    The effect of temperature and enzyme concentration in the transesterification process of synthetic microalgae oil

    Throughout the world, the fossil fuel has supplied around the 80% of the energetic requirements, in Colombia alone 95.1% of energetic demand is made by the transportation sector solely, supplied by oil, kerosene, gasoline and diesel, this sector has an extremely small participation with biofuel of 3%, which is represented only by biodiesel. Microalgae had been proposed as biofactories with a remarkable third generation biofuels production. The culture of the microorganism comprehends interesting characteristics as countless environments where its natural growth could be replicated in fresh, salty and even sewage waters, with a higher growth rate and a higher oil production. The implementation of enzymes in the transesterification process have generated a good curiosity in the field, due to its mild reactions conditions, lesser energetic requirements, a high standard in the selection of the enzymes with the objective of avoiding the formation of soaps, creating in this way cleaner products and sub-products, in which the separation of the phases biodiesel/glycerol, give the possibility to recuperate the bio catalyzer and high output of reactions. However, the high volume of medium required to obtain lipids is one of the major drawbacks to test the viability of these enzymes. The present study aims to design an enzymatic transesterification process for the production of biodiesel form synthetic Chlorella oil. The synthetic oil was designed according to the lipid profile of C 16:0, C16:1, C18:0, C18:1, C18:2 and C18:3 from Chlorella spp CHL2 cultured on Bold Basal media under limited concentrations of NaNO3. The enzymatic transesterification efficiency was evaluated by the implementation of a 22 experimental factorial design (temperature and lipase concentration) under a 3: 1 molar ratio of alcohol:oil and a fixed reaction time of 6 hours. The obtained results show that, in order to obtain superior yields of biodiesel (>91%) the transesterification process must be carried out under temperature conditions close to 38°C and lipase concentrations of 5%

    Multiobjective optimization of classifiers by means of 3-D convex Hull based evolutionary algorithms

    The receiver operating characteristic (ROC) and detection error tradeoff (DET) curves are frequently used in the machine learning community to analyze the performance of binary classifiers. Recently, the convex-hull-based multiobjective genetic programming algorithm was proposed and successfully applied to maximize the convex hull area for binary classification problems by minimizing false positive rate and maximizing true positive rate at the same time using indicator-based evolutionary algorithms. The area under the ROC curve was used for the performance assessment and to guide the search. Here we extend this research and propose two major advancements: Firstly we formulate the algorithm in detection error tradeoff space, minimizing false positives and false negatives, with the advantage that misclassification cost tradeoff can be assessed directly. Secondly, we add complexity as an objective function, which gives rise to a 3D objective space (as opposed to a 2D previous ROC space). A domain specific performance indicator for 3D Pareto front approximations, the volume above DET surface, is introduced, and used to guide the indicator -based evolutionary algorithm to find optimal approximation sets. We assess the performance of the new algorithm on designed theoretical problems with different geometries of Pareto fronts and DET surfaces, and two application-oriented benchmarks: (1) Designing spam filters with low numbers of false rejects, false accepts, and low computational cost using rule ensembles, and (2) finding sparse neural networks for binary classification of test data from the UCI machine learning benchmark.     Trinucleotide repeats in 202 families with ataxia: a small expanded (CAG)n allele at the SCA17 locus

    BACKGROUND: Ten neurodegenerative disorders characterized by spinocerebellar ataxia (SCA) are known to be caused by trinucleotide repeat (TNR) expansions. However, in some instances the molecular diagnosis is considered indeterminate because of the overlap between normal and affected allele ranges. In addition, the mechanism that generates expanded alleles is not completely understood. OBJECTIVE: To examine the clinical and molecular characteristics of a large group of Portuguese and Brazilian families with ataxia to improve knowledge of the molecular diagnosis of SCA. PATIENTS AND METHODS: We have (1) assessed repeat sizes at all known TNR loci implicated in SCA; (2) determined frequency distributions of normal alleles and expansions; and (3) looked at genotype-phenotype correlations in 202 unrelated Portuguese and Brazilian patients with SCA. Molecular analysis of TNR expansions was performed using polymerase chain reaction amplification. RESULTS: Patients from 110 unrelated families with SCA showed TNR expansions at 1 of the loci studied. Dominantly transmitted cases had (CAG)(n) expansions at the Machado-Joseph disease gene (MJD1) (63%), at SCA2 (3%), the gene for dentatorubropallidoluysian atrophy (DRPLA) (2%), SCA6 (1%), or SCA7 (1%) loci, or (CTG)(n) expansions at the SCA8 (2%) gene, whereas (GAA)(n) expansions in the Freidreich ataxia gene (FRDA) were found in 64% of families with recessive ataxia. Isolated patients also had TNR expansions at the MJD1 (6%), SCA8 (6%), or FRDA (8%) genes; in addition, an expanded allele at the TATA-binding protein gene (TBP), with 43 CAGs, was present in a patient with ataxia and mental deterioration. Associations between frequencies of SCA2 and SCA6 and a frequency of large normal alleles were found in Portuguese and Brazilian individuals, respectively. Interestingly, no association between the frequencies of DRPLA and large normal alleles was found in the Portuguese group. CONCLUSIONS: Our results show that (1) a significant number of isolated cases of ataxia are due to TNR expansions; (2) expanded DRPLA alleles in Portuguese families may have evolved from an ancestral haplotype; and (3) small (CAG)(n) expansions at the TBP gene may cause SCA17

    Assessment of pollution risk ascribed to Santa Margarida Military Camp activities (Portugal)

    Santa Margarida Military Camp (S.M.M.C.) is the only one Portuguese military training area, including firing ranges for tactical military manoeuvres of mechanised divisions. For this reason, various negative effects on the environment were expected due to the military activities, as the Military Camp’s area is classified as a high vulnerability area to pollution of its multilayer porous aquifers. The aim of this study was to identify and characterise local/regional geochemical impacts caused by the continuing military training activities performed at S.M.M.C. in the course of 52 years. An overview of the geochemical research issues as a basis for risk assessment is presented. A special attention has been put on the quality of local and regional surface waters, shallow groundwaters and groundwaters. Local soils and sediments as well as fragments of shells and bullets were sampled and analysed. The results so far obtained, indicated that none pollution effects were a consequence of the military training activities. Till now, the geochemical signatures such as, high levels of K, Cl and NO3 in waters, detected in particular sites, should be faced as tracers of diffuse pollution ascribed to urban waste disposal and cattle breading

    A whole genome screen for association with multiple sclerosis in portuguese patients

    Multiple sclerosis (MS) is common in Europe affecting up to 1:500 people. In an effort to identify genes influencing susceptibility to the disease, we have performed a population-based whole genome screen for association. In this study, 6000 microsatellite markers were typed in separately pooled DNA samples from MS patients (n = 188) and matched controls (n = 188). Interpretable data was obtained from 4661 of these markers.     Cervical cancer screening: target age bracket, screening frequency and screening method: review of recent evidence and comparison with the Portuguese performance indicator

    Esta revisão teve por objetivo avaliar a força de evidência do indicador de desempenho português relativo ao rastreio do Câncer do Colo do Útero (CCU): (1) limites etários das mulheres da população geral que o devem realizar, a (2) periodicidade com que deve ser realizado e (3) qual o melhor exame de rastreio. Foram pesquisados os seguintes termos MeSH: vaginal smears, age groups, periodicity, methods, uterine cervical cancer. Foram excluídos os artigos que não abordavam o objetivo da investigação ou que não fossem redigidos em Inglês, Português ou Espanhol. Para interpretar os artigos selecionados foi utilizada a classificação SORT. Foram encontrados 197 artigos, dos quais seleccionados 9: 1 revisão sistemática (RS), 1 estudo clínico controlado aleatorizado, 2 estudos observacionais retrospectivos e 5 normas de orientação clínica (NOC). Os autores optaram por incluir nesta revisão mais 4 NOCs e 2 RSs por considerarem ser relevantes para a população Portuguesa, apesar de não resultarem da pesquisa efectuada. Os estudos sugerem realização do rastreio entre os 21 e 25 até aos 65 anos, com uma periodicidade trienal usando a citologia convencional. Existe ainda controvérsia no que toca aos 3 objetivos deste artigo (limites etários, frequência e método).The scope of this review was to assess the strength of evidence of Portuguese performance indicators on Cervical Cancer screening: (1) age group of the women that should be screened for cervical cancer; (2) frequency of screening; and (3) the best method for screening. The following MeSH terms were searched: vaginal smears, age groups, periodicity, methods, uterine cervical cancer. Articles not reflecting the study objectives or not available in English, Portuguese or Spanish were excluded. The SORT classification was used to rate the articles selected.Of the 197 articles found, 9 that met all study criteria were selected for inclusion in this review. These included 1 systematic review, 1 randomized controlled clinical trial, 2 retrospective studies and 5 clinical guidelines. The authors also chose to include 4 clinical guidelines and two systematic reviews relevant to the Portuguese population even though they did not appear in the initial search of the literature. The studies suggest screening women between the ages of 21 to 25 years and 65 years of age, once every three years using conventional cytology. There is still controversy regarding the three objectives of this study (target age bracket, screening frequency and screening method)

    Buffering effects of soil seed banks on plant community composition in response to land use and climate

    Aim Climate and land use are key determinants of biodiversity, with past and ongoing changes posing serious threats to global ecosystems. Unlike most other organism groups, plant species can possess dormant life‐history stages such as soil seed banks, which may help plant communities to resist or at least postpone the detrimental impact of global changes. This study investigates the potential for soil seed banks to achieve this. Location Europe. Time period 1978–2014. Major taxa studied Flowering plants. Methods Using a space‐for‐time/warming approach, we study plant species richness and composition in the herb layer and the soil seed bank in 2,796 community plots from 54 datasets in managed grasslands, forests and intermediate, successional habitats across a climate gradient. Results Soil seed banks held more species than the herb layer, being compositionally similar across habitats. Species richness was lower in forests and successional habitats compared to grasslands, with annual temperature range more important than mean annual temperature for determining richness. Climate and land‐use effects were generally less pronounced when plant community richness included seed bank species richness, while there was no clear effect of land use and climate on compositional similarity between the seed bank and the herb layer. Main conclusions High seed bank diversity and compositional similarity between the herb layer and seed bank plant communities may provide a potentially important functional buffer against the impact of ongoing environmental changes on plant communities. This capacity could, however, be threatened by climate warming. Dormant life‐history stages can therefore be important sources of diversity in changing environments, potentially underpinning already observed time‐lags in plant community responses to global change. However, as soil seed banks themselves appear, albeit less, vulnerable to the same changes, their potential to buffer change can only be temporary, and major community shifts may still be expected
