    O impacto da poliquimioterapia no perfil epidemiológico da hanseníase em Juiz de Fora, Brasil

    We investigated the impact of multidrug therapy (MDT) on the epidemiological pattern of leprosy in Juiz de Fora, Brazil, from 1978 to 1995. Evaluation of 1,283 medical charts was performed according to the treatment regimen used in two different periods. Following the introduction of MDT in 1987, prevalence of leprosy decreased from 22 patients/10,000 inhabitants to 5.2 patients/10,000 inhabitants in 1995. Incidence rate of leprosy was lower in period II (1987-1995) than in period I (1978-1986). Decreasing prevalence and incidence appear to be related to drug efficacy rather than decreased case identification, since both self-referred and professionally referred treatment increased markedly from period I to period II. For both periods, multibacillary leprosy was the most frequent clinical form of the disease (±68%), and the main infection risk factor identified was household contact. Leprosy is predominantly manifested in adults, but an increase in the number of very old and very young patients was observed in period II. The MDT program has been effective both in combating leprosy and in promoting awareness of the disease.Investigamos o impacto da poliquimioterapia (PQT) no perfil epidemiológico da hanseníase em Juiz de Fora, Brasil, de 1978 a 1995. Fizemos uma avaliação de 1.283 prontuários, de acordo com o esquema terapêutico adotado em dois diferentes períodos. Desde a introdução da PQT, em 1987, a prevalência da hanseníase caiu de 22 pacientes/dez mil habitantes para 5,2 pacientes/dez mil habitantes em 1995. A incidência da doença foi menor no período II (1987-1995) em comparação ao período I (1978-1986). A diminuição da prevalência e da incidência está mais relacionada à eficácia das drogas que a uma queda na identificação da infecção, já que tanto a procura espontânea quanto os encaminhamentos aumentaram significativamente do período I para o período II. Em ambos os períodos, a forma clínica mais freqüente foi a multibacilar (±68%), e o contato intradomicilicar foi o maior fator de risco conhecido associado à infecção. Um aumento de pacientes muito velhos ou muito novos foi observado no período II. Os resultados indicam que o esquema PQT tem sido eficaz no combate à hanseníase e tem aumentado a conscientização e o conhecimento da doença

    Mineral content and volatile profiling of Prunus avium L. (sweet cherry) y-Products from Fundão Region (Portugal)

    Large amounts of Prunus avium L. by-products result from sweet cherry production and processing. This work aimed to evaluate the mineral content and volatile profiling of the cherry stems, leaves, and flowers of the Saco cultivar collected from the Fundão region (Portugal). A total of 18 min erals were determined by ICP-MS, namely 8 essential and 10 non-essential elements. Phosphorus (P) was the most abundant mineral, while lithium (Li) was detected in trace amounts. Three different preparations were used in this work to determine volatiles: hydroethanolic extracts, crude extracts, and aqueous infusions. A total of 117 volatile compounds were identified using HS-SPME/GC MS, distributed among different chemical classes: 31 aldehydes, 14 alcohols, 16 ketones, 30 esters, 4 acids, 4 monoterpenes, 3 norisoprenoids, 4 hydrocarbons, 7 heterocyclics, 1 lactone, 1 phenol, and 2 phenylpropenes. Benzaldehyde, 4-methyl-benzaldehyde, hexanal, lilac aldehyde, and 6-methyl 5-hepten-2-one were the major volatile compounds. Differences in the types of volatiles and their respective amounts in the different extracts were found. This is the first study that describes the mineral and volatile composition of Portuguese sweet cherry by-products, demonstrating that they could have great potential as nutraceutical ingredients and natural flavoring agents to be used in the pharmaceutical, cosmetic, and food industries.info:eu-repo/semantics/publishedVersio

    Study of the optical properties of TeO2-PbO-TiO2 glass system

    We describe the preparation and some optical properties of high refractive index TeO2-PbO-TiO2 glass system. Highly homogeneous glasses were obtained by agitating the mixture during the melting process in an alumina crucible. The characterization was done by X-ray diffraction, Raman scattering, light absorption and linear refractive index measurements. The results show a change in the glass structure as the PbO content increases: the TeO4 trigonal bipyramids characteristics of TeO2 glasses transform into TeO3 trigonal pyramids. However, the measured refractive indices are almost independent of the glass composition.     Influence of drying temperature on the chemical constituents of jaboticaba (Plinia Jaboticaba (Vell.) Berg) skin

    Jaboticaba is a fruit native to Brazil. Its skin represents up to 43% of the fruit and contains high levels of fiber, minerals and phenolic compounds. The use of the skin waste adds value to the fruit. However, one of the drawbacks of skin storage is the high water content, which requires drying processes to preserve the skin without leading to the loss of nutrients and antioxidants. The influence of different drying temperatures on the levels of nutrients and antioxidants was investigated. Jaboticaba (Plinia jaboticaba (Vell.) Berg, genotype Sabará) skins were lyophilized or dried at three temperatures (30, 45, and 60ºC, using food dryers). The skins were then ground, stored (protected from light) and subjected to analysis of proximate composition, vitamin C, phytate, polyphenols, anthocyanins and antioxidant activity. The drying process had little effect on the proximate composition of the flour, presenting significant difference only for crude protein, fiber and non-nitrogenous extract. The greatest preservation of chemical constituents occurs in the lyophilized jaboticaba skins. Among the drying temperatures tested, however, the skins dried at 45 and60°C had more highly preserved nutritional substances and antioxidants

    Metabolic profiling and antibacterial activity of Eryngium pristis Cham. & Schltdl. - prospecting for its use in the treatment of bacterial infections

    Morbidity and mortality of the infected patients by multidrug-resistant bacteria have increased, emphasizing the urgency of fi ght for the discovery of new innovative antibiotics. In this sense, natural products emerge as valuable sources of bioactive compounds. Among the biodiversity, Eryngium pristis Cham. & Schltdl. (Apiaceae Lindl.) is traditionally used to treat thrush and ulcers of throat and mouth, as diuretic and emmenagogue, but scarcely known as an antimicrobial agent. With this context in mind, the goals of this study were to investigate the metabolic profi le and the antibacterial activity of ethanolic extract (EE-Ep) and hexane (HF-Ep), dichloromethane (DF-Ep), ethyl acetate (EAF-Ep) and butanol (BF-Ep) fractions from E. pristis leaves. Gas Chromatography-Mass Spectrometry (GC-MS) was performed to stablish the metabolic profi le and revealed the presence of 12 and 14 compounds in EAF-Ep and HF-Ep, respectively. β-selinene, spathulenol, globulol, 2-methoxy-4-vinylphenol, α-amyrin, β-amyrin, and lupeol derivative were some of phytochemicals identifi ed. The antibacterial activity was determined by Minimal Inhibitory Concentration (MIC) using the broth micro-dilution against eight ATCC® and fi ve methicillin-resistant Staphylococcus aureus (MRSA) clinical strains. HF-Ep was the most eff ective (MIC ≤ 5,000 μg/μL), being active against the largest part of tested Gram-positive and Gram-negative bacterial strains, including MRSA, with exception of Escherichia coli (ATCC 25922) and Pseudomonas aeruginosa (ATCC 9027) and (ATCC 27853). These results suggest that E. pristis is a natural source of bioactive compounds for the search of new antibiotics which can be an interesting therapeutic approach to recover patients mainly infected by MRSA strains.info:eu-repo/semantics/publishedVersio

Essa importante parceria tem o objetivo de contribuir para a formação continuada dos profissionais da educação brasileiros que atuam na área de alfabetização, somando-se aos vários esforços que têm sido envidados pelo Ministério da Educação (MEC) para elevar a qualidade dos processos de alfabetização no Brasil e, consequentemente, os seus resultados.     APROVEITAMENTO DO SUBPRODUTO DO PROCESSAMENTO DE PUPUNHA E AVALIAÇÃO DOS COMPOSTOS FENÓLICOS

    Introdução e objetivos: A pupunheira (Bactris gasipaes Kunth), é uma palmeira cultivada, principalmente, para produção de palmito. A industrialização gera grande quantidade de subprodutos, que contêm quantidades significativas de nutrientes e compostos importantes na alimentação. Seu aproveitamento é uma alternativa para diminuir o impacto ambiental e aumentar rentabilidade. Os fenólicos são compostos importantes, sua quantificação revela o poder antioxidante, qualidade do alimento e dos potenciais benefícios à saúde. O objetivo deste estudo, foi avaliar o teor de fenólicos totais do subproduto de pupunha in natura e de sua farinha. Metodologia: As bainhas residuais resultantes do processamento do palmito de pupunha, foram obtidas em indústria de palmito. O preparo da farinha foi realizado de acordo com o proposto por Simas et al. (2010)1, com modificações. O conteúdo de fenólicos totais dos extratos foi determinado de acordo com o método de Zielinski e Kozlowska (2000)2. Os resultados foram submetidos a análise de variança em Software SISVAR®, utilizando o Teste de Tuckey e o de t-Student, a 5% de significância. Resultados e discussões: Os resultados dos fenólicos totais das bainhas in natura, revelam que os extratos etéreo, etanólico e aquoso, apresentaram concentrações variáveis de polifenóis e diferiram estatisticamente (p<0,05). O extrato aquoso no resíduo in natura e na farinha (191,72 e 192,44 mg EAG.100g-1 amostra) apresentaram maiores teores de fenólicos, e foram diferentes estatisticamente (p<0,05). Destaca-se, ainda, o extrato etanólico da farinha e do resíduo in natura (192,37 e 191,15 mg EAG.100g-1 amostra), ao contrário do extrato etéreo (190,99 e 191,97 mg EAG.100g-1 amostra, respectivamente). Conclusões: A bainha residual de pupunha in natura possui quantidade significativa de compostos fenólicos, e o processamento aplicado para a obtenção de farinha promove a manutenção deste conteúdo

    The Combination of Gefitinib With ATRA and ATO Induces Myeloid Differentiation in Acute Promyelocytic Leukemia Resistant Cells

    In approximately 15% of patients with acute myeloid leukemia (AML), total and phosphorylated EGFR proteins have been reported to be increased compared to healthy CD34(+) samples. However, it is unclear if this subset of patients would benefit from EGFR signaling pharmacological inhibition. Pre-clinical studies on AML cells provided evidence on the pro-differentiation benefits of EGFR inhibitors when combined with ATRA or ATO in vitro. Despite the success of ATRA and ATO in the treatment of patients with acute promyelocytic leukemia (APL), therapy-associated resistance is observed in 5-10% of the cases, pointing to a clear need for new therapeutic strategies for those patients. In this context, the functional role of EGFR tyrosine-kinase inhibitors has never been evaluated in APL. Here, we investigated the EGFR pathway in primary samples along with functional in vitro and in vivo studies using several APL models. We observed that total and phosphorylated EGFR (Tyr992) was expressed in 28% and 19% of blast cells from APL patients, respectively, but not in healthy CD34(+) samples. Interestingly, the expression of the EGF was lower in APL plasma samples than in healthy controls. The EGFR ligand AREG was detected in 29% of APL patients at diagnosis, but not in control samples. In vitro, treatment with the EGFR inhibitor gefitinib (ZD1839) reduced cell proliferation and survival of NB4 (ATRA-sensitive) and NB4-R2 (ATRA-resistant) cells. Moreover, the combination of gefitinib with ATRA and ATO promoted myeloid cell differentiation in ATRA- and ATO-resistant APL cells. In vivo, the combination of gefitinib and ATRA prolonged survival compared to gefitinib- or vehicle-treated leukemic mice in a syngeneic transplantation model, while the gain in survival did not reach statistical difference compared to treatment with ATRA alone. Our results suggest that gefitinib is a potential adjuvant agent that can mitigate ATRA and ATO resistance in APL cells. Therefore, our data indicate that repurposing FDA-approved tyrosine-kinase inhibitors could provide new perspectives into combination therapy to overcome drug resistance in APL patients
