    Partitioning of starter bacteria and added exogenous enzyme activities between curd and whey during Cheddar cheese manufacture

    peer-reviewedPartitioning of starter bacteria and enzyme activities was investigated at different stages of Cheddar cheese manufacture using three exogenous commercial enzyme preparations added to milk or at salting. The enzyme preparations used were: Accelase AM317, Accelase AHC50, Accelerzyme CPG. Flow cytometric analysis indicated that AHC50 or AM317 consisted of permeabilised or dead cells and contained a range of enzyme activities. The CPG preparation contained only carboxypeptidase activity. Approximately 90% of starter bacteria cells partitioned with the curd at whey drainage. However, key enzyme activities partitioned with the bulk whey in the range of 22%–90%. An increased level of enzyme partitioning with the curd was observed for AHC50 which was added at salting, indicating that the mode of addition influenced partitioning. These findings suggest that further scope exists to optimise both bacterial and exogenous enzyme incorporation into cheese curd to accelerate ripening.Department of Agriculture, Food and the Marin

    Development of error criteria for adaptive multi-element polynomial chaos approaches

    International audienceThis paper presents and compares different methodologies to create an adaptive stochastic space partitioning in polynomial chaos applications which use a multi-element approach. To implement adaptive partitioning, Wan and Karniadakis first developed a criterion based on the relative error in local variance. We propose here two different error criteria: one based on the residual error and the other on the local variance discontinuity created by partitioning. The methods are applied to classical differential equations with long-term integration difficulties, including the Kraichnan–Orszag three-mode problem, and to simple linear and nonlinear mechanical systems whose stochastic dynamic responses are investigated. The efficiency and robustness of the approaches are investigated by comparison with Monte-Carlo simulations. For the different examples considered, they show significantly better convergence characteristics than the original error criterion used

    A three-dimensional macroscopic fundamental diagram for mixed bi-modal urban networks

    Recent research has studied the existence and the properties of a macroscopic fundamental diagram (MFD) for large urban networks. The MFD should not be universally expected as high scatter or hysteresis might appear for some type of networks, like heterogeneous networks or freeways. In this paper, we investigate if aggregated relationships can describe the performance of urban bi-modal networks with buses and cars sharing the same road infrastructure and identify how this performance is influenced by the interactions between modes and the effect of bus stops. Based on simulation data, we develop a three-dimensional vehicle MFD (3D-vMFD) relating the accumulation of cars and buses, and the total circulating vehicle flow in the network. This relation experiences low scatter and can be approximated by an exponential-family function. We also propose a parsimonious model to estimate a three-dimensional passenger MFD (3D-pMFD), which provides a different perspective of the flow characteristics in bi-modal networks, by considering that buses carry more passengers. We also show that a constant Bus-Car Unit (BCU) equivalent value cannot describe the influence of buses in the system as congestion develops. We then integrate a partitioning algorithm to cluster the network into a small number of regions with similar mode composition and level of congestion. Our results show that partitioning unveils important traffic properties of flow heterogeneity in the studied network. Interactions between buses and cars are different in the partitioned regions due to higher density of buses. Building on these results, various traffic management strategies in bi-modal multi-region urban networks can then be integrated, such as redistribution of urban space among different modes, perimeter signal control with preferential treatment of buses and bus priority
