    Searching for Stability as we Age: The PCA-Biplot Approach

    Principal component analysis (PCA) has been successfully applied to gait data; however, interpretation of the components is challenging. An alternative is to use a graphical display called biplot that gives insights into relationships and trends of data sets. Our goal was to demonstrate the sensitivity of gait variables to aging in elderly women with PCA-biplot. One hundred fifty-one elderly females (71.6±5.0 yrs), 152 adults (44.7±5.4 yrs) and 150 young (21.7±4.1 yrs) participated in the study. Gait spatial and temporal parameters were collected using a computerized carpet. PCA-biplot, discriminant analysis and MANOVA were used in the analysis. PCA-biplot revealed that elderly females walked with lower velocity, shorter step length, reduced swing time, higher cadence, and increased double support time compared to the other two groups. The greatest distances between the groups were along the variable step length with the elderly group showing a decrease of 8.4 cm in relation to the younger group. The discriminant function confirmed the importance of principal component 2 for group separation. Because principal component 2 was heavily weighted by step length and swing time, it represents a measure of stability. As women age they seek a more stable gait by decreasing step length, swing time, and velocity. PCA-biplot highlighted the importance of the variable step length in distinguishing between women of different age groups. It is well-known that as we age we seek a more stable gait. The PCA-biplot emphasized that premise and gave further important insights into relationships and trends of this complex data set

    Structural study of Cu2x_{2-x}Se alloys produced by mechanical alloying

    The crystalline structures of superionic high temperature copper selenides Cu2x_{2-x}Se (0x0.250 \le x \le 0.25) produced by Mechanical Alloying were investigated using X-ray diffraction (XRD) technique. The measured XRD patterns showed the presence of the peaks corresponding to the crystalline superionic high temperature α\alpha-Cu2_2Se phase in the as-milled sample, and its structural data were determined by means of a Rietveld refinement procedure. After a heat treatment in argon at 200^\circC for 90 h, this phase transforms to the superionic high temperature α\alpha-Cu1.8_{1.8}Se phase, whose structural data where also determined through the Rietveld refinement. In this phase, a very low occupation of the trigonal 32(f) sites (3\sim 3%) by Cu ions is found. In order to explain the evolution of the phases in the samples, two possible mechanisms are suggested: the high mobility of Cu ions in superionic phases and the intense diffusive processes in the interfacial component of samples produced by Mechanical Alloying.Comment: 2 figures, submitted to Acta Crystallographic

    Structure and Mechanism of Dimer-Monomer Transition of a Plant Poly(A)-Binding Protein upon RNA Interaction: Insights into Its Poly(A) Tail Assembly

    Poly(A)-binding proteins (PABPs) play crucial roles in mRNA biogenesis, stability, transport and translational control in most eukaryotic cells. Although animal PABPs are well-studied proteins, the biological role, three-dimensional structure and RNA-binding mode of plant PABPs remain largely uncharacterized. Here, we report the structural features and RNA-binding mode of a Citrus sinensis PABP (CsPABPN1). CsPABPN1 has a domain architecture of nuclear PABPs (PABPNs) with a single RNA recognition motif (RRM) flanked by an acidic N-terminus and a GRPF-rich C-terminus. The RRM domain of CsPABPN1 displays virtually the same three-dimensional structure and poly(A)-binding mode of animal PABPNs. However, while the CsPABPN1 RRM domain specifically binds poly(A), the full-length protein also binds poly(U). CsPABPN1 localizes to the nucleus of plant cells and undergoes a dimer–monomer transition upon poly(A) interaction. We show that poly(A) binding by CsPABPN1 begins with the recognition of the RNA-binding sites RNP1 and RNP2, followed by interactions with residues of the β2 strands, which stabilize the dimer, thus leading to dimer dissociation. Like human PABPN1, CsPABPN1 also seems to form filaments in the presence of poly(A). Based on these data, we propose a structural model in which contiguous CsPABPN1 RRM monomers wrap around the RNA molecule creating a superhelical structure that could not only shield the poly(A) tail but also serve as a scaffold for the assembly of additional mRNA processing factors

    Leguminosas quando consorciadas com o café e usadas como adubação verde podem contribuir fornecendo nitrogênio e proteção ao solo pela adição de matéria orgânica. O trabalho foi conduzido com o objetivo de avaliar o efeito do uso de leguminosas no sistema de produção de café, no segmento de agricultura familiar, visando promover a implantação e a manutenção dos cafezais de forma técnica e economicamente sustentável. O experimento foi conduzido no período de novembro de 2000 a abril de 2003, em delineamento experimental de blocos ao acaso em esquema de parcelas subdivididas, com quatro repetições. Os tratamentos utilizados nas parcelas foram as leguminosas (Stizolobium atterrimum, Cajanus cajan, Canavalia endiformis e Flemingia congesta) e a testemunha (sem leguminosa) e, nas subparcelas, duas doses de N (0 e 22 g de N por cova). A Flemingia congesta e a Mucuna aterrima foram as leguminosas que mais influenciaram positivamente a produtividade dos cafeeiros, independente da adubação nitrogenada. Em relação à testemunha, o aumento em produção foi de 109% quando utilizou-se a Flemingia congesta e 52% com a Mucuna aterrima. A Flemingia congesta foi também a leguminosa que melhor controlou as invasoras, dado o volume de fitomassa produzida e a possibilidade de 2 cortes durante um período de doze meses, evidenciando o potencial desta leguminosa na formação de novos cafezais no Acre. Por outro lado, a Canavalia ensiformis, leguminosa que é normalmente utilizada nas entrelinhas dos cafeeiros pelos cafeicultores, do Acre e de outras regiões produtoras, neste trabalho influenciou negativamente a altura das plantas, diâmetro da copa e crescimento dos cafeeiros

Based on the fact that organic fertilization can improve soil quality, we compared the effects of conventional and organic soil management on microbial activity and mycorrhization of seedless grape crops. We measured glomerospores number, most probable number (MPN) of propagules, richness of arbuscular mycorrhizal fungi (AMF) species, AMF root colonization, EE-BRSP production, carbon microbial biomass (C-MB), microbial respiration, fluorescein diacetate hydrolytic activity (FDA) and metabolic coefficient (qCO2). The organic management led to an increase in all variables with the exception of EE-BRSP and qCO2. Mycorrhizal colonization increased from 4.7% in conventional crops to 15.9% in organic crops. Spore number ranged from 4.1 to 12.4 per 50 g-1 soil in both management systems. The most probable number of AMF propagules increased from 79 cm-3 soil in the conventional system to 110 cm-3 soil in the organic system. Microbial carbon, CO2 emission, and FDA activity were increased by 100 to 200% in the organic crop. Thirteen species of AMF were identified, the majority in the organic cultivation system. Acaulospora excavata, Entrophospora infrequens, Glomus sp.3 and Scutellospora sp. were found only in the organically managed crop. S. gregaria was found only in the conventional crop.     Engineering Branch-and-Cut Algorithms for the Equicut Problem

    A minimum equicut of an edge-weighted graph is a partition of the nodes of the graph into two sets of equal size such hat the sum of the weights of edges joining nodes in different partitions is minimum. We compare basic linear and semidefnite relaxations for the equicut problem, and and that linear bounds are competitive with the corresponding semidefnite ones but can be computed much faster. Motivated by an application of equicut in theoretical physics, we revisit an approach by Brunetta et al. and present an enhanced branch-and-cut algorithm. Our computational results suggest that the proposed branch-andcut algorithm has a better performance than the algorithm of Brunetta et al.. Further, it is able to solve to optimality in reasonable time several instances with more than 200 nodes from the physics application

    Pre-M Phase-promoting Factor Associates with Annulate Lamellae in Xenopus Oocytes and Egg Extracts

    We have used complementary biochemical and in vivo approaches to study the compartmentalization of M phase-promoting factor (MPF) in prophase Xenopus eggs and oocytes. We first examined the distribution of MPF (Cdc2/CyclinB2) and membranous organelles in high-speed extracts of Xenopus eggs made during mitotic prophase. These extracts were found to lack mitochondria, Golgi membranes, and most endoplasmic reticulum (ER) but to contain the bulk of the pre-MPF pool. This pre-MPF could be pelleted by further centrifugation along with components necessary to activate it. On activation, Cdc2/CyclinB2 moved into the soluble fraction. Electron microscopy and Western blot analysis showed that the pre-MPF pellet contained a specific ER subdomain comprising "annulate lamellae" (AL): stacked ER membranes highly enriched in nuclear pores. Colocalization of pre-MPF with AL was demonstrated by anti-CyclinB2 immunofluorescence in prophase oocytes, in which AL are positioned close to the vegetal surface. Green fluorescent protein-CyclinB2 expressed in oocytes also localized at AL. These data suggest that inactive MPF associates with nuclear envelope components just before activation. This association may explain why nuclei and centrosomes stimulate MPF activation and provide a mechanism for targeting of MPF to some of its key substrates
