    Teste rápido de aglutinação utilizando partículas de látex para a detecção de anticorpos anti-cisticercos em amostras de líquido cefalorraquidiano (LCR)

    Simple and rapid latex-based diagnostic tests have been used for detecting specific antigens or antibodies in several diseases. In this article, we present the preliminary results obtained with a latex agglutination test (LAT) for diagnosing neurocysticercosis by detection of antibodies in CSF. A total of 43 CSF samples were assayed by the LAT: 19 CSF samples from patients with neurocysticercosis and 24 CSF samples from patients with other neurologic disorders (neurosyphilis, n = 8; neurotoxoplasmosis, n = 3; viral meningitis, n = 4, chronic headache, n = 9). The LAT exhibited 89.5% sensitivity and 75% specificity. The use of LAT seems to be an additional approach for the screening of neurocysticercosis with advantage of simplicity and rapidity. Further studies could be performed using purified antigens and serum samples.Testes diagnósticos simples e rápidos baseados na aglutinação de partículas de látex têm sido utilizados para a pequisa de antígenos ou anticorpos específicos em muitas doenças. No presente trabalho, é descrito um teste de aglutinação em lâmina para a pesquisa de anticorpos contra cisticercos de Taenia solium, utilizando partículas de látex revestidas com um extrato bruto do parasita. Anticorpos anti-cisticercos foram pesquisados em 19 amostras de LCR de pacientes com neurocisticercose e em 24 amostras de LCR de pacientes com outros problemas neurológicos (neurosífilis, n = 8; neurotoxoplasmose, n = 3; meningite viral, n = 4; cefaléia crônica, n = 9). O teste de aglutinação apresentou sensibilidade e especificidade de 89,5% e 75%, respectivamente. O teste de aglutinação para cisticercose idealizado é simples, rápido e barato. Essas características tornam o teste um meio promissor de expansão e simplificação do imunodiagnósico da neurocisticercose. Estudos futuros poderiam testar a sensibilização das partículas de látex com antígenos de cisticercos purificados e a pesquisa de anticorpos em amostras de soros

    The Brazilian Ceremony in Honor of Body Donors: An opportunity to express gratitude and reflect on medical education

    Introduction: Memorial ceremonies are a way for institutions to show their gratitude for the gesture made by body donors and are an opportunity for students to reflect on the ethical paradigm shift. Therefore, this study aims to describe the memorial ceremony at the Federal University of Health Sciences of Porto Alegre and assess the perception of both students and the donors’ relatives regarding the event. Material and methods: In 2016, a questionnaire was applied to donors’ relatives with questions regarding structure of the ceremony, ethical training of the students and importance of body donation. Another questionnaire was applied to the first-year students of the medical school, with questions about understanding the use of bodies, and their thoughts on death and relevant ethical issues. Results: Fifty questionnaires were answered by family members and 98% agreed that the ceremony was an opportunity to feel closer to the donors and 100% said that the ceremony is important in the ethical training of students. Among students, 68 questionnaires were answered, and all affirmed that the event had a positive impact on the perception of physician-patient relationship and personal growth, and 86.7% reported enhanced empathy. Conclusion: The memorial ceremony seems to encourage students to reflect on ethical issues, because at the same time that the donors’ relatives perceive an attitude of care and respect on the part of the students, the students state a heightened sense of commitment and responsibility regarding their learning and professional formation, which depends on the gesture made by the donors

    Is a Genome a Codeword of an Error-Correcting Code?

    Since a genome is a discrete sequence, the elements of which belong to a set of four letters, the question as to whether or not there is an error-correcting code underlying DNA sequences is unavoidable. The most common approach to answering this question is to propose a methodology to verify the existence of such a code. However, none of the methodologies proposed so far, although quite clever, has achieved that goal. In a recent work, we showed that DNA sequences can be identified as codewords in a class of cyclic error-correcting codes known as Hamming codes. In this paper, we show that a complete intron-exon gene, and even a plasmid genome, can be identified as a Hamming code codeword as well. Although this does not constitute a definitive proof that there is an error-correcting code underlying DNA sequences, it is the first evidence in this direction

    Flowability of inert particle beds with fruit pulp: effects on the drying in spouted bed

    In this work six types of inert particles were characterized and analyzed for drying tropical fruit pulps. The repose angle was determined with and without the addition of water and pulp of various tropical fruits. The bed flowability was related to the particle properties and chemical composition of pulps. The influence of the repose angle on the drying performance was analyzed. It was also verified that the composition of pulps influenced the bed flowability. The global analysis showed that the pulps with high lipids and insoluble solids content and low reducing sugar content improved the bed flowability. The results obtained with the drying of the fruit pulps using low-density polystyrene granules as inert particles showed that high flowabilities lead to lower minimum spout flow rates, but do not necessarily lead to the highest powder production.Neste trabalho, foram caracterizados seis tipos de material inerte, utilizados na secagem de polpa de frutas em leito de jorro. Determinou-se o ângulo de repouso das partículas, com e sem adição de água e de polpa de diversas frutas tropicais. Correlacionou-se a escoabilidade com as propriedades das partículas e com a composição química das polpas. Analisou-se a influência do ângulo de repouso sobre o desempenho do secador, no que se refere à produção. Os resultados mostraram que, em geral, as polpas com elevadas concentrações de gordura e sólidos insolúveis e baixos teores de açúcares redutores, facilitam a escoabilidade. Uma análise dos resultados obtidos na secagem de polpa de frutas tropicais, utilizando-se partículas de poliestireno de baixa densidade, como material inerte, mostrou que, embora a escoabilidade permita a obtenção de menores vazões de jorro mínimo, em relação ao desempenho do secador, pode não favorecer uma produção maior de pó.47548

    Absence of Fas-L aggravates renal injury in acute Trypanosoma cruzi infection

    Trypanosoma cruzi infection induces diverse alterations in immunocompetent cells and organs, myocarditis and congestive heart failure. However, the physiological network of disturbances imposed by the infection has not been addressed thoroughly. Regarding myocarditis induced by the infection, we observed in our previous work that Fas-L-/- mice (gld/gld) have very mild inflammatory infiltration when compared to BALB/c mice. However, all mice from both lineages die in the early acute phase. Therefore, in this work we studied the physiological connection relating arterial pressure, renal function/damage and cardiac insufficiency as causes of death. Our results show that a broader set of dysfunctions that could be classified as a cardio/anaemic/renal syndrome is more likely responsible for cardiac failure and death in both lineages.     Regulatory T Cells Phenotype in Different Clinical Forms of Chagas' Disease

    CD25High CD4+ regulatory T cells (Treg cells) have been described as key players in immune regulation, preventing infection-induced immune pathology and limiting collateral tissue damage caused by vigorous anti-parasite immune response. In this review, we summarize data obtained by the investigation of Treg cells in different clinical forms of Chagas' disease. Ex vivo immunophenotyping of whole blood, as well as after stimulation with Trypanosoma cruzi antigens, demonstrated that individuals in the indeterminate (IND) clinical form of the disease have a higher frequency of Treg cells, suggesting that an expansion of those cells could be beneficial, possibly by limiting strong cytotoxic activity and tissue damage. Additional analysis demonstrated an activated status of Treg cells based on low expression of CD62L and high expression of CD40L, CD69, and CD54 by cells from all chagasic patients after T. cruzi antigenic stimulation. Moreover, there was an increase in the frequency of the population of Foxp3+ CD25HighCD4+ cells that was also IL-10+ in the IND group, whereas in the cardiac (CARD) group, there was an increase in the percentage of Foxp3+ CD25High CD4+ cells that expressed CTLA-4. These data suggest that IL-10 produced by Treg cells is effective in controlling disease development in IND patients. However, in CARD patients, the same regulatory mechanism, mediated by IL-10 and CTLA-4 expression is unlikely to be sufficient to control the progression of the disease. These data suggest that Treg cells may play an important role in controlling the immune response in Chagas' disease and the balance between regulatory and effector T cells may be important for the progression and development of the disease. Additional detailed analysis of the mechanisms on how these cells are activated and exert their function will certainly give insights for the rational design of procedure to achieve the appropriate balance between protection and pathology during parasite infections
