    Cytotoxic Activity of Non-specific Lipid Transfer Protein (nsLTP1) from Ajwain \u3cem\u3e(Trachyspermum ammi)\u3c/em\u3e Seeds

    Background Trachyspermum ammi, commonly known as Ajwain, is a member of the Apiaceae family. It is a therapeutic herbal spice with diverse pharmacological properties, used in traditional medicine for various ailments. However, all previous studies were conducted using small molecule extracts, leaving the protein’s bioactivity undiscovered. Aim The current study aimed to demonstrate the cytotoxic activity of Ajwain non-specific lipid transfer protein (nsLTP1) in normal breast (MCF10A), breast cancer (MCF-7), and pancreatic cancer (AsPC-1) cell lines. Also, to evaluate its structural stability in human serum as well as at high temperature conditions. Methods The cytotoxic activity of Ajwain nsLTP1 was evaluated in MCF-7 and AsPC-1 cell lines using MTT assay. Annexin V-FITC and PI staining were used to detect the early apoptotic and late apoptotic cells. The role of nsLTP1 in inducing apoptosis was further studied by quantifying Bcl-2, Bax, Caspase-3, Survivin, EGFR, and VEGF genes expression using RT-PCR. CD spectroscopy analyzed the nsLTP1 conformational changes after thermal treatment for structure stability determination. The RP-HPLC was used to analyze the nsLTP1 degradation rate in human serum at different time intervals incubated at 37 °C. Results Ajwain nsLTP1 showed a potent cytotoxic effect in MCF-7 and AsPC-1. The IC50 value obtained in MCF-7 was 8.21 μM, while for AsPC-1 4.17 μM. The effect of nsLTP1 on stimulating apoptosis revealed that the proportions of apoptotic cells in both cell lines were relatively increased depending on the concentration. The apoptotic cells percentage at 20 μM was in MCF-7 71% (***P \u3c 0.001) and AsPC-1 88% (***P \u3c 0.001). These results indicate that nsLTP1 might efficaciously induce apoptosis in multiple types of cancerous cells. Genes expression in MCF-7 and AsPC-1 showed significant upregulation in Bax and Caspase-3 and downregulation in Bcl-2, Survivin, EGFR, and VEGF protein. The CD analysis of nsLTP1 showed a significant thermostable property. In serum, nsLTP1 showed a slow degradation rate, indicating high stability with a half-life of ~ 8.4 h. Conclusion Our results revealed the potential anticancer activity of Ajwain nsLTP1 and its mechanism in inducing apoptosis. It further exhibited thermostable properties at high temperatures and in human serum, which suggested this protein as a promising anticancer agent

    Prevalence of Black Quarter in Association with Patho-Morphological Alterations in Affected Tissues at Different Zones of Punjab-Pakistan

    Background: Blackleg or Black Quarter (BQ) is a serious bacterial disease caused by Clostridium chauvoei. It causes edematous and gaseous changes in skeletal muscles of animals. The study was designed to find prevalence of BQ in 6 districts of Punjab, Pakistan from June 2018 to June 2019.Methods: Animals were randomly monitored, and selected in each union council regardless of age, sex and species, but lameness and hyperthermia were the selection parameters. A proforma was used to record the experimental readings of each animal. A total of 1500 swab samples from bovines (cattle and buffaloes) were processed through conventional PCR and culture sensitivity tests to examine the comparative diagnostic efficacy and suitability of the test.Results: Sensitivity and specificity of culture tests were 87.6% and 34.19% respectively, whereas PCR demonstrated 81% and 47.3% results for the said aspects, respectively. Alpha toxin gene (Ccta) was amplified at 52.2oC yielding an amplicon size of 1400 base pairs. Area wise and season wise prevalence of each animal was also determined. The prevalence of BQ in cattle was significantly higher (P 0.05) in Lahore (0%) and Nankana Sahib (8%) which belonged to the canal irrigated zones.Conclusion: There was higher prevalence of BQ in different areas of Punjab, Pakistan that show the higher risk of disease transmission. A systematic surveillance system is essential to regularly monitor the disease incidence and prevalence in these areas of Pakistan.Keywords: Black Quarter; Prevalence; Pathological; Ruminants;  Zone

    Climate change risk perception and adaptation to climate smart agriculture are required to increase wheat production for food security

    Climate change poses a serious risk to wheat farmers in many regions of the world. The present study was conducted in the Sialkot District, Punjab, Pakistan, to investigate climate change trends during the past thirty years and to determine farmers’ knowledge and perceptions about climate change. The study also addresses the impacts of climate change on wheat production, current adaptation strategies, and limitations in adaptations to climate-smart agriculture (CSA) through a questionnaire-based survey. The historical weather data from the past thirty years indicated an increase in the mean annual minimum and maximum temperature and a decrease in annual total precipitation. Wheat productivity during the past thirty years showed an increasing trend but it was inconsistent. The respondents’ perception of climate change indicated that the literate farmers and those with broad farming experience were more knowledgeable about the climatic effects on wheat production. However, the survey results showed that the age of the farmers did not affect their perceptions. The current management practices are primarily based on prior experiences (70%) and traditional practices (30%). The standard management practices to increase farm productivity include an increase in fertilizer use (70%), a decrease in manure use (24%), and intercropping or switching to other crop cultivations (60%). The farmers stated that their reasons for limited adaptation to climate smart farm practices (CSFP) were due to their lack of knowledge and skills (86%), lack of modern technologies (74%), economic constraints (78%), politics (86%), and social influences (74%). Based on the survey results, the study suggests that addressing these gaps can increase farm-level wheat productivity to increase resilience. This can be achieved by introducing stateof- the-art farming practices through farmer training and by providing institutional services with a focus on climate-specific farm consultation services, leading to climate-smart agricultural practices for improved food security. Highlights - Literate farmers are more aware of climate change as compared to illiterate farmers. - The farmers emphasized the increase in both the summer and winter temperature. - Rainfall is identified as a major climate threat in the study area. - The farmers identified that the highest impact of climate change occurred during the harvest phase of wheat. - The farmers stated that the limited adoption of climate smart agricultural practices is due to lack of knowledge and technological, economic, and other gaps

    Novel CNGA3 and CNGB3 mutations in two Pakistani families with achromatopsia

    PURPOSE: To identify the genetic defect in two Pakistani families with autosomal recessive achromatopsia. METHODS: Two families (RP26 and RP44) were originally diagnosed with retinal dystrophy based upon their medical history. To localize the causative genes in these families, homozygosity mapping was performed using Affymetrix 10K single nucleotide polymorphism (SNP) arrays. Sequence analysis was used to find the mutations in candidate genes cyclic nucleotide-gated channel alpha-3 (CNGA3; family RP26) and cyclic nucleotide-gated channel beta-3 (CNGB3; family RP44). Control individuals were analyzed by allele-specific PCR for the CNGA3 mutation and BstXI restriction analysis for the CNGB3 mutation. After genetic analysis, clinical diagnosis was re-evaluated by electroretinography and color vision testing. During the course of this study, selected affected members of family RP26 were given pink glasses as supportive therapy. RESULTS: Sequence analysis of the positional candidate genes identified a novel missense mutation in CNGA3 (c.822G>T; p.R274S) in family RP26, and a novel CNGB3 frameshift mutation (c.1825delG; p.V609WfsX9) in family RP44. Clinical re-evaluation after genetic analysis revealed that both families have segregating autosomal recessive achromatopsia. CONCLUSIONS: Genetic analysis of two Pakistani families with retinal disease enabled the establishment of the correct diagnosis of achromatopsia. Two novel mutations were identified in CNGA3 and CNGB3 that are both specifically expressed in cone photoreceptors. Re-evaluation of the clinical status revealed that both families had achromatopsia. The use of pink glasses in patients was helpful in reducing photophobia and enabled rod-mediated vision

    Prognostic model to predict postoperative acute kidney injury in patients undergoing major gastrointestinal surgery based on a national prospective observational cohort study.

    Background: Acute illness, existing co-morbidities and surgical stress response can all contribute to postoperative acute kidney injury (AKI) in patients undergoing major gastrointestinal surgery. The aim of this study was prospectively to develop a pragmatic prognostic model to stratify patients according to risk of developing AKI after major gastrointestinal surgery. Methods: This prospective multicentre cohort study included consecutive adults undergoing elective or emergency gastrointestinal resection, liver resection or stoma reversal in 2-week blocks over a continuous 3-month period. The primary outcome was the rate of AKI within 7 days of surgery. Bootstrap stability was used to select clinically plausible risk factors into the model. Internal model validation was carried out by bootstrap validation. Results: A total of 4544 patients were included across 173 centres in the UK and Ireland. The overall rate of AKI was 14·2 per cent (646 of 4544) and the 30-day mortality rate was 1·8 per cent (84 of 4544). Stage 1 AKI was significantly associated with 30-day mortality (unadjusted odds ratio 7·61, 95 per cent c.i. 4·49 to 12·90; P < 0·001), with increasing odds of death with each AKI stage. Six variables were selected for inclusion in the prognostic model: age, sex, ASA grade, preoperative estimated glomerular filtration rate, planned open surgery and preoperative use of either an angiotensin-converting enzyme inhibitor or an angiotensin receptor blocker. Internal validation demonstrated good model discrimination (c-statistic 0·65). Discussion: Following major gastrointestinal surgery, AKI occurred in one in seven patients. This preoperative prognostic model identified patients at high risk of postoperative AKI. Validation in an independent data set is required to ensure generalizability

    Prevalence and antibiotic resistance of Salmonella spp. in South Punjab-Pakistan.

    Present study aimed at investigating the magnitude of the prevalence and antibiotic resistance among four Salmonella spp. i.e., S. typhi, S. paratyphi A, S. paratyphi B and S. typhimurium. Raw milk and environment samples were collected from the five districts of southern part of the province of Punjab in Pakistan i.e., Multan, Bahawalpur, Lodhran, Dera Ghazi Khan and Muzaffargarh. Extent of antibiotic resistance was also determined and classified as resistant, intermediate and susceptible. District-wise prevalence data on Salmonella spp. in milk and environmental samples indicated higher S. typhi, S. paratyphi B and S. typhimurium count in Bahawalpur, D.G. Khan and Muzaffargarh districts, respectively. Amongst 13 tested antibiotics, chloramphenicol and ofloxacin were found to be the most susceptible against Salmonella spp. Increased emergence of antibacterial resistance was noted with respect to the type of antibiotics among Salmonella spp. isolates. The study suggests serious interventions to be practiced by the farmers and raw milk vendors in animal husbandry and milk marketing, respectively to curb the burden of Salmonella spp. prevalence in milk. Further, active engagement of animal health division and enforcement agencies to ensure sagacious use of antibiotics at farm level may also help in containment of antimicrobial resistance in Salmonella spp
