    Management discriminant properties in semiarid soils

    The physical properties of coarse - textured soils in semiarid regions often deteriorate with use. We hypostatized that the changes in the physical properties of the soil were related to the cropping sistem employed. 5urface sampled of 52 Entic Haplustolls under three diferents uses (24 under continuous cultivation), 18 under rotation wuth grass Ieys (R), and 10 virgin soils M were analyzed for c1ay, silt, organic malter anrd water content, bulk density, compaction and aggregate stability. Data were analyzed statistically using principal components, canonical variables, and discriminant functions. A satisfactory segregation of the soils according to discriminant properties (coarse organic matter, aggregate stability, and suceptibility lo compaction) was obtained. The model developed satisfactory classified the soils under different uses (100% R, 83% e, and 88% V). Principal component analysis also showed that bulck density, compaction, and wet aggregate salability are related lo organic matter content.     Influencias del material original y la acción antrópica sobre características morfológicas, físicas y contenidos de materia orgánica de suelos cultivados y vírgenes de la región semiárida pampeana central (Argentina)

    En este estudio se evaluó la incidencia de las características heredadas del material original y aquellas condicionadas por el laboreo sobre propiedades morfológicas, físicas (granulometría, densidad aparente y poros) y contenidos de materia orgánica de horizontes A de suelos cultivados y vírgenes de la región semiárida pampeana central argentina. Este estudio se llevó a cabo sobre 12 perfiles de zonas planas, media loma alta y baja de dos toposecuencias cultivadas y dos vírgenes. Los resultados muestran que las diferencias textura les entre perfiles determinan la proporción de poros chicos y medianos < 10 µ) y posiblemente de materia orgánica, mientras que el laboreo condiciona la densidad aparente y los poros grandes (10-1000 u) de los diferentes horizontes A. Los espesores del solum hasta la acumulación secundaria de CaCo y la acumulación residual de arena son, las características morfológicas que más fehacientemente indicaron la degradación mientras que los espesores de los horizontes superiores y las características morfológicas dependientes de los contenidos de materia orgánica principalmente color y estructura) no son indicadores confiables.Estudio financiado por SECYT dentro del Plan de Expansión de Fronteras Agropecuarias.     FcpB Is a Surface Filament Protein of the Endoflagellum Required for the Motility of the Spirochete Leptospira

    International audienceThe spirochete endoflagellum is a unique motility apparatus among bacteria. Despite its critical importance for pathogenesis, the full composition of the flagellum remains to be determined. We have recently reported that FcpA is a novel flagellar protein and a major component of the sheath of the filament of the spirochete Leptospira. By screening a library of random transposon mutants in the spirochete Leptospira biflexa, we found a motility-deficient mutant harboring a disruption in a hypothetical gene of unknown function. Here, we show that this gene encodes a surface component of the endoflagellar filament and is required for typical hook- and spiral-shaped ends of the cell body, coiled structure of the endoflagella, and high velocity phenotype. We therefore named the gene fcpB for flagellar-coiling protein B. fcpB is conserved in all members of the Leptospira genus, but not present in other organisms including other spirochetes. Complementation of the fcpB− mutant restored the wild-type morphology and motility phenotypes. Immunoblotting with anti-FcpA and anti-FcpB antisera and cryo-electron microscopy of the filament indicated that FcpB assembled onto the surface of the sheath of the filament and mostly located on the outer (convex) side of the coiled filament. We provide evidence that FcpB, together with FcpA, are Leptospira-specific novel components of the sheath of the filament, key determinants of the coiled and asymmetric structure of the endoflagella and are essential for high velocity. Defining the components of the endoflagella and their functions in these atypical bacteria should greatly enhance our understanding of the mechanisms by which these bacteria produce motility

    Desmosterol Incorporation Into Ram Sperm Membrane Before Cryopreservation Improves in vitro and in vivo Fertility

    Pregnancy rates in ewes are markedly low after cervical insemination with frozen-thawed sperm. Sensitivity of ram sperm to freeze-thawing is related to the lipid composition of the membrane, particularly to its low sterol content. Recently, we proved that sterol content of ram sperm can be increased by treatment with methyl-β-cyclodextrin-sterol complexes and we provided mechanistic based evidence on the differential behavior of cholesterol and desmosterol in the ram sperm membrane. In the present study, we evaluated the role of increasing cholesterol and desmosterol content of ram sperm before cryopreservation, on the extent and distribution of sterols, cryocapacitation status, acrosome integrity, DNA damage associated with apoptosis and fertility competence in vitro and in vivo of post-thawed sperm. After freeze-thawing, similar levels of sterol content were evidenced in control sperm cells and in those pre-incubated with either cholesterol or desmosterol. Still, moderately higher levels of sterols were registered in treated sperm compared to the control, indicating no physiological excess of sterols after thawing or sterol losses that exceed the control. Live cell imaging of fluorescent cholesterol evidenced the presence of sperm sub-populations differentially affected by freeze-thawing. Similar unimodal frequency profiles were observed between sterol-enriched groups, while the control exhibited a sub-population of sperm compatible with low sterol content. Tyrosine phosphorylation was significantly lower when ram sperm incorporated cholesterol compared to the control. No difference in this capacitation parameter was found between the latter and desmosterol-enriched sperm. The percentage of sperm with damaged acrosomes post-thawing, assessed by a fluorescent lectin, was reduced in sperm that incorporated sterols before freezing, irrespective of the sterol class. These results suggest that sterols exert a stabilizing effect on the acrosome. No differences were found in levels of apoptotic DNA fragmentation among experimental groups. As to fertility trials, desmosterol-enriched sperm gave rise to higher rates of in vitro activated oocytes by heterologous fertilization and to significantly lower pregnancy loss in vivo. Our research provides new insights on sterol incorporation into ram sperm prior to cryopreservation, in particular on the additional benefit of incorporating desmosterol as a strategy to improve fertility outcome.Fil: Carro, Maria de Las Mercedes.     The Effect of Regional Analgesia on Vascular Tone in Hip Arthroplasty Patients

    Background: While it is assumed that neuraxial analgesia and pain management may beneficially influence perioperative hemodynamics, few studies provided data quantifying such effects and none have assessed the potential contribution of the addition of a nerve block. Questions/purposes: This clinical trial compared the visual analog scale (VAS) scores and measurement of arterial tone using augmentation index of patients who received combined spinal-epidural (CSE) only to patients who received both CSE and lumbar plexus block. Methods: After obtaining written consent, 92 patients undergoing total hip arthroplasty were randomized to receive either CSE or CSE with lumbar plexus block (LPB). Perioperative pain and arterial tone were measured using VAS scores and augmentation index (AI) respectively, at baseline and at various times postoperatively. Results: After the exclusion of 2 patients, 44 patients received CSE alone and 46 patients received CSE and LPB. Patient demographics and perioperative characteristics were similar in both groups. AI continuously decreased after placement of a CSE with or without LBP, beyond full resolution of neuraxial and peripheral blockade. Although the LPB group demonstrated a statistically significant reduction of VAS pain scores in the postanesthesia care unit (PACU; P \u3c 0.05), overall, the addition of a LPB did not significantly reduce the AI when compared to the control group. Conclusion: The addition of a LPB provided better pain control in the PACU but did not reduce the AI, compared to the control group. We conclude that the addition of a LPB may have limited ability to affect arterial tone in the presence of a continuous infusion of epidural analgesics. In summary, the addition of a LPB in patients undergoing total hip arthroplasty is clinically effective and provided better pain control, especially in the immediate postoperative period. The continuous decrease on the AI in both groups beyond the full resolution of the neuroaxial and LPB will require further studies

    Comment on ‘‘Coxibs Refocus Attention on the Cardiovascular Risks of Non-Aspirin NSAIDs’’

    In a recent issue of the American Journal of Cardiovascular Drugs, Thomas et al. analyzed the cardiovascular safety of cyclo-oxygenase (COX)-2 inhibitors (coxibs). In the Recommendations section of the paper, the authors stated that, based on the results of the PRECISION (Prospective Randomized Evaluation of Celecoxib Integrated Safety versus Ibuprofen Or Naproxen) trial, celecoxib at moderate doses is not inferior to ibuprofen or naproxen in terms of cardiovascular safety. The PRECISION investigators seem to imply that celecoxib is a ‘cardiovascular safe’ drug, and we would like to challenge this statement with a more detailed analysis of the PRECISION trial.Facultad de Ciencias Médica

    Bayesian estimation of nonsynonymous/synonymous rate ratios for pairwise sequence comparisons.

    The nonsynonymous/synonymous rate ratio (ω = d(N)/d(S)) is an important measure of the mode and strength of natural selection acting on nonsynonymous mutations in protein-coding genes. The simplest such analysis is the estimation of the d(N)/d(S) ratio using two sequences. Both heuristic counting methods and the maximum-likelihood (ML) method based on a codon substitution model are widely used for such analysis. However, these methods do not have nice statistical properties, as the estimates can be zero or infinity in some data sets, so that their means and variances are infinite. In large genome-scale comparisons, such extreme estimates (either 0 or ∞) of ω and sequence distance (t) are common. Here, we implement a Bayesian method to estimate ω and t in pairwise sequence comparisons. Using a combination of computer simulation and real data analysis, we show that the Bayesian estimates have better statistical properties than the ML estimates, because the prior on ω and t shrinks the posterior of those parameters away from extreme values. We also calculate the posterior probability for ω > 1 as a Bayesian alternative to the likelihood ratio test. The new method is computationally efficient and may be useful for genome-scale comparisons of protein-coding gene sequences
