    Trace Element Composition of Stream Sediments an Integrating Factor for Water Quality

    Bottom sediments, suspended sediments, and water were sampled along 130 miles of the Buffalo River in northern Arkansas. The water and acid extracts of the suspended sediments and the minus 95 mesh fraction of the bottom sediments were analyzed by atomic absorption spectrometry. All samples were analyzed for Na, K, Mg, Ca, Zn, Cd, Cu, Pb, Fe, Co, Cr, Ni, and Mn. Selected bottom samples also were analyzed by As, Hg, and Zr. Zr was determined by x-ray fluorescence. Li and Sr were determined for selected water and suspended sediment samples. There is a general decrease downstream in Fe, Cu, Cr, Ni, Mn, Pb, K, and Na in the bottom sediments as the drainage area increases in carbonate rock and decreases in shale. The elements Mg, Ca, Zn, and Cd increase in bottom sediments downstream. The values for these elements in the water, especially the major elements, also correspond closely with the geology of the region. Tributaries are sites of abrupt rise and fall of metal values, within a few miles, from background to anomalously high values to background, especially tributaries draining Zn and Pb mineralized areas. The bottom sediments are mainly quartz and chert grains. These grains apparently are coated with hydrous iron oxide which acts as a sorbent for many of the elements and is a dominant transport mechanism for acid extractable Co, Cr, Ni, Cu, Mn, and K. Other acid extractable metals, particularly Mg, Ca, Zn, Cd, and Pb, are mostly in clastic grains. Graphic representation of the Langmuir equation for Mn is consistent with adsorption of Mn by iron in both bottom sediments and suspended sediments. On the basis of the volume of water collected, all the elements except Fe are more concentrated in the water than in the suspended sediments. Fe concentration of the suspended sediments increases with increasing flow because the suspended load is increased. The Mn/Fe ratio of the suspended sediments is approximately equal to or greater than that of the bottom sediments. The Mn/Fe ratio of suspended sediments relative to that of the bottom sediments increases downstream, possibly because of an autocatalytic effect of Mn precipitation. The relationship between sediment and water concentrations is not clear from the data because of the restricted concentration ranges for some elements in the suspended sediment and water. The sediment from the Buffalo River can be used to estimate grossly the concentration of elements in the water

    Role of PINCH and Its Partner Tumor Suppressor Rsu-1 in Regulating Liver Size and Tumorigenesis

    Particularly interesting new cysteine-histidine-rich protein (PINCH) protein is part of the ternary complex known as the IPP (integrin linked kinase (ILK)-PINCH-Parvin-α) complex. PINCH itself binds to ILK and to another protein known as Rsu-1 (Ras suppressor 1). We generated PINCH 1 and PINCH 2 Double knockout mice (referred as PINCH DKO mice). PINCH2 elimination was systemic whereas PINCH1 elimination was targeted to hepatocytes. The genetically modified mice were born normal. The mice were sacrificed at different ages after birth. Soon after birth, they developed abnormal hepatic histology characterized by disorderly hepatic plates, increased proliferation of hepatocytes and biliary cells and increased deposition of extracellular matrix. After a sustained and prolonged proliferation of all epithelial components, proliferation subsided and final liver weight by the end of 30 weeks in livers with PINCH DKO deficient hepatocytes was 40% larger than the control mice. The livers of the PINCH DKO mice were also very stiff due to increased ECM deposition throughout the liver, with no observed nodularity. Mice developed liver cancer by one year. These mice regenerated normally when subjected to 70% partial hepatectomy and did not show any termination defect. Ras suppressor 1 (Rsu-1) protein, the binding partner of PINCH is frequently deleted in human liver cancers. Rsu-1 expression is dramatically decreased in PINCH DKO mouse livers. Increased expression of Rsu-1 suppressed cell proliferation and migration in HCC cell lines. These changes were brought about not by affecting activation of Ras (as its name suggests) but by suppression of Ras downstream signaling via RhoGTPase proteins. In conclusion, our studies suggest that removal of PINCH results in enlargement of liver and tumorigenesis. Decreased levels of Rsu-1, a partner for PINCH and a protein often deleted in human liver cancer, may play an important role in the development of the observed phenotype. © 2013 Donthamsetty et al

    Canton Connections: A University-Community Partnership for Post-Disaster Revitalization

    Back-to-back hurricanes prompted the creation of a partnership between Western Carolina University and an affected community in western North Carolina. The partnership was designed to promote the economic, social, and cultural revitalization of the community while creating opportunities for civic engagement and enriched student learning. The principal stakeholders in the partnership were the university and the municipal government, representing the community at large. The partners undertook several projects over a three-year period as part of a comprehensive, multifaceted initiative. In this article, the authors discuss the benefits and impact of the projects on participants and the community. They also share the insights gained and lessons learned from the initiative and comment briefly on factors inherent in effective university-community partnerships

    Oct4 Is Crucial for Transdifferentiation of Hepatocytes to Biliary Epithelial Cells in an in Vitro Organoid Culture Model

    BACKGROUND: Hepatocyte to biliary transdifferentiation has been documented in various models of bile duct injury. In this process, mature hepatocytes transform into mature biliary epithelial cells by acquiring biliary phenotypic markers. Several signaling pathways including PI3 kinase, Notch, Hes1, Sox9, and Hippo are shown to be involved in the process. However, if Oct4 is involved in hepatocyte to biliary transdifferentiation is unknown. METHODS: We investigated the role of Oct4 in hepatocyte to biliary transdifferentiation utilizing an in vitro organoid culture system as a model of transdifferentiation. Oct4 was inhibited using adenovirus containing Oct4 shRNA. Hepatocyte specific HNF-4α and biliary specific HNF-1β & CK19 expression were assessed to gauge the extent of transdifferentiation. RESULTS: Oct4 was induced during hepatocyte to biliary transdifferentiation. Oct4 inhibition significantly downregulated the appearance of biliary cells from hepatocytes. This was accompanied by a significant downregulation of signaling pathways including Notch, Sox9, and Hippo. CONCLUSION: Our findings suggest that Oct4 is crucial for hepatocyte to biliary transdifferentiation and maturation and that it acts upstream of Notch, Sox9, and Hippo signaling in this model. This finding identifies new signaling through Oct4 in plasticity between hepatocytes and biliary epithelial cells, which can be potentially utilized to identify new strategies in chronic biliary diseases

    Formation of a rotating hole from a close limit head-on collision

    Realistic black hole collisions result in a rapidly rotating Kerr hole, but simulations to date have focused on nonrotating final holes. Using a new solution of the Einstein initial value equations we present here waveforms and radiation for an axisymmetric Kerr-hole-forming collision starting from small initial separation (the ``close limit'' approximation) of two identical rotating holes. Several new features are present in the results: (i) In the limit of small separation, the waveform is linear (not quadratic) in the separation.     To provide geoidal topography over the world oceans, a radar altimeter carried by earth satellite is planned. Ground truth calibration will be provided by a grid comprised of the equatorial belt and meridional traverses along the 30°W and 150°W meridians. Ground truth topography is derived from gravity values measured along these traverses.     A First Attempt to Bring Computational Biology into Advanced High School Biology Classrooms

    Computer science has become ubiquitous in many areas of biological research, yet most high school and even college students are unaware of this. As a result, many college biology majors graduate without adequate computational skills for contemporary fields of biology. The absence of a computational element in secondary school biology classrooms is of growing concern to the computational biology community and biology teachers who would like to acquaint their students with updated approaches in the discipline. We present a first attempt to correct this absence by introducing a computational biology element to teach genetic evolution into advanced biology classes in two local high schools. Our primary goal was to show students how computation is used in biology and why a basic understanding of computation is necessary for research in many fields of biology. This curriculum is intended to be taught by a computational biologist who has worked with a high school advanced biology teacher to adapt the unit for his/her classroom, but a motivated high school teacher comfortable with mathematics and computing may be able to teach this alone. In this paper, we present our curriculum, which takes into consideration the constraints of the required curriculum, and discuss our experiences teaching it. We describe the successes and challenges we encountered while bringing this unit to high school students, discuss how we addressed these challenges, and make suggestions for future versions of this curriculum.We believe that our curriculum can be a valuable seed for further development of computational activities aimed at high school biology students.     An Analysis of Private School Closings

    We add to the small literature on private school supply by exploring exits of K-12 private schools. We find that the closure of private schools is not an infrequent event, and use national survey data from the National Center for Education Statistics to study closures of private schools. We assume that the probability of an exit is a function of excess supply of private schools over the demand, as well as the school's characteristics such as age, size, and religious affiliation.     Work-Related Mental Health and Job Performance: Can Mindfulness Help?

    Work-related mental health issues such as work-related stress and addiction to work impose a significant health and economic burden to the employee, the employing organization, and the country of work more generally. Interventions that can be empirically shown to improve levels of work-related mental health – especially those with the potential to concurrently improve employee levels of work performance – are of particular interest to occupational stakeholders. One such broad-application interventional approach currently of interest to occupational stakeholders in this respect is mindfulness-based interventions (MBIs). Following a brief explication of the mindfulness construct, this paper critically discusses current research directions in the utilization of mindfulness in workplace settings and assesses its suitability for operationalization as an organization-level work-related mental health intervention. By effecting a perceptual-shift in the mode of responding and relating to sensory and cognitive-affective stimuli, employees that undergo mindfulness training may be able to transfer the locus of control for stress from external work conditions to internal metacognitive and attentional resources. Therefore, MBIs may constitute cost-effective organization-level interventions due to not actually requiring any modifications to human resource management systems and practises. Based on preliminary empirical findings and on the outcomes of MBI studies with clinical populations, it is concluded that MBIs appear to be viable interventional options for organizations wishing to improve the mental health of their employees
