    AbstractOur recent results on autocrine nerve growth factor (NGF) synthesis in B lymphocytes, which directly regulates the expression and release of calcitonin gene-related peptide (CGRP), a neuropeptide known to down-regulate immune response, led us to propose an anti-inflammatory action of NGF. In the present work, we investigated whether the endogenous synthesis of NGF can regulate the expression of CGRP in other antigen-presenting cells, such as monocytes, and whether this may have a functional effect. Our data indicate that human monocytes synthesize basal levels of NGF and CGRP and that, following lipopolysaccharide (LPS) stimulation, NGF and CGRP expression are both up-regulated. When endogenous NGF is neutralized, the up-regulation of CGRP expression induced by LPS is inhibited. The expression of membrane molecules involved in T-cell activation such as human leukocyte antigen-DR (HLA-DR) and CD86 is affected by endogenous NGF, and similar effects were obtained using a CGRP1 receptor antagonist. In addition, NGF deprivation in LPS-treated monocytes significantly decreases interleukin 10 (IL-10) synthesis. Our findings indicate that endogenous NGF synthesis has a functional role and may represent a physiologic mechanism to down-regulate major histocompatibility complex (MHC) class II and CD86 expression and alter the development of immune responses

    Nerve growth factor is an autocrine survival factor for memory B lymphocytes

    AbstractProduction of nerve growth factor (NGF) was assessed in cultures of human T and B lymphocytes and macrophages. NGF was constitutively produced by B cells only, which also expressed surface p140trk-A and p75NGFR molecules and hence efficiently bound and internalized the cytokine. Neutralization of endogenous NGF caused disappearance of Bcl-2 protein and apoptotic death of resting lymphocytes bearing surface IgG or IgA, a population comprising memory cells, while surface IgM/IgD “virgin” B lymphocytes were not affected. In vivo administration of neutralizing anti-NGF antibodies caused strong reduction in the titer of specific IgG in mice immunized with tetanus toxoid, nitrophenol, or arsonate and reduced numbers of surface IgG or IgA B lymphocytes. Thus, NGF is an autocrine survival factor for memory B lymphocytes

    I programmi di identificazione precoce delle ipoacusie infantili permette di intraprendere rapidamente l’iter riabilitativo e protesico appropriato; nonostante ciò appare ancora elevato il numero di pazienti che non aderiscono al programma terapeutico. L’obiettivo di questo articolo è presentare i risultati di un’analisi strategica che prende in considerazione i punti di forza, i punti di debolezza, le opportunità e i rischi del processo di follow up audiologico/protesico/linguistico di bambini affetti da ipoacusia bilaterale permanente. Il follow up in questione, coinvolgendo bambini piccoli, implica il coinvolgimento di professionisti specializzati e inseriti in un team multidisciplinare e una gestione plurisettoriale complessa e prolungata. Nell’ambito del progetto finanziato dal Ministero della Salute Italiano CCM 2013 denominato “Programma regionale di identificazione, intervento e presa in carico precoci per la prevenzione dei disturbi comunicativi nei bambini con deficit uditivo” lo scopo di quest’analisi è stata quella di offrire delle raccomandazioni per armonizzare i criteri di valutazione dei risultati terapeutici e trarre spunto da questi per successivamente fornire delle linee guida riguardanti le metodiche più adeguate nella valutazione del bambino con ipoacusia permanente

    Genomic NGFB variation and multiple sclerosis in a case control study

    <p>Abstract</p> <p>Background</p> <p>Nerve growth factor β (NGFB) is involved in cell proliferation and survival, and it is a mediator of the immune response. ProNGF, the precursor protein of NGFB, has been shown to induce cell death via interaction with the p75 neurotrophin receptor. In addition, this neurotrophin is differentially expressed in males and females. Hence NGFB is a good candidate to influence the course of multiple sclerosis (MS), much like in the murine model of experimental autoimmune encephalomyelitis (EAE).</p> <p>Methods</p> <p>Ten single nucleotide polymorphisms (SNPs) were genotyped in the <it>NGFB </it>gene in up to 1120 unrelated MS patients and 869 controls. Expression analyses were performed for selected MS patients in order to elucidate the possible functional relevance of the SNPs.</p> <p>Results</p> <p>Significant association of NGFB variations with MS is evident for two SNPs. <it>NGFB </it>mRNA seems to be expressed in sex- and disease progression-related manner in peripheral blood mononuclear cells.</p> <p>Conclusion</p> <p>NGFB variation and expression levels appear as modulating factors in the development of MS.</p

    Differentiation of normal and cancer cells induced by sulfhydryl reduction: biochemical and molecular mechanisms

    We examined the morphological, biochemical and molecular outcome of a nonspecific sulfhydryl reduction in cells, obtained by supplementation of N-acetyl-L-cysteine (NAC) in a 0.1-10 mM concentration range. In human normal primary keratinocytes and in colon and ovary carcinoma cells we obtained evidences for: (i) a dose-dependent inhibition of proliferation without toxicity or apoptosis; (ii) a transition from a proliferative mesenchymal morphology to cell-specific differentiated structures; (iii) a noticeable increase in cell-cell and cell-substratum junctions; (iv) a relocation of the oncogenic beta-catenin at the cell-cell junctions; (v) inhibition of microtubules aggregation; (vi) upregulation of differentiation-related genes including p53, heat shock protein 27 gene, N-myc downstream-regulated gene 1, E-cadherin, and downregulation of cyclooxygenase-2; (vii) inhibition of c-Src tyrosine kinase. In conclusion, a thiol reduction devoid of toxicity as that operated by NAC apparently leads to terminal differentiation of normal and cancer cells through a pleiade of converging mechanisms, many of which are targets of the recently developed differentiation therapy
