TRPM8 Activation via 3-Iodothyronamine Blunts VEGF-Induced Transactivation of TRPV1 in Human Uveal Melanoma Cells
In human uveal melanoma (UM), tumor enlargement is associated with increases in aqueous humor vascular endothelial growth factor-A (VEGF-A) content that induce neovascularization. 3-Iodothyronamine (3-T1AM), an endogenous thyroid hormone metabolite, activates TRP melastatin 8 (TRPM8), which blunts TRP vanilloid 1 (TRPV1) activation by capsaicin (CAP) in human corneal, conjunctival epithelial cells, and stromal cells. We compare here the effects of TRPM8 activation on VEGF-induced transactivation of TRPV1 in an UM cell line (92.1) with those in normal primary porcine melanocytes (PM) since TRPM8 is upregulated in melanoma. Fluorescence Ca2+-imaging and planar patch-clamping characterized functional channel activities. CAP (20 μM) induced Ca2+ transients and increased whole-cell currents in both the UM cell line and PM whereas TRPM8 agonists, 100 μM menthol and 20 μM icilin, blunted such responses in the UM cells. VEGF (10 ng/ml) elicited Ca2+ transients and augmented whole-cell currents, which were blocked by capsazepine (CPZ; 20 μM) but not by a highly selective TRPM8 blocker, AMTB (20 μM). The VEGF-induced current increases were not augmented by CAP. Both 3-T1AM (1 μM) and menthol (100 μM) increased the whole-cell currents, whereas 20 μM AMTB blocked them. 3-T1AM exposure suppressed both VEGF-induced Ca2+ transients and increases in underlying whole-cell currents. Taken together, functional TRPM8 upregulation in UM 92.1 cells suggests that TRPM8 is a potential drug target for suppressing VEGF induced increases in neovascularization and UM tumor growth since TRPM8 activation blocked VEGF transactivation of TRPV1
Characterization of temperature-sensitive transient receptor potential channels in cultivated uveal melanoma cells (92.1)
Das Aderhautmelanom ist der häufigste maligne intraokulare Tumor im
Erwachsenenalter mit einer Inzidenz von 5.1 Fällen pro 100.000 Einwohner. Die
Letalität geht vornehmlich von dessen hoher Metastasierungsrate aus. Dabei ist
am häufigsten die Leber, gefolgt von Lunge, Knochen und der Haut betroffen.
Die Diagnose ist oft ein in einer Routineuntersuchung gestellter
Zufallsbefund, da dieser Tumor sich lange asymptomatisch entwickelt. Bisher
konnte die Mortalitätsrate durch keine neue Behandlungsmöglichkeit gesenkt
werden. Wie bei allen Typen von Neoplasien können Störungen der zellulären
Homöostase zu einer Progression des Tumors führen. Ein wichtiger sekundärer
Botenstoff in diesem Zusammenhang ist Calcium, weil es in viele Signalwege der
Zelle involviert ist. Zur Familie der calciumregulierenden Kanäle gehören die
Transient-Rezeptor-Potential-Ionenkanäle (TRPs), die in Neoplasien
fehlreguliert sein können. In dieser Studie wurden in einer malignen
Aderhautmelanomzelllinie und in gesunden Aderhautmelanozyten die funktionale
Expression der TRP-Subtypen TRP-Vanilloidrezeptor 1 (TRPV1, Capsaicinrezeptor)
und TRP-Melastatinrezeptor 8 (TRPM8, Mentholrezeptor) ermittelt und
verglichen. Hierfür wurden hochsensitive Methoden wie Calcium Imaging und die
planare Patch-Clamp-Technik verwendet. In den gesunden Aderhautmelanozyten
ließen sich mit dem TRPV1-Agonisten Capsaicin (CAP) induzierte Ca2+-Einströme
messen. Dagegen waren keine Ströme nach TRPM8-Stimulation mit Icilin zu
erkennen. Im Vergleich dazu zeigten die malignen Aderhautmelanozyten CAP und
Icilin induzierte Ca2+-Einströme sowie erhöhte Ganzzellströme. Die
Stromantwortmuster sind charakteristisch für TRPM8- und TRPV1-Expression.
Zusätzlich konnten die Calcium- und Stromantworten durch die TRPV1- und
TRPM8-spezifischen Blocker Capsazepin (CPZ) und BCTC unterdrückt werden. Die
Identifizierung von signifikanten Unterschieden in der funktionalen Expression
dieser beiden Kanäle eröffnet neue Möglichkeiten der gezielten Entwicklung
neuer Behandlungsmethoden unter möglicher Schonung von gesundem Gewebe.
Dennoch können diese Erkenntnisse nur sehr vorsichtig auf dem lebenden
menschlichen Organismus übertragen werden. Zum einen wurden in dieser Studie
als Kontrollgruppe als Modell keine menschlichen Aderhautmelanozyten
verwendet, sondern Zellen aus dem Schwein isoliert. Zum anderen ist es ein
allgemein anerkannter Fakt, dass man In-vitro-Daten nur schlecht auf einen
lebenden Organismus übertragen kann. Es braucht weitere Untersuchungen, um
diese Erkenntnisse zu verifizieren und daraus einen klinischen Nutzen zu
ziehen.Uveal melanoma is the most common malignant intraocular cancer among adults
with an incidence of 5.1 cases per 100.000 inhabitants. The case fatality rate
is accounted for by the high risk of metastasis. The most affected organs are
the liver, followed by lung, bone and skin. The diagnosis is often found in a
routine examination because of the long asymptomatically development of this
cancer. To diagnose this disease different techniques can be used, which until
have not reduced the mortality rate. As with all kinds of neoplasia,
dysfunctions of cellular homoestasis may lead to a progression of the cancer.
An important second messenger is calcium because it is involved in many
signalling pathways within the cell. The family of calcium-regulated channels
involves transient receptor potential channels (TRPs), which are often
dysregulated in neoplasia. In this study, we compared the malignant uveal
melanoma cell line 92.1 with healthy uveal melanocytes from pigs for
functional expression of the TRP subtypes TRPV1 (vanilloid receptor, capsaicin
receptor) and TRPM8 (melastatin receptor, menthol receptor). For the
investigations, we used high sensitive functional assays like calcium imaging
and planar patch-clamping. In healthy uveal melanocytes capsaicin, a TRPV1
agonist, induced increases of intracellular calcium and whole-cell currents
whereas the TRPM8 agonist icilin did not clearly induce calcium regulation in
these cells. In contrast such calcium increase could be stimulated by both,
capsaicin and icilin in malignant uveal melanoma cells. The changes in ion
current behaviour indicate functional TRPM8 and TRPV1 expression since their
activation could be blocked with either capsazepine (CPZ, TRPV1blocker) or
BCTC (TRPM8 blocker). The identification of significant differences in the
functional expression of both channels opens new treatment options. Nether the
less these new findings can not be easily transferred to a living organism. On
the one side, we used uveal cells out of a pig eye. On the other hand, it is
problematic to compare in vitro data with a complex human being. There is a
need for much more research before a clinical benefit can be achieved
