    “We Are a Very Happy Family”: 19th-Century Familial Power Dynamics

    This article examines the roles of family members in the mid-19th century in America, using the Willard family as a case study. Ultimately, this thick description of the Willard family demonstrates that power within the family structure was neither intrinsic nor static; moreover, one person did not control the family and its decisions at all times. Instead, each family member, depending upon circumstances, situations, and his or her own nature, negotiated and laid claim to power through various sources of authority. Josiah Willard’s authority stemmed from his role as father and husband; society crowned him king of the household, and he had the physical size to maintain it. His wife, Mary Hill Willard, often won power struggles through love and through demonstration of her moral superiority. The children, Oliver, Frances, and Mary, exhibited their power through a variety of acts—negotiation, playing to their positions as inferiors within the family, and simple rebellion. In these ways, the Willard family provides us with a template for understanding many of the middle-class families of the mid-19th century as well as the power dynamic between parents and their children

    The assessment of the Spacelab carrier induced contaminant environment was continued, and the ability of Spacelab to meet established contamination control criteria for the space transportation system program was determined. The primary areas considered included: (1) updating, refining, and improving the Spacelab contamination computer model and contamination analysis methodology, (2) establishing the resulting adjusted induced environment predictions for comparison with the applicable criteria, (3) determining the Spacelab design and operational requirements necessary to meet the criteria, (4) conducting mission feasibility analyses of the combined Spacelab/Orbiter contaminant environment for specific proposed mission and payload mixes, and (5) establishing a preliminary Spacelab mission support plan as well as model interface requirements; A summary of those activities conducted to date with respect to the modelling, analysis, and predictions of the induced environment, including any modifications in approach or methodology utilized in the contamination assessment of the Spacelab carrier, was presented

    Experimental investigation of contamination prevention techniques to cryogenic surfaces on board orbiting spacecraft

    Within the simulation limitations of on-orbit conditions, it was demonstrated that a helium purge system could be an effective method for reducing the incoming flux of contaminant species. Although a generalized purge system was employed in conjunction with basic telescope components, the simulation provided data that could be used for further modeling and design of a specific helium injection system. Experimental telescope pressures required for 90% attenuation appeared to be slightly higher (factor of 2 to 5). Cooling the helium purge gas and telescope components from 300 to 140 K had no measurable effect on stopping efficiency of a given mass flow of helium from the diffuse injector

    A preliminary assessment of the computer interface requirements of the Spacelab configuration contamination computer model was conducted to determine the compatibility of the program, as presently formatted, with the computer facilities at MSFC. The necessary Spacelab model modifications are pointed out. The MSFC computer facilities and their future plans are described, and characteristics of the various computers as to availability and suitability for processing the contamination program are discussed. A listing of the CDC 6000 series and UNIVAC 1108 characteristics is presented so that programming requirements can be compared directly and differences noted

    Eco-evolutionary dynamics of clonal multicellular life cycles

    The evolution of multicellular life cycles is a central process in the course of the emer-gence of multicellularity. The simplest multicellular life cycle is comprised of the growth of the propagule into a colony and its fragmentation to give rise to new propagules. The majority of theo-retical models assume selection among life cycles to be driven by internal properties of multicellular groups, resulting in growth competition. At the same time, the influence of interactions between groups on the evolution of life cycles is rarely even considered. Here, we present a model of colo-nial life cycle evolution taking into account group interactions. Our work shows that the outcome of evolution could be coexistence between multiple life cycles or that the outcome may depend on the initial state of the population – scenarios impossible without group interactions. At the same time, we found that some results of these simpler models remain relevant: evolutionary stable strategies in our model are restricted to binary fragmentation – the same class of life cycles that contains all evolutionarily optimal life cycles in the model without interactions. Our results demonstrate that while models neglecting interactions can capture short- term dynamics, they fall short in predicting the population- scale picture of evolution

    This AVD High Impact Fellows Program Team Project developed high school level learning modules on renewable energy. The learning modules focus on the collection, analysis and graphing of real-world data and the making of assessments and projections of the use of renewable energy at the school location. The project focuses primarily on solar energy but all other major alternative energy sources are also briefly addressed. The modules focus on solar energy because it is the only feasible alternative energy source for the region. The ultimate goal of the project is to produce a presentation for the school principal making recommendations for implementation and energy return on investment. The final form of the instructional material is a lesson plan for a full curriculum unit in an AP Environmental Science. The materials focus on fostering inquiry and technological integration

    Induction of Granulysin and Perforin Cytolytic Mediator Expression in 10-Week-Old Infants Vaccinated with BCG at Birth

    Background. While vaccination at birth with Mycobacterium bovis Bacilli Calmette-Guérin (BCG) protects against severe childhood tuberculosis, there is no consensus as to which components of the BCG-induced immune response mediate this protection. However, granulysin and perforin, found in the granules of cytotoxic T lymphocytes and Natural Killer (NK) cells, can kill intracellular mycobacteria and are implicated in protection against Mycobacterium tuberculosis. Methods. We compared the cellular expression of granulysin and perforin cytolytic molecules in cord blood and peripheral blood from 10-week-old infants vaccinated at birth with either Japanese or Danish BCG, administered either intradermally or percutaneously. Results. In cord blood, only CD56+ NK cells expressed granulysin and perforin constitutively. These cytolytic mediators were upregulated in CD4+ and CD8+ cord blood cells by ex vivo stimulation with BCG but not with PPD. Following BCG vaccination of neonates, both BCG and PPD induced increased expression of granulysin and perforin by CD4+ and CD8+ T cells. There was no difference in expression of cytolytic molecules according to vaccination route or strain. Conclusions. Constitutive expression of perforin and granulysin by cord blood NK-cells likely provides innate immunity, while BCG vaccination-induced expression of these cytolytic mediators may contribute towards protection of the neonate against tuberculosis

    Low expression of the putative tumour suppressor spinophilin is associated with higher proliferative activity and poor prognosis in patients with hepatocellular carcinoma

    BACKGROUND: Spinophilin, a multifunctional intracellular scaffold protein, is reduced in certain types of cancer and is regarded as a novel putative tumour suppressor protein. However, the role of spinophilin in hepatocellular carcinoma (HCC) has never been explored before. METHODS: In this study, we determined for the first time the expression pattern of spinophilin in human HCC by immunohistochemistry and quantitative reverse transcriptase–PCR analysis. In addition, we performed immunohistochemical analysis of p53, p14(ARF) and the proliferation marker Ki-67. Kaplan–Meier curves and multivariate Cox proportional models were used to study the impact on clinical outcome. Small interfering RNA (siRNA) was used to silence spinophilin and to explore the effects of reduced spinophilin expression on cellular growth. RESULTS: In our study, complete loss of spinophilin immunoreactivity was found in 44 of 104 HCCs (42.3%) and reduced levels were found in an additional 37 (35.6%) cases. After adjusting for other prognostic factors, multivariate Cox regression analysis identified low expression of spinophilin as an independent prognostic factor with respect to disease-free (hazard ratio (HR)=1.8; 95% confidence interval (CI)=1.04–3.40; P=0.043) and cancer-specific survival (HR=2.0; CI=1.1–3.8; P=0.025). Reduced spinophilin expression significantly correlated with higher Ki-67 index in HCC (P=0.014). Reducing spinophilin levels by siRNA induced a higher cellular growth rate and increased cyclin D2 expression in tumour cells (P<0.05). CONCLUSION: This is the first study of the expression pattern and distribution of spinophilin in HCC. According to our data, the loss of spinophilin is associated with higher proliferation and might be useful as a prognostic marker in patients with HCC

    Increased neutrophil-lymphocyte ratio is a poor prognostic factor in patients with primary operable and inoperable pancreatic cancer

    Background: The neutrophil-lymphocyte ratio (NLR) has been proposed as an indicator of systemic inflammatory response. Previous findings from small-scale studies revealed conflicting results about its independent prognostic significance with regard to different clinical end points in pancreatic cancer (PC) patients. Therefore, the aim of our study was the external validation of the prognostic significance of NLR in a large cohort of PC patients. Methods: Data from 371 consecutive PC patients, treated between 2004 and 2010 at a single centre, were evaluated retrospectively. The whole cohort was stratified into two groups according to the treatment modality. Group 1 comprised 261 patients with inoperable PC at diagnosis and group 2 comprised 110 patients with surgically resected PC. Cancer-specific survival (CSS) was assessed using the Kaplan–Meier method. To evaluate the independent prognostic significance of the NLR, the modified Glasgow prognostic score (mGPS) and the platelet-lymphocyte ratio univariate and multivariate Cox regression models were applied. Results: Multivariate analysis identified increased NLR as an independent prognostic factor for inoperable PC patients (hazard ratio (HR)=2.53, confidence interval (CI)=1.64–3.91, P<0.001) and surgically resected PC patients (HR=1.61, CI=1.02–2.53, P=0.039). In inoperable PC patients, the mGPS was associated with poor CSS only in univariate analysis (HR=1.44, CI=1.04–1.98). Conclusion: Risk prediction for cancer-related end points using NLR does add independent prognostic information to other well-established prognostic factors in patients with PC, regardless of the undergoing therapeutic modality. Thus, the NLR should be considered for future individual risk assessment in patients with PC
