    Monte Carlo Simulation of the Three-dimensional Ising Spin Glass

    We study the 3D Edwards-Anderson model with binary interactions by Monte Carlo simulations. Direct evidence of finite-size scaling is provided, and the universal finite-size scaling functions are determined. Using an iterative extrapolation procedure, Monte Carlo data are extrapolated to infinite volume up to correlation length \xi = 140. The infinite volume data are consistent with both a continuous phase transition at finite temperature and an essential singularity at finite temperature.     MICROORGANISMS ON HUMAN-COMPUTER INTERFACES: AN EXAMINATION OF AUTOMATED TELLER MACHINES AND CYBERCAFÉS IN ILESA AND IJEBU-JESA, NIGERIA.

    This study examined ATMs and keyboards in public cybercafés of Ilesa & Ijebu-jesa, Nigeria for the presence of bacteria of health importance.Ten ATMs in Ilesa and 10 cybercafés each in Ilesa & Ijebu-jesa were sampled at random and cultured by standard procedures. Total bacterial counts and coliform counts were recorded. The susceptibility of isolates to commercially available antibiotics was assayed by the disk diffusion method.Total bacterial counts from ATM surfaces in Ilesa ranged from 2.80 x 105 CFU/Ml to 1.90 x 105 CFU/Ml and the mean count was 2.08 x 105 CFU/Ml. The mean heterotrophic count of keyboards from Ilesa was 2.01 x 105 CFU/Ml while the mean coliform count was 1.50 x  103 CFU/Ml. In Ijebu-jesa, the maximum bacterial count was observed on the 4th keyboard sampled; also the only keyboard from which coliform bacteria was isolated (1.00 x 103 CFU/Ml). The mean heterotrophic count of keyboards in this area was 2.42 x 105 CFU/Ml. Staphylococcus aureus made up 50 % of isolates from ATMs in Ilesa, 90 % of those from cybercafés in Ilesa and 20 % of cybercafés in Ijebu-jesa. Pseudomonas sp. was 10 % of isolates from ATMs surfaces in Ilesa and keyboards in Ijebu-jesa cybercafés and 30 % of those from keyboards in Ilesa. Isolates were 53 % susceptible to common antibiotics. The study concluded there was high presence of bacteria on the surfaces of ATMs and keyboards in Ilesa and Ijebu-jesa, some of which were potentially pathogenic and drug resistant

    Pioneers in CNS inhibition: 2. Charles Sherrington and John Eccles on inhibition in spinal and supraspinal structures

    This article reviews the contributions of the English neurophysiologist, Charles Scott Sherrington [1857–1952], and his Australian PhD trainee and collaborator, John Carew Eccles [1903–1997], to the concept of central inhibition in the spinal cord and brain. Both were awarded Nobel Prizes; Sherrington in 1932 for “discoveries regarding the function of neurons,” and Eccles in 1963 for “discoveries concerning the ionic mechanisms involved in excitation and inhibition in central portions of the nerve cell membrane.” Both spoke about central inhibition at their Nobel Prize Award Ceremonies. The subsequent publications of their talks were entitled “Inhibition as a coordinative factor” and “The ionic mechanism of postsynaptic inhibition”, respectively. Sherrington's work on central inhibition spanned 41 years (1893–1934), and for Eccles 49 years (1928–1977). Sherrington first studied central inhibition by observing hind limb muscle responses to electrical (peripheral nerve) and mechanical (muscle) stimulation. He used muscle length and force measurements until the early 1900s and electromyography in the late 1920s. Eccles used these techniques while working with Sherrington, but later employed extracellular microelectrode recording in the spinal cord followed in 1951 by intracellular recording from spinal motoneurons. This considerably advanced our understanding of central inhibition. Sherrington's health was poor during his retirement years but he nonetheless made a small number of largely humanities contributions up to 1951, one year before his death at the age of 94. In contrast, Eccles retained his health and vigor until 3 years before his death and published prolifically on many subjects during his 22 years of official retirement. His last neuroscience article appeared in 1994 when he was 91. Despite poor health he continued thinking about his life-long interest, the mind-brain problem, and was attempting to complete his autobiography in the last years of his life

    The structure of a resuscitation-promoting factor domain from Mycobacterium tuberculosis shows homology to lysozymes

    Resuscitation-promoting factor (RPF) proteins reactivate stationary-phase cultures of (G+C)-rich Gram-positive bacteria including the causative agent of tuberculosis, Mycobacterium tuberculosis. We report the solution structure of the RPF domain from M. tuberculosis Rv1009 (RpfB) solved by heteronuclear multidimensional NMR. Structural homology with various glycoside hydrolases suggested that RpfB cleaved oligosaccharides. Biochemical studies indicate that a conserved active site glutamate is important for resuscitation activity. These data, as well as the presence of a clear binding pocket for a large molecule, indicate that oligosaccharide cleavage is probably the signal for revival from dormancy

    Catalysis by hen egg-white lysozyme proceeds via a covalent intermediate

    Hen egg-white lysozyme (HEWL) was the first enzyme to have its three-dimensional structure determined by X-ray diffraction techniques(1). A catalytic mechanism, featuring a long-lived oxo-carbenium-ion intermediate, was proposed on the basis of model-building studies(2). The `Phillips' mechanism is widely held as the paradigm for the catalytic mechanism of beta -glycosidases that cleave glycosidic linkages with net retention of configuration of the anomeric centre. Studies with other retaining beta -glycosidases, however, provide strong evidence pointing to a common mechanism for these enzymes that involves a covalent glycosyl-enzyme intermediate, as previously postulated(3). Here we show, in three different cases using electrospray ionization mass spectrometry, a catalytically competent covalent glycosyl-enzyme intermediate during the catalytic cycle of HEWL. We also show the three-dimensional structure of this intermediate as determined by Xray diffraction. We formulate a general catalytic mechanism for all retaining beta -glycosidases that includes substrate distortion, formation of a covalent intermediate, and the electrophilic migration of C1 along the reaction coordinate

    Relationship between craving and personality in treatment-seeking women with substance-related disorders

    BACKGROUND: Individual differences may impact susceptibility to addiction. The impact of personality features on drug craving, however, has not been studied, particularly in women. METHODS: Ninety-five treatment-seeking women with substance dependence, abstinent for at least 5 and no more than 21 days, were investigated regarding the correlation between personality factors and craving. Personality was assessed using the Temperament and Character Inventory (TCI), the NEO Personality Inventory Revised (NEO-PI-R), and the Barratt Impulsiveness Scale version 11 (BIS-11). Cravings were assessed through the Pennsylvania Craving Scale (PCS), and the Craving Questionnaire (CQ). Anxiety and depressive symptomatology were also recorded. RESULTS: Craving scores were positively correlated with depression and negatively correlated with number of days abstinent from substance use. Also, craving scores were positively associated with the novelty-seeking factor from the TCI and the total score on the BIS-11, and negatively associated with the conscientiousness and agreeableness facets of the NEO-PI-R. CONCLUSION: Findings suggest that personality features, particularly impulsiveness, can be important predictors of craving in women, which has important implications for treatment planning
