P-D Effects on the Reliability of Oil Offshore Jacket Platforms in Mexico

    Get PDF
Given the important economic consequences of an oil platform failure, all the aspects of its structural behavior and safety issues need to be carefully considered. In particular, P-D effects on the deck legs of marine offshore jacket platforms may be relevant when the deck height and the vertical load are significant. In this paper, the impact of the moment amplification, due to lenderness of the deck legs, on the platform safety is examined and appraised from he viewpoint of the structural reliability. The formulation is applied to a typical tall deck marine platform under the environmental loading at the Bay of Campeche, Mexico, and its reliability index is calculated with and without the P-D effect. The results presented herein may be sed to improve the current practice in the design and assessment of offshore marine platforms in Mexico and to update the current version of the code

Development of dependable controllers in the context of machines design

    Get PDF
Proceedings of ICMD 2013In the domain of machines' design, one of the most important issues to solve is related with the controller's design, mainly, guaranteeing that the machine will behave as expected. In order to achieve a dependable controller, some steps can be considered, such as the formalization of its specification - before being translated to the program that will be inserted in the controller device - and the respective analysis and verification. Nowadays, some formal analysis techniques, such as formal verification, are used to achieve this purpose. The dependability of a controller, however, is impacted by its execution context. This paper proposes an approach for the formal verification of the specification of mechatronic system's controllers, which considers, on the formal verification tasks, the behavior of the plant and the behavior of the Human Machine Interface of the Mechatronic system. Some conclusions are extrapolated for other systems of the same kind

    Constructing Self-Dual Strings

We present an ADHMN-like construction which generates self-dual string solutions to the effective M5-brane worldvolume theory from solutions to the Basu-Harvey equation. Our construction finds a natural interpretation in terms of gerbes, which we develop in some detail. We also comment on a possible extension to stacks of multiple M5-branes.Comment: 1+19 pages, presentation improved, minor corrections, published versio

The spectrum of mutations and molecular pathogenesis of hemophilia A in 181 Portuguese patients

    Get PDF
Disease-causing alterations within the F8 gene were identified in 177 hemophilia A families of Portuguese origin. The spectrum of non-inversion F8 mutations in 101 families included 67 different alterations, namely: 36 missense, 8 nonsense and 4 splice site mutations, as well as 19 insertions/deletions. Thirty-four of these mutations are novel. Molecular modeling allowed prediction of the conformational changes introduced by selected amino acid substitutions and their correlation with the patients' phenotypes. The relatively frequent, population-specific, missense mutations together with de novo alterations can lead to significant differences in the spectrum of F8 mutations among different populationsThis study was partially supported by Fundação para a Ciência e a Tecnologia: research grant PBIC/C/SAU/1588/92 and Programa de Financiamento Plurianual do CIGM

Evolutionary trajectories in rugged fitness landscapes

We consider the evolutionary trajectories traced out by an infinite population undergoing mutation-selection dynamics in static, uncorrelated random fitness landscapes. Starting from the population that consists of a single genotype, the most populated genotype \textit{jumps} from a local fitness maximum to another and eventually reaches the global maximum. We use a strong selection limit, which reduces the dynamics beyond the first time step to the competition between independent mutant subpopulations, to study the dynamics of this model and of a simpler one-dimensional model which ignores the geometry of the sequence space. We find that the fit genotypes that appear along a trajectory are a subset of suitably defined fitness \textit{records}, and exploit several results from the record theory for non-identically distributed random variables. The genotypes that contribute to the trajectory are those records that are not \textit{bypassed} by superior records arising further away from the initial population. Several conjectures concerning the statistics of bypassing are extracted from numerical simulations. In particular, for the one-dimensional model, we propose a simple relation between the bypassing probability and the dynamic exponent which describes the scaling of the typical evolution time with genome size. The latter can be determined exactly in terms of the extremal properties of the fitness distribution.Comment: Figures in color; minor revisions in tex

NOA36 Protein Contains a Highly Conserved Nucleolar Localization Signal Capable of Directing Functional Proteins to the Nucleolus, in Mammalian Cells

NOA36/ZNF330 is an evolutionarily well-preserved protein present in the nucleolus and mitochondria of mammalian cells. We have previously reported that the pro-apoptotic activity of this protein is mediated by a characteristic cysteine-rich domain. We now demonstrate that the nucleolar localization of NOA36 is due to a highly-conserved nucleolar localization signal (NoLS) present in residues 1–33. This NoLS is a sequence containing three clusters of two or three basic amino acids. We fused the amino terminal of NOA36 to eGFP in order to characterize this putative NoLS. We show that a cluster of three lysine residues at positions 3 to 5 within this sequence is critical for the nucleolar localization. We also demonstrate that the sequence as found in human is capable of directing eGFP to the nucleolus in several mammal, fish and insect cells. Moreover, this NoLS is capable of specifically directing the cytosolic yeast enzyme polyphosphatase to the target of the nucleolus of HeLa cells, wherein its enzymatic activity was detected. This NoLS could therefore serve as a very useful tool as a nucleolar marker and for directing particular proteins to the nucleolus in distant animal species

Geometric Phases and Mielnik's Evolution Loops

The cyclic evolutions and associated geometric phases induced by time-independent Hamiltonians are studied for the case when the evolution operator becomes the identity (those processes are called {\it evolution loops}). We make a detailed treatment of systems having equally-spaced energy levels. Special emphasis is made on the potentials which have the same spectrum as the harmonic oscillator potential (the generalized oscillator potentials) and on their recently found coherent states.Comment: 11 pages, harvmac, 2 figures available upon request; CINVESTAV-FIS GFMR 11/9
