    Sensors for product characterization and quality of specialty crops—A review

    This review covers developments in non-invasive techniques for quality analysis and inspection of specialty crops, mainly fresh fruits and vegetables, over the past decade up to the year 2010. Presented and discussed in this review are advanced sensing technologies including computer vision, spectroscopy, X-rays, magnetic resonance, mechanical contact, chemical sensing, wireless sensor networks and radiofrequency identification sensors. The current status of different sensing systems is described in the context of commercial application. The review also discusses future research needs and potentials of these sensing technologies. Emphases are placed on those technologies that have been proven effective or have shown great potential for agro-food applications. Despite significant progress in the development of non-invasive techniques for quality assessment of fruits and vegetables, the pace for adoption of these technologies by the specialty crop industry has been slow

    Development of soft computing and applications in agricultural and biological engineering

    Soft computing is a set of “inexact” computing techniques, which are able to model and analyze very complex problems. For these complex problems, more conventional methods have not been able to produce cost-effective, analytical, or complete solutions. Soft computing has been extensively studied and applied in the last three decades for scientific research and engineering computing. In agricultural and biological engineering, researchers and engineers have developed methods of fuzzy logic, artificial neural networks, genetic algorithms, decision trees, and support vector machines to study soil and water regimes related to crop growth, analyze the operation of food processing, and support decision-making in precision farming. This paper reviews the development of soft computing techniques. With the concepts and methods, applications of soft computing in the field of agricultural and biological engineering are presented, especially in the soil and water context for crop management and decision support in precision agriculture. The future of development and application of soft computing in agricultural and biological engineering is discussed

    Computer vision based classification of fruits and vegetables for self-checkout at supermarkets

    The field of machine learning, and, in particular, methods to improve the capability of machines to perform a wider variety of generalised tasks are among the most rapidly growing research areas in today’s world. The current applications of machine learning and artificial intelligence can be divided into many significant fields namely computer vision, data sciences, real time analytics and Natural Language Processing (NLP). All these applications are being used to help computer based systems to operate more usefully in everyday contexts. Computer vision research is currently active in a wide range of areas such as the development of autonomous vehicles, object recognition, Content Based Image Retrieval (CBIR), image segmentation and terrestrial analysis from space (i.e. crop estimation). Despite significant prior research, the area of object recognition still has many topics to be explored. This PhD thesis focuses on using advanced machine learning approaches to enable the automated recognition of fresh produce (i.e. fruits and vegetables) at supermarket self-checkouts. This type of complex classification task is one of the most recently emerging applications of advanced computer vision approaches and is a productive research topic in this field due to the limited means of representing the features and machine learning techniques for classification. Fruits and vegetables offer significant inter and intra class variance in weight, shape, size, colour and texture which makes the classification challenging. The applications of effective fruit and vegetable classification have significant importance in daily life e.g. crop estimation, fruit classification, robotic harvesting, fruit quality assessment, etc. One potential application for this fruit and vegetable classification capability is for supermarket self-checkouts. Increasingly, supermarkets are introducing self-checkouts in stores to make the checkout process easier and faster. However, there are a number of challenges with this as all goods cannot readily be sold with packaging and barcodes, for instance loose fresh items (e.g. fruits and vegetables). Adding barcodes to these types of items individually is impractical and pre-packaging limits the freedom of choice when selecting fruits and vegetables and creates additional waste, hence reducing customer satisfaction. The current situation, which relies on customers correctly identifying produce themselves leaves open the potential for incorrect billing either due to inadvertent error, or due to intentional fraudulent misclassification resulting in financial losses for the store. To address this identified problem, the main goals of this PhD work are: (a) exploring the types of visual and non-visual sensors that could be incorporated into a self-checkout system for classification of fruits and vegetables, (b) determining a suitable feature representation method for fresh produce items available at supermarkets, (c) identifying optimal machine learning techniques for classification within this context and (d) evaluating our work relative to the state-of-the-art object classification results presented in the literature. An in-depth analysis of related computer vision literature and techniques is performed to identify and implement the possible solutions. A progressive process distribution approach is used for this project where the task of computer vision based fruit and vegetables classification is divided into pre-processing and classification techniques. Different classification techniques have been implemented and evaluated as possible solution for this problem. Both visual and non-visual features of fruit and vegetables are exploited to perform the classification. Novel classification techniques have been carefully developed to deal with the complex and highly variant physical features of fruit and vegetables while taking advantages of both visual and non-visual features. The capability of classification techniques is tested in individual and ensemble manner to achieved the higher effectiveness. Significant results have been obtained where it can be concluded that the fruit and vegetables classification is complex task with many challenges involved. It is also observed that a larger dataset can better comprehend the complex variant features of fruit and vegetables. Complex multidimensional features can be extracted from the larger datasets to generalise on higher number of classes. However, development of a larger multiclass dataset is an expensive and time consuming process. The effectiveness of classification techniques can be significantly improved by subtracting the background occlusions and complexities. It is also worth mentioning that ensemble of simple and less complicated classification techniques can achieve effective results even if applied to less number of features for smaller number of classes. The combination of visual and nonvisual features can reduce the struggle of a classification technique to deal with higher number of classes with similar physical features. Classification of fruit and vegetables with similar physical features (i.e. colour and texture) needs careful estimation and hyper-dimensional embedding of visual features. Implementing rigorous classification penalties as loss function can achieve this goal at the cost of time and computational requirements. There is a significant need to develop larger datasets for different fruit and vegetables related computer vision applications. Considering more sophisticated loss function penalties and discriminative hyper-dimensional features embedding techniques can significantly improve the effectiveness of the classification techniques for the fruit and vegetables applications

    Exploring Hyperspectral Imaging and 3D Convolutional Neural Network for Stress Classification in Plants

    Hyperspectral imaging (HSI) has emerged as a transformative technology in imaging, characterized by its ability to capture a wide spectrum of light, including wavelengths beyond the visible range. This approach significantly differs from traditional imaging methods such as RGB imaging, which uses three color channels, and multispectral imaging, which captures several discrete spectral bands. Through this approach, HSI offers detailed spectral signatures for each pixel, facilitating a more nuanced analysis of the imaged subjects. This capability is particularly beneficial in applications like agricultural practices, where it can detect changes in physiological and structural characteristics of crops. Moreover, the ability of HSI to monitor these changes over time is advantageous for observing how subjects respond to different environmental conditions or treatments. However, the high-dimensional nature of hyperspectral data presents challenges in data processing and feature extraction. Traditional machine learning algorithms often struggle to handle such complexity. This is where 3D Convolutional Neural Networks (CNNs) become valuable. Unlike 1D-CNNs, which extract features from spectral dimensions, and 2D-CNNs, which focus on spatial dimensions, 3D CNNs have the capability to process data across both spectral and spatial dimensions. This makes them adept at extracting complex features from hyperspectral data. In this thesis, we explored the potency of HSI combined with 3D-CNN in agriculture domain where plant health and vitality are paramount. To evaluate this, we subjected lettuce plants to varying stress levels to assess the performance of this method in classifying the stressed lettuce at the early stages of growth into their respective stress-level groups. For this study, we created a dataset comprising 88 hyperspectral image samples of stressed lettuce. Utilizing Bayesian optimization, we developed 350 distinct 3D-CNN models to assess the method. The top-performing model achieved a 75.00\% test accuracy. Additionally, we addressed the challenge of generating valid 3D-CNN models in the Keras Tuner library through meticulous hyperparameter configuration. Our investigation also extends to the role of individual channels and channel groups within the color and near-infrared spectrum in predicting results for each stress-level group. We observed that the red and green spectra have a higher influence on the prediction results.     Revelation of Yin-Yang Balance in Microbial Cell Factories by Data Mining, Flux Modeling, and Metabolic Engineering

    The long-held assumption of never-ending rapid growth in biotechnology and especially in synthetic biology has been recently questioned, due to lack of substantial return of investment. One of the main reasons for failures in synthetic biology and metabolic engineering is the metabolic burdens that result in resource losses. Metabolic burden is defined as the portion of a host cells resources either energy molecules (e.g., NADH, NADPH and ATP) or carbon building blocks (e.g., amino acids) that is used to maintain the engineered components (e.g., pathways). As a result, the effectiveness of synthetic biology tools heavily dependents on cell capability to carry on the metabolic burden. Although genetic modifications can effectively engineer cells and redirect carbon fluxes toward diverse products, insufficient cell ATP powerhouse is limited to support diverse microbial activities including product synthesis. Here, I employ an ancient Chinese philosophy (Yin-Yang) to describe two contrary forces that are interconnected and interdependent, where Yin represents energy metabolism in the form of ATP, and Yang represents carbon metabolism. To decipher Yin-Yang balance and its implication to microbial cell factories, this dissertation applied metabolic engineering, flux analysis, data mining tools to reveal cell physiological responses under different genetic and environmental conditions. Firstly, a combined approach of FBA and 13C-MFA was employed to investigate several engineered isobutanol-producing strains and examine their carbon and energy metabolism. The result indicated isobutanol overproduction strongly competed for biomass building blocks and thus the addition of nutrients (yeast extract) to support cell growth is essential for high yield of isobutanol. Based on the analysis of isobutanol production, \u27Yin-Yang\u27 theory has been proposed to illustrate the importance of carbon and energy balance in engineered strains. The effects of metabolic burden and respiration efficiency (P/O ratio) on biofuel product were determined by FBA simulation. The discovery of energy cliff explained failures in bioprocess scale-ups. The simulation also predicted that fatty acid production is more sensitive to P/O ratio change than alcohol production. Based on that prediction, fatty acid producing strains have been engineered with the insertion of Vitreoscilla hemoglobin (VHb), to overcome the intracellular energy limitation by improving its oxygen uptake and respiration efficiency. The result confirmed our hypothesis and different level of trade-off between the burden and the benefit from various introduced genetic components. On the other side, a series of computational tools have been developed to accelerate the application of fluxomics research. Microbesflux has been rebuilt, upgraded, and moved to a commercial server. A platform for fluxomics study as well as an open source 13C-MFA tool (WUFlux) has been developed. Further, a computational platform that integrates machine learning, logic programming, and constrained programming together has been developed. This platform gives fast predictions of microbial central metabolism with decent accuracy. Lastly, a framework has been built to integrate Big Data technology and text mining to interpret concepts and technology trends based on the literature survey. Case studies have been performed, and informative results have been obtained through this Big Data framework within five minutes. In summary, 13C-MFA and flux balance analysis are only tools to quantify cell energy and carbon metabolism (i.e., Yin-Yang Balance), leading to the rational design of robust high-producing microbial cell factories. Developing advanced computational tools will facilitate the application of fluxomics research and literature analysis
