    Mutagenic and antimutagenic potentials of fruit juices of five medicinal plants in Allium cepa L.: Possible influence of DPPH free radical scavengers

    Mutagenic and antimutagenic activities of freeze dried fruit juices (FDFJ) of Morinda elliptica Ridl. (Rubiaceae), Morinda citrifolia L. (Rubiaceae), Averrhoa bilimbi L. (Oxalidaceae), Phyllantus acidus (L.) Skeels (Phyllantaceae) and Myristica fragrans Houtt. (Myristicaceae) in Allium cepa L was evaluated. Testing the mutagenic activity, onions were suspended in solution of different concentrations of FDFJ alone in tap water for 48 h. Thereafter, root tips were prepared and observed for dividing cells and chromosomal aberrations using a light microscope. Antimutagenicity screening was similar to the mutagenic evaluation, except that the solution of FDFJ was combined with 0.1% cylophosphamide-CP. Free radicals (2,2-diphenyl-1-picrylhyrazyl) scavenging activity of the FDFJ was tested using butylated hydroxylanisole (BHA) and butylated hydroxytoluene (BHT) as the standards and their phenolic contents were evaluated by comparing with gallic acid equivalents (GAE). The free radicals scavenging power of M. fragrans at 1 mg/ml was almost similar to that of BHA and BHT and its phenolic content was 21 ± 6.0 mg GAE/g, the highest among the tested juices. A. cepa cell division was inhibited in a dose-dependent manner by the FDFJ of M. fragrans, while the induced chromosomal aberrations were non dose dependent. The cytotoxicity and chromosomal aberrations of CP were suppressed throughout the tested concentrations of M. fragrans, unlike the effects of other four juices. These results suggest that the observed activities of FDFJ of M. fragrans may be due to the quantity and quality of phenolic compounds with antioxidant activity, suggesting its use in preventing the DNAdamaging effects of mutagens.Key words: Allium cepa, antimutagenicity, antioxidants, cyclophosphamide, mutagenicity

    Preliminary Phytochemical and Physicochemical Characterization of Gynura segetum (Lour) Merr (Compositae) Leaf

    Purpose: To carry out phytochemical and physicochemical studies on the leaves of Gynura segetum.Methods: Preliminary phytochemical and physicochemical studies using chromatographic techniques,were carried out, Parameters evaluated include ash value, loss on drying and extractive value, amongst others. .Results: Phytochemical results revealed that the leaves of Gynura segetum contain alkaloid, terpene, flavonoid, tannin and saponin. Mean ash value (%) was 13.5 (total), 2.49 (acid-insoluble ash) and 6.14 (water-soluble ash). Loss on drying was 10.70 % while extractive value was 20.60 % in water and 14.13 % in ethanol. Thin layer chromatography (TLC) analysis indicates the presence of rutin in the methanol extract of G. segetum and this was supported by high performance liquid chromatography (HPLC) studies.Conclusion: Useful quantitative and descriptive data essential for identifying and characterizing the plant for the purpose of quality control are presented.Keywords: Gynura segetum, Quality control, Extractive, Ash valu

    Evaluation of Biological Activities of Extracts and Chemical Constituents of Mimusops elengi

    Purpose: To isolate some compounds from the leaves and bark of Mimusops elengi, and examine them for their antibacterial and anti-inflammatory properties.Experimental: The compounds were isolated from the leaf and bark chloroform extracts using column chromatography, and characterized using physical and spectroscopic methods. The isolated compounds and their respective extracts were tested for antibacterial activity by micro-dilution antibacterial assay, and for anti-inflammatory activity by cyclooxygenase inhibitory assay.Results: of the compounds isolated include spinasterol (1), ursolic acid (2) and 3β, 6β, 19α, 23- tetrahydroxyurs-12-en-28-oic acid (3) from the leaves; and taraxerol (4) and spinasterol β-Dglucopyranoside (5) from the bark. A majority of the samples showed good activity against Staphylococcus aureus (9.7 ─ 78.0 μg/mL), while moderate activity was observed against Gramnegative bacteria (78.0 ─ 156 μg/mL). Strong COX inhibition was observed for the leaf extract, and (1); selective COX-2 inhibition for (2) and (3); and selective COX-1 inhibition for bark extract, (4) and (5).Conclusion: This is the first report describing the anti-inflammatory potential of M. elengi on the basis of its isolated constituents.     The antiinflammatory potential of phenolic compounds from Emblica officinalis L. in rat

    Antiinflammatory effects of phenolic compounds from Emblica officinalis were evaluated in carrageenan and cotton pellet induced acute and chronic inflammatory animal model. Fractions of E. officinalis containing free (FPEO) and bounded (BPEO) phenolic compounds were assessed by HPLC technique. The free and bound phenolic compounds were studied for their acute and chronic antiinflammatory activity at dose level of 20 and 40 mg/kg. The carrageenan induced acute inflammation was assessed by measuring rat paw volume at different time of intervals. Further, cotton pellet induced chronic inflammation was assessed by granulomatous tissue mass estimation along with the estimation of tissue biomarker changes (i.e. lipid peroxidation, reduced glutathione, myeloperoxidase and plasma extravasation). The results indicated that in both acute and chronic inflammation, FPEO and BPEO show reduction in the inflammation, but significant effects was observed only at high doses of both fractions which was comparable to diclofenac treated group. In conclusion, phenolic compounds of E. officinalis may serve as potential herbal candidate for amelioration of acute and chronic inflammation due to their modulatory action of free radicals

    Induction of apoptosis of human primary osteoclasts treated with extracts from the medicinal plant Emblica officinalis

    <p>Abstract</p> <p>Background</p> <p>Osteoclasts (OCs) are involved in rheumatoid arthritis and in several pathologies associated with bone loss. Recent results support the concept that some medicinal plants and derived natural products are of great interest for developing therapeutic strategies against bone disorders, including rheumatoid arthritis and osteoporosis. In this study we determined whether extracts of <it>Emblica officinalis </it>fruits display activity of possible interest for the treatment of rheumatoid arthritis and osteoporosis by activating programmed cell death of human primary osteoclasts.</p> <p>Methods</p> <p>The effects of extracts from <it>Emblica officinalis </it>on differentiation and survival of human primary OCs cultures obtained from peripheral blood were determined by tartrate-acid resistant acid phosphatase (TRAP)-positivity and colorimetric MTT assay. The effects of <it>Emblica officinalis </it>extracts on induction of OCs apoptosis were studied using TUNEL and immunocytochemical analysis of FAS receptor expression. Finally, <it>in vitro </it>effects of <it>Emblica officinalis </it>extracts on NF-kB transcription factor activity were determined by gel shift experiments.</p> <p>Results</p> <p>Extracts of <it>Emblica officinalis </it>were able to induce programmed cell death of mature OCs, without altering, at the concentrations employed in our study, the process of osteoclastogenesis. <it>Emblica officinalis </it>increased the expression levels of Fas, a critical member of the apoptotic pathway. Gel shift experiments demonstrated that <it>Emblica officinalis </it>extracts act by interfering with NF-kB activity, a transcription factor involved in osteoclast biology. The data obtained demonstrate that <it>Emblica officinalis </it>extracts selectively compete with the binding of transcription factor NF-kB to its specific target DNA sequences. This effect might explain the observed effects of <it>Emblica officinalis </it>on the expression levels of interleukin-6, a NF-kB specific target gene.</p> <p>Conclusion</p> <p>Induction of apoptosis of osteoclasts could be an important strategy both in interfering with rheumatoid arthritis complications of the bone skeleton leading to joint destruction, and preventing and reducing osteoporosis. Accordingly, we suggest the application of <it>Emblica officinalis </it>extracts as an alternative tool for therapy applied to bone diseases.</p
