Optimization of total carotenoids extraction from pequi pulp (Caryocar brasiliense Camb.) using a mixture planning method/ Otimização da extração de carotenoides totais da polpa de pequi (Caryocar brasiliense Camb.) usando método de planejamento de misturas
The pequi is a native fruit of the Brazilian Savanna and the consumption of its carotenoid-rich pulp has been associated with several health benefits. However, there are considerable differences in carotenoid content reported for this fruit, mainly due to the diversity of extraction methods used in the recovery of these compounds from pequi pulp. The objective of this study was to evaluate the efficiency of different organic solvents and mixtures in the extraction of total carotenoids from pequi pulp. For the carotenoids extraction, pure solvents and the mixtures of acetone, ethyl alcohol and petroleum ether were used. The mixture planning was used to determine the proportions of the solvents in each treatment. To our knowledge, this is the first study to identify that the most efficient treatment for total carotenoid extraction from pequi pulp is pure petroleum ether solvent (234.40 μg/g). In contrast, ethyl alcohol was the solvent that extracted the lowest concentration of total carotenoids (124.03 μg/g). Pure acetone, one of the most used solvents in the extraction of carotenoids from pequi, showed intermediate efficiency (166.07 μg/g). Thus, pure petroleum ether is the solvent recommended for total carotenoid extraction, mainly in the samples rich in carotenoid and lipids, such as pequi pulp
Coincidence or correlation?
Rationale:Multiple Endocrine Neoplasia type 1 (MEN1) is a familial syndrome that results from the disruption of a tumor suppressor protein called MENIN. Its management is challenging, as MEN1 affects different endocrine tissues and predisposes to both benign and malignant tumors. MENIN-deficient cells have recently been recognized to play a role in triggering autoimmunity. Herein, we present a case of MEN1 with multiple endocrine and autoimmune disorders.Patient concerns:A 50 years old female with a 25 years history of complicated nephrolithiasis presented with primary hyperparathyroidism.Diagnoses:Over several decades, she was diagnosed with recurrent primary hyperparathyroidism, autoimmune thyroiditis, multinodular goiter, pernicious anemia, metastatic gastric type 1 neuroendocrine tumor, macroprolactinemia, gonadotropin deficiency, mucosa-associated lymphoid tissue lymphoma of the thyroid gland, positive anti-calcium sensor receptor antibodies, and BRCA 1/2-negative invasive breast cancer. The autoimmune regulator gene was sequenced, but no pathogenic variants were found. Next-generation sequencing revealed both a pathogenic MEN1 mutation and a benign CDC73 gene variant. Familial genetic screening revealed a large kindred with multiple carriers of one or both genetic variants (MEN1 = 19; CDC73 = 7).Interventions:The patient underwent surgical excision of three parathyroid glands, total thyroidectomy and breast tumorectomy plus tamoxifen, and monthly injections of octreotide. The patient and family members with the MEN1 mutation are under a life-long surveillance program for MEN1 prototypic tumors.Outcomes:The patient was stable and alive during a 24-years follow-up period.Lessons:With the present case, the authors highlight a new interplay between MENIN and the immune system, which may have implications for future targeted life-long surveillance and treatment of MEN1 patients.publishersversionpublishe

Pentalogy of Fallot in a Crioulo Foal
Background: Cardiac anomalies account for 3.5% of congenital defects in horses. Tetralogy of Fallot (TOF) is characterized by ventricular septal defect, pulmonary stenosis, dextroposition the aorta and right ventricular hypertrophy secondary.Pentalogy of Fallot (POF) is a variation and is characterized by a defect in the right atrioventricular valve or persistence of the ductus arteriosus, in addition to the other defects observed in tetralogy. Affected animals usually have stunted growth, exercise intolerance, weakness, lethargy, cyanosis, dyspnea and syncope. The aim of this study was to describe a case of POF, as diagnosed in a Crioulo foal in southern Brazil.Case: Clinical signs were observed at the ffth day of life and were characterized by cyanosis, intense tracheal stertor, tachycardia, marked heart murmur, HR 160 bpm, RR 80 breaths/min, T 39.8°C and syncope. These signs worsened with physical activity. Endoscopic evaluation of the upper airways revealed no anatomical conformation changes or tissue dysfunction. A cardiac ultrasound showed a loss of continuity of the interventricular septum, right ventricular wall thickening, and a hyperechoic appearance of the area where the right atrioventricular valve (tricuspid) should be. The crises have become more severe and more frequent, the animal began to show signs of ischemia, such as loss of motor coordination and limited mobility. The foal died at 15 days old. At necropsy the liver was congested and had a nutmeg appearance, there was foam in the trachea, and the lungs were edematous. There was hydropericardium, and the heart was increased in size with a globoid shape. The heart had thickening of the walls of the right and left ventricle and pulmonary artery stenosis. There was also an oval orifce 2.5 cm in diameter in the interventricular septum connecting the ventricles (ventricular septal defect), an overriding aorta and aplasia of the tricuspid valve. Histologically the liver had centrilobular to mediozonal necrosis, and there was pulmonary edema and congestion.Discussion: The diagnosis of pentalogy of Fallot was based on the clinical signs, the ultrasound results and the macroscopic lesions observed at necropsy. Tetralogy of Fallot is one of the most common congenital cardiac malformations observed in horses, however, in a study of congenital malformations in this species, only 3.5% had cardiac malformations out ofthe 608 foals examined. The prevalence of heart defects in horses compared with other domestic species is relatively low (0.1% - 0.5%). In this case, the malformation was probably sporadic because the foal was the only animal to be born with this malformation on the farm. In the Crioulo breed, congenital defect reports are scarce. In a retrospective study of equine diseases in southern Brazil over a period of 35 years, malformations were not observed in 708 horses of this breed, whichis the predominant breed in the region. In the present case, the fact that the mare had another three healthy foals with the same stallion reduces the possibility that this was a case of hereditary origin. It is likely that this case is of a sporadic nature with no chance of determining the cause. The differential diagnoses in animals with lethargy, cyanosis, and exerciseintolerance or syncope include complicated or multiple cardiac defects. There are no previous reports of TOF and POF in the Crioulo breed in Brazil. Keywords: Pentalogia de Fallot, cardiac ultrasound, cardiac malformation, horse
Paramagnetic epoxy resin
This work illustrates that macrocycles can be used as crosslinking agents for curing epoxy resins, provided that
they have appropriate organic functionalities. As macrocycles can complex metal ions in their structure, this curing reaction
allows for the introduction of that metal ion into the resin network. As a result, some characteristic physical properties of the
metallomacrocycle could be transferred to the new material. The bisphenol A diglycidyl ether (BADGE, n = 0) and hemin
(a protoporphyrin IX containing the Fe(III) ion, and an additional chloride ligand) have been chosen. The new material has
been characterized by differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), Fourier Transform Infrared
(FT-IR), Nuclear Magnetic Resonance (NMR), Transmission Electron Microscopy (TEM), and magnetic susceptibility
measurements). Fe(III) remains in the high-spin state during the curing process and, consequently, the final material exhibits
the magnetic characteristics of hemin. The loss of the chlorine atom ligand during the cure of the resin allows that Fe(III)
can act as Lewis acid, catalyzing the crosslinking reactions. At high BADGE n = 0/hemin ratios, the formation of ether and
ester bonds occurs simultaneously during the process. The authors thank the Ministerio de Ciencia y Tecnología, Spain, Project MAT2010-61721 for financial support. ECV also thanks Dr. Senén Paz and GAIRESA for a scholarshipS
Identificación y caracterización molecular de virus Orf en mujeres de Sudamérica expuestas laboralmente
Contagious Ecthyma (CE) is a severe exanthematous dermatitis caused by the Orf virus (ORFV) that mainly affects domestic small ruminants such as sheep and goats. It is a worldwide-distributed occupational zoonosis, particularly infecting those in close contact with animals or animal products such as shepherds, farmers and veterinarians, among others. In the present work, we report the first human CE case confirmed in Argentina. A phylogenetic analysis based on four gene sequences of the isolated strain responsible for the disease showed that this isolate grouped with other ORFV sequences that caused reported CE cases in sheep from the same Argentine province. We also sequenced a sample from a Chilean human case reported in 2017, whose phylogenetic analysis showed that it groups together with other Argentine isolates from locations close to the border with Chile.Fil: Peralta, Andrea Verónica. Modulation of cathelicidins, IFNβ and TNFα by bovine alpha-herpesviruses is dependent on the stage of the infectious cycle
Production of antimicrobial peptides cathelicidins, interferons and cytokines is an important feature in airway epithelial host defense. The innate immune response to alpha-herpesvirus infection at the sites of primary replication has not been fully studied. Thus, the aim of this study was to determine the expression of innate immune components, cathelicidins, IFNβ, TNFα and TNF receptors (TNFRI and TNFRII) during acute infection and reactivation of bovine herpesvirus type 1 (BoHV-1) and 5 (BoHV-5) in the respiratory tract and lymphoid tissue of their natural host. We found that BoHV infection modulates mainly the expression of BMAP28, a key cathelicidin in cattle. It was downregulated by both viruses in retropharyngeal lymph nodes of acutely infected-calves, and it was accompanied by a lower expression of IFNβ, TNFα and TNFRI. BoHV-5 showed a pronounced role in the downregulation of BMAP28, even in nasal mucosa and lung. However, during reactivation, BoHV-5 upregulated both BMAP28 and IFNβ in retropharyngeal lymph nodes. Acute replication induced also TNFα mRNA and protein synthesis, and expression of TNFRI and II was positively regulated during both acute infection and reactivation, particularly in the trachea. Moreover, BMAP27 was detected during BoHV-1 reactivation suggesting a potential role at this stage. Thus, cathelicidins are implicated in alpha-herpesvirus infections of the bovine respiratory system and the response is distinct during BoHV-1 and BoHV-5 acute infection and reactivation. This demonstrates that these viruses modulate differentially the components of innate immune response, possibly influencing their pathogenesis. This study provides an initial pilot analysis of factors that might be implicated in alpha-herpesvirus infection of the bovine respiratory systemEEA BalcarceFil: Burucúa, Mercedes María.
Submetida à terapia cirúrgica para ressecção da lesão e seguimento com mesilato de imatinibe (Glivec®).Gastrointestinal stromal tumors (GIST) are rare, with unpredictable behavior, most of them asymptomatic or nonspecific symptoms. They may arise in any place of digestive tube, and the standard treatment is the complete surgical resection, however recurrences and metastases are frequent. The following case is a report from an elderly patient with growing and painful abdominal mass on physical examination, and the result of complementary imaging tests suggests that this is GIST. Submitted to surgical therapy for resection of the lesion and follow-up with imatinib mesylate (Glivec®)
