    A unified constitutive model for asymmetric tension and compression creep-ageing behaviour of naturally aged Al-Cu-Li alloy

    A set of unified constitutive equations is presented that predict the asymmetric tension and compression creep behaviour and recently observed double primary creep of pre-stretched/naturally aged aluminium-cooper-lithium alloy AA2050-T34. The evolution of the primary micro- and macro-variables related to the precipitation hardening and creep deformation of the alloy during creep age forming (CAF) are analysed and modelled. Equations for the yield strength evolution of the alloy, including an initial reversion and subsequent strengthening, are proposed based on a theory of concurrent dissolution, re-nucleation and growth of precipitates during artificial ageing. We present new observations of so-called double primary creep during the CAF process. This phenomenon is then predicted by introducing effects of interacting microstructures, including evolving precipitates, diffusing solutes and dislocations, into the sinh-law creep model. In addition, concepts of threshold creep stress σth and a microstructure-dependant creep variable H, which behave differently under different external stress directions, are proposed and incorporated into the creep model. This enables prediction of the asymmetric tension and compression creep-ageing behaviour of the alloy. Quantitative transmission electron microscopy (TEM) and related small-angle X-ray scattering (SAXS) analysis have been carried out for selected creep-aged samples to assist the development and calibration of the constitutive model. A good agreement has been achieved between the experimental results and the model. The model has the potential to be applied to creep age forming of other heat-treatable aluminium alloys

    Robotic surgery for gynecologic cancers: indications, techniques and controversies.

    Minimally invasive surgery for gynecologic cancers is associated with fewer postoperative complications including less blood loss and quicker recovery time compared to traditional laparotomy. The robotic platform has allowed patients access to minimally invasive surgery due to its increased utilization by gynecologic oncologists. Many surgeons have embraced the robotic platform due to its technological advances over traditional laparoscopy including high-definition 3D optics, wristed instrumentation, camera stability and improved ergonomics. While robotic surgery continues as a mainstay in the management of gynecologic cancers, it remains controversial in regards to its cost effectiveness and more recently, its long-term impact on clinical and oncologic outcomes. A strong component of the justification of this surgical platform is based on extrapolated data from traditional laparoscopy despite limited prospective randomized trials for robotic-assisted surgery. In this review, we highlight the use of robotic surgery in the management of gynecologic cancers in special populations: fertility sparing patients, the morbidly obese, the elderly, and patients with a favorable response to neoadjuvant chemotherapy

    Response to arXiv:0811.3876 "Comment on a recent conjectured solution of the three dimensional Ising model" by Wu et al

    This is a Response to a recent Comment [F.Y. Wu et al., Phil. Mag. 88, 3093 (2008), arXiv:0811.3876] on the conjectured solution of the three-dimensional (3D) Ising model [Z.D. Zhang, Phil. Mag. 87, 5309 (2007), arXiv:0705.1045]. Several points are made: 1) Conjecture 1, regarding the additional rotation, is understood as performing a transformation for smoothing all the crossings of the knots; 2) The weight factors in Conjecture 2 are interpreted as a novel topologic phase; 3) The conjectured solution and its low- and high-temperature expansions are supported by the mathematical theorems for the analytical behavior of the Ising model.     Semiparametric Multivariate Accelerated Failure Time Model with Generalized Estimating Equations

    The semiparametric accelerated failure time model is not as widely used as the Cox relative risk model mainly due to computational difficulties. Recent developments in least squares estimation and induced smoothing estimating equations provide promising tools to make the accelerate failure time models more attractive in practice. For semiparametric multivariate accelerated failure time models, we propose a generalized estimating equation approach to account for the multivariate dependence through working correlation structures. The marginal error distributions can be either identical as in sequential event settings or different as in parallel event settings. Some regression coefficients can be shared across margins as needed. The initial estimator is a rank-based estimator with Gehan's weight, but obtained from an induced smoothing approach with computation ease. The resulting estimator is consistent and asymptotically normal, with a variance estimated through a multiplier resampling method. In a simulation study, our estimator was up to three times as efficient as the initial estimator, especially with stronger multivariate dependence and heavier censoring percentage. Two real examples demonstrate the utility of the proposed method

    Definition of the σW regulon of Bacillus subtilis in the absence of stress

    Bacteria employ extracytoplasmic function (ECF) sigma factors for their responses to environmental stresses. Despite intensive research, the molecular dissection of ECF sigma factor regulons has remained a major challenge due to overlaps in the ECF sigma factor-regulated genes and the stimuli that activate the different ECF sigma factors. Here we have employed tiling arrays to single out the ECF σW regulon of the Gram-positive bacterium Bacillus subtilis from the overlapping ECF σX, σY, and σM regulons. For this purpose, we profiled the transcriptome of a B. subtilis sigW mutant under non-stress conditions to select candidate genes that are strictly σW-regulated. Under these conditions, σW exhibits a basal level of activity. Subsequently, we verified the σW-dependency of candidate genes by comparing their transcript profiles to transcriptome data obtained with the parental B. subtilis strain 168 grown under 104 different conditions, including relevant stress conditions, such as salt shock. In addition, we investigated the transcriptomes of rasP or prsW mutant strains that lack the proteases involved in the degradation of the σW anti-sigma factor RsiW and subsequent activation of the σW-regulon. Taken together, our studies identify 89 genes as being strictly σW-regulated, including several genes for non-coding RNAs. The effects of rasP or prsW mutations on the expression of σW-dependent genes were relatively mild, which implies that σW-dependent transcription under non-stress conditions is not strictly related to RasP and PrsW. Lastly, we show that the pleiotropic phenotype of rasP mutant cells, which have defects in competence development, protein secretion and membrane protein production, is not mirrored in the transcript profile of these cells. This implies that RasP is not only important for transcriptional regulation via σW, but that this membrane protease also exerts other important post-transcriptional regulatory functions

    Burden of upper gastrointestinal symptoms in patients prescribed dabigatran for stroke prevention

    BACKGROUND: Dabigatran, a non-vitamin K antagonist oral anticoagulant, has been shown to prevent stroke in patients with non-valvular atrial fibrillation. Nonetheless, studies show that 10%-30% of those prescribed dabigatran experience dyspepsia that may eventually lead to discontinuation of therapy and loss of clinical benefit. AIM: To evaluate the gastrointestinal tolerability of dabigatran utilizing a validated questionnaire, as well as determining subsequent non-compliance and drug discontinuation. METHOD: This is an observational study. All patients were assessed by a validated questionnaire, Hong Kong dyspepsia index, prior to drug prescription and again 4 weeks later. RESULTS: In this study, 115 patients with non-valvular atrial fibrillation (mean age: 74.6 ± 11.4 years; mean CHA2DS2-VASc score was 3.39 ± 1.59) were prescribed dabigatran. At baseline, the mean Hong Kong dyspepsia index was 12.9 ± 1.6 and nine patients had significant dyspepsia (Hong Kong dyspepsia index ⩾ 16). After 4 weeks, the mean Hong Kong dyspepsia index was similar at 12.6 ± 1.9 (p = 0.23). There was no change in Hong Kong dyspepsia index after initiation of dabigatran in 59 (51.3%) patients, and improvement in 37 (32.2%). Only 19 (16.5%) patients had worsening of Hong Kong dyspepsia index, and among these 19 patients, only 1 patient (0.9%) discontinued dabigatran due to significant dyspepsia. CONCLUSION: Worsening of dyspepsia with dabigatran 110 mg twice daily was uncommon with correct drug administration and clear instructions provided. Systematic assessment of dyspeptic symptoms using a validated questionnaire (i.e. Hong Kong dyspepsia index) before and after treatment initiation allows a more objective comparison of dyspeptic symptoms

    China’s rising hydropower demand challenges water sector

    Demand for hydropower is increasing, yet the water footprints (WFs) of reservoirs and hydropower, and their contributions to water scarcity, are poorly understood. Here, we calculate reservoir WFs (freshwater that evaporates from reservoirs) and hydropower WFs (the WF of hydroelectricity) in China based on data from 875 representative reservoirs (209 with power plants). In 2010, the reservoir WF totaled 27.9 × 109 m3 (Gm3), or 22% of China’s total water consumption. Ignoring the reservoir WF seriously underestimates human water appropriation. The reservoir WF associated with industrial, domestic and agricultural WFs caused water scarcity in 6 of the 10 major Chinese river basins from 2 to 12 months annually. The hydropower WF was 6.6 Gm3 yr−1 or 3.6 m3 of water to produce a GJ (109 J) of electricity. Hydropower is a water intensive energy carrier. As a response to global climate change, the Chinese government has promoted a further increase in hydropower energy by 70% by 2020 compared to 2012. This energy policy imposes pressure on available freshwater resources and increases water scarcity. The water-energy nexus requires strategic and coordinated implementations of hydropower development among geographical regions, as well as trade-off analysis between rising energy demand and water use sustainability

    Probe set algorithms: is there a rational best bet?

    Affymetrix microarrays have become a standard experimental platform for studies of mRNA expression profiling. Their success is due, in part, to the multiple oligonucleotide features (probes) against each transcript (probe set). This multiple testing allows for more robust background assessments and gene expression measures, and has permitted the development of many computational methods to translate image data into a single normalized "signal" for mRNA transcript abundance. There are now many probe set algorithms that have been developed, with a gradual movement away from chip-by-chip methods (MAS5), to project-based model-fitting methods (dCHIP, RMA, others). Data interpretation is often profoundly changed by choice of algorithm, with disoriented biologists questioning what the "accurate" interpretation of their experiment is. Here, we summarize the debate concerning probe set algorithms. We provide examples of how changes in mismatch weight, normalizations, and construction of expression ratios each dramatically change data interpretation. All interpretations can be considered as computationally appropriate, but with varying biological credibility. We also illustrate the performance of two new hybrid algorithms (PLIER, GC-RMA) relative to more traditional algorithms (dCHIP, MAS5, Probe Profiler PCA, RMA) using an interactive power analysis tool. PLIER appears superior to other algorithms in avoiding false positives with poorly performing probe sets. Based on our interpretation of the literature, and examples presented here, we suggest that the variability in performance of probe set algorithms is more dependent upon assumptions regarding "background", than on calculations of "signal". We argue that "background" is an enormously complex variable that can only be vaguely quantified, and thus the "best" probe set algorithm will vary from project to project

    Quasi-static stop band with flexural metamaterial having zero rotational stiffness

    Metamaterials realizing stop bands have attracted much attentions recently since they can break-through the well-known mass law. However, achieving the stop band at extremely low frequency has been still a big challenge in the fields of elastic metamaterials. In this paper, we propose a new metamaterial based on the idea of the zero rotational stiffness, to achieve extremely low frequency stop band for flexural elastic waves. Unlike the previous ways to achieve the stop band, we found that the zero rotational stiffness can provide a broad stop band at extremely low frequency, which starts from even almost zero frequency. To achieve the zero rotational stiffness, we propose a new elastic metamaterial consisting of blocks and links with the hinge connection. Analytic developments as well as numerical simulations evidence that this new metamaterial can exhibit extremely low and broad stop band, even at the quasi-static ranges. In addition, the metamaterial is shown to exhibit the negative group velocity at extremely low frequency ranges, as well as the quasi-static stop band, if it is properly designed.ope
