    Epigenetics override pro-inflammatory PTGS transcriptomic signature towards selective hyperactivation of PGE 2 in colorectal cancer

    This is an Open Access article distributed under the terms of the Creative Commons Attribution License.-- et al.[Background]: Misregulation of the PTGS (prostaglandin endoperoxide synthase, also known as cyclooxygenase or COX) pathway may lead to the accumulation of pro-inflammatory signals, which constitutes a hallmark of cancer. To get insight into the role of this signaling pathway in colorectal cancer (CRC), we have characterized the transcriptional and epigenetic landscapes of the PTGS pathway genes in normal and cancer cells. [Results]: Data from four independent series of CRC patients (502 tumors including adenomas and carcinomas and 222 adjacent normal tissues) and two series of colon mucosae from 69 healthy donors have been included in the study. Gene expression was analyzed by real-time PCR and Affymetrix U219 arrays. DNA methylation was analyzed by bisulfite sequencing, dissociation curves, and HumanMethylation450K arrays. Most CRC patients show selective transcriptional deregulation of the enzymes involved in the synthesis of prostanoids and their receptors in both tumor and its adjacent mucosa. DNA methylation alterations exclusively affect the tumor tissue (both adenomas and carcinomas), redirecting the transcriptional deregulation to activation of prostaglandin E 2 (PGE 2 ) function and blockade of other biologically active prostaglandins. In particular, PTGIS, PTGER3, PTGFR, and AKR1B1 were hypermethylated in more than 40 % of all analyzed tumors. [Conclusions]: The transcriptional and epigenetic profiling of the PTGS pathway provides important clues on the biology of the tumor and its microenvironment.     Effect of forcing vine regrowth on 'Tempranillo' (Vitis vinifera L.) berry development and quality in Extremadura

    In warmer regions, fruit ripening in the wine grape tends to take place during the hottest part of the growing season. This can have negative consequences on the qualitative characteristics of the grape berries at harvest. Forcing vines to regrow can be an aggressive but effective technique to delay the harvest date, but needs to be evaluated carefully in each growing condition. In 2017, in an experimental vineyard in Extremadura, forcing was conducted 3 (F1 treatment) and 22 (F2 treatment) days after anthesis (May 18 and June 6) by hedging growing shoots to seven nodes and removing summer laterals, leaves and primary clusters. Vines grown using conventional practices were used for the Control treatment. Forcing delayed the harvest date from August 22 (Control) to September 14 (F1) and October 19 (F2). Shifting the berry growth period modified the duration of the different fruit development stages. Compared to the Control treatment, the F1 and F2 berries were smaller at harvest, but had similar skin weight percentages; however, the seed weight percentage of the F2 berries was higher. The differences in grape composition observed at harvest between the various treatments were further accentuated in the wines. At harvest, the F2 berries had significantly higher total polyphenol and anthocyanin content than the Control and F1 berries, which had similar values. In the wines, both F1 and F2 characteristics differed considerably from the Control, most notably in the high F2 tannin concentration. These preliminary results from the first year of study indicate the potential of this technique to obtain wine grapes with very different characteristics, offering new viticultural perspectives in warm climate areas

    CD44-high neural crest stem-like cells are associated with tumour aggressiveness and poor survival in neuroblastoma tumours

    BACKGROUND: Neuroblastoma is a paediatric tumour originated from sympathoadrenal precursors and characterized by its heterogeneity and poor outcome in advanced stages. Intra-tumoral cellular heterogeneity has emerged as an important feature in neuroblastoma, with a potential major impact on tumour aggressiveness and response to therapy. CD44 is an adhesion protein involved in tumour progression, metastasis and stemness in different cancers; however, there has been controversies about the significance of CD44 expression in neuroblastoma and its relationship with tumour progression. METHODS: We have performed transcriptomic analysis on patient tumour samples studying the outcome of patients with high CD44 expression. Adhesion, invasion and proliferation assays were performed in sorted CD44high neuroblastoma cells. Tumoursphere cultures have been used to enrich in undifferentiated stem-like cells and to asses self-renewal and differentiation potential. We have finally performed in vivo tumorigenic assays on cell line-derived or Patient-derived xenografts. FINDINGS: We show that high CD44 expression is associated with low survival in high-grade human neuroblastoma, independently of MYCN amplification. CD44 is expressed in a cell population with neural crest stem-like features, and with the capacity to generate multipotent, undifferentiated tumourspheres in culture. These cells are more invasive and proliferative in vitro. CD44 positive cells obtained from tumours are more tumorigenic and metastatic, giving rise to aggressive neuroblastic tumours at high frequency upon transplantation. INTERPRETATION: We describe an unexpected intra-tumoural heterogeneity within cellular entities expressing CD44 in neuroblastoma, and propose that CD44 has a role in neural crest stem-like undifferentiated cells, which can contribute to tumorigenesis and malignancy in this type of cancer.     Estimation of kinetic parameters in a chromatographic separation model via Bayesian inference

    A modelagem de processos de adsorção tem sido empregada com frequência nas indústrias químicas, petroquímicas e refinarias, por exemplo para separação e purificação de misturas em unidade de Leito Móvel Simulado (LMS). Na representação matemática do modelo, a determinação de parâmetros é um passo importante para o projeto de condições cromatográficas para a separação contínua, em processos do tipo LMS. Este trabalho tem por objetivo a análise de estimativa de parâmetros em processos de adsorção, usando um sistema cromatográfico com uma coluna, para a separação das substâncias Glicose e Frutose. Investiga-se o uso da abordagem Bayesiana, através de métodos de Monte Carlo via Cadeias de Markov (MCMC), assim como o uso da abordagem da máxima verossimilhança, utilizando duas técnicas estocásticas diferentes, o Algoritmo de Colisão de Partículas (PCA - Particle Collision Algorithm), e o Algoritmo de Otimização por Enxame de Partículas (PSO - Particle Swarm Optimization) para executar a tarefa de minimização da função objetivo. Diferentes casos são apresentados com o objetivo de analisar a significância estatística das estimativas obtidas para os parâmetros, fazendo-se uma comparação crítica entre a solução via inferência Bayesiana e via minimização da função objetivo com métodos estocásticos. Os resultados obtidos demonstram que o uso da abordagem Bayesiana fornece uma proposta vantajosa para a estimativa de parâmetros em transferência de massa, oferecendo resultados com maior riqueza de informação estatística.The modeling of adsorption processes appears quite frequently in the chemical industry, petrochemical plants and refineries, for example for separation and purification of mixtures in Simulated Moving Bed (SMB) units. In the mathematical formulation, the accurate determination of the model parameters is an important step for the design of chromatographic conditions for continuous separation in SMB processes. This work is aimed at the estimation of the model parameters in adsorption processes, using a chromatographic column for the separation of glucose and fructose. The Bayesian framework for inverse problems is investigated through the implementation of Markov Chain Monte Carlo methods (MCMC) and a critical comparison against the classical Maximum Likelihood approach, with the minimization of the objective function via two different stochastic techniques, namely the Particle Collision Algorithm (PCA), and the Particle Swarm Optimization (PSO) is performed. Different cases are presented in order to investigate the statistical significance of the estimates obtained, and perform comparisons between the solution via Bayesian inference and via the minimization of the objective function with the stochastic methods. The results demonstrate that the Bayesian approach employs less computational effort to achieve estimates with comparable statistical information.Peer Reviewe

    Effects of n-3 polyunsaturated fatty acids on cardiac ion channels

All rights reserved. it is reproduced with permissionDietary n-3 polyunsaturated fatty acids (PUFAs) have been reported to exhibit antiarrhythmic properties, and these effects have been attributed to their capability to modulate ion channels. In the present review, we will focus on the effects of PUFAs on a cardiac sodium channel (Nav1.5) and two potassium channels involved in cardiac atrial and ventricular repolarization (Kv) (Kv1.5 and Kv11.1). n-3 PUFAs of marine (docosahexaenoic, DHA and eicosapentaenoic acid, EPA) and plant origin (alpha-linolenic acid, ALA) block Kv1.5 and Kv11.1 channels at physiological concentrations. Moreover, DHA and EPA decrease the expression levels of Kv1.5, whereas ALA does not. DHA and EPA also decrease the magnitude of the currents elicited by the activation of Nav1.5 and calcium channels. These effects on sodium and calcium channels should theoretically shorten the cardiac action potential duration (APD), whereas the blocking actions of n-3 PUFAs on Kv channels would be expected to produce a lengthening of cardiac action potential.     The Origin of Amerindians and the Peopling of the Americas According to HLA Genes: Admixture with Asian and Pacific People

    The classical three-waves theory of American peopling through Beringia was based on a mixed anthropological and linguistic methodology. The use of mtDNA, Y chromosome and other DNA markers offers different results according to the different markers and methodologies chosen by different authors. At present, the peopling of Americas remains uncertain, regarding: time of population, number of peopling waves and place of peopling entrance among other related issues. In the present review, we have gathered most available HLA data already obtained about First Native American populations, which raise some doubts about the classical three waves of American peopling hypothesis. In summary, our conclusions are: 1) North West Canadian Athabaskans have had gene flow with: a) close neighboring populations, b) Amerindians, c) Pacific Islanders including East Australians and d) Siberians; 2) Beringia was probably not the only entrance of people to America: Pacific Ocean boat trips may have contributed to the HLA genetic American profile (or the opposite could also be true); 3) Amerindians entrance to America may have been different to that of Athabaskans and Eskimos and Amerindians may have been in their lands long before Athabaskans and Eskimos because they present and altogether different set of HLA-DRB1 allele frequencies; 4) Amerindians show very few “particular alleles”, almost all are shared with other Amerindians, Athabaskans and Pacific Islanders, including East Australians and Siberians; 5) Our results do not support the three waves model of American peopling, but another model where the people entrance is not only Beringia, but also Pacific Coast. Reverse migration (America to Asia) is not discarded and different movements of people in either direction in different times are supported by the Athabaskan population admixture with Asian-Pacific population and with Amerindians, 6) HLA variability is more common than allele veriability in Amerindians. Finally, it is shown that gene genealogy analises should be completed with allele frequency analyses in population relatednes and migrations studies

    Síndrome post-colecistectomía: análisis de una cohorte prospectiva

    Introducción. El síndrome post-colecistectomía fue descrito hace más de 70 años y se define como la presencia de síntomas gastrointestinales que pueden persistir después de la colecistectomía o aparecer como nuevos síntomas. Este síndrome, poco conocido, puede manifestarse desde pocos meses hasta varios años luego de la cirugía y ocurre entre 5 % y 40 % de los pacientes. Método. Estudio de cohorte prospectiva, que incluyó pacientes a quienes se les realizó colecistectomía laparoscópica en tres unidades quirúrgicas de Bogotá, D.C., Colombia, durante un período de ocho meses, con seguimientos periódicos durante dos años. El síndrome post-colecistectomía se definió como la presencia de al menos un síntoma gastrointestinal a partir del primer mes de la cirugía. Resultados. Se incluyeron 380 pacientes de los cuales 183 (48,2 %) desarrollaron el síndrome. La edad tuvo diferencia estadísticamente significativa entre aquellos que lo desarrollaron y los que no (p=0,024). La diarrea fue el único síntoma preoperatorio asociado al desarrollo del síndrome. El síndrome post-colecistectomía se encontró en el 42,1 % de los pacientes al primer mes de seguimiento y en el 17,4 % al segundo año. Conclusiones. El síndrome post-colecistectomía ocurre en un porcentaje importante de pacientes. En concordancia con la literatura, se encontró que la diarrea prequirúrgica es un factor de riesgo independiente para el desarrollo del síndrome. Se recomienda un adecuado seguimiento de los pacientes después de una colecistectomía

    8-PropargylaminoBODIPY: unprecedented blue-emitting pyrromethene dye. Synthesis, photophysics and laser properties

    Highly emitting 8-propargylaminoBODIPY (8-PAB) 2 was prepared in 94% yield.     Bichromatic laser emission from dipyrromethene dyes incorporated into solid polymeric media

    9 pages, 9 figures.Bichromatic laser emission from dipyrromethene-based solid-state dye lasers is reported. The dependence of this dual emission on different factors and its origin and causes are discussed in the light of different models proposed in the literature. Our experimental results indicate that the long-wavelength emission can be explained in terms of reabsorption/reemission effects and inhomogeneous broadening of the S0-S1 transition. The short-wavelength emission corresponds to the usual S0-S1 transition and dominates at low dye concentration.This work was supported by Project Nos.
