    Improving the Process Ability of Poly (3-Hydroxybutyrate) by a Polyphenolic Natural Additive

    The severe environmental issues generated by the non-biodegradability of most polymeric materials has fostered the search for more eco-friendly solutions. Polyhydroxybutyrate (PHB), produced by bacterial cells and easily hydrolyzed is one of appealing choice but its properties are still not competitive with those of standard polymers. Among others, one reason is related to the fact that the high processing temperature (due to the high crystallization point) is responsible for severe thermal degradation. In this paper, the effect of Tannic Acid (TA) as a thermal stabilizing processing agent for HPB is investigated by rheological as well as calorimetric techniques under conditions similar to those used in real processing applications. The results show that TA is effective in improving the thermal stability and processability of PHB, while keeping its biodegradable attitude. Compared to neat PHB, TA containing samples show a lower and delayed thermal degradation. FTIR measurements support the hypothesis that stabilization is determined by TA/PHB crosslinking

    Characterization of blood redox status of early and mid-late lactating dairy cows

    The effect of the stage of lactation on blood redox homeostasis of bovine and buffalo cows was evaluated. The investigation was carried out on early lactating and mid-late lactating cows, reared in a farm located in Campania (southern Italy). Plasma concentration of α-tocopherol and ascorbate, the total antioxidant capacity (TAC), glutathione peroxidase (GPx), and superoxide dismutase activities were higher (P < 0.01) in mid-late lactating cows, thus suggesting a higher consumption of antioxidants during early lactation. Plasma concentration of protein-bound carbonyls (PC) and nitrotyrosine (N-Tyr), and the level of lipid hydroperoxides (LPO) were higher (P < 0.01) in early lactating cows, thus suggesting that lipid peroxidation and peroxynitrite production are crucial in determining oxidative modifications in plasma. TAC was positively correlated with ascorbate concentration (P < 0.03), and negatively correlated with PC concentration (P < 0.002), and ascorbate was negatively correlated with PC (P < 0.03) in mid-late lactating group. These findings demonstrate that circulating ascorbate plays a major role in preventing protein modifications induced by carbonyls, and that ascorbate scavenging effect is impaired during early lactation. We calculated a protein oxidative stress index as the ratio (PC + N-Tyr)/TAC multiplied by 100, and we found that this parameter was higher (P < 0.0001) in early lactating cows. Therefore, it could be useful for assessing the extent of protein oxidative damage in relation to the whole antioxidant status. Further, we suggest that the LPO/GPx ratio multiplied by 100 might be used as lipid oxidative stress index in lactating cows. This index was higher (P < 0.0001) in early lactating cows, and might represent a standard parameter for evaluating the lipid damage depending on a deficiency of the enzymatic antioxidant defence. These parameters are proposed for a possible effective description of physiological changes associated with lactation

    The 2020 underwater archaeological research in the inlet of Torre Santa Sabina - Baia dei Camerini (Municipality of Carovigno, Brindisi, Italy ) represented the first phase of the pilot intervention of the Interreg Italia-Croatia UnderwaterMuse project, which aims to enhance and make accessible the huge underwater heritage of the areas involved through the creation of submerged archaeological parks and the narrative and communicative use of the virtual reality. The Puglia Region - Department of Tourism, Economics of Culture and Valorization of the Territory, partner of the UnderwaterMuse project, launching these researches aimed at valorisation, has involved the three regional Universities thanks to an agreement. The fruitful synergy between the various actors involved and the support of the territory and the community have allowed the achievement of the objectives of this campaign, preliminary to the broader and more articulated intervention foreseen for the next year. Thanks to the collaboration with the Polytechnic of Turin, the entire stretch of coast was mapped with drone flights, in order to reconstruct the coastal landscape in the various phases, starting from the Bronze Age. Targeted interventions were carried out on the Roman wreck of the imperial age, beached and abandoned at the ancient shore and now submerged due to the relative rise in sea level, and on the remains of on-board equipment of a ship of the Serenissima, the Galea Magna (1598). Another important focus was represented by the stratigraphy of cargos resulting from the various shipwreck episodes, accumulated at the foot of the western cliff

    [Italiano]:Il volume presenta contributi scientifici sul tema della menzogna nelle sue più diversificate ‘manifestazioni’ testuali e declinazioni ‘comunicative’, potendo essere intesa come immaginazione, invenzione, finzione, artificio, dissimulazione, deformazione ma anche come manipolazione.Il volume si articola in tre macrosezioni: letteraria, filologica-linguistica e artistica. La sezione letteraria contiene una riflessione ramificata che offre uno sguardo multiprospettico. La menzogna è analizzata dapprima come tema per poi tramutarsi in oggetto di studio della critica letteraria, giungendo, infine, in un mutevole paesaggio geoculturale. La seconda sezione, quella filologica-linguistica, segue invece un principio diacronico: si indaga la menzogna a partire dalla tradizione manoscritta fino alla comunicazione digitale. La terza e ultima sezione, che conferma la natura interdisciplinare e transmediale del volume, è dedicata alle strategie di rappresentazione e dissimulazione in rapporto alle diverse forme d’arte, quali il teatro, le serie TV, la docufiction, nonché le arti performative. Il volume si chiude con un’appendice che contiene le riproduzioni delle opere pittoriche di alcuni artisti, presentate in occasione della mostra che ha affiancato la Graduate Conference ./[English]: The volume presents a collection of papers focussing on the topic of “lies” in its different textual ‘manifestations’ and ‘communicative’ declinations, and can be understood in terms of imagination, invention, artifice, dissimulation, disfiguration and manipulation. The volume is divided into three macro-sections: literature, philology and linguistics, and the arts. The literary section contains a ramification of reflections, which provides a multi-perspective view. “Lies” are analyzed as a topic and become an object of literary criticism, entering into a mutable geo-cultural landscape. The second section of philology and linguistics adopts, instead, a diachronic approach: investigations on “lies” range from the manuscript tradition to digital communication. The third and last section, which confirms the interdisciplinary and transmedial nature of this volume, is dedicated to strategies of representation and dissimulation in relation to the different forms of art, including theatre, television series, docufiction and other performing arts.     Clinical, Pathological and Prognostic Features of Rare BRAF Mutations in Metastatic Colorectal Cancer (mCRC): A Bi-Institutional Retrospective Analysis (REBUS Study)

    Simple SummarySomatic BRAF mutations occur in approximately 10% of metastatic colorectal cancers (mCRCs) and, according to the involved codon, are classified as V600E and in non-V600, accounting for 80% and 20%, respectively. Being the most frequent mutation, the BRAF V600E mutation has been extensively investigated and up to now its clinical, pathological and molecular phenotype and its prognostic impact have been clearly described. On the contrary, evidence concerning BRAF non-V600 is weaker. We retrospectively evaluated 537 mCRC patients treated at two Italian Institutions. This study corroborates and strengthens available evidence concerning phenotype and prognostic performance of BRAF non-V600 compared to BRAF V600E and BRAF wild-type mCRCs. This deeper insight on rare BRAF non-V600 mutated mCRC is a primary issue in the precision oncology era, since the wider application of NGS is expected to increase the identification of those aberrations.Recently, retrospective analysis began to shed light on metastatic colorectal cancers (mCRCs) harboring rare BRAF non-V600 mutations, documenting a distinct phenotype and a favorable prognosis. This study aimed to confirm features and prognosis of rare BRAF non-V600 mCRCs compared to BRAF V600E and BRAF wild-type mCRCs treated at two Italian Institutions. Overall, 537 cases were retrospectively evaluated: 221 RAS/BRAF wild-type, 261 RAS mutated, 46 BRAF V600E and 9 BRAF non-V600. Compared to BRAF V600E mCRC, BRAF non-V600 mCRC were more frequently left-sided, had a lower tumor burden and displayed a lower grade and an MMR proficient/MSS status. In addition, non-V600 mCRC patients underwent more frequently to resection of metastases with radical intent. Median overall survival (mOS) was significantly longer in the non-V600 compared to the V600E group. At multivariate analysis, only age &lt; 65 years and ECOG PS 0 were identified as independent predictors of better OS. BRAF V600E mCRCs showed a statistically significant worse mOS when compared to BRAF wild-type mCRCs, whereas no significant difference was observed between BRAF non-V600 and BRAF wild-type mCRCs. Our study corroborates available evidence concerning incidence, clinicopathologic characteristics and prognosis of BRAF-mutated mCRCs

    The dual targeting of insulin and insulin-like growth factor 1 receptor enhances the mTOR inhibitor-mediated antitumor efficacy in hepatocellular carcinoma

    Deregulation of mTOR and IGF pathways is frequent in hepatocellular carcinoma (HCC), thus mTOR and IGF1R represent suitable therapeutic targets in HCC. The aim of this study was to evaluate the effects of mTOR inhibitors (mTORi) and OSI-906, blocker of IGF1R/IR, on HCC cell proliferation, viability, migration and invasion, and alpha-fetoprotein (α-FP) secretion. In HepG2 and HuH-7 we evaluated, the expression of mTOR and IGF pathway components; the effects of Sirolimus, Everolimus, Temsirolimus and OSI-906 on cell proliferation; the effects of Sirolimus, OSI-906, and their combination, on cell secretion, proliferation, viability, cell cycle, apoptosis, invasion and migration. Moreover, intracellular mechanisms underlying these cell functions were evaluated in both cell lines. Our results show that HepG2 and HuH-7 present with the same mRNA expression profile with high levels of IGF2. OSI-906 inhibited cell proliferation at high concentration, while mTORi suppressed cell proliferation in a dose-time dependent manner in both cell lines. The co-treatment showed an additive inhibitory effect on cell proliferation and viability. This effect was not related to induction of apoptosis, but to G0/G1 phase block. Moreover, the co-treatment prevented the Sirolimus-induced AKT activation as escape mechanism. Both agents demonstrated to be differently effective in inhibiting α-FP secretion. Sirolimus, OSI-906, and their combination, blocked cell migration and invasion in HuH-7. These findings indicate that, co-targeting of IGF1R/IR and mTOR pathways could be a novel therapeutic approach in the management of HCC, in order to maximize antitumoral effect and to prevent the early development of resistance mechanisms

    New hadrons as ultra-high energy cosmic rays

    Ultra-high energy cosmic ray (UHECR) protons produced by uniformly distributed astrophysical sources contradict the energy spectrum measured by both the AGASA and HiRes experiments, assuming the small scale clustering of UHECR observed by AGASA is caused by point-like sources. In that case, the small number of sources leads to a sharp exponential cutoff at the energy E<10^{20} eV in the UHECR spectrum. New hadrons with mass 1.5-3 GeV can solve this cutoff problem. For the first time we discuss the production of such hadrons in proton collisions with infrared/optical photons in astrophysical sources. This production mechanism, in contrast to proton-proton collisions, requires the acceleration of protons only to energies E<10^{21} eV. The diffuse gamma-ray and neutrino fluxes in this model obey all existing experimental limits. We predict large UHE neutrino fluxes well above the sensitivity of the next generation of high-energy neutrino experiments. As an example we study hadrons containing a light bottom squark. These models can be tested by accelerator experiments, UHECR observatories and neutrino telescopes.Comment: 17 pages, revtex style; v2: shortened, as to appear in PR

    A great deal of evidences indicate that impaired fetal growth and in utero exposure to risk factors, especially maternal hypercholesterolemia, may be relevant for human pathophysiological signs of atherosclerosis and subsequent development of cardiovascular disease (CVD) during different life stages. Despite the underlying mechanisms of fetal programming are still unknown, epigenetics has been suggested as one of the possible explanations for the associations between intrauterine risk factors and CVD development. Indeed, a lot of translational studies support the hypothesis that epigenetic changes are related to increased CVD risk although it is still not possible to establish a direct causality in humans. Notably, epigenetic modifications can be reversible through therapeutic approaches employing histone deacetylase inhibitors, histone acetyltransferase inhibitors and commonly used drugs like statins. Thus, the whole comprehension of these mechanisms will provide in the next future the rationale for the development of novel tools to be used in the primary prevention and therapy of CVD

    Quantum numbers of the X(3872)X(3872) state and orbital angular momentum in its ρ0Jψ\rho^0 J\psi decay

    Angular correlations in B+X(3872)K+B^+\to X(3872) K^+ decays, with X(3872)ρ0J/ψX(3872)\to \rho^0 J/\psi, ρ0π+π\rho^0\to\pi^+\pi^- and J/ψμ+μJ/\psi \to\mu^+\mu^-, are used to measure orbital angular momentum contributions and to determine the JPCJ^{PC} value of the X(3872)X(3872) meson. The data correspond to an integrated luminosity of 3.0 fb1^{-1} of proton-proton collisions collected with the LHCb detector. This determination, for the first time performed without assuming a value for the orbital angular momentum, confirms the quantum numbers to be JPC=1++J^{PC}=1^{++}. The X(3872)X(3872) is found to decay predominantly through S wave and an upper limit of 4%4\% at 95%95\% C.L. is set on the fraction of D wave.Comment: 16 pages, 4 figure
