    Design principles of hair-like structures as biological machines

    Hair-like structures are prevalent throughout biology and frequently act to sense or alter interactions with an organism's environment. The overall shape of a hair is simple: a long, filamentous object that protrudes from the surface of an organism. This basic design, however, can confer a wide range of functions, owing largely to the flexibility and large surface area that it usually possesses. From this simple structural basis, small changes in geometry, such as diameter, curvature and inter-hair spacing, can have considerable effects on mechanical properties, allowing functions such as mechanosensing, attachment, movement and protection. Here, we explore how passive features of hair-like structures, both individually and within arrays, enable diverse functions across biology. Understanding the relationships between form and function can provide biologists with an appreciation for the constraints and possibilities on hair-like structures. Additionally, such structures have already been used in biomimetic engineering with applications in sensing, water capture and adhesion. By examining hairs as a functional mechanical unit, geometry and arrangement can be rationally designed to generate new engineering devices and ideas

    Intrinsic differentiation potential of adolescent human tendon tissue: an in-vitro cell differentiation study

    BACKGROUND: Tendinosis lesions show an increase of glycosaminoglycan amount, calcifications, and lipid accumulation. Therefore, altered cellular differentiation might play a role in the etiology of tendinosis. This study investigates whether adolescent human tendon tissue contains a population of cells with intrinsic differentiation potential. METHODS: Cells derived from adolescent non-degenerative hamstring tendons were characterized by immunohistochemistry and FACS-analysis. Cells were cultured for 21 days in osteogenic, adipogenic, and chondrogenic medium and phenotypical evaluation was carried out by immunohistochemical and qPCR analysis. The results were compared with the results of similar experiments on adult bone marrow-derived stromal cells (BMSCs). RESULTS: Tendon-derived cells stained D7-FIB (fibroblast-marker) positive, but α-SMA (marker for smooth muscle cells and pericytes) negative. Tendon-derived cells were 99% negative for CD34 (endothelial cell marker), and 73% positive for CD105 (mesenchymal progenitor-cell marker). In adipogenic medium, intracellular lipid vacuoles were visible and tendon-derived fibroblasts showed upregulation of adipogenic markers FABP4 (fatty-acid binding protein 4) and PPARG (peroxisome proliferative activated receptor γ). In chondrogenic medium, some cells stained positive for collagen 2 and tendon-derived fibroblasts showed upregulation of collagen 2 and collagen 10. In osteogenic medium Von Kossa staining showed calcium deposition although osteogenic markers remained unaltered. Tendon-derived cells and BMCSs behaved largely comparable, although some distinct differences were present between the two cell populations. CONCLUSION: This study suggests that our population of explanted human tendon cells has an intrinsic differentiation potential. These results support the hypothesis that there might be a role for altered tendon-cell differentiation in the pathophysiology of tendinosis

    A hypomorphic Cbx3 allele causes prenatal growth restriction and perinatal energy homeostasis defects

    Mammals have three HP1 protein isotypes HP1β (CBX1), HP1γ (CBX3) and HP1α (CBX5) that are encoded by the corresponding genes Cbx1, Cbx3 and Cbx5. Recent work has shown that reduction of CBX3 protein in homozygotes for a hypomorphic allele (Cbx3 hypo) causes a severe postnatal mortality with around 99% of the homozygotes dying before weaning. It is not known what the causes of the postnatal mortality are. Here we show that Cbx3 hypo/hypo conceptuses are significantly reduced in size and the placentas exhibit a haplo-insufficiency. Late gestation Cbx3 hypo/hypo placentas have reduced mRNA transcripts for genes involved in growth regulation, amino acid and glucose transport. Blood vessels within the Cbx3 hypo/hypo placental labyrinth are narrower than wild-type. Newborn Cbx3 hypo/hypo pups are hypoglycemic, the livers are depleted of glycogen reserves and there is almost complete loss of stored lipid in brown adipose tissue (BAT). There is a 10-fold reduction in expression of the BAT-specific Ucp1 gene, whose product is responsible for non-shivering themogenesis. We suggest that it is the small size of the Cbx3 hypo/hypo neonates, a likely consequence of placental growth and transport defects, combined with a possible inability to thermoregulate that causes the severe postnatal mortality

    Measurement of differential top-quark-pair production cross sections in pp collisions at √s = 7TeV

    Normalised differential top-quark-pair production cross sections are measured in pp collisions at a centre-of-mass energy of 7 TeVat the LHC with the CMS detector using data recorded in 2011 corresponding to an integrated luminosity of 5. 0 fb-1. The measurements are performed in the lepton+jets decay channels (e+jets and μ+jets) and the dilepton decay channels (e+}e-, μ+μ-, and μ±e∓). The tt̄ differential cross section is measured as a function of kinematic properties of the final-state charged leptons and jets associated to b quarks, as well as those of the top quarks and the tt̄ system. The data are compared with several predictions from perturbative QCD calculations up to approximate next-to-next-to-leading-order precision. No significant deviations from the standard model are observed. © 2013 CERN for the benefit of the CMS collaboration

    Panic and perjury: a psychosocial exploration of agency

    The aim of this article is to explore the predicament of one man in difficult circumstances, in order to produce a psychosocial analysis that could contribute to a social psychological understanding of agency. After a brief review of the problem of dualism in theorizing agency and relevant developments in theories of self, and a critique of assumptions about unitary rational subjects, we emphasize the effects of unconscious conflict on choice and agency. We also identify the importance of including people's biographically motivated investments in specific discursive positions. Vince, a middle-aged, working-class man from the north of England, the subject of our case study, had already been faced with difficult choices in order to hold on to a job that was bad for him. Now the choice appeared to be taken out of his hands by an illness without a discernible organic base and with no prospect of improvement that, for 5 months, had forced him to be on sick leave. Our approach in this paper, based on interpretation of material from two interviews, is to detail the multiple and contradictory meanings of Vince's job to him. In each of three areas — the daily experience of the job, the meaning of having a respectable job, and Vince's relationship with his boss — we illustrate the power of a psychosocial analysis to escape the limitations of both voluntaristic and deterministic accounts of individual action. In conclusion we define the agent of choice in Vince's case as the divided psychosocial subject of unconscious conflict; a subject located in social realities mediated not only by social discourses but by psychic defences
