    Yttria-Doped Zirconia as Solid Electrolyte for Fuel-Cell Applications

    7.3-10 mol% yttria-doped zirconia (YDZ) was studied with emphasis on its long-term stability as solid electrolyte. The decomposition of common 8.5YDZ (950 °C) was detected by analytical TEM. As second issue, the microstructural and chemical properties of nanocrystalline 7.3YDZ thin films were investigated. Metastable t\u27\u27-YDZ was found to precipitate in nanoscaled regions in YDZ up to 10 mol% yttria. Furthermore, a revised boundary of the c+t phase field, in which YDZ decomposes, is presented

    Characterization of the Isothermal and Thermomechanical Fatigue Behavior of a Duplex Steel Considering the Alloy Microstructure

    Isothermal and thermomechanical fatigue behavior of duplex stainless steel (DSS) X2CrNiMoN22-5-3 was investigated. The aim of this work was to understand the fatigue behavior by correlation of the isothermal and thermomechanical fatigue behavior with microstructural observations. Fatigue tests at plastic-strain-amplitude of 0.2% were carried out at 20, 300 and 600 °C, while in-phase (IP) and out-of-phase (OP) thermomechanical fatigue (TMF) experiments were performed between 300 and 600 °C. During the 20 °C fatigue test, a continuous softening was observed. Transmission electron microscopy examinations reveal pronounced planar slip behavior in austenite. At 300 °C, deformation concentrates in the ferrite, where strong interactions between Crx_{x}N and dislocations were observed that explain the pronounced cyclic hardening. DSS studied exhibits softening throughout the whole isothermal fatigue test at 600 °C. In ferrite, during the 600 °C fatigue test, the G phase, γ′ austenite precipitated, and an unordered dislocation arrangement was observed. The stress responses of the TMF tests can be correlated to those of the isothermal fatigue tests. In IP mode, a positive mean stress resulted in premature failure. No γ′ austenite but the formation of subgrains in the ferrite phase was observed after TMF tests. The plastic deformation of the austenite at high temperatures results in an unordered dislocation arrangement

    Butterfly gyroid nanostructures as a time-frozen glimpse of intracellular membrane development

    The formation of the biophotonic gyroid material in butterflywing scales is an exceptional feat of evolutionary engineering of functional nanostructures. It is hypothesized that this nanostructure forms by chitin polymerization inside a convolutedmembrane of corresponding shape in the endoplasmic reticulum. However, this dynamic formation process, including whether membrane folding and chitin expression are simultaneous or sequential processes, cannot yet be elucidated by in vivo imaging. We report an unusual hierarchical ultrastructure in the butterfly Thecla opisena that, as a solid material, allows high-resolution three-dimensional microscopy. Rather than the conventional polycrystalline spacefilling arrangement, a gyroid occurs in isolated facetted crystallites with a pronounced size gradient.When interpreted as a sequence of time-frozen snapshots of the morphogenesis, this arrangement provides insight into the formation mechanisms of the nanoporous gyroid material as well as of the intracellular organelle membrane that acts as the template

    A modern look at a medieval bilayer metal leaf: nanotomography of Zwischgold

    Many European sculptures and altarpieces from the Middle Ages were decorated with Zwischgold, a bilayer metal leaf with an ultra-thin gold face backed by silver. Zwischgold corrodes quickly when exposed to air, causing the surface of the artefact to darken and lose gloss. The conservation of such Zwischgold applied artefacts has been an obstinate problem. We have acquired quantitative, 3D nanoscale images of Zwischgold samples from 15th century artefacts and modern materials using ptychographic X-ray computed tomography (PXCT), a recently developed coherent diffractive imaging technique, to investigate the leaf structure and chemical state of Zwischgold. The measurements clearly demonstrate decreasing density (increasing porosity) of the leaf materials and their corrosion products, as well as delamination of the leaves from their substrate. Each of these effects speak to typically observed issues in the conservation of such Zwischgold applied artefacts. Further, a rare variant of Zwischgold that contains extremely thin multiple gold layers and an overlapping phenomenon of Zwischgold with other metal leaves are observed through PXCT. As supportive data, scanning electron microscopy (SEM) and scanning transmission electron microscopy (STEM) coupled with energy dispersive X-ray analysis (EDX) were performed on the medieval samples

    Mechanism and treatment for learning and memory deficits in mouse models of Noonan syndrome.

    In Noonan syndrome (NS) 30-50% of subjects show cognitive deficits of unknown etiology and with no known treatment. Here, we report that knock-in mice expressing either of two NS-associated mutations in Ptpn11, which encodes the nonreceptor protein tyrosine phosphatase Shp2, show hippocampal-dependent impairments in spatial learning and deficits in hippocampal long-term potentiation (LTP). In addition, viral overexpression of an NS-associated allele PTPN11(D61G) in adult mouse hippocampus results in increased baseline excitatory synaptic function and deficits in LTP and spatial learning, which can be reversed by a mitogen-activated protein kinase kinase (MEK) inhibitor. Furthermore, brief treatment with lovastatin reduces activation of the GTPase Ras-extracellular signal-related kinase (Erk) pathway in the brain and normalizes deficits in LTP and learning in adult Ptpn11(D61G/+) mice. Our results demonstrate that increased basal Erk activity and corresponding baseline increases in excitatory synaptic function are responsible for the LTP impairments and, consequently, the learning deficits in mouse models of NS. These data also suggest that lovastatin or MEK inhibitors may be useful for treating the cognitive deficits in NS
