    The Function of a Nail: An Archaeological Examination of Three 18th- and 19th-Century Eastern Pequot Reservation Homes in Southeastern Connecticut

    This thesis examines three indigenous households excavated on the Eastern Pequot reservation in North Stonington, Connecticut. Architectural artifact and spatial analyses, combined with historical documents, are utilized to understand reservation building practices of Native Americans navigating colonialism in the 18th and 19th century. The homes are small in design with at least one window and one stone chimney each. They all possessed cellars, but not all are stone-lined. Nails and window glass serve as the primary architectural artifact classes in this work, with an emphasis on their manufacture and modification. Examining nail and glass type, quantity, modification, and spatial patterns facilitates discussion on the forms Eastern Pequot homes took and how they entered the archaeological record. Furthermore, historical records combined with archaeological evidence suggest repairs were made to the homes or materials recycled from them. Documents highlight the relationship between the overseer and tribal members on the reservation and suggest overseers played an active role in Eastern Pequot home maintenance, at least in the 19th century. Results indicate that when these homes entered the archaeological record, they were intentionally demolished, although perhaps not immediately after the residents left. This historical and material evidence offers insight on Eastern Pequot strategies to navigating reservation life during colonialism of the 18th and 19th century

    Optimization with artificial intelligence in additive manufacturing: a systematic review

    In situations requiring high levels of customization and limited production volumes, additive manufacturing (AM) is a frequently utilized technique with several benefits. To properly configure all the parameters required to produce final goods of the utmost quality, AM calls for qualified designers and experienced operators. This research demonstrates how, in this scenario, artificial intelligence (AI) could significantly enable designers and operators to enhance additive manufacturing. Thus, 48 papers have been selected from the comprehensive collection of research using a systematic literature review to assess the possibilities that AI may bring to AM. This review aims to better understand the current state of AI methodologies that can be applied to optimize AM technologies and the potential future developments and applications of AI algorithms in AM. Through a detailed discussion, it emerges that AI might increase the efficiency of the procedures associated with AM, from simulation optimization to in-process monitoring

    Pomegranate fruit (PF) is a fruit rich in nutraceuticals. Nonedible parts of the fruit, especially peels, contain high amounts of bioactive components that have been largely used in traditional medicine, such as the Chinese, Unani, and Ayurvedic ones, for treating several diseases. Polyphenols such as anthocyanins, tannins, flavonoids, phenolic acids, and lignans are the major bioactive molecules present in PF. Therefore, PF is considered a source of natural multifunctional agents that exert simultaneously antioxidant, anti-inflammatory, antitumor, antidiabetic, cardiovascular, and neuroprotective activities. Recently, several studies have reported that the nutraceuticals contained in PF (seed, peel, and juice) have a potential beneficial role in Alzheimer's disease (AD). Research suggests that the neuroprotective effect of PF is mostly due to its potent antioxidant and anti-inflammatory activities which contribute to attenuate the neuroinflammation associated with AD. Despite the numerous works conducted on PF, to date the mechanism by which PF acts in combatting AD is not completely known. Here, we summarize all the recent findings (in vitro and in vivo studies) related to the positive effects that PF and its bioactive components can have in the neurodegeneration processes occurring during AD. Moreover, considering the high biotransformation characteristics of the nutraceuticals present in PF, we propose to consider the chemical structure of its active metabolites as a source of inspiration to design new molecules with the same beneficial effects but less prone to be affected by the metabolic degradation process

    Promising Support Coming from Nature: Antioxidant and Anti-Inflammatory Potential of Castanea sativa Wood Distillate on Skin Cells

    Tissue homeostasis, function recovery, and protection mechanisms are boosted by the balanced and timely control of inflammation and oxidative stress. Nowadays, many natural products and bio-derivates exhibit antioxidant and anti-inflammatory activity, supporting medical care and tissue wellness against inflammation, oxidative stress, and inflammaging. Castanea sativa wood distillate (WD) is a bio-derivative used as a corroborant and biofertilizer in agriculture. Based on the safety profile of low concentrations of WD on human cells, the present study aims to assess the anti-inflammatory and antioxidant activity of WD on different cell types in the integumentary system. Human keratinocytes, mucosal epithelium, dermal fibroblasts, and endothelial cells were exposed to WD, and the concentrations devoid of pro-apoptotic potential were profiled. Then, the effect of nontoxic doses of WD revealed an anti-inflammatory effect, observed through the immunodetection of prostanoid cascade markers in experimentally induced inflammation. A reduction in endothelial hyperpermeability was evidenced by the immunofluorescence analysis of cell–cell adhesion proteins, VE-cadherin and ZO-1. In addition, WD buffered the exogenously produced oxidative stress. On the whole, WD showed both anti-inflammatory and antioxidant activities on the various cell types, preserving endothelial barrier integrity. Overall, this study supports the involvement of this bio-derivative in novel exploitable fields, such as therapeutic dermatological applications for human and animal medical care

    Autonomous Service Drones for Multimodal Detection and Monitoring of Archaeological Sites

    Constant detection and monitoring of archaeological sites and objects have always been an important national goal for many countries. The early identification of changes is crucial to preventive conservation. Archaeologists have always considered using service drones to automate collecting data on and below the ground surface of archaeological sites, with cost and technical barriers being the main hurdles against the wide-scale deployment. Advances in thermal imaging, depth imaging, drones, and artificial intelligence have driven the cost down and improved the quality and volume of data collected and processed. This paper proposes an end-to-end framework for archaeological sites detection and monitoring using autonomous service drones. We mount RGB, depth, and thermal cameras on an autonomous drone for low-altitude data acquisition. To align and aggregate collected images, we propose two-stage multimodal depth-to-RGB and thermal-to-RGB mosaicking algorithms. We then apply detection algorithms to the stitched images to identify change regions and design a user interface to monitor these regions over time. Our results show we can create overlays of aligned thermal and depth data on RGB mosaics of archaeological sites. We tested our change detection algorithm and found it has a root mean square error of 0.04. To validate the proposed framework, we tested our thermal image stitching pipeline against state-of-the-art commercial software. We cost-effectively replicated its functionality while adding a new depth-based modality and created a user interface for temporally monitoring changes in multimodal views of archaeological sites

    Low-dose aspirin for primary prevention of cardiovascular events in postmenopausal women with type-2 diabetes: The prescriptive approach in the real world

    Background: The long-Term efficacy of low-dose aspirin for primary prevention of cardiovascular (CV) events in postmenopausal women with type-2 diabetes is controversial. Therefore, it is recommended only on an individual basis, recommendation of grade C. Methods: We enrolled 275 consecutive postmenopausal women with type-2 diabetes, without an increased bleeding risk and without preexisting CV disease as coronary artery disease, stroke, and peripheral vascular disease, but with a high risk assessed by score >10%, aged 60-69 years. All were receiving aspirin (75-100 mg daily), aspirin group (AG). 170 postmenopausal women with type-2 diabetes and without preexisting cardiovascular (CV) disease, but not on aspirin treatment, despite a high risk assessed by score >10%, were control group (CG). Mean age was 66 ± 4 years for AG and 65 ± 7 years for CG. Our goal was to identify the prevalence of low-dose aspirin prescriptions in these populations according to different clinical conditions. Results: Women with only high risk were 41/275 (15%) on AG and 72/170 (42.3%) on CG, Chi-squared 41, Odds ratio 0.2, c.i. 95%, P < 0.0001. Women affected by metabolic syndrome were 105/275 (38.1%) on AG and 47/170 (27.6%) on CG, Chi-squared 5.1, Odds ratio 1.6, c.i. 95%, P < 0.02. Women affected by metabolic cardiomyopathy were 111/275 (40.3%) on AG and 44/170 (25.9%) on CG, Chi-squared 8, Odds ratio 1.8, c.i. 95%, P < 0.004. Women affected by diabetic cardiomyopathy were 18/275 (6.6%) on AG and 7/170 (4.2%) on CG, Chi-squared 1.2, Odds ratio 16, c.i. 95%, P < 0.2 n.s. Conclusions: Low-dose aspirin in our population is prescribed preferentially in postmenopausal women with type-2 diabetes when affected by metabolic syndrome or metabolic cardiomyopathy, at the opposite women with only high risk have lower chance to receive aspirin

    Transthyretin complexes with curcumin and bromo-estradiol: Evaluation of solubilizing multicomponent mixtures

    Crystallographic structure determination of protein–ligand complexes of transthyretin (TTR) has been hindered by the low affinity of many compounds that bind to the central cavity of the tetramer. Because crystallization trials are carried out at protein and ligand concentration that approach the millimolar range, low affinity is less of a problem than the poor solubility of many compounds that have been shown to inhibit amyloid fibril formation. To achieve complete occupancy in co-crystallization experiments, the minimal requirement is one ligand for each of the two sites within the TTR tetramer. Here we present a new strategy for the co-crystallization of TTR using high molecular weight polyethylene glycol instead of high ionic strength precipitants, with ligands solubilized in multicomponent mixtures of compounds. This strategy is applied to the crystallization of TTR complexes with curcumin and 16a-bromo-estradiol. Here we report the crystal structures with these compounds and with the ferulic acid that results from curcumin degradation
