    A least-squares implicit RBF-FD closest point method and applications to PDEs on moving surfaces

    The closest point method (Ruuth and Merriman, J. Comput. Phys. 227(3):1943-1961, [2008]) is an embedding method developed to solve a variety of partial differential equations (PDEs) on smooth surfaces, using a closest point representation of the surface and standard Cartesian grid methods in the embedding space. Recently, a closest point method with explicit time-stepping was proposed that uses finite differences derived from radial basis functions (RBF-FD). Here, we propose a least-squares implicit formulation of the closest point method to impose the constant-along-normal extension of the solution on the surface into the embedding space. Our proposed method is particularly flexible with respect to the choice of the computational grid in the embedding space. In particular, we may compute over a computational tube that contains problematic nodes. This fact enables us to combine the proposed method with the grid based particle method (Leung and Zhao, J. Comput. Phys. 228(8):2993-3024, [2009]) to obtain a numerical method for approximating PDEs on moving surfaces. We present a number of examples to illustrate the numerical convergence properties of our proposed method. Experiments for advection-diffusion equations and Cahn-Hilliard equations that are strongly coupled to the velocity of the surface are also presented

    Purified Human Pancreatic Duct Cell Culture Conditions Defined by Serum-Free High-Content Growth Factor Screening

    The proliferation of pancreatic duct-like CK19+ cells has implications for multiple disease states including pancreatic cancer and diabetes mellitus. The in vitro study of this important cell type has been hampered by their limited expansion compared to fibroblast-like vimentin+ cells that overgrow primary cultures. We aimed to develop a screening platform for duct cell mitogens after depletion of the vimentin+ population. The CD90 cell surface marker was used to remove the vimentin+ cells from islet-depleted human pancreas cell cultures by magnetic-activated cell sorting. Cell sorting decreased CD90+ cell contamination of the cultures from 34±20% to 1.3±0.6%, yielding purified CK19+ cultures with epithelial morphology. A full-factorial experimental design was then applied to test the mitogenic effects of bFGF, EGF, HGF, KGF and VEGF. After 6 days in test conditions, the cells were labelled with BrdU, stained and analyzed by high-throughput imaging. This screening assay confirmed the expected mitogenic effects of bFGF, EGF, HGF and KGF on CK19+ cells and additionally revealed interactions between these factors and VEGF. A serum-free medium containing bFGF, EGF, HGF and KGF led to CK19+ cell expansion comparable to the addition of 10% serum. The methods developed in this work should advance pancreatic cancer and diabetes research by providing effective cell culture and high-throughput screening platforms to study purified primary pancreatic CK19+ cells

    Experimental Analysis of the Laser-Induced Instruction Skip Fault Model

    International audienceMicrocontrollers storing valuable data or using security functions are vulnerable to fault injection attacks. Among the various types of faults, instruction skips induced at runtime proved to be effective against identification routines or encryption algorithms. Several research works assessed a fault model that consists in a single instruction skip, i.e. the ability to prevent one chosen instruction in a program from being executed. This assessment is used to design countermeasures able to withstand a single instruction skip. We question this fault model on experimental basis and report the possibility to induce with a laser an arbitrary number of instruction skips. This ability to erase entire sections of a firmware has strong implications regarding the design of counter- measures

    Evidence for reducing cancer-specific mortality due to screening for breast cancer in Europe: A systematic review

    Background: The aim of this study was to quantify the impact of organised mammography screening on breast cancer mortality across European regions. Therefore, a systematic review was performed including different types of studies from all European regions and stringently used clearly defined quality appraisal to summarise the best evidence. Methods: Six databases were searched including Embase, Medline and Web of Science from inception to March 2018. To identify all eligible studies which assessed the effect of organised screening on breast cancer mortality, two reviewers independently applied predefined inclusion and exclusion criteria. Original studies in English with a minimum follow-up of five years that were randomised controlled trials (RCTs) or observational studies were included. The Cochrane risk of bias instrument and the Newcastle–Ottawa Scale were used to assess the risk of bias. Results: Of the 5015 references initially retrieved, 60 were included in the final analysis. Those comprised 36 cohort studies, 17 case–control studies and 7 RCTs. None were from Eastern Europe. The quality of the included studies varied: Nineteen of these studies were of very good or good quality. Of those, the reduction in breast cancer mortality in attenders versus non-attenders ranged between 33% and 43% (Northern Europe), 43%–45% (Southern Europe) and 12%–58% (Western Europe). The estimates ranged between 4% and 31% in invited versus non-invited.     Morphology of the digital sheath in horses

    Introduction: The digital sheath is located in the palmar (plantar) face of the distal limb. It consists of a synovium, divided into intimate and vascularised supportive layers, and ligaments that strengthen the tendon sheath in its palmar (plantar) face. The synovium surrounds the digital flexor tendons during their passage within the sheath. The synovium fluid, produced by the filtering of the blood and by the intima cells, allows lubrification within the sheath. Lameness, originating from digital sheath pathology is not rare, and it is important, for any equine practitioner, to know its normal morphology. The aim of this study is to precise the morphology of the digital sheath, particularly its synovium because the literature is not unanimous. Methods: The digitals tips of 7 "sound" horses were collected. Samples were taken within the 3 annular ligaments (Proximal, Digital Proximal, Digital Distal). The digital flexor tendons were sampled with the visceral sheath of the synovial membrane at the level of the proximal sesamoĂŻd bones and just proximal to the medium scutum. All these samples were embedded in tissue-tek and freezed for cryosectioning. The proximal and distal recessus of the synovial membrane were also sampled, fixed in formalin and embedded in paraffin. Sections were stained with haematoxylin/eosin. One sample of the synovial membrane was taken for electronic microscopy and embedded in? Results: Annular ligaments are composed of dense connective tissue but are transversal strengthening of the fascia rather than true ligaments. The digital flexor tendons showed the typical organisation of the collagen network. Type III fibrocytes were observed within the deep digital flexor tendon. The synovial membrane showed two layers: an intima (with visceral and parietal sheath) with fibroblast-like cells and macrophagic cells, and a sub-intima composed of fibrous tissue that was sometime so thickened that it was difficult to cut! The recessus contained large synovial fringe with adipose tissue. Conclusion: This study permitted to precise the morphology of the digital sheath in horse which may help to better understand the pathological changes
