Engineering Synthetic TAL Effectors with Orthogonal Target Sites
The ability to engineer biological circuits that process and respond to complex cellular signals has the potential to impact many areas of biology and medicine. Transcriptional activator-like effectors (TALEs) have emerged as an attractive component for engineering these circuits, as TALEs can be designed de novo to target a given DNA sequence. Currently, however, the use of TALEs is limited by degeneracy in the site-specific manner by which they recognize DNA. Here, we propose an algorithm to computationally address this problem. We apply our algorithm to design 180 TALEs targeting 20 bp cognate binding sites that are at least 3 nt mismatches away from all 20 bp sequences in putative 2 kb human promoter regions. We generated eight of these synthetic TALE activators and showed that each is able to activate transcription from a targeted reporter. Importantly, we show that these proteins do not activate synthetic reporters containing mismatches similar to those present in the genome nor a set of endogenous genes predicted to be the most likely targets in vivo. Finally, we generated and characterized TALE repressors comprised of our orthogonal DNA binding domains and further combined them with shRNAs to accomplish near complete repression of target gene expression
Mots clés: kystes, protozoaires flagellés, agriculture maraîchère, bas-fonds et zone urbaineEnglish AbstractObjectives: To evaluate the abundance of environmental forms of flagellated protozoa in waste water that is exploited for urban wetland agricultural practices in Yaoundé.Methodology and results: A study was carried out from November 2013 to April 2014, with bioenvironmental and bio-statistical assessment from 2015-2016. Sampling took place on monthly basis. The flagellated protozoan cysts were identified by lugol coloration after concentrating the samples according to the method -of flotation using zinc sulphate. Biological analysis of water showed the presence of flagellated protozoan cysts with average densities of (101 ± 70 cysts/L) for Giardia intestinalis, Chilomastix mesnili (34  ± 20 cysts/L), of Retortamonas intestinalis (3 ± 6 cysts/L) and of Enteromonas hominis (1 ± 3 cysts/L). Generally, the highest cysts densities were registered during the short rainy season (172 ± 108 cysts/L). The Statistical analysis showed positive correlations (P ≤ 0, 05) between the density of these cysts and physic-chemical variables such as suspended solids, oxydability and turbidity. This testifies the importance of colloidal or suspended particles in the transportation of micro-organisms in water. Conclusion and application of results: In view of these results and the importance of urban agricultural practices in Yaoundé, plants must be well washed and properly cooked before consumption in order to avoid a health risk that is limited to the spread of this waterborne pathogen in this metropoly. Keywords: Cyst, flagellated protozoa, wetland agricultural, dregs and urban zon

Etude De L’efficacité Du Cashew Nut Shell Liquid (CNSL) De Anacardium Occidentale L. Extrait À Froid Sur Le Contrôle De Anopheles Gambiae S.L Résistant Aux Pyréthrinoïdes
In order to combat the vectors of malaria, it is necessary to know their behavior with regard to the insecticide products envisaged. The present study is aimed at the CNSL efficiency test extracted from the seed hull of Anacardium occidentale L. for the control of pyrethroid-resistant Anopheles gambiae s.l. Sensitivity tests were carried out on the larvae of stages 2, 3 and 4 of An. gambiae of the wild and Kis-kdr strains carrying the Kdr gene. The "Kisumu" strain devoid of resistance gene served as a control. The mortalities were read after 24 hours of exposure. LD50 and DL90 were determined using the log-probit method of dose determination corresponding to a proportion. The high LD50 determined is 0.0042 mg / ml and corresponds to the LD50 of the larvae of stage 2 of the wild strain. The low LD50 is determined on stage 2 larvae "Kisumu". The high DL90 is determined on stage 4 larvae of the wild strain. The low LD90 determined corresponds to stage 2 larvae "Kisumu". The comparison of the LD between the different strains shows a low sensitivity to CNSL of the larvae of resistant strains compared to the control strain, indicating the influence of the resistance factor kdr on the CNSL efficiency. However, the low-dose CNSL larvicidal activity on the larvae of the kdr-carrying strains provides an alternative advantage in the management of vector resistance to pyrethroid
