    Optimización superficial de alambres de ortodoncia de Ni-Ti superelástico mediante nitruración gaseosa. Parte II: Cuantificación de la mejora de la nanodureza y el coeficiente de fricción

    La mejora del deslizamiento entre el alambre de ortodoncia y el bracket es uno de los principales problemas en la terapia ortodoncica. Este estudio presenta la nitruración gaseosa como un eficaz método de mejora del deslizamiento de los alambres ortodóncicos de Ni-Ti superelástico (composiciones cercanas a la equiatómica) respecto al bracket, principal inconveniente de estas aleaciones en la aplicación ortodóncica. Mediante microscopía óptica y electrónica de barrido se comprobó la existencia de una capa superficial nitrurada, así como la morfología de las fases presentes. Asimismo, se realizaron microanálisis químicos de las capas exteriores mediante técnica de energía dispersiva de rayos X. En todas las muestras nitruradas la dureza aumentó con la severidad del tratamiento y se alcanzaron valores de hasta 1111HV en comparación con los valores de 323 HV del alambre sin tratar. Se diseñaron ensayos de tipo pin on disk adaptados a las peculiaridades de los alambres, que mostraron un descenso del coeficiente de fricción. El endurecimiento y mejora de la fricción en la superficie del material se justifican por la formación de las capas nitruradas.Peer Reviewe

    Interaction of glutathione transferase from horse erythrocytes with 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole

    7-Chloro-4-nitrobenzo-2-oxa-1,3-diazole reacts with two thiol groups of the dimeric horse erythrocyte glutathione transferase at pH 5.0, with strong inactivation reversible on dithiothreitol treatment. The inactivation kinetic follows a biphasic pattern, similar to that caused by other thiol reagents as recently reported. Both S-methylglutathione and 1-chloro-2,4-dinitrobenzene protect the enzyme from inactivation. Analysis of the reactive SH group-containing peptide gives the sequence Ala-Ser-Cys-Leu-Tyr, identical with that of the peptide that contains the reactive cysteine 47 of the human placental transferase. In the presence of glutathione, the enzyme is not inactivated by this reagent, but it catalyzes its conjugation to glutathione. At higher pH values, 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole reacts with 2 tyrosines/dimer and lysines, as well as with cysteines. Reaction with lysine seems essentially without effect on activity; whether the reactive tyrosines are important for activity could not be determined using this reagent only. However, 2 tyrosines among the 4 that are nitrated by tetranitro-methane are important for activity

    Glycine-Stabilized Zinc Gluconate has similar bioavailability than Zinc Sulfate in a zinc fortified probiotic food

    Objective: In this study, we evaluated zinc gluconate stabilized with glycine (GZ) and sulfate (SZ) in fermented milk as vehicle. Zinc bioavailability was evaluated in an animal model (Sprague Dawley rats) for both zinc sources in the vehicle with a probiotic (Lactobacillus casei DN114001). Results: For growth parameters, higher weight gain and femur weight values were observed when probiotic and zinc were provided together, independent of the source (weight gain: SZ 81.4g±4.0g; GZ 81.8g±4.0g and 70.2g±12.5g without the probiotic; femur weight: SZ 0.51g ±0.05g; GZ 0.52g±0.05g and 0.42g±0.03g without the probiotic). Femur zinc content was higher for zinc gluconate stabilized with glycine in the presence of the probiotic (97.04ppm±8.40ppm), and the results were similar for zinc sulfate with or without probiotic (84.51ppm±2.44ppm and 84.94ppm±2.28ppm, respectively). Serum antioxidant capacity and immune cellular response were also evaluated by using free radical scavenging assays and a T cell proliferation assay respectively. The free radical scavenging assay showed a tendency to increase with zinc provision, and the highest proliferation index was observed for glycine-stabilized zinc gluconate and the probiotic. Conclusion: The results indicate that the combination of zinc (as glycine-stabilized zinc gluconate) and a probiotic may be beneficial for the evaluated parameters.Fil: Tesan, Fiorella Carla.     Genetic and Physiological Analysis of Iron Biofortification in Maize Kernels

    BACKGROUND: Maize is a major cereal crop widely consumed in developing countries, which have a high prevalence of iron (Fe) deficiency anemia. The major cause of Fe deficiency in these countries is inadequate intake of bioavailable Fe, where poverty is a major factor. Therefore, biofortification of maize by increasing Fe concentration and or bioavailability has great potential to alleviate this deficiency. Maize is also a model system for genomic research and thus allows the opportunity for gene discovery. Here we describe an integrated genetic and physiological analysis of Fe nutrition in maize kernels, to identify loci that influence grain Fe concentration and bioavailability. METHODOLOGY: Quantitative trait locus (QTL) analysis was used to dissect grain Fe concentration (FeGC) and Fe bioavailability (FeGB) from the Intermated B73 Ă— Mo17 (IBM) recombinant inbred (RI) population. FeGC was determined by ion coupled argon plasma emission spectroscopy (ICP). FeGB was determined by an in vitro digestion/Caco-2 cell line bioassay. CONCLUSIONS: Three modest QTL for FeGC were detected, in spite of high heritability. This suggests that FeGC is controlled by many small QTL, which may make it a challenging trait to improve by marker assisted breeding. Ten QTL for FeGB were identified and explained 54% of the variance observed in samples from a single year/location. Three of the largest FeGB QTL were isolated in sister derived lines and their effect was observed in three subsequent seasons in New York. Single season evaluations were also made at six other sites around North America, suggesting the enhancement of FeGB was not specific to our farm site. FeGB was not correlated with FeGC or phytic acid, suggesting that novel regulators of Fe nutrition are responsible for the differences observed. Our results indicate that iron biofortification of maize grain is achievable using specialized phenotyping tools and conventional plant breeding techniques

    Researching COVID to enhance recovery (RECOVER) tissue pathology study protocol: Rationale, objectives, and design.

    ImportanceSARS-CoV-2 infection can result in ongoing, relapsing, or new symptoms or organ dysfunction after the acute phase of infection, termed Post-Acute Sequelae of SARS-CoV-2 (PASC), or long COVID. The characteristics, prevalence, trajectory and mechanisms of PASC are poorly understood. The objectives of the Researching COVID to Enhance Recovery (RECOVER) tissue pathology study (RECOVER-Pathology) are to: (1) characterize prevalence and types of organ injury/disease and pathology occurring with PASC; (2) characterize the association of pathologic findings with clinical and other characteristics; (3) define the pathophysiology and mechanisms of PASC, and possible mediation via viral persistence; and (4) establish a post-mortem tissue biobank and post-mortem brain imaging biorepository.MethodsRECOVER-Pathology is a cross-sectional study of decedents dying at least 15 days following initial SARS-CoV-2 infection. Eligible decedents must meet WHO criteria for suspected, probable, or confirmed infection and must be aged 18 years or more at the time of death. Enrollment occurs at 7 sites in four U.S. states and Washington, DC. Comprehensive autopsies are conducted according to a standardized protocol within 24 hours of death; tissue samples are sent to the PASC Biorepository for later analyses. Data on clinical history are collected from the medical records and/or next of kin. The primary study outcomes include an array of pathologic features organized by organ system. Causal inference methods will be employed to investigate associations between risk factors and pathologic outcomes.DiscussionRECOVER-Pathology is the largest autopsy study addressing PASC among US adults. Results of this study are intended to elucidate mechanisms of organ injury and disease and enhance our understanding of the pathophysiology of PASC

    Using ethical orientations to explain administrative pressure to practice unethically: A pilot study

    The promotion of students’ welfare is one of the central professional and ethical responsibilities of school psychology practitioners. However, some practitioners are subjected to pressure from administrators to engage in behavior that runs counter to ethical mandates and may be detrimental to students’ well-being. The phenomenon of administrative pressure to violate ethical standards might be explained by professionals’ adherence to disparate ethical philosophies, with school psychologists prioritizing the protection of individual rights and school principals valuing the “good of the many.” This pilot study explored the feasibility of using a scenario-based instrument in drawing out and comparing the dominant ethical perspectives of school psychologists and school principals. Participants (N = 56) consisted of 35 school psychologists and 21 school principals who completed a survey measure that included six ethical dilemmas, each featuring a conflict between the rights of an individual student and benefits to the larger student body. Participants were required to select the more ethical of two solutions from options reflecting either Kantian or utilitarian considerations. The results of this investigation point to the possibility that school professionals endorse conflicting ethical philosophies under certain circumstances. Although both professional groups tended to favor a Kantian framework, school principals were comparatively more supportive of utilitarian principles. The incorporation of ethical vignettes into research aimed at identifying incompatibilities in ideological preferences seems to hold promise as an investigative technique. Implications for practice are offered with an emphasis on suggestions for bridging the gap between professional considerations of administrators and school psychologists

    Improving Schools’ Readiness for Involvement in Suicide Prevention: An Evaluation of the Creating Suicide Safety in Schools (CSSS) Workshop

    Schools have an important role to play in combatting suicide, a significant public health problem that disproportionately affects adolescents and young adults. Schools can work to reduce youth suicidality by adopting policies that align with best practice recommendations pertaining to suicide prevention, intervention, and postvention. This study examined the impact of a one-day training, the Creating Suicide Safety in Schools (CSSS) workshop, on the readiness of school personnel to improve their schools’ suicide-related policies and procedures. Participants (N = 562) consisted predominantly of school-based mental health professionals working in communities of low or mixed socioeconomic status in New York State. Survey data were collected according to a one-group pre-test—post-test design with a 3-month follow-up. Workshop participants demonstrated improvements from pre-test to post-test in their attitudes about the importance of school-based suicide prevention, knowledge of best practices, perceptions of administrative support, and feelings of empowerment to work collaboratively to enhance their schools’ suicide safety. At follow-up, participants reported barriers to implementing changes, most commonly in the form of insufficient time and stigma surrounding the topic of suicide. The results of this study provide preliminary evidence for the effectiveness of the CSSS workshop as a promising method for improving schools’ suicide safety, yet additional research using randomized controlled trials needs to be conducted
