    A common garden design reveals population-specific variability in potential impacts of hybridisation between populations of farmed and wild Atlantic salmon, Salmo salar L

    Released individuals can have negative impacts on native populations through various mechanisms; including competition, disease transfer and introduction of maladapted gene-complexes. Previous studies indicate that the level of farmed Atlantic salmon introgression in native populations is population-specific. However few studies have explored the potential role of population diversity or river characteristics, such as temperature, on the consequences of hybridisation. We compared freshwater growth of multiple families derived from two farmed, five wild, and two F1 hybrid salmon populations at three contrasting temperatures (7°C, 12°C, and 16°C) in a common garden experiment. As expected, farmed salmon outgrew wild salmon at all temperatures, with hybrids displaying intermediate growth. However, differences in growth were population-specific and some wild populations performed better than others relative to the hybrid and farmed populations at certain temperatures. Therefore, the competitive balance between farmed and wild salmon may depend both on the thermal profile of the river and the genetic characteristics of the respective farmed and wild strains. While limited to F1 hybridisation, the present study shows the merits in adopting a more complex spatially resolved approach to risk management of local populations

    Deposition of ammonium and nitrate in the roots of maize seedlings supplied with different nitrogen salts

    This study measured total osmolarity and concentrations of NH4+, NO3–, K+, soluble carbohydrates, and organic acids in maize seminal roots as a function of distance from the apex, and NH4+ and NO3– in xylem sap for plants receiving NH4+ or NO3– as a sole N-source, NH4+ plus NO3–, or no nitrogen at all. The disparity between net deposition rates and net exogenous influx of NH4+ indicated that growing cells imported NH4+ from more mature tissue, whereas more mature root tissues assimilated or translocated a portion of the NH4+ absorbed. Net root NO3– influx under Ca(NO3)2 nutrition was adequate to account for pools found in the growth zone and provided twice as much as was deposited locally throughout the non-growing tissue. In contrast, net root NO3– influx under NH4NO3 was less than the local deposition rate in the growth zone, indicating that additional NO3– was imported or metabolically produced. The profile of NO3– deposition rate in the growth zone, however, was similar for the plants receiving Ca(NO3)2 or NH4NO3. These results suggest that NO3– may serve a major role as an osmoticant for supporting root elongation in the basal part of the growth zone and maintaining root function in the young mature tissues

    Moral Disengagement Mechanisms Predict Cyber Aggression Among Emerging Adults

    The Internet has given rise to many new communication tools (e.g., social media, text messaging), which, while beneficial in many respects, have become a means for aggressing against others. As evidence of the adverse correlates of cyber aggression mounts, improved understanding of the mechanisms that facilitate electronic aggression is needed. Moral disengagement (i.e., cognitive processes through which individuals disengage from their moral values) has been shown to predict cyber aggression when assessed as a unitary construct. The present study investigated the eight moral disengagement mechanisms measured by the Moral Disengagement Measure (Detert et al., 2008) and their relationships to four types of cyber aggression perpetration assessed with the Cyberbullying Experiences Survey (i.e., malice, public humiliation, deception, and unwanted contact; Doane et al., 2013). Emerging adults (N = 404, 58.67% women) aged 18 to 29 (M = 25.16, SD = 2.76) recruited through Amazon.com’s MTurk website completed measures online, and data were analyzed via path analysis. Each type of cyber aggression perpetration was predicted by different moral disengagement mechanisms. Advantageous comparison and dehumanization were the strongest predictors, and dehumanization was the only mechanism to predict all forms of cyber aggression. These findings provide support for the role of these mechanisms in cyber aggression and suggest that examining moral disengagement mechanisms individually may help to improve our understanding of cyber aggression among emerging adults. Further clinical and research implications are discussed

    Assailants or Saints?: Racial, Ethnic, and Gender Depictions on a Social Media Based City News Website

    A large literature indicates that Black males are overrepresented as criminals in traditional newspapers and broadcast news. However, little scholarly attention has been paid to online-only city newspapers. The authors conducted a content analysis of a social media based city news website in a Southeastern state. Multiple coders assessed 8,142 stories that ran over the course of three years and found that, in line with previous research, Black males were disproportionately portrayed as criminals, were more likely to have mugshots accompanying their stories, and were more likely to have their race mentioned in the text of the story than any other demographic group. Furthermore, the website interface design exacerbated the portrayal of Black male criminality. The authors also found that White females were most likely to be portrayed as philanthropists and award winners. Our results offer strong support for scapegoat and power structure theories, and limited support for racial threat and market share theories. We argue that intersectional theoretical and methodological approaches are necessary to understand media portrayals of race/ethnicity, gender, social class, and other important social characteristics

    Calcein Staining as a Tool to Investigate Coccolithophore Calcification

    Despite the oceanographic and geological significance of coccolithophores, the cellular mechanisms that underlie the intracellular production and subsequent secretion of their CaCO3 coccoliths remain poorly understood. Tools for labeling coccoliths and coccospheres in order to track their production would be of great value. We therefore evaluated the use of calcein, a derivative of fluorescein, as a method to fluorescently label coccoliths. The calcein method readily labeled pre-existing coccospheres in a range of coccolithophore species, including diploid and haploid life history phases, without compromising the coccolith structure. Calcite staining was verified though epifluorescence and confocal microscopy, and both stained and unstained cells and coccoliths were readily distinguished using flow cytometry. The fluorescence of stained coccoliths was retained for >3 days allowing us to confirm their polar secretion by distinguishing pre-existing coccoliths from the accumulation and distribution of non-fluorescent coccoliths produced after calcein exposure. The calcein treatment had no significant effect on photosynthetic physiology, external calcite morphology, or growth rates of the cells over an 8-day period. The calcein staining method therefore represents a simple non-invasive, non-toxic optical technique to ‘tag’ calcium carbonate coccoliths and track their production in response to environmental manipulations or pharmacological treatments. Moreover, calcein staining of the coccosphere allowed for heterogenous patterns of calcification, growth, and cell division to be detected in a population of cells. This is the first description of the use of calcein to stain the biomineral structures of calcifying phytoplankton and this approach has the potential to be applied to detailed cytological investigations as well as high-throughput analysis of cultured cells or field populations

    Protein fiber linear dichroism for structure determination and kinetics in a low-volume, low-wavelength couette flow cell

    High-resolution structure determination of soluble globular proteins relies heavily on x-ray crystallography techniques. Such an approach is often ineffective for investigations into the structure of fibrous proteins as these proteins generally do not crystallize. Thus investigations into fibrous protein structure have relied on less direct methods such as x-ray fiber diffraction and circular dichroism. Ultraviolet linear dichroism has the potential to provide additional information on the structure of such biomolecular systems. However, existing systems are not optimized for the requirements of fibrous proteins. We have designed and built a low-volume (200 μL), low-wavelength (down to 180 nm), low-pathlength (100 μm), high-alignment flow-alignment system (couette) to perform ultraviolet linear dichroism studies on the fibers formed by a range of biomolecules. The apparatus has been tested using a number of proteins for which longer wavelength linear dichroism spectra had already been measured. The new couette cell has also been used to obtain data on two medically important protein fibers, the all-β-sheet amyloid fibers of the Alzheimer's derived protein Aβ and the long-chain assemblies of α1-antitrypsin polymers

    The epigenetic clock is correlated with physical and cognitive fitness in the Lothian Birth Cohort 1936

    Background: The DNA methylation-based 'epigenetic clock' correlates strongly with chronological age, but it is currently unclear what drives individual differences. We examine cross-sectional and longitudinal associations between the epigenetic clock and four mortality-linked markers of physical and mental fitness: lung function, walking speed, grip strength and cognitive ability. Methods: DNA methylation-based age acceleration (residuals of the epigenetic clock estimate regressed on chronological age) were estimated in the Lothian Birth Cohort 1936 at ages 70 (n=920), 73 (n=299) and 76 (n=273) years. General cognitive ability, walking speed, lung function and grip strength were measured concurrently. Cross-sectional correlations between age acceleration and the fitness variables were calculated. Longitudinal change in the epigenetic clock estimates and the fitness variables were assessed via linear mixed models and latent growth curves. Epigenetic age acceleration at age 70 was used as a predictor of longitudinal change in fitness. Epigenome-wide association studies (EWASs) were conducted on the four fitness measures. Results: Cross-sectional correlations were significant between greater age acceleration and poorer performance on the lung function, cognition and grip strength measures (r range: -0.07 to -0.05, P range: 9.7 x 10 to 0.024). All of the fitness variables declined over time but age acceleration did not correlate with subsequent change over 6 years. There were no EWAS hits for the fitness traits. Conclusions: Markers of physical and mental fitness are associated with the epigenetic clock (lower abilities associated with age acceleration). However, age acceleration does not associate with decline in these measures, at least over a relatively short follow-up

    Experiences of learning through collaborative evaluation from a masters programme in professional education

    This paper presents findings from a collaborative evaluation project within a masters programme in professional education. The project aimed to increase knowledge of research methodologies and methods through authentic learning where participants worked in partnership with the tutor to evaluate the module which they were studying. The project processes, areas of the course evaluated and the data collection methods are outlined. The findings focus on key themes from evaluating the effectiveness of using a collaborative evaluation approach, including: enhanced student engagement; creativity of the collaborative evaluation approach; equality between the tutor and students; and enhanced research skills. Discussion focuses on the outcomes and effectiveness of the project and tutor reflections on adopting a collaborative approach. This paper highlights lessons from the project relevant to those interested in staff-student partnership approaches and those facilitating postgraduate learning and teaching programmes and educational research courses
