    Tensor-valued diffusion MRI in under 3 minutes: An initial survey of microscopic anisotropy and tissue heterogeneity in intracranial tumors

    Purpose: To evaluate the feasibility of a 3-minute b-tensor encoding protocol for diffusion MRI-based assessment of the microscopic anisotropy and tissue heterogeneity in a wide range of intracranial tumors. Methods: B-tensor encoding was performed in 42 patients with intracranial tumors (gliomas, meningiomas, adenomas, metastases). Microscopic anisotropy and tissue heterogeneity were evaluated by estimating the anisotropic kurtosis (MKAMK_A) and isotropic kurtosis (MKIMK_I), respectively. An extensive imaging protocol was compared with a faster 3-minute protocol. Results: The fast imaging protocol yielded parameters with characteristics in terms of bias and precision similar to the full protocol. Glioblastomas had lower microscopic anisotropy than meningiomas (MKA=0.29±0.06(MK_A = 0.29 \pm 0.06 versus 0.45±0.08,p=0.003)0.45\pm0.08, p = 0.003). Metastases had higher tissue heterogeneity (MKI=0.57±0.07)(MK_I = 0.57\pm0.07) than both the glioblastomas (0.44±0.06,p<0.001)(0.44\pm0.06, p < 0.001) and meningiomas (0.46±0.06,p=0.03)(0.46\pm0.06, p = 0.03). Conclusion: Evaluation of the microscopic anisotropy and tissue heterogeneity in intracranial tumor patients is feasible in clinically relevant times frames.Comment: Submitted to Magnetic Resonance in Medicin

    Relationships among intramammary health, udder and teat characteristics, and productivity of extensively managed ewes

    Mastitis is an economically important disease and its subclinical state is difficult to diagnose, which makes mitigation more challenging. The objectives of this study were to screen clinically healthy ewes in order to 1) identify cultivable microbial species in milk, 2) evaluate somatic cell count (SCC) thresholds associated with intramammary infection, and 3) estimate relationships between udder and teat morphometric traits, SCC, and ewe productivity. Milk was collected from two flocks in early (\u3c5 \u3ed) and peak (30 to 45 d) lactation to quantify SCC (n = 530) and numerate cultivable microbial species by culture-based isolation followed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS; n = 243) identification. Within flock and lactation stage, 11% to 74% (mean = 36%) of samples were culture positive. More than 50 unique identifications were classified by MALDI-TOF MS analysis, and Bacillus licheniformis (18% to 27%), Micrococcus flavus (25%), Bacillus amyloliquefaciens (7% to 18%), and Staphylococcus epidermidis (26%) were among the most common within flock and across lactation stage. Optimum SCC thresholds to identify culture-positive samples ranged from 175 × 103 to 1,675 × 103 cells/mL. Ewe productivity was assessed as total 120-d adjusted litter weight (LW120) and analyzed within flock with breed, parity, year, and the linear covariate of log10 SCC (LSCC) at early or peak lactation. Although dependent on lactation stage and year, each 1-unit increase in LSCC (e.g., an increase in SCC from 100 × 103 to 1,000 × 103 cells/mL) was predicted to decrease LW120 between 9.5 and 16.1 kg when significant. Udder and teat traits included udder circumference, teat length, teat placement, and degree of separation of the udder halves. Correlations between traits were generally low to moderate within and across lactation stage and most were not consistently predictive of ewe LSCC. Overall, the frequencies of bacteria-positive milk samples indicated that subclinical mastitis (SCM) is common in these flocks and can impact ewe productivity. Therefore, future research is warranted to investigate pathways and timing of microbial invasion, genomic regions associated with susceptibility, and husbandry to mitigate the impact of SCM in extensively managed ewes

    The Diabetes Remission Clinical Trial (DiRECT): protocol for a cluster randomised trial

    Background: Despite improving evidence-based practice following clinical guidelines to optimise drug therapy, Type 2 diabetes (T2DM) still exerts a devastating toll from vascular complications and premature death. Biochemical remission of T2DM has been demonstrated with weight loss around 15kg following bariatric surgery and in several small studies of non-surgical energy-restriction treatments. The non-surgical Counterweight-Plus programme, running in Primary Care where obesity and T2DM are routinely managed, produces &gt;15 kg weight loss in 33 % of all enrolled patients. The Diabetes UK-funded Counterpoint study suggested that this should be sufficient to reverse T2DM by removing ectopic fat in liver and pancreas, restoring first-phase insulin secretion. The Diabetes Remission Clinical Trial (DiRECT) was designed to determine whether a structured, intensive, weight management programme, delivered in a routine Primary Care setting, is a viable treatment for achieving durable normoglycaemia. Other aims are to understand the mechanistic basis of remission and to identify psychological predictors of response. Methods/Design: Cluster-randomised design with GP practice as the unit of randomisation: 280 participants from around 30 practices in Scotland and England will be allocated either to continue usual guideline-based care or to add the Counterweight-Plus weight management programme, which includes primary care nurse or dietitian delivery of 12-20weeks low calorie diet replacement, food reintroduction, and long-term weight loss maintenance. Main inclusion criteria: men and women aged 20-65years, all ethnicities, T2DM 0-6years duration, BMI 27-45 kg/m2. Tyneside participants will undergo Magnetic Resonance (MR) studies of pancreatic and hepatic fat, and metabolic studies to determine mechanisms underlying T2DM remission. Co-primary endpoints: weight reduction ≥ 15 kg and HbA1c &lt;48 mmol/mol at one year. Further follow-up at 2 years. Discussion: This study will establish whether a structured weight management programme, delivered in Primary Care by practice nurses or dietitians, is a viable treatment to achieve T2DM remission. Results, available from 2018 onwards, will inform future service strategy

    Reducing Impacts of Double-crested Cormorants to Natural Resources in Central New York: A Review of a Collaborative Research, Management, and Monitoring Program

    Double-crested Cormorants (Phalacrocorax auritus) have used central New York waters for breeding and stopover habitats during migration since 1984. In response to public concern over Oneida Lake, the United States Department of Agriculture Wildlife Services and the New York State Department of Environmental Conservation initiated an integrated research, management and monitoring program aimed at mitigating cormorant impacts to fisheries and other natural resources in 1998. The history of this program was reviewed and efforts to reduce negative impacts of the Double-crested Cormorant population in central New York described. Management was successful, as demonstrated by a substantial decrease in cormorant use of Oneida Lake during spring, summer and fall seasons, and the apparent recovery of certain sportfish populations. Research identified cormorant movement patterns within and among water bodies and documented cormorant responses to hazing and other management techniques. The cormorant management program in central New York was intended to keep cormorant use of Oneida Lake at a level that prevents unsustainable impacts to fisheries populations

    Time-Dependent Partition-Free Approach in Resonant Tunneling Systems

    An extended Keldysh formalism, well suited to properly take into account the initial correlations, is used in order to deal with the time-dependent current response of a resonant tunneling system. We use a \textit{partition-free} approach by Cini in which the whole system is in equilibrium before an external bias is switched on. No fictitious partitions are used. Besides the steady-state responses one can also calculate physical dynamical responses. In the noninteracting case we clarify under what circumstances a steady-state current develops and compare our result with the one obtained in the partitioned scheme. We prove a Theorem of asymptotic Equivalence between the two schemes for arbitrary time-dependent disturbances. We also show that the steady-state current is independent of the history of the external perturbation (Memory Loss Theorem). In the so called wide-band limit an analytic result for the time-dependent current is obtained. In the interacting case we propose an exact non-equilibrium Green function approach based on Time Dependent Density Functional Theory. The equations are no more difficult than an ordinary Mean Field treatment. We show how the scattering-state scheme by Lang follows from our formulation. An exact formula for the steady-state current of an arbitrary interacting resonant tunneling system is obtained.     BET inhibitors induce apoptosis through a MYC independent mechanism and synergise with CDK inhibitors to kill osteosarcoma cells

    Osteosarcoma (OS) survival rates have plateaued in part due to a lack of new therapeutic options. Here we demonstrate that bromodomain inhibitors (BETi), JQ1, I-BET151, I-BET762, exert potent anti-tumour activity against primary and established OS cell lines, mediated by inhibition of BRD4. Strikingly, unlike previous observations in long-term established human OS cell lines, the antiproliferative activity of JQ1 in primary OS cells was driven by the induction of apoptosis, not cell cycle arrest. In further contrast, JQ1 activity in OS was mediated independently of MYC downregulation. We identified that JQ1 suppresses the transcription factor FOSL1 by displacement of BRD4 from its locus. Loss of FOSL1 phenocopied the antiproliferative effects of JQ1, identifying FOSL1 suppression as a potential novel therapeutic approach for OS. As a monotherapy JQ1 demonstrated significant anti-tumour activity in vivo in an OS graft model. Further, combinatorial treatment approaches showed that JQ1 increased the sensitivity of OS cells to doxorubicin and induced potent synergistic activity when rationally combined with CDK inhibitors. The greater level of activity achieved with the combination of BETi with CDK inhibitors demonstrates the efficacy of this combination therapy. Taken together, our studies show that BET inhibitors are a promising new therapeutic for OS
