Empirical Modeling of Used Nuclear Fuel Radiation Emissions for Safeguards Purposes
For nuclear nonproliferation safeguards, the ability to characterize used nuclear fuel (UNF) is a vital process. Fuel characterization allows for independent verification by inspectors of operator declarations of the special nuclear material flow and nuclear related activities within a facility, and an estimation of fissile material remaining in a fuel assembly. Current methods to verify this information rely heavily on non-destructive assay techniques, such as gamma spectroscopy and neutron detection measurements. While these measurements are effective tools for estimating a specific characteristic of the fuel, such as burnup or cooling time, they often require an accurate estimation of a select few isotopes in the fuel. This requirement means that the characterization is based on a very small amount of information that is contained in radiation emissions. To help overcome this limitation, this work investigates the use of empirical modeling to predict the burnup, initial enrichment, and cooling time of a Westinghouse 17x17 UNF assembly. This technique utilizes the entire spectrum of gamma emissions and gross neutron counts to predict each output to explore the full suite of information contained in these signatures. Three primary parametric modeling techniques are investigated for their performance in modeling this system: Ordinary Least Squares Regression (OLS), Principal Component Regression (PCR), and Partial Least Squares Regression (PLS). The models created are evaluated based on their root mean square percent error and condition number. The uncertainty of the best performing model is then quantified to understand the prediction interval of the predicted characterization and how this compares to the uncertainty of current measurement and characterization techniques. The PLS models are able to provide the best predictions while being stable. The PCR models have a consistent trade-off between accurate prediction results and stability. The OLS model provides fairly accurate results but is highly unstable due to correlations in the input data. The best model is the PLS model based on cross validation because it is stable, yields the lowest RMSPE values for burnup and enrichment predictions, and yields the second lowest percent of cooling time predictions that are more than 1 year away from the actual value. When used with the validation data set, this model yields RMSPE values of 0.42%, 1.39% and 4.61% for the burnup, enrichment, and cooling time, respectively. The total uncertainty of the predictions of this model are calculated to be 0.220 GWd/MTU, 0.051% U-235, and 0.694 years, for the burnup, enrichment, and cooling time, respectively
Modeling Temporal Trends in Aphid Vector Dispersal and Cucumber Mosaic Virus Epidemics in Snap Bean
Cucumber mosaic virus (CMV) has become a major limiting factor in snap bean production in the Great Lakes region of North America, and epidemics have occurred more frequently since the soybean aphid, Aphis glycines Matsumura, was introduced. Major aphid vectors of CMV epidemics were identified by statistically relating their temporal dispersal trends to the incidence of CMV. Alates were monitored weekly using water pan traps in 74 snap bean fields in New York and Pennsylvania from 2002 to 2006. Plants were tested for CMV by ELISA one time during late bloom in 2002 and 2003 and weekly over the season from 2004 to 2006. Principal vectors of CMV included Acyrthosiphon pisum (Harris), A. glycines, Aphis gossypii Glover, and Therioaphis trifolii (Monell). Among these, A. glycines and T. trifolii were likely responsible for severe CMV epidemics because they were among the most abundant species captured, they efficiently transmit CMV, and their dispersal activity was positively correlated with periods when CMV incidence was highest. Moreover, because high numbers of A. glycines and T. trifolii disperse during July and August, snap bean fields planted beyond late June are at risk for infection during early vegetative stages and are subsequently more at risk for yield loss. In contrast, plantings up to late June are less likely to become infected during early developmental stages and should escape yield loss because major vectors are dispersing infrequently. Overcoming ABCG2-mediated drug resistance with imidazo-[1,2-b]-pyridazine-based Pim1 kinase inhibitors
Purpose Multidrug efflux pumps such as ABCG2 confer drug resistance to a number of cancer types, leading to poor prognosis and outcome. To date, the strategy of directly inhibiting multidrug efflux pumps in order to overcome drug resistance in cancer has been unsuccessful. An alternative strategy is to target proteins involved in the regulation of multidrug efflux pump activity or expression. Pim1 kinase has been demonstrated to phosphorylate ABCG2, promote its oligomerisation and contribute to its ability to confer drug resistance. Methods In the present manuscript, imidazo-pyridazine-based inhibitors of Pim1 were examined for their ability to overcome ABCG2-mediated drug resistance. Drug efficacy was measured as a cytotoxic response or an effect on transport by ABCG2. Protein expression patterns were assessed using western immuno-blotting. Results The two Pim1 inhibitors increased the potency of flavopiridol, mitoxantrone, topotecan and doxorubicin, specifically in ABCG2-expressing cells. This effect was associated with an increase in the cellular accumulation of [3H]-mitoxantrone, suggesting direct impairment of the transporter. However, prolonged pre-incubation with the studied inhibitors greatly enhanced the effect on mitoxantrone accumulation. The inhibitors caused a significant time-dependent reduction in the expression of ABCG2 in the resistant cells, an effect that would improve drug efficacy. Conclusion Consequently, it appears that the Pim1 inhibitors display a dual-mode effect on ABCG2-expressing cancer cells. This may provide a powerful new strategy in overcoming drug resistance by targeting proteins that regulate expression of efflux pumps

WHO global research priorities for antimicrobial resistance in human health
The WHO research agenda for antimicrobial resistance (AMR) in human health has identified 40 research priorities to be addressed by the year 2030. These priorities focus on bacterial and fungal pathogens of crucial importance in addressing AMR, including drug-resistant pathogens causing tuberculosis. These research priorities encompass the entire people-centred journey, covering prevention, diagnosis, and treatment of antimicrobial-resistant infections, in addition to addressing the overarching knowledge gaps in AMR epidemiology, burden and drivers, policies and regulations, and awareness and education. The research priorities were identified through a multistage process, starting with a comprehensive scoping review of knowledge gaps, with expert inputs gathered through a survey and open call. The priority setting involved a rigorous modified Child Health and Nutrition Research Initiative approach, ensuring global representation and applicability of the findings. The ultimate goal of this research agenda is to encourage research and investment in the generation of evidence to better understand AMR dynamics and facilitate policy translation for reducing the burden and consequences of AMR
Enrichment dynamics for advanced reactor HALEU support
Transitioning to High Assay Low Enriched Uranium-fueled reactors will alter the material requirements of the current nuclear fuel cycle, in terms of the mass of enriched uranium and Separative Work Unit capacity. This work simulates multiple fuel cycle scenarios using Cyclus to compare how the type of the advanced reactor deployed and the energy growth demand affect the material requirements of the transition to High Assay Low Enriched Uranium-fueled reactors. Fuel cycle scenarios considered include the current fleet of Light Water Reactors in the U.S. as well as a no-growth and a 1% growth transition to either the Ultra Safe Nuclear Corporation Micro Modular Reactor or the X-energy Xe-100 reactor from the current fleet of U.S. Light Water Reactors. This work explored parameters of interest including the number of advanced reactors deployed, the mass of enriched uranium sent to the reactors, and the Separative Work Unit capacity required to enrich natural uranium for the reactors. Deploying Micro Modular Reactors requires a higher average mass and Separative Work Unit capacity than deploying Xe-100 reactors, and a lower enriched uranium mass and a higher Separative Work Unity capacity than required to fuel Light Water Reactors before the transition. Fueling Xe-100 reactors requires less enriched uranium and Separative Work Unit capacity than fueling Light Water Reactors before the transition
Association between emergency department attendances, sociodemographic factors and long-term health conditions in the population of Norfolk and Waveney, England: Cross sectional study
IntroductionDemand for urgent and emergency health care in England has grown over the last decade, for reasons that are not clear. Changes in population demographics may be a cause. This study investigated associations between individualsâ characteristics (including socioeconomic deprivation and long-term health conditions (LTC)) and the frequency of emergency department (ED) attendances in the Norfolk and Waveney subregions of the East of England.MethodsThe study population was people who were registered with 91 of 106 Norfolk and Waveney general practices during one year, from April 1, 2022, to March 31, 2023. Linked primary and secondary care and geographical data included each individualâs sociodemographic characteristics, number of ED attendances during the same year, and, for some individuals, LTCs and number of general practice (GP) appointments. Associations between these factors and ED attendance were estimated using Poisson regression models.Results1,027,422 individuals were included, of whom 57.4% had GP data on the presence or absence of LTC, and 43.1% had both LTC and general practitioner appointment data. In the total population, ED attendances were more frequent in individuals aged under five years (adjusted incidence rate ratio (IRR) 1.25, 95% confidence interval 1.23 to 1.28) compared to 15â35 years; living in more socioeconomically deprived areas (IRR 0.61 (0.60 to 0.63) for the least deprived compared to the most deprived; and living closer to the nearest ED. Among individuals with LTC data, each additional LTC was also associated with increased ED attendances (IRR 1.16 (1.15 to 1.16)). Among individuals with LTC and GP appointment data, each additional GP appointment was also associated with increased ED attendances (IRR 1.03 (1.026 to 1.027)).ConclusionsIn the Norfolk and Waveney population, ED attendance rates were higher for young children and individuals living in more deprived areas and closer to EDs. In individuals with LTC and GP appointment data, both factors were also associated with higher ED attendance
