Bridging the gap to nontoxic fungal control
Doutoramento em Biologia - Instituto Superior de AgronomiaBlad is an edible, Lupinus cotyledon-derived 173 amino acid residue polypeptide which occurs
naturally and undissociated as part of a soluble 210 kDa oligomer, hereby termed Bladcontaining
oligomer (BCO). BCO is isolated during growth of Lupinus seedlings and is a
breakdown product of β-conglutin catabolism. In this work, BCO inherent, potent and wide
spectrum antifungal activity was demonstrated under in vitro and greenhouse conditions.
Several field trials showed that BCO exhibits an equal or better performance than the
conventional chemicals over a large variety of crops/fungal pathogens. BCO antifungal activity
was also showed to extend to human pathogens, including both yeasts and filamentous fungi.
In vitro tests demonstrated a higher efficacy on a molar basis when compared to the best
chemical fungicides tested, with an absence of mammal toxicity or genotoxicity. In addition to
the lectin and catalytic activities which allow BCO to target the fungal cell wall, this work
revealed other highly complex and multitarget mode(s) of action. BCO enters fungal cells
causing several morphological alterations and seems to essentially act through inhibition of
metal ion homeostasis resulting in an apoptotic cell death. In an attempt to provide a structural
characterization of Blad, data from electronic microscopy, dynamic light scattering, SAXS and
X-ray crystallography were obtained from heterologous expressed Blad, however the results
were not conclusive. Last but not least, this work also aimed at the development of a “super-
Blad”: a fusion protein consisting of Blad and two selected antibacterial peptides, SP10-5 and
Sub5. The resulting chimeras not only maintained a strong antibacterial activity but were also
able to retain the ability to inhibit the growth of both yeast and filamentous fungi.
In summary, the overall approach of this thesis considered phytopathogens and human
pathogens as belonging to the same scientific area and therefore aimed at producing a uniform
natural multitarget, step-stone antifungal solutionN/

TUBERCULOSIS IN CHILDREN: NEW FORMS OF DIAGNOSIS
Introdução: A tuberculose ainda é um grave problema de saúde pública em Portugal. Para diminuir o número de casos de tuberculose ativa em populações de baixa e intermédia incidência, é necessário um diagnóstico rápido e um tratamento eficaz. A prova tuberculínica é o método de rastreio recomendado, mas as suas limitações são conhecidas. Em 2001, foi aprovado o primeiro de diversos interferon gamma release assays, considerados úteis no diagnóstico de infeção latente por Mycobacterium tuberculosis, amplamente utilizados na abordagem da tuberculose nos adultos.
Objectivos: Sumarizar a informação disponível sobre os interferon gamma release assays, nomeadamente no que diz respeito à técnica; às vantagens no diagnóstico da infeção latente por Mycobacterium tuberculosis; à sensibilidade e especificidade quando aplicados à população pediátrica; à caracterização de fatores interferentes; e ao seu significado na monitorização do tratamento com antituberculosos.
Desenvolvimento: Os interferon gamma release assays são testes imunologicamente seletivos, desenvolvidos com base em antigénios do Mycobacterium tuberculosis. Têm sido aplicados na Pediatria, em regiões com diferentes taxas de prevalência de tuberculose, de forma a compará-los com a prova tuberculínica relativamente à sensibilidade e especificidade.
Conclusões: A utilização destes testes como forma de rastreio em Pediatria apresenta limitações. São necessários estu- dos a nível nacional que permitam mostrar de que forma a prova tuberculínica e os interferon gamma release assays se devem articular. Atualmente, os interferon gamma release assays ape- nas complementam a prova tuberculínica

A Pessoa em Situação Crítica (PSC) na transição do Desmame Ventilatório (DV)
experiencia um conjunto de adaptações necessárias perante os requisitos da
mudança do suporte ventilatório utilizando a tecnologia, para a respiração
espontanea.
Conceptualizar o DV como uma transição, permite caraterizá-lo como um processo
de mudança dinâmico e complexo, introduzindo temporalidade e sistematização,
considerando o impacto dessa mudança na condição de saúde da PSC e família. O
DV evidencia uma transição que progride de acordo com os padrões de respostas de
adaptação da PSC às exigências de cada etapa, revelando a suscetibilidade da PSC
ao risco de danos, de incidentes, ao sofrimento, e consequentemente revelando o
risco de uma transição que aumenta o grau de vulnerabilidade da PSC.
Considerando a natureza da problemática, tendo como foco de enfermagem a PSC
e família, em transição, o referencial teórico norteador do pensamento foi a teoria de
médio alcance de Afaf Meleis, a Teoria das Transições. A intervenção de enfermagem
surge como condição facilitadora de uma transição saudável, através da compreensão
abrangente da experiência da transição, perante a situação crítica da pessoa e família.
As intervenções centradas na natureza da transição e nos padrões de resposta da
PSC, têm como finalidade diminuir a vulnerabilidade da PSC através da prevenção e
redução do risco, e minimizar o impacto dos fatores de risco modificáveis na
experiência de transição.
A elaboração deste relatório expressa o percurso de desenvolvimento de
competências de mestre e de enfermeiro especialista, considerando as experiências,
as reflexões, as atividades desenvolvidas nos contextos de estágio, assim como as
competências adquiridas ao longo do curso de mestrado na área de especialização à
PSC. A realização de uma revisão integrativa da literatura, permitiu sustentar uma
prática baseada na evidência, e em simultâneo, com a reflexão e análise crítica sobre
as experiências vivenciadas na prática clínica, permitiu a intervenção especializada
de enfermagem
A equidade na indeminização dos danos não patrimoniais
This study examines the quantification of compensation for non-pecuniary
damage, awarded by means of judicial decisions based on equity, and seeks to verify
whether such calculation safeguards legal certainty and predictability when applying the
law, as well as whether it observes the principles of proportionality and equality.
Firstly, the limits for discretionary judgment permitted to the judge were
determined, by evaluating the criteria established under the law. Then, by examining the
grounds of the judicial decisions in cases that had been selected beforehand, this study
sought to detect operation modes in concrete considerations of equity used by judges.
The examination of the grounds on which these judicial decisions are based
permitted the comprehension of the calculation method used in each case and the
observation that the criteria of compensatory nature, such as the extent of the damage
and the respective consequences, assumed a primary role. Despite discrepancies in
viewpoints with regard to certain issues of law, the jurisprudence examined reveals that
great care is taken to consider the solutions reached in similar cases, in an attempt to
ensure that the different criteria applied in the quantification of compensation are given
uniform relevance. The comparison of decisions, reported to cases with similar legal
contours, did not reveal relevant discrepancies in the calculation criteria used, nor are
they disproportionate regarding the amount of compensation awarded, which means that
resorting to equity, in determining the compensation to be awarded due to nonpecuniary
damage, does not jeopardize legal certainty or predictability when applying
the law, and observes the principle of proportionality, which is anchored in the
constitutional principle of equality.
The study performed, led to the conclusion that the grounds on which judicial
decisions are based, by itemising the elements which are taken into account and the
criteria adopted by the judge, allow these to be taken into consideration in similar cases,
contributing towards uniform interpretation and application of the law, ensuring legal
certainty and predictability when resorting to equity while quantifying compensation.O presente trabalho incide sobre a quantificação da indemnização por danos não patrimoniais, operada em decisão judicial proferida com recurso à equidade, e tem o propósito de verificar se tal cálculo salvaguarda a certeza e a segurança na aplicação do direito, bem como se respeita os princípios da proporcionalidade e da igualdade.
Procedeu-se à determinação dos limites da margem de discricionariedade
conferida ao julgador, mediante a apreciação dos critérios estatuídos no direito positivo.
De seguida, através da análise da fundamentação de decisões judiciais, proferidas num âmbito previamente definido de casos, procurou-se detetar modos de funcionamento do concreto juízo de equidade utilizados pelo julgador.
A apreciação da fundamentação dessas decisões permitiu conhecer o método de cálculo em cada caso utilizado e constatar que os critérios de natureza compensatória, como sejam a extensão do dano e respetivas consequências, assumiram clara primazia.
Apesar de existirem entendimentos divergentes quanto a determinadas questões de direito, a jurisprudência analisada revela uma preocupação constante com a consideração das soluções dadas a casos análogos, numa procura de uniformização da relevância a atribuir aos diversos critérios de cálculo da indemnização. O cotejo entre decisões, reportadas a casos com contornos jurídicos similares, não evidenciou divergências relevantes nos critérios de cálculo utilizados, nem desproporção nos valores indemnizatórios obtidos, o que permitiu considerar que o recurso à equidade, na determinação da indemnização por danos não patrimoniais, não contende com a certeza e a segurança na aplicação do direito e respeita o princípio da proporcionalidade que integra o princípio constitucional da igualdade.
O estudo realizado permitiu concluir que a fundamentação das decisões judiciais, ao discriminar os elementos atendidos e os critérios seguidos pelo julgador, permite a respetiva consideração em casos análogos e, assim, contribui para a uniformização na interpretação e na aplicação do direito e confere certeza e segurança ao recurso à equidade na quantificação indemnizatória
O design de calçado como ponte entre a cultura produtiva e a cultura urbana
Mestrado em Design Integrado na Escola Superior de Tecnologia e Gestão do Instituto Politécnico de Viana do CasteloA presente investigação insere-se no âmbito do design de calçado e tem
como objetivo propor uma nova abordagem no presente sector. O projeto
baseia-se no desenvolvimento e materialização de um produto na área do
calçado, de forma a integrar a produção local em conexão com as novas
tecnologias emergentes, com o intuito de enaltecer e explorar um projeto que
pensa local para agir global. O estudo assume, então, como referência
projetual, a cultura urbana underground fundamentada nos comportamentos
sociais atuais. Para tal, o presente documento subdividisse em 4 partes
essenciais para uma correta e eficaz análise contextual e produtiva.
Na primeira parte demonstram-se os valores e as potencialidades inerentes à
industria do calçado, de forma a dinamizar a sua capacidade produtiva em
interação com novas abordagens técnicas e materiais.
Na segunda parte salienta-se a importância do design enquanto agente
impulsionador de novas descobertas e valores para além do contexto
nacional. Deste modo, destaca-se a importância do Coolhunting como
metodologia capaz de gerar uma nova abordagem a ser implementada na
industria do calçado e, consequentemente, na disciplina do design.
Na terceira parte interpreta-se o design como um processo experimental ao
nível de materiais e de tecnologias e para ser materializado segundo os
valores inerentes à indústria do calçado.
Por último, a quarta parte, refere-se à materialização final de um modelo de
calçado, por parte da empresa Project ID, de forma a enaltecer e a dinamizar
os valores inerentes ao respetivo sector.
Com a presente investigação pretende-se demonstrar a importância da
ligação entre diferentes indústrias de forma a originar a partilha de
conhecimentos com base numa criatividade empresarial sustentável, dando
origem a um novo produto fruto da experimentação inerente às distintas
áreas do designThe current investigation fits into the shoe design scope and has the goal to
propose a new approach to the current field. The project is based on the
development and materialization of a product inserted in the show industry, so
that in can integrate the local production in connection with the emerging new
technologies, aiming to amplify and explore a project that thinks local to act
global.
Therefore, the study assumes the urban underground culture grounded in the
new social behaviors as projetual reference. To do so, the document is
divided in 4 essential parts for an effective productive and contextual analysis.
In the first part it demonstrates the values and capabilities related to the shoe
industry, in a way to streamline its productive capacity in interaction with new
technical and material approaches.
In the second part it is highlighted the design importance while boosting driver
of new discovers and values behind the national context. Thus, it is
highlighted the importance of Coolhunting as methodology, capable of
generating a new approach to be implemented in the shoe industry and,
therefore, in the design discipline.
In the third part design is interpreted as an experimental process regarding
the materials and technologies, also to be materialized according to its values
related to shoe industry.
Lastly, the fourth part, it refers to the final materialization of shoe prototype, by
the company Project ID, so that the values related to the respective field are
expressed.
With the current investigation it is intended to demonstrate the importance of
connecting the opposite sectors and industries, creating a share of know-how
based in the sustainable corporate creativity, giving birth to a new product, as
a result of an experimentation related to both fields of application.

Fusion proteins towards fungi and bacteria in plant protection
Research ArticleIn agriculture, although fungi are considered the foremost problem, infections by bacteria also cause significant economical
losses. The presence of different diseases in crops often leads to a misuse of the proper therapeutic, or the combination of
different diseases forces the use of more than one pesticide. This work concerns the development of a ‘super-Blad’: a
chimeric protein consisting of Blad polypeptide, the active ingredient of a biological fungicide already on the market, and two
selected peptides, SP10-5 and Sub5, proven to possess biological potential as antibacterial agents. The resulting chimeric
protein obtained from the fusion of Blad with SP10-5 not only maintained strong antibacterial activity, especially against
Xanthomonas spp. and Pseudomonas syringae, but was also able to retain the ability to inhibit the growth of both yeast and
filamentous fungi. However, the antibacterial activity of Sub5 was considerably diminished when fused with Blad, which
seems to indicate that not all fusion proteins behave equally. These newly designed drugs can be considered promising
compounds for use in plant protection. A deeper and focused development of an appropriate formulation may result in a

Autobiographical narratives in Williams Syndrome: structural, process and content dimensions
Williams syndrome (WS) is a neurodevelopmental disorder, with an
intriguing cognitive phenotype. Previous studies found support for an atypical
profile of fictional narrative production in WS (Gonçalves et al. The British Journal
of Developmental Disabilities, 56(111), 89-109, 2010). This study aimed at testing if
the same profile is observed when individuals with WS tell autobiographical
narratives. Using a new scoring system, structural (coherence), process (complexity)
and content (multiplicity) aspects of autobiographical narrative production were
analyzed in WS. Results suggest that individuals with WS produce autobiographical
narratives that are significantly less coherent and complex than typically developing
controls, although similar to controls in terms of content diversity. These findings
point out to deficits in autobiographical narrative production in WS, suggesting the
relative preservation of the social dimension of narrative production. These results
also support the hypothesis that fictional and autobiographical narrative production

m5U54 tRNA hypomodification by lack of TRMT2A drives the generation of tRNA-derived small RNAs
Transfer RNA (tRNA) molecules contain various post-transcriptional modifications that are crucial for tRNA stability, translation efficiency, and fidelity. Besides their canonical roles in translation, tRNAs also originate tRNA-derived small RNAs (tsRNAs), a class of small non-coding RNAs with regulatory functions ranging from translation regulation to gene expression control and cellular stress response. Recent evidence indicates that tsRNAs are also modified, however, the impact of tRNA epitranscriptome deregulation on tsRNAs generation is only now beginning to be uncovered. The 5-methyluridine (m5U) modification at position 54 of cytosolic tRNAs is one of the most common and conserved tRNA modifications among species. The tRNA methyltransferase TRMT2A catalyzes this modification, but its biological role remains mostly unexplored. Here, we show that TRMT2A knockdown in human cells induces m5U54 tRNA hypomodification and tsRNA formation. More specifically, m5U54 hypomodification is followed by overexpression of the ribonuclease angiogenin (ANG) that cleaves tRNAs near the anticodon, resulting in accumulation of 5′tRNA-derived stress-induced RNAs (5′tiRNAs), namely 5′tiRNA-GlyGCC and 5′tiRNA-GluCTC, among others. Additionally, transcriptomic analysis confirms that down-regulation of TRMT2A and consequently m5U54 hypomodification impacts the cellular stress response and RNA stability, which is often correlated with tiRNA generation. Accordingly, exposure to oxidative stress conditions induces TRMT2A down-regulation and tiRNA formation in mammalian cells. These results establish a link between tRNA hypomethylation and ANG-dependent tsRNAs formation and unravel m5U54 as a tRNA cleavage protective mark.publishe
