    Strain evolution in GaN Nanowires: from free-surface objects to coalesced templates

    Top-down fabricated GaN nanowires, 250 nm in diameter and with various heights, have been used to experimentally determine the evolution of strain along the vertical direction of 1-dimensional objects. X-ray diffraction and photoluminescence techniques have been used to obtain the strain profile inside the nanowires from their base to their top facet for both initial compressive and tensile strains. The relaxation behaviors derived from optical and structural characterizations perfectly match the numerical results of calculations based on a continuous media approach. By monitoring the elastic relaxation enabled by the lateral free-surfaces, the height from which the nanowires can be considered strain-free has been estimated. Based on this result, NWs sufficiently high to be strain-free have been coalesced to form a continuous GaN layer. X-ray diffraction, photoluminescence and cathodoluminescence clearly show that despite the initial strain-free nanowires template, the final GaN layer is strained

    Molecular gas and dust in Arp 94: The formation of a recycled galaxy in an interacting system

    We present new results for the molecular gas, dust emission and the ionized gas in J1023+1952, an HI rich intergalactic star-forming tidal dwarf galaxy candidate. It is located at the projected intersection of two faint stellar tidal streams wrapped around the interacting pair of galaxies NGC 3227/6 (Arp~94). Using the IRAM 30m telescope, emission from CO(1-0) and CO(2-1) was detected across the entire extent of the neutral hydrogen cloud associated with J1023+1952, a region of the size of 8.9x5.9kpc,aswellasinthenuclearregionandouterdiskofNGC3227.ThemoleculargasisfoundtobeabundantovertheentireHIcloud,withH2toHIgasmassratiosbetween0.5and1.7.NewSpitzermidinfraredobservationsat3.6,4.5,5.8,8.0,15and24mushowthatyoungSFisrestrictedtothesouthernpartofthecloud.DespitetherelativelyuniformH x 5.9 kpc, as well as in the nuclear region and outer disk of NGC 3227. The molecular gas is found to be abundant over the entire HI cloud, with H2-to-HI gas mass ratios between 0.5 and 1.7. New Spitzer mid-infrared observations at 3.6, 4.5, 5.8, 8.0, 15 and 24mu show that young SF is restricted to the southern part of the cloud. Despite the relatively uniform H_2andHIcolumndensityacrossthecloud,youngSFoccursonlywherethevelocitydispersionintheCOandHIisafactorof and HI column density across the cloud, young SF occurs only where the velocity dispersion in the CO and HI is a factor of \simtwo lower (FWHM of 30 - 70 \kms) than elsewhere in the cloud (FWHM of 80 - 120 \kms). Thus the kinematics of the gas, in addition to its column density, seems to be a crucial factor in triggering SF. Optical/infrared spectral energy distributions (SEDs) and Halpha photometry confirm that all the knots are young, with a tentative age sequence from the south-west (oldest knots) to the north-east (youngest). Optical spectroscopy of the brightest SF region allowed us to determine the metallicity (12+log(O/H) = 8.6 +- 0.2) and the extinction (A_B$=2.4). This shows that J1023+1952 is made from metal-enriched gas which is inconsistent with the hypothesis that it represents a pre-existing dwarf galaxy. Instead, it must be formed from recycled, metal-enrichd gas, expelled from NGC 3227 or NGC 3226 in a previous phase of the interaction.Comment: accepted for publication in ApJ, 20 pages, 11 figure

    Quantum well engineering in InGaN/GaN core-shell nanorod structures

    We report the ability to control relative InN incorporation in InGaN/GaN quantum wells (QWs) grown on the semi-polar and non-polar facets of a core-shell nanorod LED structure by varying the growth conditions. A study of the cathodoluminescence emitted from series of structures with different growth temperatures and pressures for the InGaN QW layer revealed that increasing the growth pressure had the effect of increasing InN incorporation on the semi-polar facets, while increasing the growth temperature improves the uniformity of light emission from the QWs on the non-polar facets.</p

    Telomere lengths in human oocytes, cleavage stage embryos and blastocysts

    Telomeres are repeated sequences that protect the ends of chromosomes and harbour DNA-repair proteins. Telomeres shorten during each cell division in the absence of telomerase. When telomere length becomes critically short, cell senescence occurs. Telomere length therefore reflects both cellular ageing and capacity for division. We have measured telomere length in human germinal vesicle (GV) oocytes and pre-implantation embryos, by quantitative fluorescence in-situ hybridisation (Q-FISH), providing baseline data towards our hypothesis that telomere length is a marker of embryo quality. The numbers of fluorescent foci suggest that extensive clustering of telomeres occurs in mature GV stage oocytes, and in pre-implantation embryos. When calculating average telomere length by assuming that each signal presents one telomere, the calculated telomere length decreased from the oocyte to the cleavage stages, and increased between the cleavage stages and the blastocyst (11.12 vs 8.43 vs 12.22kb respectively, p<0.001). Other methods of calculation, based upon expected maximum and minimum numbers of telomeres, confirm that telomere length in blastocysts is significantly longer than cleavage stages. Individual blastomeres within an embryo showed substantial variation in calculated average telomere length. This study implies that telomere length changes according to the stage of pre-implantation embryo development

    Reflectionless tunneling in ballistic normal-metal--superconductor junctions

    We investigate the phenomenon of reflectionless tunneling in ballistic normal-metal--superconductor (NS) structures, using a semiclassical formalism. It is shown that applied magnetic field and superconducting phase difference both impair the constructive interference leading to this effect, but in a qualitatively different way. This is manifested both in the conductance and in the shot noise properties of the system considered. Unlike diffusive systems, the features of the conductance are sharp, and enable fine spatial control of the current, as well as single channel manipulations. We discuss the possibility of conducting experiments in ballistic semiconductor-superconductor structures with smooth interfaces and some of the phenomena, specific to such structures, that could be measured. A general criterion for the barrier at NS interfaces, though large, to be effectively transparent to pair current is obtained.Comment: published versio

    A novel hotspot specific isothermal amplification method for detection of the common PIK3CA p.H1047R breast cancer mutation

    Breast cancer (BC) is a common cancer in women worldwide. Despite advances in treatment, up to 30% of women eventually relapse and die of metastatic breast cancer. Liquid biopsy analysis of circulating cell-free DNA fragments in the patients’ blood can monitor clonality and evolving mutations as a surrogate for tumour biopsy. Next generation sequencing platforms and digital droplet PCR can be used to profile circulating tumour DNA from liquid biopsies; however, they are expensive and time consuming for clinical use. Here, we report a novel strategy with proof-of-concept data that supports the usage of loop-mediated isothermal amplification (LAMP) to detect PIK3CA c.3140 A > G (H1047R), a prevalent BC missense mutation that is attributed to BC tumour growth. Allele-specific primers were designed and optimized to detect the p.H1047R variant following the USS-sbLAMP method. The assay was developed with synthetic DNA templates and validated with DNA from two breast cancer cell-lines and two patient tumour tissue samples through a qPCR instrument and finally piloted on an ISFET enabled microchip. This work sets a foundation for BC mutational profiling on a Lab-on-Chip device, to help the early detection of patient relapse and to monitor efficacy of systemic therapies for personalised cancer patient management

    Persistence of anticancer activity in berry extracts after simulated gastrointestinal digestion and colonic fermentation

    Fruit and vegetable consumption is associated at the population level with a protective effect against colorectal cancer. Phenolic compounds, especially abundant in berries, are of interest due to their putative anticancer activity. After consumption, however, phenolic compounds are subject to digestive conditions within the gastrointestinal tract that alter their structures and potentially their function. However, the majority of phenolic compounds are not efficiently absorbed in the small intestine and a substantial portion pass into the colon. We characterized berry extracts (raspberries, strawberries, blackcurrants) produced by in vitro-simulated upper intestinal tract digestion and subsequent fecal fermentation. These extracts and selected individual colonic metabolites were then evaluated for their putative anticancer activities using in vitro models of colorectal cancer, representing the key stages of initiation, promotion and invasion. Over a physiologically-relevant dose range (0–50 µg/ml gallic acid equivalents), the digested and fermented extracts demonstrated significant anti-genotoxic, anti-mutagenic and anti-invasive activity on colonocytes. This work indicates that phenolic compounds from berries undergo considerable structural modifications during their passage through the gastrointestinal tract but their breakdown products and metabolites retain biological activity and can modulate cellular processes associated with colon cancer

    The prevalence and incidence of mental ill-health in adults with autism and intellectual disabilities

    The prevalence, and incidence, of mental ill-health in adults with intellectual disabilities and autism were compared with the whole population with intellectual disabilities, and with controls, matched individually for age, gender, ability-level, and Down syndrome. Although the adults with autism had a higher point prevalence of problem behaviours compared with the whole adult population with intellectual disabilities, compared with individually matched controls there was no difference in prevalence, or incidence of either problem behaviours or other mental ill-health. Adults with autism who had problem behaviours were less likely to recover over a two-year period than were their matched controls. Apparent differences in rates of mental ill-health are accounted for by factors other than autism, including Down syndrome and ability level
