsies of brachioradialis muscle, taken at fasting and insulin concentrations that induced half-maximal (GDR50) and maximal (GDR100) GDR in each subject, were used to examine the phosphorylation of Akt-Thr308, Akt-473, and pras40, in vivo biomarkers for Akt activity. RESULTS: Pre-operatively, insulin-stimulated GDR was lower in the obese compared to the lean individuals (P<0.001). Weight-loss of 29.9 ± 4 kg after surgery significantly improved GDR50 (P=0.004) but not GDR100 (P=0.3). These subjects still remained significantly more insulin resistant than the lean individuals (p<0.001). Weight loss increased insulin-stimulated skeletal muscle Akt-Thr308 and Akt-Ser473 phosphorylation, P=0.02 and P=0.03 respectively (MANCOVA), and Akt activity towards the substrate PRAS40 (P=0.003, MANCOVA), and in contrast to GDR, were fully normalised after the surgery (obese vs lean, P=0.6, P=0.35, P=0.46, respectively). CONCLUSIONS: Our data show that although Akt activity substantially improved after surgery, it did not lead to a full restoration of insulin-stimulated glucose disposal. This suggests that a major defect downstream of, or parallel to, Akt signalling remains after significant weight-loss.Diabetes Research and Wellness FoundatioMedical Research Council (MRC)Astra Zenec

Serotonin and corticosterone rhythms in mice exposed to cigarette smoke and in patients with COPD:implication for COPD-associated neuropathogenesis

    Get PDF
The circadian timing system controls daily rhythms of physiology and behavior, and disruption of clock function can trigger stressful life events. Daily exposure to cigarette smoke (CS) can lead to alteration in diverse biological and physiological processes. Smoking is associated with mood disorders, including depression and anxiety. Patients with chronic obstructive pulmonary disease (COPD) have abnormal circadian rhythms, reflected by daily changes in respiratory symptoms and lung function. Corticosterone (CORT) is an adrenal steroid that plays a considerable role in stress and anti-inflammatory responses. Serotonin (5-hydroxytryptamine; 5HT) is a neurohormone, which plays a role in sleep/wake regulation and affective disorders. Secretion of stress hormones (CORT and 5HT) is under the control of the circadian clock in the suprachiasmatic nucleus. Since smoking is a contributing factor in the development of COPD, we hypothesize that CS can affect circadian rhythms of CORT and 5HT secretion leading to sleep and mood disorders in smokers and patients with COPD. We measured the daily rhythms of plasma CORT and 5HT in mice following acute (3 d), sub-chronic (10 d) or chronic (6 mo) CS exposure and in plasma from non-smokers, smokers and patients with COPD. Acute and chronic CS exposure affected both the timing (peak phase) and amplitude of the daily rhythm of plasma CORT and 5HT in mice. Acute CS appeared to have subtle time-dependent effects on CORT levels but more pronounced effects on 5HT. As compared with CORT, plasma 5HT was slightly elevated in smokers but was reduced in patients with COPD. Thus, the effects of CS on plasma 5HT were consistent between mice and patients with COPD. Together, these data reveal a significant impact of CS exposure on rhythms of stress hormone secretion and subsequent detrimental effects on cognitive function, depression-like behavior, mood/anxiety and sleep quality in smokers and patients with COPD

On Holographic description of the Kerr-Newman-AdS-dS black holes

In this paper, we study the holographic description of the generic four-dimensional non-extremal Kerr-Newman-AdS-dS black holes. We find that if focusing on the near-horizon region, for the massless scalar scattering in the low-frequency limit, there exists hidden conformal symmetry on the solution space. Similar to the Kerr case, this suggests that the Kerr-Newman-AdS-dS black hole is dual to a two-dimensional CFT with central charges cL=cR=6a(r++r)kc_L=c_R=\frac{6a(r_++r_\ast)}{k} and temperatures TL=k(r+2+r2+2a2)4πaΞ(r++r),TR=k(r+r)4πaΞT_L=\frac{k(r_+^2+r_\ast^2+2a^2)}{4\pi a\Xi(r_++r_\ast)}, T_R=\frac{k(r_+-r_\ast)}{4\pi a\Xi}. The macroscopic Bekenstein-Hawking entropy could be recovered from the microscopic counting in dual CFT via the Cardy formula. Using the Minkowski prescription, we compute the real-time correlators of the scalar, photon and graviton in near horizon geometry of near extremal Kerr-AdS-dS black hole. In all these cases, the retarded Green's function and the corresponding absorption cross section are in perfect match with CFT prediction. We further discuss the low-frequency scattering of a charged scalar by a Kerr-Newman-AdS-dS black hole and find the dual CFT description.Comment: 22 pages; minor corrections, conlusion unchanged, references added;published versio

Extended RDF: Computability and Complexity Issues

ERDF stable model semantics is a recently proposed semantics for ERDF ontologies and a faithful extension of RDFS semantics on RDF graphs. In this paper, we elaborate on the computability and complexity issues of the ERDF stable model semantics. Based on the undecidability result of ERDF stable model semantics, decidability under this semantics cannot be achieved, unless ERDF ontologies of restricted syntax are considered. Therefore, we propose a slightly modified semantics for ERDF ontologies, called ERDF #n- stable model semantics. We show that entailment under this semantics is, in general, decidable and also extends RDFS entailment. Equivalence statements between the two semantics are provided. Additionally, we provide algorithms that compute the ERDF #n-stable models of syntax-restricted and general ERDF ontologies. Further, we provide complexity results for the ERDF #nstable model semantics on syntax-restricted and general ERDF ontologies. Finally, we provide complexity results for the ERDF stable model semantics on syntax-restricted ERDF ontologies

An inhibitory pull-push circuit in frontal cortex.

    Get PDF
Push-pull is a canonical computation of excitatory cortical circuits. By contrast, we identify a pull-push inhibitory circuit in frontal cortex that originates in vasoactive intestinal polypeptide (VIP)-expressing interneurons. During arousal, VIP cells rapidly and directly inhibit pyramidal neurons; VIP cells also indirectly excite these pyramidal neurons via parallel disinhibition. Thus, arousal exerts a feedback pull-push influence on excitatory neurons-an inversion of the canonical push-pull of feedforward input

Optical one-way quantum computing with a simulated valence-bond solid

One-way quantum computation proceeds by sequentially measuring individual spins (qubits) in an entangled many-spin resource state. It remains a challenge, however, to efficiently produce such resource states. Is it possible to reduce the task of generating these states to simply cooling a quantum many-body system to its ground state? Cluster states, the canonical resource for one-way quantum computing, do not naturally occur as ground states of physical systems. This led to a significant effort to identify alternative resource states that appear as ground states in spin lattices. An appealing candidate is a valence-bond-solid state described by Affleck, Kennedy, Lieb, and Tasaki (AKLT). It is the unique, gapped ground state for a two-body Hamiltonian on a spin-1 chain, and can be used as a resource for one-way quantum computing. Here, we experimentally generate a photonic AKLT state and use it to implement single-qubit quantum logic gates.Comment: 11 pages, 4 figures, 8 tables - added one referenc

New Insights into Properties of Large-N Holographic Thermal QCD at Finite Gauge Coupling at (the Non-Conformal/Next-to) Leading Order in N

In the context of [1]'s string theoretic dual of large-N thermal QCD-like theories at finite gauge/string coupling (as part of the `MQGP' limit of [2]), we discuss the following. First, up to LO in N, using the results of [3], we show that the local T^3 of [2] is the T^2-invariant sLag of [3] in a resolved conifold. This, together with the results of [4], shows that for a (predominantly resolved or deformed) resolved warped deformed conifold, the local T^3 of [2] in the MQGP limit, is the T^2-invariant sLag of [3] justifying the construction of the delocalized SYZ type IIA mirror of the type IIB background of [1]. Then, using the prescription of [5], we obtain the temperature dependence of the thermal (and electrical) conductivity working up to leading order in N (the number of D3-branes), and upon comparison with [6] show that the results mimic a 1+1-dimensional Luttinger liquid with impurities. Further, including sub-leading non-conformal terms in the metric determined by M (the number of fractional D-branes = the number of colors = 3 in the IR after the end of a Seiberg duality cascade), by looking at respectively the scalar, vector and tensor modes of metric perturbations and using [7]'s prescription of constructing appropriate gauge-invariant perturbations, we obtain respectively the speed of sound, the diffusion constant and the shear viscosity \eta (and \eta/s) including the non-conformal O((g_s M^2) (g_s N_f)/N<<1)-corrections, N_f being the number of flavor D7-branes.Comment: 1+75 pages, LaTeX; Some corrections in Tc-related calculations, results unchange

Gender-dependent differences in plasma matrix metalloproteinase-8 elevated in pulmonary tuberculosis.

Tuberculosis (TB) remains a global health pandemic and greater understanding of underlying pathogenesis is required to develop novel therapeutic and diagnostic approaches. Matrix metalloproteinases (MMPs) are emerging as key effectors of tissue destruction in TB but have not been comprehensively studied in plasma, nor have gender differences been investigated. We measured the plasma concentrations of MMPs in a carefully characterised, prospectively recruited clinical cohort of 380 individuals. The collagenases, MMP-1 and MMP-8, were elevated in plasma of patients with pulmonary TB relative to healthy controls, and MMP-7 (matrilysin) and MMP-9 (gelatinase B) were also increased. MMP-8 was TB-specific (p<0.001), not being elevated in symptomatic controls (symptoms suspicious of TB but active disease excluded). Plasma MMP-8 concentrations inversely correlated with body mass index. Plasma MMP-8 concentration was 1.51-fold higher in males than females with TB (p<0.05) and this difference was not due to greater disease severity in men. Gender-specific analysis of MMPs demonstrated consistent increase in MMP-1 and -8 in TB, but MMP-8 was a better discriminator for TB in men. Plasma collagenases are elevated in pulmonary TB and differ between men and women. Gender must be considered in investigation of TB immunopathology and development of novel diagnostic markers

Interleukin-1 regulates multiple atherogenic mechanisms in response to fat feeding

Background: Atherosclerosis is an inflammatory process that develops in individuals with known risk factors that include hypertension and hyperlipidaemia, influenced by diet. However, the interplay between diet, inflammatory mechanisms and vascular risk factors requires further research. We hypothesised that interleukin-1 (IL-1) signaling in the vessel wall would raise arterial blood pressure and promote atheroma. Methodology/Principal Findings: Apoe(-/-) and Apoe(-/-)/IL-1R1(-/-) mice were fed high fat diets for 8 weeks, and their blood pressure and atherosclerosis development measured. Apoe(-/-)/IL-R1(-/-) mice had a reduced blood pressure and significantly less atheroma than Apoe(-/-) mice. Selective loss of IL-1 signaling in the vessel wall by bone marrow transplantation also reduced plaque burden (p<0.05). This was associated with an IL-1 mediated loss of endothelium-dependent relaxation and an increase in vessel wall Nox 4. Inhibition of IL-1 restored endothelium-dependent vasodilatation and reduced levels of arterial oxidative stress. Conclusions/Significance: The IL-1 cytokine system links atherogenic environmental stimuli with arterial inflammation, oxidative stress, increased blood pressure and atherosclerosis. This is the first demonstration that inhibition of a single cytokine can block the rise in blood pressure in response to an environmental stimulus. IL-1 inhibition may have profound beneficial effects on atherogenesis in man
