    Relationship between physical attributes and heat stress in dairy cattle from different genetic groups.

    Dairy cattle raised under harsh conditions have to adapt and prevent heat stress. The aim of this study was to evaluate physical characteristics and their association with heat tolerance in different genetic groups of dairy cattle. Thickness of the skin and coat, length and number of hairs, body measurements, as well as physiological parameters and body temperatures by infrared thermography were determined in 19 Holstein and 19 Girolando (½ and ¾ Holstein) cows. The Holstein cattle were less tolerant to heat stress than Girolando (GH50 and GH75 Holstein), because of the difficulty in dissipating heat due to the larger body size, as well as thicker and longer hairs. The correlations between physical characteristics, physiological parameters, and thermographic measurements prove to be inconsistent among genetic groups and therefore are not predictive of heat tolerance, while the regressions of morphometric characteristics on physiological and thermographic measures were not significant. Thus, the physical characteristics were not good predictors of physiological indices and thermographic temperature and so should not be used

    Impact of two new non-conventional yeasts, Candida oleophila and Starmerella lactis-condensi, isolated from sugar-rich substrates, on Frappato wine aroma

    The interest of non-Saccharomyces yeasts in wine fermentation increased constantly in last years. This study reports for the first time the enological potential of two strains Starmerella lactis-condensi MN412 and Candida oleophila YS209. In an innovative way, these strains were used in winemaking to improve floral and fruity aroma of Frappato red wine, which has not been explored. The enological performances of the two non-Saccharomyces strains were compared to a wine strain of Starmerella bacillaris, namely Cz3, previously characterized in winemaking conditions. In these three cases, the non-Saccharomyces strain was sequentially inoculated with S. cerevisiae wine strain NF213, used as control. The St. lactis-condensi MN412 was isolated from Sicilian manna, a sugar-rich matrix, extracted from Fraxinus angustifolia trees (Oleaceae). The strain C. oleophila YS209 was isolated from honey by-products. Microbiological counts showed the ability of MN412 and YS209 to maintain high counts up to 6 days of alcoholic fermentation. Regarding chemical parameters, Cz3 showed the highest glycerol production. Analysis of VOCs revealed that the trials with non-Saccharomyces yeasts were characterized by a higher concentration of esters that contributed positively to the fruity aroma of the wines. The sensory analysis confirmed that the use of MN412 and YS209 impacted positively the final wines in terms of fruity and floral intensity, respectively, while did not generate sensory defects. In conclusion, non-conventional yeasts represent strategy to improve floral-fruity freshness of wine aroma and sugar-rich matrices such as manna ash and honey might represent novel ecological niches as source of potential oenological yeast

    The Mechanisms of Codon Reassignments in Mitochondrial Genetic Codes

    Many cases of non-standard genetic codes are known in mitochondrial genomes. We carry out analysis of phylogeny and codon usage of organisms for which the complete mitochondrial genome is available, and we determine the most likely mechanism for codon reassignment in each case. Reassignment events can be classified according to the gain-loss framework. The gain represents the appearance of a new tRNA for the reassigned codon or the change of an existing tRNA such that it gains the ability to pair with the codon. The loss represents the deletion of a tRNA or the change in a tRNA so that it no longer translates the codon. One possible mechanism is Codon Disappearance, where the codon disappears from the genome prior to the gain and loss events. In the alternative mechanisms the codon does not disappear. In the Unassigned Codon mechanism, the loss occurs first, whereas in the Ambiguous Intermediate mechanism, the gain occurs first. Codon usage analysis gives clear evidence of cases where the codon disappeared at the point of the reassignment and also cases where it did not disappear. Codon disappearance is the probable explanation for stop to sense reassignments and a small number of reassignments of sense codons. However, the majority of sense to sense reassignments cannot be explained by codon disappearance. In the latter cases, by analysis of the presence or absence of tRNAs in the genome and of the changes in tRNA sequences, it is sometimes possible to distinguish between the Unassigned Codon and Ambiguous Intermediate mechanisms.     Neutrinos Angra experiment: commissioning and first operational measurements

    The Neutrinos Angra Experiment has completed a major step by finishing the comissioning of the detector and the data acquisition system at the experimental site located in the Angra dos Reis nuclear power plant. The experiment consists of a water-based detector and associated electronics, both designed with the goal of detecting the electron antineutrinos produced by the nuclear reactor. The detection is possible due to the Inverse Beta Decay, where the final products in the water are photons in the UV-to-visible range of the spectrum. The assembled detector comprises three active volumes filled with water: (i) a cubic target detector for electron antineutrinos, covered by 32 8-inches PMTs, (ii) a lateral layer surrounding the target (veto) equipped with 4 PMTs and (iii) a third volume covering the top of both, also equipped with 4~PMTs. In the present document the main features of the detector assembly as well as the integration of the readout electronics on-site are reported. Finally, some operational characteristics are shown based on straightforward analysis of the first measurements performed during the last months with the fully working detector

    Elevated temperature-humidity index induces physiological, blood and milk alterations in Holstein cows in a more pronounced manner than in ½ and ¾ Holstein × Gir.

    Bos taurus taurus and Bos taurus indicus cattle subspecies present different capabilities in coping with situations of elevated temperatures, the latter being more tolerant to heat stress. Thus, some breeding programs crossed these subspecies to produce a high producing yet heat-tolerant breed (Girolando). Nineteen Holstein (H100) and 19 Girolando cows [(½ Holstein × Gir (H50) and ¾ Holstein × Gir (H75)] with similar milk production were used in a six-day experiment to evaluate the consequences of heat stress due to shade deprivation on their physiological, blood and milk traits. Cows were exposed to a non-shaded environment between morning (06:00h; GMT -3:00) and evening milking (14:30h; GMT -3:00) with access to water ad libitum. Procedures were conducted before morning and evening milkings. Physiological parameters related to mechanisms of heat dissipation were measured, as well as the milk composition. Blood traits were evaluated. The temperature-humidity index (THI) was calculated. Statistical procedures included analysis of variance, correlation, and principal factors. THI was elevated during the trial and negatively impacted physiological, milk, and blood parameters in H100, H75, and H50. Alterations in physiology, milk stability, milk composition, and blood traits were more pronounced in H100. Holstein cows presented changes in physiological parameters in a more pronounced manner and in some milk and blood traits related to the reduced capability of this breed in dealing with elevated THI. The similarity in milk production levels excludes this parameter as a justification for differences in heat tolerance, with genetic composition being the main reason for these results
