    Exploring the role of the WNT5A rs566926 polymorphism and its interactions in non-syndromic orofacial cleft: a multicenter study in Brazil

    Associations between the WNT5A rs566926 variant and non-syndromic orofacial cleft (NSOC) have been reported in different populations. Objective:This study aimed to investigate the role of the rs566926 single nucleotide polymorphism (SNP) in WNT5A and its interactions with SNPs in BMP4, FGFR1, GREM1, MMP2, and WNT3 in the occurrence of NSOC in a Brazilian population. Methodology: A case-control genetic association study was carried out involving participants from four regions of Brazil, totaling 801 patients with non-syndromic cleft lip with or without cleft palate (NSCL±P), 273 patients with cleft palate only (NSCPO), and 881 health volunteers without any congenital condition (control). Applying TaqMan allelic discrimination assays, we evaluated WNT5A rs566926 in an ancestry-structured multiple logistic regression analysis, considering sex and genomic ancestry as covariates. Interactions between rs566926 and variants in genes involved in the WNT5A signaling pathway (BMP4, FGFR1, GREM1, MMP2, and WNT3) were also explored. Results: WNT5A rs566926 was significantly associated with an increased risk of NSCL±P, particularly due to a strong association with non-syndromic cleft lip only (NSCLO), in which the C allele increased the risk by 32% (OR: 1.32, 95% CI: 1.04–1.67, p=0.01). According to the proportions of European and African genomic ancestry, the association of rs566926 reached significant levels only in patients with European ancestry. Multiple interactions were detected between WNT5A rs566926 and BMP4 rs2071047, GREM1 rs16969681 and rs16969862, and FGFR1 rs7829058. Conclusion: The WNT5A rs566926 polymorphism was associated with NSCL±P, particularly in individuals with NSCLO and high European ancestry. Epistatic interactions involving WNT5A rs566926 and variants in BMP4, GREM1, and FGFR1 may contribute to the risk of NSCL±P in the Brazilian population

    Dinâmica da matéria orgânica de um neossolo quartzarênico de cerrado convertido para cultivo em sucessão de soja e milheto

    O uso e o manejo de Neossolos Quartzarênicos podem influenciar a matéria orgânica do solo (MOS), mas a magnitude dessas alterações ainda é pouco pesquisada nas condições de Cerrado. O objetivo deste trabalho foi estudar as variações que ocorrem na MOS em um Neossolo Quartzarênico, por consequência da conversão da vegetação original para o uso agrícola e cultivo sequencial de soja e milheto em sucessão. Em uma área de cultivo comercial em Alto Garças, MT, foram coletadas amostras de solo nas profundidades de 0-5, 0-10, 0-20 e 0-40 cm, em talhões que estavam sob sucessão soja-milheto durante 1, 2, 6, 8 e 10 anos e, como referência da vegetação original, amostras foram tomadas em área adjacente sob Cerrado nativo. Foram determinados os teores de carbono orgânico total (COT), carbono e nitrogênio total e o teor de carbono nas frações ácidos húmicos, ácidos fúlvicos e na matéria orgânica leve (MOL), além da relação isotópica do carbono na MOS. A conversão do Cerrado para cultivo agrícola elevou o teor de COT do solo, sobretudo em razão do aporte de MOL e fragmentos de carvão. O manejo desse solo com a sucessão soja-milheto, mesmo com o uso de gradagens pesadas, não causou reduções nos teores de COT, ao longo do tempo. Contudo, observou-se variação na distribuição do carbono nas diferentes frações da MOS ao longo do tempo de cultivo, indicando alterações significativas na qualidade da MOS. Notou-se aumento no percentual de carbono na forma de MOL e redução dos teores de carbono na forma de ácidos fúlvicos ao longo do tempo de cultivo, sugerindo a perda seletiva dos compostos mais solúveis. A relação isotópica do carbono do solo aumentou com o tempo de cultivo, indicando maior contribuição do carbono derivado do milheto para a manutenção dos teores de MOS. Os atributos da MOS relativos à camada de 0-20 cm foram mais sensíveis às variações em razão do tempo de cultivo do que os mesmos atributos avaliados nas demais camadas estudadas. As variáveis relacionadas ao fracionamento da MOS evidenciaram-se mais sensíveis às variações por causa do tempo de cultivo do que simplesmente do teor de COT do solo, apresentando o potencial de uso dessas variáveis como indicadores de qualidade do solo.The use and management of Quartzipsamments can influence soil organic matter (SOM), but the magnitude of these variations for the specific conditions in the Cerrado (savanna-like vegetation) is still poorly studied. The aim of this work was to study the variations in SOM in a Quartzipsamment, after the conversion of native vegetation to cropland (soybean-millet rotation). Soil was sampled from 0-5, 0-10, 0-20 and 0-40 cm depths of a commercial field in Alto Garças, Mato Grosso, Brazil, of plots under soybean- millet rotation in plots converted to croplands for 1, 2, 6, 8, and 10 years. Additionally, soil samples were taken from an adjacent area under native Cerrado (savanna) as a reference of the original vegetation. Total organic carbon (TOC), total C, total N, C content in humic and fulvic acids, light organic matter (LOM), and the SOM carbon isotope ratio were determined. The conversion of Cerrado to agriculture increased the soil TOC content, mainly due to the contribution of LOM and charcoal fragments. The management of this soil with soybean- millet rotation, despite the use of heavy disking, did not reduce the TOC levels over time. However a variation in the C distribution in the different SOM fractions was observed over time, indicating significant changes in the SOM quality. Over time, an increase was observed in light organic matter and a reduction in C contents in the form of fulvic acids, suggesting a selective loss of the more soluble C-compounds. The isotope ratio of soil C increased with time, indicating a higher contribution of C derived from millet, maintaining the SOM levels. The SOM properties in the 0-20 cm layer were more sensitive to variations as a function of cultivation time than the same properties in the other layers. Soil organic matter fractions were more sensitive to variations in function of cultivation time than the soil TOC content, showing the potential use of these variables as soil quality indicators

    Desempenho de plantas de cobertura em sobressemeadura na cultura da soja

    The objective of this work was to evaluate the establishing capacity of cover crops, oversown onto soybean. The soybean was planted at different seasons, so that in the R7 stage (physiologic maturation) different climatic conditions would be present for cover crop implantation and growth. The experiment was carried out in Rio Verde, GO, Brazil, in the summer crop season 2005/2006, on a Rhodic Ferralsol. Six cover crops were evaluated: Brachiaria brizantha, B. ruziziensis, B. decumbens, Eleusine coracana, Pennisetum glaucum, and the hybrid Cober Crop [Sorghum bicolor (L.) Moench x Sorghum sudanense Piper Stapf]. Fallow treatment was used as control. Soybean was planted at four sowing seasons: 10/27/2005, 11/10/2005, 11/24/2005 and 12/14/2005. Oversowing was carried out manually, by broadcasting at the R7 stage of soybean. Except for the second sowing season, in which weed competition reduced the mean stand of cover crops, all the remaining sowing seasons of soybean resulted in satisfactory initial cover crop growth, as given by: emergence, crop height, soil cover, and biomass accumulation. Oversowing with B. ruziziensis, B. brizantha, B. decumbens and the hybrid Cober Crop show higher potential to produce mulch, during the off-season in Cerrado.O objetivo deste trabalho foi avaliar a capacidade de estabelecimento de plantas de cobertura em sobressemeadura na cultura soja.     Nutrient cycling by cover crops and yield of soybean and rice in no-tillage

    O objetivo deste trabalho foi avaliar a ciclagem de nutrientes por plantas de cobertura e a sua influência sobre o desempenho da rotação entre arroz de terras altas e soja. As culturas foram implantadas sob plantio direto, em Latossolo Vermelho distroférrico, na região do Cerrado de Goiás. As plantas de cobertura foram semeadas mecanicamente, também em plantio direto, após a colheita da soja (25/3/2008) e do arroz (7/4/2009). Utilizou-se o delineamento de blocos ao acaso, com parcelas subdivididas no tempo e quatro repetições. Os tratamentos consistiram das seguintes espécies de plantas de cobertura, além do pousio: Urochloa ruziziensis, U. brizantha, Pennisetum glaucum e U. ruziziensis + Cajanus cajan. A avaliação dos nutrientes remanescentes na palhada foi feita aos: 0, 15, 30, 60, 90 e 120 dias a partir da data da dessecação das plantas de cobertura. Urochloa ruziziensis e U. brizantha apresentaram maior eficiência no acúmulo e na liberação de nutrientes, principalmente quanto ao potássio. Urochloa ruziziensis é a espécie mais indicada como planta de cobertura antecessora à cultura do arroz de terras altas, em plantio direto. No entanto, nenhuma espécie de cobertura afeta significativamente a produtividade de grãos da soja.The objective of this work was to evaluate nutrient cycling by cover crops and its influence on the performance of the rotation between upland rice and soybean. The crops were sown under no‑tillage, in a Rhodic Haplustox, in the Cerrado region of the state of Goiás, Brazil. The cover crops were sown mechanically, also under no‑tillage, after the harvest of soybean (3/25/2008) and rice (4/7/2009). A randomized complete block design was used, split‑plotted in time, with four replicates. The treatments consisted of the following cover crop species, besides fallow: Urochloa ruziziensis, U. brizantha, Pennisetum glaucum and U. ruziziensis + Cajanus cajan. The evaluation of the remaining nutrient in straw was performed at 0, 15, 30, 60, 90, and 120 days from the desiccation date of the cover crops. Urochloa ruziziensis and U. brizantha were the most efficient in the accumulation and release of nutrients, especially potassium. Urochloa ruziziensis is the most suited species for use as a cover crop previous to upland rice cultivation under no‑tillage. However, none of the cover crop species significantly affect soybean grain yield

    SiO2-Ag Composite as a Highly Virucidal Material: A Roadmap that Rapidly Eliminates SARS-CoV-2

    COVID-19, as the cause of a global pandemic, has resulted in lockdowns all over the world since early 2020. Both theoretical and experimental efforts are being made to find an effective treatment to suppress the virus, constituting the forefront of current global safety concerns and a significant burden on global economies. The development of innovative materials able to prevent the transmission, spread, and entry of COVID-19 pathogens into the human body is currently in the spotlight. The synthesis of these materials is, therefore, gaining momentum, as methods providing nontoxic and environmentally friendly procedures are in high demand. Here, a highly virucidal material constructed from SiO2-Ag composite immobilized in a polymeric matrix (ethyl vinyl acetate) is presented. The experimental results indicated that the as-fabricated samples exhibited high antibacterial activity towards Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) as well as towards SARS-CoV-2. Based on the present results and radical scavenger experiments, we propose a possible mechanism to explain the enhancement of the biocidal activity. In the presence of O2 and H2O, the plasmon-assisted surface mechanism is the major reaction channel generating reactive oxygen species (ROS). We believe that the present strategy based on the plasmonic effect would be a significant contribution to the design and preparation of efficient biocidal materials. This fundamental research is a precedent for the design and application of adequate technology to the next-generation of antiviral surfaces to combat SARS-CoV-2

    Engineering polycotton fiber surfaces, with an timicrobial activity against S. aureus, E. Coli, C. albicans and SARS-CoV-2

    Pathogenic microorganisms are becoming a potential threat to the health of human beings and the environment worldwide. In this present study, we have developed a polycotton fiber, in which by incorporation and functionalization of aggregated Ag NPs are achieved by using the pad-dry-cure meth- od. Upon contact, this coating shows antimicrobial activity against S. aureus, E. Coli, C. albicans and SARS-CoV-2. The polycotton AgNP, inhibiting nearly of the virus was able to prevent cross-infections, and does not causes allergies or photoirritation, proving the safety of its use. To the best of our knowledge, this is the first report of an antimicrobial coating that could rapidly reduce the infective load of bacteria, fungi, and inhibit SARS--CoV-2. Taken together, the antimicrobial coating reported herein holds great promise to be developed for further application in healthcare settings
