Regulatory framework for advanced therapy medicinal products in Europe and United States

    Get PDF
Advanced therapy medicinal products (ATMPs) are a fast-growing field of innovative therapies. The European Union (EU) and the United States (US) are fostering their development. For both regions, ATMPs fall under the regulatory framework of biological products, which determines the legal basis for their development. Sub-classifications of advanced therapies are different between regions, while in EU, there are four major groups, i.e., gene therapy, somatic cell therapy, tissue-engineered therapies, and combined advanced therapies; in US, the sub-classification covers two major groups of products, i.e., gene therapy and cellular therapy. The inclusion criteria that define a gene therapy are equivalent in both regions, and the exclusion criteria are directly related to the indications of the product. In the EU, there is a clear differentiation between cell- and tissue-based products regarding their classification as advanced therapies or coverage by other legal frameworks, whereas in US, there is a broader classification about whether or not these products can be categorized as biologic products. Both in EU and in US, in order to classify a cell- or a tissue-based product as an advanced therapy, it must be ensured that the processing of the cells implies a manipulation that alters their biological characteristics, although the term of manipulation in US differentiates between structural and non-structural cells and tissues. The regulatory terminology used to define ATMPs and their sub-classification reveals some differences between EU and US

Exploring the relevance of the type of horizontal separator to optimize the extraction efficacy for the Arbequina variety

The productivity of virgin olive oil depends not only on agronomic factors but also on the technological factors of the extraction process. The ‘Arbequina’ variety has extractability problems, which is a challenge for master millers anywhere. This work aims to evaluate the behavior of different decanters and seeks to modulate the effect of some processing parameters and their interactions with oil extraction efficiency in the case of ‘Arbequina.’ Fruit characteristics, processing parameters, and extractability were collected over 10 years from 38 decanters that belong to five different brands. The results have shown that fruit moisture is the most relevant factor for oil extractability, especially over 52%. Furthermore, extractability is positively correlated with malaxing temperature, addition of water, and total fat content in the fruit. However, the results show that before applying a regulation, the type of decanter must be considered. The model used in this study has allowed us to optimize the regulations for each type of decanter to reduce oil losses within the pomace, achieving an extraction efficiency within the range of 78%–91.5%. In fact, the best extraction efficiency results (91.5%) were obtained by processing at temperatures >26°C and water injection of 5%.The author(s) declare financial support was received for the research, authorship, and/or publication of this article. Research on university teachers today requires greater depth regarding the relationship between personality, teaching and burnout. The latter is a well-studied phenomenon in teachers of other educational stages. There are many investigations that have delved into the effect of burnout in teachers, given that this population is especially vulnerable to developing it due to the affective and emotional components inherent to the professional profile. On the other hand, digital competition is another phenomenon that has been exposed in recent years. In this sense, different studies refer to the different skills related to the use of tools and technologies in the classroom context. For the development of the study, a battery of questionnaires has been used, including; the 16PF Personality Questionnaire, the Maslach Burnout Inventory (MBI) and the Digital Competence Questionnaire of Spanish Higher Education Teachers. The results from the data analysis using the statistical program SPSS showed that there are no significant differences in terms of gender and burnout. On the other hand, a positive relationship has been observed between depersonalization and emotional exhaustion with the feeling of self-sufficiency. In addition, a positive relationship is observed between the nervous tension scale with depersonalization. Regarding digital competence, a positive relationship has also been found between personal fulfillment with the use of ICT in the classroom and also with a positive attitude towards ICT. The need to expand research regarding personality in university teachers is observed. In the same way as the study of burnout in this educational stage and the importance of good digital competence in current higher education teachers.La investigación sobre docentes universitarios, requiere a día de hoy una mayor profundidad con respecto a la relación entre la personalidad, la docencia y el burnout. Purpose: This study aims to assess inappropriate prescribing (IP) to elderly patients during the month prior to hospitalization and to compare different IP criteria. Methods: An observational, prospective and multicentric study was carried out in the internal medicine services of seven Spanish hospitals. Patients aged 75 years and older were randomly selected after hospital admission for a year. To assess potentially inappropriate medicines (PIMs), the Beers and STOPP criteria were used and to assess Potentially Prescribing Omissions (PPOs), the START criteria and ACOVE-3 medicine quality indicators were used. An analysis to assess factors associated with IP was performed. Results: 672 patients [median age (Q1_Q3) 82 (79-86) years, 55.9% female] were included. Median prescribed medicines in the month prior to hospitalization were 10(Q1-Q3 7-13). The prevalence of IP was 87.6%, and 54.3% of patients had PIMs and PPOs concurrently. A higher prevalence rate of PIMs was predicted using the STOPP criteria than with the Beers criteria (p<.001) and a higher prevalence of PPOs using the ACOVE-3 criteria than using the START criteria (p<.001) was observed. Polypharmacy (≥10 medicines) was the strongest predictor of IP [OR=11.34 95% confidence interval (CI) 4.96-25.94], PIMs [OR=14.16, 95% CI 6.44-31.12], Beers-listed PIMs [OR=8.19, 95% CI 3.01-22.28] and STOPP-listed PIMs [OR=8.21, 95% CI 3.47-19.44]. PIMs was the strongest predictor of PPOs [OR=2.79, 95% CI 1.81-4.28]. Conclusions: A high prevalence of polypharmacy and PIMs and PPOs were reported. More than half the patients had simultaneous PIMs and PPOs. The related factors to PIMs and PPOs were different

Phase-mediated magnon chaos-order transition in cavity optomagnonics

Magnon as a quantized spin wave has attracted extensive attentions in various fields of physics, such as magnon spintronics, microwave photonics, and cavity quantum electrodynamics. Here, we explore theoretically magnon chaos-order transition in cavity optomagnonics, which is still remains largely unexplored in this emerging field. We find that the evolution of the magnon experiences the transition from order to period-doubling bifurcation and finally enter chaos by adjusting the microwave driving power. Different from the normal chaos, the magnon chaos-order transition proposed here is phase-mediated. Beyond their fundamental scientific significance, our results will contribute to the comprehending of nonlinear phenomena and chaos in optomagnonical system, and may find applications in chaos-based secure communication

Influence of the ripening stage and extraction conditions on the phenolic fingerprint of 'Corbella' extra-virgin olive oil

The ancient 'Corbella' olive variety from the center-north of Catalonia is being recovered to obtain quality extra-virgin olive oil (EVOO) with unique organoleptic properties. The aim of this work was to determine the effect of agronomic and technical factors on the phenolic fingerprint of EVOO and to establish the optimum harvesting time and crushing and malaxation conditions for 'Corbella' olives. Therefore, three different ripening indices (0.3, 1.2, and 3.2) and three crushing temperatures (10, 18, and 25 OC) were studied. Additionally, a factorial design to optimize the phenolic concentration of the EVOO was developed, applying a range of sieve diameters (4 and 6 mm), and malaxation time (30 and 60 min) and temperature (27, 32, and 37 °C). The phenolic profile was analyzed by ultra-high performance liquid chromatography coupled to mass spectrometry in a tandem detector. The level of secoiridoids, the major phenolic compounds in the oil, was higher when using olives harvested earlier. Oleuropein aglycone and ligstroside aglycone were degraded during crushing at high temperatures, resulting in the formation of oleacein and oleocanthal. The best processing conditions in terms of total phenolic content were found to be 30 min of malaxation at 37 OC, the crushing size not having any affect.
