    An optical fiber measurement system design on tool radial vibration

    The effects of tool radial vibration bring not only poor surface quality, inferior dimensional accuracy, but also disproportionate tool wear or tool breakage and excessive noise. Therefore, online measurement and monitoring of tool vibration are necessary. In order to monitor the tool vibration, an optical fiber measurement system was design in this pater. Firstly, the structure and basic principle of the optical fiber sensor was given; secondly, the light intensity to voltage converter circuit was introduced; then, an experiment platform was built for verify the feasibility of the optical measuring system, and the result shows that the radial vibration of a smooth 10 mm diameter shaft can be measured quickly

    Testing for high-dimensional white noise

    Testing for multi-dimensional white noise is an important subject in statistical inference. Such test in the high-dimensional case becomes an open problem waiting to be solved, especially when the dimension of a time series is comparable to or even greater than the sample size. To detect an arbitrary form of departure from high-dimensional white noise, a few tests have been developed. Some of these tests are based on max-type statistics, while others are based on sum-type ones. Despite the progress, an urgent issue awaits to be resolved: none of these tests is robust to the sparsity of the serial correlation structure. Motivated by this, we propose a Fisher's combination test by combining the max-type and the sum-type statistics, based on the established asymptotically independence between them. This combination test can achieve robustness to the sparsity of the serial correlation structure,and combine the advantages of the two types of tests. We demonstrate the advantages of the proposed test over some existing tests through extensive numerical results and an empirical analysis.Comment: 84 page

    Short-term lifestyle education on obesity reduction in adolescents

    BackgroundsObesity is increasing in adolescents in China. However, the awareness of obesity and prevention on related risk factors were not well known. We aim to assess the effectiveness of short-term health education intervention on obesity in Chinese adolescents.MethodsIn this study, 42 primary and secondary schools from Qingdao were randomly divided into the education and control groups. A total of 11,739 adolescents was included in the current study. The logistic regression was employed to assess odds ratio (OR) of education intervention on overweight and obesity prevalence adjusting for covariates.ResultsThe baseline prevalence of overweight and obesity was significantly higher in urban than in rural areas and in boys than in girls. After 1 year lifestyle intervention, the proportion of students with awareness of obesity was higher, meanwhile age-adjusted mean values of weight, body mass index, duration of watching TV and doing homework were lower in education group than control group. The corresponding figures were 43.6 [95% CI (confidence intervals); 43.3–43.9] kg versus 44.3 (95% CI; 44.0–44.6) kg, 18.6 (95% CI; 18.5–18.7) kg/m2 versus 18.9 (95% CI; 18.8–19.1) kg/m2, 1.3 (95% CI; 1.2–1.3) hours/d versus 1.4 (95% CI; 1.3–1.4) hours/d, and 1.5 (95% CI; 1.4–1.5) hours/d versus 1.8 (95% CI, 1.7–1.8) hours/d. The multivariable adjusted OR for combined prevalence of overweight and obesity was 0.85 (95% CI, 0.76–0.96) in education group as compared with control group.ConclusionShort-term health education intervention results in significantly higher reductions in obesity parameters and improvement in awareness in Chinese adolescents

    Imaging and Pathological Features of Percutaneous Cryosurgery on Normal Lung Evaluated in a Porcine Model

    Background and objective Lung cancer is one of the most commonly occurring malignancies and frequent causes of death in the world. Cryoablation is a safe and alternative treatment for unresectable lung cancer. Due to the lung being gas-containing organ and different from solid organs such as liver and pancreas, it is difficult to achieve the freezing range of beyond the tumor edge 1 cm safety border. The aim of this study is to examine the effect of different numbers of freeze cycles on the effectiveness of cryoablation on normal lung tissue and to create an operation guideline that gives the best effect. Methods Six healthy Tibetan miniature pigs were given a CT scan and histological investigation after percutaneous cryosurgery. Cryoablation was performed as 2 cycles of 10 min of active freezing in the left lung; each freeze followed by a 5 min thaw. In the right lung, we performed the same 2 cycles of 5 min of freezing followed by 5 min of thawing. However, for the right lung, we included a third cycle of consisting of 10 min of freezing followed by 5 min of thawing. Three cryoprobes were inserted into the left lung and three cryoprobes in the right lung per animal, one in the upper and two in the lower lobe, so as to be well away from each other. Comparison under the same experimental condition was necessary. During the experiment, observations were made regarding the imaging change of ice-ball. The lungs were removed postoperatively at 3 intervals: 4 h, 3 d of postoperation and 7 d of postoperation, respectively, to view microscopic and pathological change. Results The ice-ball grew gradually in relation to the increase in time, and the increase in number of cycles. The size of the cryolesion (hypothesis necrotic area) in specimens, over time, became larger in size than the size of the ice-ball during operation, regardless of whether 2 or 3 freeze-thaw cycles were performed. The area of necrosis was gradually increased over the course of time. The hypothesis necrotic area was equal to necrosis area 3 d after cryosurgery. Conclusion Percutaneous cryoablation of the lung can achieve complete ablation of target tissue. The freezing technique may be different depending on the individual circumstances of each tumor. In technology, 3 freeze-thaw cycles are recommended, and the range of cryoablation’s effective diameter may be not necessarily beyond the tumor edge at least 1 cm safe border during cryosurgery

    Quantitative proteomics analysis reveals an important role of the transcriptional regulator UidR in the bacterial biofilm formation of Aeromonas hydrophila

    IntroductionBacterial biofilm is a well-known characteristic that plays important roles in diverse physiological functions, whereas the current intrinsic regulatory mechanism of its formation is still largely unknown. MethodsIn the present study, a label-free based quantitative proteomics technology was conducted to compare the differentially expressed proteins (DEPs) between ΔuidR and the wild-type strain in the biofilm state.ResultsThe results showed that the deletion of gene uidR encoding a TetR transcriptional regulator significantly increased the biofilm formation in Aeromonas hydrophila. And there was a total of 220 DEPs, including 120 up-regulated proteins and 100 down-regulated proteins between ΔuidR and the wild-type strain based on the quantitative proteomics. Bioinformatics analysis suggested that uidR may affect bacterial biofilm formation by regulating some related proteins in glyoxylic acid and dicarboxylic acid pathway. The expressions of selected proteins involved in this pathway were further confirmed by q-PCR assay, and the results was in accordance with the quantitative proteomics data. Moreover, the deletion of four genes (AHA_3063, AHA_3062, AHA_4140 and aceB) related to the glyoxylic acid and dicarboxylic acid pathway lead to a significant decrease in the biofilm formation. DiscussionThus, the results indicated that uidR involved in the regulatory of bacterial biofilm formation, and it may provide a potential target for the drug development and a new clue for the prevention of pathogenic A. hydrophila in the future

    A correlation analysis of HHV infection and its predictive factors in an HIV-seropositive population in Yunnan, China

    Human herpesviruses (HHVs) have a particularly high prevalence in certain high-risk populations and cause increased morbidity and mortality in patients with acquired immunodeficiency syndrome (AIDS). Screening and treating subclinical HHV infections reduce human immunodeficiency virus (HIV) infection incidence, disease progression, and transmission. However, there are few studies on HHVs, HIV coinfection rates, and their related risk factors. We aimed to clarify the prevalence of all eight HHVs in peripheral blood samples collected from HIV-positive patients, and explore the association of HHV infection in HIV-positive patients in an HIV-seropositive population in Yunnan. We recruited 121 HIV-positive patients with highly active antiretroviral therapy (HAART) and 45 healthy individuals. All the eight HHVs were detected using polymerase chain reaction and their epidemiological information and clinical data were collected and statistically analyzed. A high prevalence of HHVs (89.3%) was observed in individuals with HIV infections and with herpes simplex virus (HSV)-2 (65.3%), and HSV-1 (59.5%) being the most common. Coinfection with more than two different HHVs was more common in patients with HIV infections receiving HAART (72.7%) than in healthy controls. Older age, being married, higher HIV-1 plasma viral loads, and use of antiviral protease inhibitors were independently correlated with an increased frequency of HHVs, but we found no association with CD4 count, WHO HIV clinical stage, and HIV infection duration. Our findings are of great significance for the prevention of HHV opportunistic infection in patients with AIDS and their clinical treatment

    Replication Fork Stability Confers Chemoresistance in BRCA-deficient Cells

    Brca1- and Brca2-deficient cells have reduced capacity to repair DNA double-strand breaks (DSBs) by homologous recombination (HR) and consequently are hypersensitive to DNA damaging agents, including cisplatin and poly(ADP-ribose) polymerase (PARP) inhibitors. Here we show that loss of the MLL3/4 complex protein, PTIP, protects Brca1/2-deficient cells from DNA damage and rescues the lethality of Brca2-deficient embryonic stem cells. However, PTIP deficiency does not restore HR activity at DSBs. Instead, its absence inhibits the recruitment of the MRE11 nuclease to stalled replication forks, which in turn protects nascent DNA strands from extensive degradation. More generally, acquisition of PARPi and cisplatin resistance is associated with replication fork (RF) protection in Brca2-deficient tumor cells that do not develop Brca2 reversion mutations. Disruption of multiple proteins, including PARP1 and CHD4, leads to the same end point of RF protection, highlighting the complexities by which tumor cells evade chemotherapeutic interventions and acquire drug resistance

    High-dimensional proportionality test of two covariance matrices and its application to gene expression data

    With the development of modern science and technology, more and more high-dimensional data appear in the application fields. Since the high dimension can potentially increase the complexity of the covariance structure, comparing the covariance matrices among populations is strongly motivated in high-dimensional data analysis. In this article, we consider the proportionality test of two high-dimensional covariance matrices, where the data dimension is potentially much larger than the sample sizes, or even larger than the squares of the sample sizes. We devise a novel high-dimensional spatial rank test that has much-improved power than many existing popular tests, especially for the data generated from some heavy-tailed distributions. The asymptotic normality of the proposed test statistics is established under the family of elliptically symmetric distributions, which is a more general distribution family than the normal distribution family, including numerous commonly used heavy-tailed distributions. Extensive numerical experiments demonstrate the superiority of the proposed test in terms of both empirical size and power. Then, a real data analysis demonstrates the practicability of the proposed test for high-dimensional gene expression data
