    Splenic nerve bundle stimulation in acute and chronic inflammation

    Splenic neurovascular bundle stimulation holds potential to treat acute and chronic inflammatory conditions. In the first part of the thesis, the available literature on the interactions between the immune system and nervous system in the intestine is summarized. Then, it is shown that a specialized T-cell, that can produce the neurotransmitter acetylcholine, resides in the gut an plays a dual role in the development of experimental colitis in mice. Furthermore, electrical splenic neurovascular bundle stimulation ameliorated the outcomes of colitis in mice and reversed transcriptomic changes in the gut that were induced by colitis. The second part of the thesis focused on the translation of splenic neurovascular bundle stimulation to the human situation. It is shown that there are significant changes between murine and human innervation of the spleen. Using computed tomography (CT) images the course and the characteristics of the splenic artery were described. These data were used to develop a cuff electrode that could be used for electrical stimulation of the splenic neurovascular bundle in humans. Finally, it was demonstrated that splenic neurovascular bundle stimulation in humans was safe and feasible in a pilot study with patients that underwent esophagectomy

    Modular lifelong machine learning

    Deep learning has drastically improved the state-of-the-art in many important fields, including computer vision and natural language processing (LeCun et al., 2015). However, it is expensive to train a deep neural network on a machine learning problem. The overall training cost further increases when one wants to solve additional problems. Lifelong machine learning (LML) develops algorithms that aim to efficiently learn to solve a sequence of problems, which become available one at a time. New problems are solved with less resources by transferring previously learned knowledge. At the same time, an LML algorithm needs to retain good performance on all encountered problems, thus avoiding catastrophic forgetting. Current approaches do not possess all the desired properties of an LML algorithm. First, they primarily focus on preventing catastrophic forgetting (Diaz-Rodriguez et al., 2018; Delange et al., 2021). As a result, they neglect some knowledge transfer properties. Furthermore, they assume that all problems in a sequence share the same input space. Finally, scaling these methods to a large sequence of problems remains a challenge. Modular approaches to deep learning decompose a deep neural network into sub-networks, referred to as modules. Each module can then be trained to perform an atomic transformation, specialised in processing a distinct subset of inputs. This modular approach to storing knowledge makes it easy to only reuse the subset of modules which are useful for the task at hand. This thesis introduces a line of research which demonstrates the merits of a modular approach to lifelong machine learning, and its ability to address the aforementioned shortcomings of other methods. Compared to previous work, we show that a modular approach can be used to achieve more LML properties than previously demonstrated. Furthermore, we develop tools which allow modular LML algorithms to scale in order to retain said properties on longer sequences of problems. First, we introduce HOUDINI, a neurosymbolic framework for modular LML. HOUDINI represents modular deep neural networks as functional programs and accumulates a library of pre-trained modules over a sequence of problems. Given a new problem, we use program synthesis to select a suitable neural architecture, as well as a high-performing combination of pre-trained and new modules. We show that our approach has most of the properties desired from an LML algorithm. Notably, it can perform forward transfer, avoid negative transfer and prevent catastrophic forgetting, even across problems with disparate input domains and problems which require different neural architectures. Second, we produce a modular LML algorithm which retains the properties of HOUDINI but can also scale to longer sequences of problems. To this end, we fix the choice of a neural architecture and introduce a probabilistic search framework, PICLE, for searching through different module combinations. To apply PICLE, we introduce two probabilistic models over neural modules which allows us to efficiently identify promising module combinations. Third, we phrase the search over module combinations in modular LML as black-box optimisation, which allows one to make use of methods from the setting of hyperparameter optimisation (HPO). We then develop a new HPO method which marries a multi-fidelity approach with model-based optimisation. We demonstrate that this leads to improvement in anytime performance in the HPO setting and discuss how this can in turn be used to augment modular LML methods. Overall, this thesis identifies a number of important LML properties, which have not all been attained in past methods, and presents an LML algorithm which can achieve all of them, apart from backward transfer

    Reasoning about quantities and concepts: studies in social learning

    We live and learn in a ‘society of mind’. This means that we form beliefs not just based on our own observations and prior expectations but also based on the communications from other people, such as our social network peers. Across seven experiments, I study how people combine their own private observations with other people’s communications to form and update beliefs about the environment. I will follow the tradition of rational analysis and benchmark human learning against optimal Bayesian inference at Marr’s computational level. To accommodate human resource constraints and cognitive biases, I will further contrast human learning with a variety of process level accounts. In Chapters 2–4, I examine how people reason about simple environmental quantities. I will focus on the effect of dependent information sources on the success of group and individual learning across a series of single-player and multi-player judgement tasks. Overall, the results from Chapters 2–4 highlight the nuances of real social network dynamics and provide insights into the conditions under which we can expect collective success versus failures such as the formation of inaccurate worldviews. In Chapter 5, I develop a more complex social learning task which goes beyond estimation of environmental quantities and focuses on inductive inference with symbolic concepts. Here, I investigate how people search compositional theory spaces to form and adapt their beliefs, and how symbolic belief adaptation interfaces with individual and social learning in a challenging active learning task. Results from Chapter 5 suggest that people might explore compositional theory spaces using local incremental search; and that it is difficult for people to use another person’s learning data to improve upon their hypothesis

    Towards computing low-makespan solutions for multi-arm multi-task planning problems

    We propose an approach to find low-makespan solutions to multi-robot multi-task planning problems in environments where robots block each other from completing tasks simultaneously. We introduce a formulation of the problem that allows for an approach based on greedy descent with random restarts for generation of the task assignment and task sequence. We then use a multi-agent path planner to evaluate the makespan of a given assignment and sequence. The planner decomposes the problem into multiple simple subproblems that only contain a single robots and a single task, and can thus be solved quickly to produce a solution for a fixed task sequence. The solutions to the subproblems are then combined to form a valid solution to the original problem. We showcase the approach on robotic stippling and robotic bin picking with up to 4 robot arms.     On the Mathematics of RNA Velocity II: Algorithmic Aspects

    In a previous paper [CSIAM Trans. Appl. Math. 2 (2021), 1-55], the authors proposed a theoretical framework for the analysis of RNA velocity, which is a promising concept in scRNA-seq data analysis to reveal the cell state-transition dynamical processes underlying snapshot data. The current paper is devoted to the algorithmic study of some key components in RNA velocity workflow. Four important points are addressed in this paper: (1) We construct a rational time-scale fixation method which can determine the global gene-shared latent time for cells. (2) We present an uncertainty quantification strategy for the inferred parameters obtained through the EM algorithm. (3) We establish the optimal criterion for the choice of velocity kernel bandwidth with respect to the sample size in the downstream analysis and discuss its implications. (4) We propose a temporal distance estimation approach between two cell clusters along the cellular development path. Some illustrative numerical tests are also carried out to verify our analysis. These results are intended to provide tools and insights in further development of RNA velocity type methods in the future.Comment: 32 pages, 5 figure

    The main objective of this doctoral thesis is the design, validation and implementation of methodologies that allow the geometric and topological modelling of navigable spaces, whether inside buildings or urban environments, to be integrated into three-dimensional geographic information systems (GIS-3D). The input data of this work will consist mainly of point clouds (which can be classified) acquired by LiDAR systems both indoors and outdoors. In addition, the use of BIM infrastructure models and cadastral maps is proposed depending on their availability. Point clouds provide a large amount of environmental information with high accuracy compared to data offered by other acquisition technologies. However, the lack of data structure and volume requires a great deal of processing effort. For this reason, the first step is to structure the data by dividing the input cloud into simpler entities that facilitate subsequent processes. For this first division, the physical elements present in the cloud will be considered, since they can be walls in the case of interior environments or kerbs in the case of exteriors. In order to generate navigation routes adapted to different mobile agents, the next objective will try to establish a semantic subdivision of space according to the functionalities of space. In the case of internal environments, it is possible to use BIM models to evaluate the results and the use of cadastral maps that support the division of the urban environment. Once the navigable space is divided, the design of topologically coherent navigation networks will be parameterized both geometrically and topologically. For this purpose, several spatial discretization techniques, such as 3D tessellations, will be studied to facilitate the establishment of topological relationships, adjacency, connectivity and inclusion between subspaces. Based on the geometric characterization and the topological relations established in the previous phase, the creation of three-dimensional navigation networks with multimodal support will be addressed and different levels of detail will be considered according to the mobility specifications of each agent and its purpose. Finally, the possibility of integrating the networks generated in a GIS-3D visualization system will be considered. For the correct visualization, the level of detail can be adjusted according to geometry and semantics. Aspects such as the type of user or transport, mobility, rights of access to spaces, etc. They must be considered at all times.El objetivo principal de esta tesis doctoral es el diseño, la validación y la implementación de metodologías que permitan el modelado geométrico y topológico de espacios navegables, ya sea de interiores de edificios o entornos urbanos, para integrarse en sistemas de información geográfica tridimensional (SIG). -3D). Los datos de partida de este trabajo consistirán principalmente en nubes de puntos (que pueden estar clasificados) adquiridas por sistemas LiDAR tanto en interiores como en exteriores. Además, se propone el uso de modelos BIM de infraestructuras y mapas catastrales en función de su disponibilidad. Las nubes de puntos proporcionan una gran cantidad de información del entorno con gran precisión con respecto a los datos ofrecidos por otras tecnologías de adquisición. Sin embargo, la falta de estructura de datos y su volumen requiere un gran esfuerzo de procesamiento. Por este motivo, el primer paso que se debe realizar consiste en estructurar los datos dividiendo la nube de entrada en entidades más simples que facilitan los procesos posteriores. Para esta primera división se considerarán los elementos físicos presentes en la nube, ya que pueden ser paredes en el caso de entornos interiores o bordillos en el caso de los exteriores. Con el propósito de generar rutas de navegación adaptadas a diferentes agentes móviles, el próximo objetivo intentará establecer una subdivisión semántica del espacio de acuerdo con las funcionalidades del espacio. En el caso de entornos internos, es posible utilizar modelos BIM para evaluar los resultados y el uso de mapas catastrales que sirven de apoyo en la división del entorno urbano. Una vez que se divide el espacio navegable, se parametrizará tanto geométrica como topológicamente al diseño de redes de navegación topológicamente coherentes. Para este propósito, se estudiarán varias técnicas de discretización espacial, como las teselaciones 3D, para facilitar el establecimiento de relaciones topológicas, la adyacencia, la conectividad y la inclusión entre subespacios. A partir de la caracterización geométrica y las relaciones topológicas establecidas en la fase anterior, se abordará la creación de redes de navegación tridimensionales con soporte multimodal y se considerarán diversos niveles de detalle según las especificaciones de movilidad de cada agente y su propósito. Finalmente, se contemplará la posibilidad de integrar las redes generadas en un sistema de visualización tridimensional 3D SIG 3D. Para la correcta visualización, el nivel de detalle se puede ajustar en función de la geometría y la semántica. Sen embargo, a falta de estrutura dos datos e a seu volume esixe un amplo esforzo de procesado. Por este motivo o primeiro paso a levar a cabo consiste nunha estruturación dos datos mediante a división da nube de entrada en entidades máis sinxelas que faciliten os procesos posteriores. Para esta primeira división consideraranse elementos físicos presentes na nube como poden ser paredes no caso de entornos interiores ou bordillos no caso de exteriores. Coa finalidade de xerar rutas de navegación adaptadas a distintos axentes móbiles, o seguinte obxectivo tratará de establecer unha subdivisión semántica do espazo de acordo as funcionalidades do espazo. No caso de entornos interiores plantease a posibilidade de empregar modelos BIM para avaliar os resultados e o uso de mapas catastrais que sirvan de apoio na división do entorno urbano. Unha vez divido o espazo navigable parametrizarase tanto xeométricamente como topolóxicamene de cara ao deseño de redes de navegación topolóxicamente coherentes. Para este fin estudaranse varias técnicas de discretización de espazos como como son as teselacións 3D co obxectivo de facilitar establecer relacións topolóxicas, de adxacencia, conectividade e inclusión entre subespazos. A partir da caracterización xeométrica e das relación topolóxicas establecidas na fase previa abordarase a creación de redes de navegación tridimensionais con soporte multi-modal e considerando varios niveis de detalle de acordo as especificacións de mobilidade de cada axente e a súa finalidade. Finalmente comtemplarase a posibilidade de integrar as redes xeradas nun sistema SIX 3D visualización tridimensional. Para a correcta visualización o nivel de detalle poderá axustarse en base a xeometría e a semántica. Aspectos como o tipo de usuario ou transporte, mobilidade, dereitos de acceso a espazos, etc. deberán ser considerados en todo momento

    Count-Free Single-Photon 3D Imaging with Race Logic

    Single-photon cameras (SPCs) have emerged as a promising technology for high-resolution 3D imaging. A single-photon 3D camera determines the round-trip time of a laser pulse by capturing the arrival of individual photons at each camera pixel. Constructing photon-timestamp histograms is a fundamental operation for a single-photon 3D camera. However, in-pixel histogram processing is computationally expensive and requires large amount of memory per pixel. Digitizing and transferring photon timestamps to an off-sensor histogramming module is bandwidth and power hungry. Here we present an online approach for distance estimation without explicitly storing photon counts. The two key ingredients of our approach are (a) processing photon streams using race logic, which maintains photon data in the time-delay domain, and (b) constructing count-free equi-depth histograms. Equi-depth histograms are a succinct representation for ``peaky'' distributions, such as those obtained by an SPC pixel from a laser pulse reflected by a surface. Our approach uses a binner element that converges on the median (or, more generally, to another quantile) of a distribution. We cascade multiple binners to form an equi-depth histogrammer that produces multi-bin histograms. Our evaluation shows that this method can provide an order of magnitude reduction in bandwidth and power consumption while maintaining similar distance reconstruction accuracy as conventional processing methods.Comment: Accepted for presentation at the 2023 International Conference on Computational Photograph

    Vulnerability of the Nigerian coast and communities to climate change induced coastal erosion

    Improving coastal resilience to climate change hazards requires understanding past shoreline changes. As the coastal population grows, evaluation and monitoring of shoreline changes are essential for planning and development. Population growth increases exposure to sea level rise and coastal hazards. Nigeria, where the study is situated, is among the top fifteen countries in the world for coastal population exposure to sea level rise. This study provided a novel lens in establishing a link between social factors and the intensifying coastal erosion along the Akwa Ibom State study coast. The mixed-method approach used in the study to assess the vulnerability of the Nigerian coast and communities to climate change-induced coastal erosion proved to be essential in gathering a wide range of data (physical, socio economic, participatory GIS maps and social learning) that contributed to a more robust and holistic assessment of coastal erosion, which is a complex issue due to the interplay between the human and natural environments. Remotely sensed data was used to examine the susceptibility and coastal evolution of Akwa Ibom State over 36 years (1984 -2020). Longer-term (1984- 2020) and short-term (2015-2020) shoreline change analyses were used to understand coastal erosion and accretion. From 1984-2020, the total average linear regression rate (LRR) was - 2.7+0.18m/yr and from 2015-2020, it was -3.94 +1.28m/yr, demonstrating an erosional trend along the study coast. Although the rate of erosion varies along the study coast, the linear regression rates (LRR) results show a predominant trend of erosion in both the short and longer term. According to the 2022 Intergovernmental Panel on Climate Change report, loss of land, loss of assets, community disruption and livelihood, loss of environmental resources, ecosystem, loss of life, or adverse health impact are all potential risks along the African coast due to climate change – this study shows that these risks are already occurring today. To quantify the anticipated future coastal erosion risk by 2040 along the study coast, the findings in this study show an overall average LRR of -2.73+ 0.99 m/yr which anticipates that coastal erosion will still be prevalent along the coast by 2040. And, given the current global climate change situation, should be expected to be much higher than the current forecasting. This study re-conceptualised the European Environmental Agency Driver-Pressure StateImpact-Response (DPSIR) model to show Hazard-Driver-Pressure-State-Impact ResponseObservation causal linkages to coastal erosion hazards. The results showed how human activities and environmental interactions have evolved through time, causing coastal erosion. Removal of vegetation cover/backstop for residential and agricultural purposes, indicate that human activities significantly contribute to the study area's susceptibility, rapid shoreline changes, and vulnerability to coastal erosion, in addition to oceanic and climate change drivers such as sea level rise and storminess. Risk perception of coastal erosion in the study area was analysed using the rhizoanalytic method proposed by Deleueze. The method demonstrates how connections and movements can be related and how data can be used to show multiplicity, mark and unmark ideas, rupture pre-conceptions and make new connections. This study shows that coastal erosion awareness is insufficient to build a long-term management plan and sustain coastal resilience. The Hino's conceptual model which provides in-depth understanding on planned retreat was used to illustrate migratory and planned retreat for the study coast where relocation has already occurred due to coastal erosion. The result fell within the Self-Reliance quadrant, indicating that people left the risk zone without government backing or retreat plans. Other coastal residents who have not relocated fell within the Hunkered Down quadrant, showing that they are willing to stay in the risk zone and cope with the threat unless the government/environmental agencies relocate them. This study shows that coastal resilience requires adaptive capacity and government support. However, multilevel governance has inhibited government-community dialogue and involvement, increasing coastal erosion vulnerability. The coastal vulnerability index to coastal erosion was calculated using the Analytical Hierarchy Process weightings. It revealed that 67.55% of the study coast falls within the high-very high vulnerability class while 32.45% is within the very low-low vulnerability class. This study developed and combined a risk perception index to coastal erosion (RPIerosion) and participatory GIS (PGIS) mapping into a novel coastal vulnerability index called the integrated coastal erosion vulnerability index (ICEVI). The case study evaluation in Akata, showed an improvement in the overall vulnerability assessment to reflect the real-world scenario, which was consistent with field data. This study demonstrated not only the presence and challenges of coastal erosion in the research area but also the relevance of involvement between the local stakeholders, government and environmental agencies. Thus, showing the potential for the perspectives of the inhabitants of these regions to inform the understanding of the resilience capacity of the people impacted, and importantly to inform future co-design and/or selection of effective adaptation methods, to better support coastal climate change resilience in these communities. Overall, the study provides a useful contribution to coastal erosion vulnerability assessments in data-scarce regions more broadly, where the mixed-methods approach used here can be applied elsewhere
