    Preliminary Evidence of Increased Hippocampal Myelin Content in Veterans with Posttraumatic Stress Disorder.

    Recent findings suggest the formation of myelin in the central nervous system by oligodendrocytes is a continuous process that can be modified with experience. For example, a recent study showed that immobilization stress increased oligodendrogensis in the dentate gyrus of adult rat hippocampus. Because changes in myelination represents an adaptive form of brain plasticity that has a greater reach in the adult brain than other forms of plasticity (e.g., neurogenesis), the objective of this "proof of concept" study was to examine whether there are differences in myelination in the hippocampi of humans with and without post-traumatic stress disorder (PTSD). We used the ratio of T1-weighted/T2-weighted magnetic resonance image (MRI) intensity to estimate the degree of hippocampal myelination in 19 male veterans with PTSD and 19 matched trauma-exposed male veterans without PTSD (mean age: 43 ± 12 years). We found that veterans with PTSD had significantly more hippocampal myelin than trauma-exposed controls. There was also found a positive correlation between estimates of hippocampal myelination and PTSD and depressive symptom severity. To our knowledge, this is the first study to examine hippocampal myelination in humans with PTSD. These results provide preliminary evidence for stress-induced hippocampal myelin formation as a potential mechanism underlying the brain abnormalities associated with vulnerability to stress

    The Role of RFamide-Related Peptide-3 in Age-Related Reproductive Decline in Female Rats.

    Reproductive senescence, the point in time when females cease to show estrous cyclicity, is associated with endocrine changes in the hypothalamus, pituitary, and gonads. However, the mechanisms triggering this transition are not well understood. To gain a better understanding of the top-down control of the transition from reproductive competence to a state of reproductive senescence, we investigated middle-aged female rats exhibiting varying degrees of reproductive decline, including individuals with normal cycles, irregular cycles, and complete cessation of cycles. We identified hormonal changes in the brain that manifest before ovarian cycles exhibit any deterioration. We found that females exhibit an increase in RFamide-related peptide-3 (RFRP3) mRNA expression in the hypothalamus in middle age prior to changes in estrous cycle length. This increase is transient and followed by subsequent decreases in kisspeptin (KiSS1) and gonadotropin-releasing hormone (GnRH) mRNA expression. Expression of RFRP3 and its receptor also increased locally in the ovaries with advancing age. While it is well known that aging is associated with decreased GnRH release and downstream disruption of the hypothalamic-pituitary-gonadal (HPG) axis, herein, we provide evidence that reproductive senescence is likely triggered by alterations in a network of regulatory neuropeptides upstream of the GnRH system

    Mass and orbit constraints of the gamma-ray binary LS 5039

    We present the results of space-based photometric and ground-based spectroscopic observing campaigns on the gamma-ray binary LS 5039. The new orbital and physical parameters of the system are similar to former results, except we found a lower eccentricity. Our MOST-data show that any broad-band optical photometric variability at the orbital period is below the 2 mmag level.     The variable stellar wind of Rigel probed at high spatial and spectral resolution

    We present a spatially resolved, high-spectral resolution (R=12000) K-band temporal monitoring of Rigel using AMBER at the VLTI. Rigel was observed in the Bracket Gamma line and its nearby continuum in 2006-2007, and 2009-2010. These unprecedented observations were complemented by contemporaneous optical high-resolution spectroscopy. We analyse the near-IR spectra and visibilities with the 1D non-LTE radiative-transfer code CMFGEN. The differential and closure phase signal exhibit asymmetries that are interpreted as perturbations of the wind. A systematic visibility decrease is observed across the Bracket Gamma. During the 2006-2007 period the Bracket Gamma and likely the continuum forming regions were larger than in the 2009-2010 epoch. Using CMFGEN, we infer a mass-loss rate change of about 20% between the two epochs. We further find time variations in the differential visibilities and phases. The 2006-2007 period is characterized by noticeable variations of the differential visibilities in Doppler position and width and by weak variations in differential and closure phase. The 2009-2010 period is much more quiet with virtually no detectable variations in the dispersed visibilities but a strong S-shape signal is observed in differential phase coinciding with a strong ejection event discernible in the optical spectra. The differential phase signal that is sometimes detected is reminiscent of the signal computed from hydrodynamical models of corotating interaction regions. For some epochs the temporal evolution of the signal suggests the rotation of the circumstellar structures.Comment: Paper accepted in the A&A journa

    Tomographic readout of an opto-mechanical interferometer

    The quantum state of light changes its nature when being reflected off a mechanical oscillator due to the latter's susceptibility to radiation pressure. As a result, a coherent state can transform into a squeezed state and can get entangled with the motion of the oscillator. The complete tomographic reconstruction of the state of light requires the ability to readout arbitrary quadratures. Here we demonstrate such a readout by applying a balanced homodyne detector to an interferometric position measurement of a thermally excited high-Q silicon nitride membrane in a Michelson-Sagnac interferometer. A readout noise of \unit{1.9 \cdot 10^{-16}}{\metre/\sqrt{\hertz}} around the membrane's fundamental oscillation mode at \unit{133}{\kilo\hertz} has been achieved, going below the peak value of the standard quantum limit by a factor of 8.2 (9 dB). The readout noise was entirely dominated by shot noise in a rather broad frequency range around the mechanical resonance.Comment: 7 pages, 5 figure

    First Stellar Abundances in the Dwarf Irregular Galaxy Sextans A

    We present the abundance analyses of three isolated A-type supergiant stars in the dwarf irregular galaxy Sextans A from high-resolution spectra the UVES spectrograph at the VLT. Detailed model atmosphere analyses have been used to determine the stellar atmospheric parameters and the elemental abundances of the stars. The mean iron group abundance was determined from these three stars to be [(FeII,CrII)/H]=-0.99+/-0.04+/-0.06. This is the first determination of the present-day iron group abundances in Sextans A. These three stars now represent the most metal-poor massive stars for which detailed abundance analyses have been carried out. The mean stellar alpha element abundance was determined from the alpha element magnesium as [alpha(MgI)/H]=-1.09+/-0.02+/-0.19. This is in excellent agreement with the nebular alpha element abundances as determined from oxygen in the H II regions. These results are consistent from star-to-star with no significant spatial variations over a length of 0.8 kpc in Sextans A. This supports the nebular abundance studies of dwarf irregular galaxies, where homogeneous oxygen abundances are found throughout, and argues against in situ enrichment. The alpha/Fe abundance ratio is [alpha(MgI)/FeII,CrII]=-0.11+/-0.02+/-0.10, which is consistent with the solar ratio. This is consistent with the results from A-supergiant analyses in other Local Group dwarf irregular galaxies but in stark contrast with the high [alpha/Fe] results from metal-poor stars in the Galaxy, and is most clearly seen from these three stars in Sextans A because of their lower metallicities. The low [alpha/Fe] ratios are consistent with the slow chemical evolution expected for dwarf galaxies from analyses of their stellar populations.Comment: 40 pages, 8 figures, accepted for publication in A

    Virological and Parasitological Characterization of Mini-LEWE Minipigs Using Improved Screening Methods and an Overview of Data on Various Minipig Breeds

    Minipigs play an important role in biomedical research and have also been used as donor animals in xenotransplantation. To serve as a donor in xenotransplantation, the animals must be free of potential zoonotic viruses, bacteria and parasites. Porcine endogenous retroviruses (PERVs) are integrated in the genome of all pigs and cannot be eliminated as most of the other pig viruses can. PERV-A and PERV-B infect human cells in cell culture and are integrated in all pigs, whereas PERV-C infects only pig cells and it is found in many, but not all pigs. Minipigs are known for a high prevalence of recombinant PERV-A/C viruses able to infect human cells (Denner and Schuurman, Viruses, 2021;13:1869). Here, Mini-LEWE minipigs are screened for the first time for pig viruses including PERV. Peripheral blood mononuclear cells (PBMCs) from 10 animals were screened using PCR-based methods (PCR, RT-PCR, and real-time PCR). In comparison with our previous screening assays, numerous improvements were introduced, e.g., the usage of gene blocks as a PCR standard and foreign RNA to control reverse transcription in RT-PCR. Using these improved detection methods, Mini-LEWE pigs were found to be negative for porcine cytomegalovirus (PCMV), porcine lymphotropic herpesviruses (PLHV-1, -2 and -3), porcine circoviruses (PCV1, 2, 3 and 4), porcine parvovirus (PPV) and hepatitis E virus (HEV). All animals carried PERV-A, PERV-B and PERV-C in their genome. PERV-A/C was not found. In contrast to all other minipig breeds (Göttingen minipigs, Aachen minipigs, Yucatan micropig, Massachusetts General Hospital miniature pigs), Mini-LEWE minipigs have less viruses and no PERV-A/C. Parasitological screening showed that none of the Mini-LEWE minipigs harbored ecto- and gastrointestinal parasites, but at least one animal tested positive for anti-Toxoplasma gondii antibodies

    Long-term spectroscopic monitoring of the Luminous Blue Variable HD160529

    We have spectroscopically monitored the galactic Luminous Blue Variable HD 160529 and obtained an extensive high-resolution data set that covers the years 1991 to 2002. During this period, the star evolved from an extended photometric minimum phase towards a new visual maximum. In several observing seasons, we covered up to four months with almost daily spectra. Our spectra typically cover most of the visual spectral range with a high spectral resolution (about 20,000 or more). This allows us to investigate the variability in many lines and on many time scales from days to years. We find a correlation between the photospheric HeI lines and the brightness of the star, both on a time scale of months and on a time scale of years. The short-term variations are smaller and do not follow the long-term trend, strongly suggesting different physical mechanisms. Metal lines also show both short-term and long-term variations in strength and also a long-term trend in radial velocity. Most of the line-profile variations can be attributed to changing strengths of lines. Propagating features in the line profiles are rarely observed. We find that the mass-loss rate of HD 160529 is almost independent of temperature, i.e. visual brightness.Comment: 14 pages, 15 figures, accepted for publication in Astronomy & Astrophysic

    On the photometric variability of blue supergiants in NGC 300 and its impact on the Flux-weighted Gravity-Luminosity Relationship

    We present a study of the photometric variability of spectroscopically confirmed supergiants in NGC 300, comprising 28 epochs extending over a period of five months. We find 15 clearly photometrically variable blue supergiants in a sample of nearly 70 such stars, showing maximum light amplitudes ranging from 0.08 to 0.23 magnitudes in the V band, and one variable red supergiant. We show their light curves, and determine semi-periods for two A2 Ia stars. Assuming that the observed changes correspond to similar variations in the bolometric luminosity, we test for the influence of this variability on the Flux-weighted Gravity--Luminosity Relationship and find a negligible effect, showing that the calibration of this relationship, which has the potential to measure extragalactic distances at the Cepheid accuracy level, is not affected by the stellar photometric variability in any significant way.Comment: 9 pages, 3 figures, 3 tables.     Congenital toxoplasmosis: Serology, PCR, parasite isolation and molecular characterization of toxoplasma gondii

    Introducción El diagnóstico de toxoplasmosis congénita (TC) en el recién nacido es muy importante porque debe recibir tratamiento siempre, sintomático o no, para evitar o aminorar las secuelas de la enfermedad. Objetivo Evaluación comparativa de los métodos disponibles en la institución para el diagnóstico de TC. Materiales y Métodos Se evaluaron métodos diagnósticos en 67 niños cuyas madres cursaron toxoplasmosis aguda durante el embarazo. Se utilizó la técnica de Sabin Feldman para IgG al nacimiento y durante el seguimiento serológico hasta el año de vida. Para determinar IgM, IgA e IgE se utilizó la técnica immunosorbent agglutination assay (ISAGA). El diagnóstico directo se realizó por reacción de polimerasa en cadena (RPC), aislamiento y caracterización molecular del parásito. Resultados La sensibilidad (S) de ISAGA IgM fue 87%, ISAGA IgA 91% y la especificidad (E) fue 100% para ambas; cuando se realizaron en conjunto, la S aumentó a 98%. La detección de IgE contribuyó al diagnóstico cuando se la detectó sólo en la sangre del neonato y no en sangre materna. Se aisló el parásito en cuatro casos de TC, uno fue genotipo II y los otros tres, genotipos “atípicos”. La S del aislamiento fue 80% y la E 100%. Conclusión Los métodos serológicos utilizados mostraron una buena eficacia diagnóstica. Un caso fue detectado sólo por el aislamiento y la caracterización molecular tiene gran valor epidemiológico.Background. Congenital toxoplasmosis diagnosis in the newborn is a very important issue due to the need for early treatment to prevent future sequels. Aim To compare available methods at the institution for the diagnosis of congenital toxoplasmosis. Material and Methods In this study we have evaluated the different diagnostic tests used in 67 congenital exposed newborns, including serological tests, PCR, parasite isolation and molecular characterization. Results The ISAGA IgM and IgA tests showed sensitivity (Se) of 87 and 91%, respectively, and specificity (Sp) of 100%. When ISAGA IgM and IgA were performed simultaneously, the Se increased to 98% and the Sp was 100%. The presence of IgE contributed to the diagnosis when it was detected in the child's serum but not in maternal blood. In four congenital infected children the parasite was isolated and genotyped: one was genotype II and the other three were “atypical” genotypes. No parasite was isolated in children without congenital toxoplasmosis. Discussion Overall, serological tests showed a good diagnostic performance although in one case they were all negative and isolation was the only tool to identify the infection. We conclude that it is essential to use all diagnostic tests in every single exposed child, including if possible, molecular characterization due to its epidemiological implication.Fil: Carral, Liliana.
