Enzyme treatment to decrease solids and improve digestion of primary sewage sludge
The aim of anaerobic digestion of primary sewage sludge is to convert the carbonaceous material contained in the solids into methane and carbon dioxide. The products of digestion are therefore gases, stabilised sludge solids which are subsequently dewatered and disposed of, and sludge liquor which is generally further treated. This investigation assessed the impact of addition of hydrolytic enzymes to anaerobic digesters. Cellulase and pronase E were added singly and in combination, and it was foundthat the mixture of the two enzymes resulted in an 80% reduction in solids (cf. 20% in the control), 93% removal of particulate chemical oxygen demand (COD) (59% in the control) and 97% total COD removal(vs. 63%). The total suspended solids (TSS) concentration was reduced by 80%, from 25 g/l to 5 g/l. Single enzymes had little or no impact on sludge solubilisation, and final COD and TSS, but all of the enzyme additions were seen to decrease the production of volatile fatty acids (VFAs). Since accumulation of VFAs can lead to digester failure, it was concluded that the enzyme additives enhanced digester performance in terms of degradation of COD, reduction in sludge solids remaining afterdigestion and improved digester stability owing to the stable prevailing pH. The results indicate that enzyme addition at full scale could be expected to lead to greater methane yields, lower strength sludge liquors and a significant reduction in the requirements for and costs of digested sludge dewatering and disposa
The effects of increased freshwater inflow on metal enrichment in selected Eastern Cape estuaries, South Africa
The concentrations of select metals (Cd, Co, Cu, Fe, Pb, Ni and Zn) within the water column and sediment of the permanently open Kariega Estuary and temporary open/closed Riet and East Kleinemonde Estuaries were investigated during a dry and a wet season. Enrichment factors (EFs), using Fe as a reference element, and baseline linear regression models for metals vs Fe were used to assess the extent of metal enrichment in the sediments. The results of the study indicate that Cd, Co Ni and Pb were enriched above baseline concentrations (1.0 < EF < 4.1) in the sediments of all three estuaries. Co, Pb and Ni enrichment in the Kariega Estuary sediments was significantly higher during the dry season, and the mean concentrations of Pb and Cd in the water column were 19-fold and 66-fold higher in the dry season. The elevated concentration of metals during the dry season could be related to accumulation of diffuse pollution from human activities within the catchment area. Conversely, inflow of freshwater into the estuary had the net effect of reducing the concentration and enrichment of these metals within the Kariega Estuary due to scouring and outflow of estuarine water and sediment into the marine environment.The temporal variations in metal concentrations and enrichment factors were less pronounced in the temporary open/closed estuaries than the Kariega Estuary. The observed trend can probably be related to the low anthropogenic impact within the catchment areas of these systems, and the relatively smaller size of the catchments. Significant spatial variations existed in metal enrichment in the sediment of both the East Kleinemonde and Riet Estuaries, with the highest degrees of enrichment occurring in the sediments from the marine environment and lower reaches.Keywords: metal enrichment, estuaries, sediment and water, South Afric
Antimicrobial Resistance in New Zealand—A One Health Perspective
Antimicrobial resistance (AMR) is an increasing global threat that affects human, animal and, often less acknowledged, environmental health. This complex issue requires a multisectoral One Health approach to address the interconnectedness of humans, animals and the natural environment. The prevalence of AMR in these reservoirs varies widely among countries and thus often requires a country-specific approach. In New Zealand (NZ), AMR and antimicrobial usage in humans are relatively well-monitored and-understood, with high human use of antimicrobials and the frequency of resistant pathogens increasing in hospitals and the community. In contrast, on average, NZ is a low user of antimicrobials in animal husbandry systems with low rates of AMR in food-producing animals. AMR in New Zealand’s environment is little understood, and the role of the natural environment in AMR transmission is unclear. Here, we aimed to provide a summary of the current knowledge on AMR in NZ, addressing all three components of the One Health triad with a particular focus on environmental AMR. PSS14 VALIDATION OF THE EYELASH SATISFACTION FOLLOW-UP QUESTIONNAIRE FOR FOLLOW-UP SELF-ASSESSMENT OF EYELASH SATISFACTION
Recent hypotheses on the action of antidepressants imply a modulation of excitatory amino acid transmission. Here, the effects of long-term antidepressant application in rats with the drug tianeptine were examined at hippocampal CA3 commissural associational (c/a) glutamate receptor ion channels, employing the whole-cell patch-clamp technique. The drug's impact was tested by subjecting rats to daily restraint stress for three weeks in combination with tianeptine treatment (10 mg/kg/day). Whereas stress increased the deactivation time-constant and amplitude of the N -methyl-d-aspartate (NMDA) receptor-mediated excitatory postsynaptic currents (EPSCs), it did not affect the alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA)/kainate receptor-mediated EPSCs. Concomitant pharmacological treatment of stressed animals with tianeptine resulted in a normalized scaling of the amplitude ratio of NMDA receptor to AMPA/kainate receptor-mediated currents and prevented the stress-induced attenuation of NMDA-EPSCs deactivation. Both paired-pulse-facilitation and frequency-dependent plasticity remained unchanged. Both in control and stressed animals, however, tianeptine treatment strengthened the slope of the input-output relation of EPSCs. The latter was mimicked by exposing hippocampal slices in vitro with 10 mum tianeptine, which rapidly increased the amplitudes of NMDA- and AMPA/kainate EPSCs. The enhancement of EPSCs could be blocked by the intracellular presence of the kinase inhibitor staurosporine (1 mum), suggesting the involvement of a postsynaptic phosphorylation cascade rather then presynaptic release mechanisms at CA3 c/a synapses. These results indicate that tianeptine targets the phosphorylation-state of glutamate receptors at the CA3 c/a synapse. This novel signal transduction mechanism for tianeptine may provide a mechanistic resolution for its neuroprotective properties and, moreover, a pharmacological trajectory for its memory enhancing and/or antidepressant activity
Individual placement and support versus individual placement and support enhanced with work-focused cognitive behaviour therapy: feasibility study for a randomised controlled trial
Introduction: Employment is a key goal for many people with long-term mental health issues. Evidence-based individual placement and support is a widely advocated approach. This study explored whether individual placement and support outcomes could be enhanced with work-focused counselling.
Method: The study was designed as a pragmatic randomised controlled trial comparing the cost-effectiveness, in severe mental illness, of work-focused intervention (intervention) as an adjunct to individual placement and support compared to individual placement and support alone (control).
Results: The original sample (330) proved impossible to attain so the design was revised to a pilot study from which information on feasibility of a full trial could be drawn. Twenty-five individuals out of 74 found paid work but no difference was found in the mean number of hours in paid employment between the intervention and control groups.
Conclusion: Results demonstrate that delivering work-focused counselling in tandem with individual placement and support is feasible and acceptable to service users. The study observed that, even during a period of recession (2010–13), individuals with mental health problems succeeded in obtaining paid employment. Any additional benefit of counselling over individual placement and support alone could not be ascertained, due mainly to the high drop-out rate from this study
The mu problem and sneutrino inflation
We consider sneutrino inflation and post-inflation cosmology in the singlet
extension of the MSSM with approximate Peccei-Quinn(PQ) symmetry, assuming that
supersymmetry breaking is mediated by gauge interaction. The PQ symmetry is
broken by the intermediate-scale VEVs of two flaton fields, which are
determined by the interplay between radiative flaton soft masses and higher
order terms. Then, from the flaton VEVs, we obtain the correct mu term and the
right-handed(RH) neutrino masses for see-saw mechanism. We show that the RH
sneutrino with non-minimal gravity coupling drives inflation, thanks to the
same flaton coupling giving rise to the RH neutrino mass. After inflation,
extra vector-like states, that are responsible for the radiative breaking of
the PQ symmetry, results in thermal inflation with the flaton field, solving
the gravitino problem caused by high reheating temperature. Our model predicts
the spectral index to be n_s\simeq 0.96 due to the additional efoldings from
thermal inflation. We show that a right dark matter abundance comes from the
gravitino of 100 keV mass and a successful baryogenesis is possible via

Effective theories of single field inflation when heavy fields matter
We compute the low energy effective field theory (EFT) expansion for
single-field inflationary models that descend from a parent theory containing
multiple other scalar fields. By assuming that all other degrees of freedom in
the parent theory are sufficiently massive relative to the inflaton, it is
possible to derive an EFT valid to arbitrary order in perturbations, provided
certain generalized adiabaticity conditions are respected. These conditions
permit a consistent low energy EFT description even when the inflaton deviates
off its adiabatic minimum along its slowly rolling trajectory. By generalizing
the formalism that identifies the adiabatic mode with the Goldstone boson of
this spontaneously broken time translational symmetry prior to the integration
of the heavy fields, we show that this invariance of the parent theory dictates
the entire non-perturbative structure of the descendent EFT. The couplings of
this theory can be written entirely in terms of the reduced speed of sound of
adiabatic perturbations. The resulting operator expansion is distinguishable
from that of other scenarios, such as standard single inflation or DBI
inflation. In particular, we re-derive how certain operators can become
transiently strongly coupled along the inflaton trajectory, consistent with
slow-roll and the validity of the EFT expansion, imprinting features in the
primordial power spectrum, and we deduce the relevant cubic operators that
imply distinct signatures in the primordial bispectrum which may soon be
