    Galactic chemical evolution

    We analyze the evolution of oxygen abundance radial gradients resulting from our chemical evolution models calculated with different prescriptions for the star formation rate (SFR) and for the gas infall rate, in order to assess their respective roles in shaping gradients. We also compare with cosmological simulations and confront all with recent observational datasets, in particular with abundances inferred from planetary nebulae. We demonstrate the critical importance in isolating the specific radial range over which a gradient is measured, in order for their temporal evolution to be useful indicators of disk growth with redshift

    Event data recording is crucial in robotics research, providing prolonged insights into a robot's situational understanding, progression of behavioral state, and resulting outcomes. Such recordings are invaluable when debugging complex robotic applications or profiling experiments ex post facto. As robotic developments mature into production, both the roles and requirements of event logging will broaden, to include serving as evidence for auditors and regulators investigating accidents or fraud. Given the growing number of high profile public incidents involving self-driving automotives resulting in fatality and regulatory policy making, it is paramount that the integrity, authenticity and non-repudiation of such event logs are maintained to ensure accountability. Being mobile cyber-physical systems, robots present new threats, and vulnerabilities beyond traditional IT: unsupervised physical system access or postmortem collusion between robot and OEM could result in the truncation or alteration of prior records. In this letter, we address immutablization of log records via integrity proofs and distributed ledgers with special considerations for mobile and public service robot deployments

    Pharmacokinetics of Mephedrone Enantiomers in Whole Blood after a Controlled Intranasal Administration to Healthy Human Volunteers

    Mephedrone, which is one of the most popular synthetic cathinones, has one chiral centre and thus exists as two enantiomers: R-(+)-mephedrone and S-(−)-mephedrone. There are some preliminary data suggesting that the enantiomers of mephedrone may display enantioselective pharmacokinetics and exhibit different neurological effects. In this study, enantiomers of mephedrone were resolved via chromatographic chiral recognition and the absolute configuration was unambiguously determined by a combination of elution order and chiroptical analysis (i.e., circular dichroism). A chiral liquid chromatography tandem mass spectrometry method was fully validated and was applied to the analysis of whole blood samples collected from a controlled intranasal administration of racemic mephedrone hydrochloride to healthy male volunteers. Both enantiomers showed similar kinetics, however, R-(+)-mephedrone had a greater mean Cmax of 48.5 ± 11.9 ng/mL and a longer mean half-life of 1.92 ± 0.27 h compared with 44.6 ± 11.8 ng/mL and 1.63 ± 0.23 h for S-(−)-mephedrone, respectively. Moreover, R-(+)-mephedrone had a lower mean clearance and roughly 1.3 times greater mean area under the curve than S-(−)-mephedrone. Significant changes in the enantiomeric ratio over time were observed, which suggest that the analytes exhibit enantioselective pharmacokinetics. Even though the clinical significance of this finding is not yet fully understood, the study confirms that the chiral nature, and consequently the enantiomeric purity of mephedrone, can be a crucial consideration when interpreting toxicological results

    Antibody Recognition of Cancer-Related Gangliosides and Their Mimics Investigated Using in silico Site Mapping

    Modified gangliosides may be overexpressed in certain types of cancer, thus, they are considered a valuable target in cancer immunotherapy. Structural knowledge of their interaction with antibodies is currently limited, due to the large size and high flexibility of these ligands. In this study, we apply our previously developed site mapping technique to investigate the recognition of cancer-related gangliosides by anti-ganglioside antibodies. The results reveal a potential ganglioside-binding motif in the four antibodies studied, suggesting the possibility of structural convergence in the anti-ganglioside immune response. The structural basis of the recognition of ganglioside-mimetic peptides is also investigated using site mapping and compared to ganglioside recognition. The peptides are shown to act as structural mimics of gangliosides by interacting with many of the same binding site residues as the cognate carbohydrate epitopes. These studies provide important clues as to the structural basis of immunological mimicry of carbohydrates
