    Oscillatory mechanisms of conscious perception and attention

    Although the prominent role of neural oscillations in perception and cognition has been continuously investigated, some critical questions remain unanswered. My PhD thesis was aimed at addressing some of them. First, can we dissociate oscillatory underpinnings of perceptual accuracy and subjective awareness? Current work would strongly suggest that this dissociation can be drawn. While the fluctuations in alpha-amplitude decide perceptual bias and metacognitive abilities, the speed of alpha activity (i.e., alpha-frequency) dictates sensory sampling, shaping perceptual accuracy. Second, how are these oscillatory mechanisms integrated during attention? The obtained results indicate that a top-down visuospatial mechanism modulates neural assemblies in visual areas via oscillatory re-alignment and coherence in the alpha/beta range within the fronto-parietal brain network. These perceptual predictions are reflected in the retinotopically distributed posterior alpha-amplitude, while perceptual accuracy is explained by the higher alpha-frequency at the to-be-attended location. Finally, sensory input, elaborated via fast gamma oscillations, is linked to specific phases of this slower activity via oscillatory nesting, enabling integration of the feedback-modulated oscillatory activity with sensory information. Third, how can we relate this oscillatory activity to other neural markers of behaviour (i.e., event-related potentials)? The obtained results favour the oscillatory model of ERP genesis, where alpha-frequency shapes the latency of early evoked-potentials, namely P1, with both neural indices being related to perceptual accuracy. On the other hand, alpha-amplitude dictates the amplitude of later P3 evoked-response, whereas both indices shape subjective awareness. Crucially, by combining different methodological approaches, including neurostimulation (TMS) and neuroimaging (EEG), current work identified these oscillatory-behavior links as causal and not just as co-occurring events. Current work aimed at ameliorating the use of the TMS-EEG approach by explaining inter-individual differences in the stimulation outcomes, which could be proven crucial in the way we design entrainment experiments and interpret the results in both research and clinical settings

    Geometric Data Analysis: Advancements of the Statistical Methodology and Applications

    Data analysis has become fundamental to our society and comes in multiple facets and approaches. Nevertheless, in research and applications, the focus was primarily on data from Euclidean vector spaces. Consequently, the majority of methods that are applied today are not suited for more general data types. Driven by needs from fields like image processing, (medical) shape analysis, and network analysis, more and more attention has recently been given to data from non-Euclidean spaces–particularly (curved) manifolds. It has led to the field of geometric data analysis whose methods explicitly take the structure (for example, the topology and geometry) of the underlying space into account. This thesis contributes to the methodology of geometric data analysis by generalizing several fundamental notions from multivariate statistics to manifolds. We thereby focus on two different viewpoints. First, we use Riemannian structures to derive a novel regression scheme for general manifolds that relies on splines of generalized Bézier curves. It can accurately model non-geodesic relationships, for example, time-dependent trends with saturation effects or cyclic trends. Since Bézier curves can be evaluated with the constructive de Casteljau algorithm, working with data from manifolds of high dimensions (for example, a hundred thousand or more) is feasible. Relying on the regression, we further develop a hierarchical statistical model for an adequate analysis of longitudinal data in manifolds, and a method to control for confounding variables. We secondly focus on data that is not only manifold- but even Lie group-valued, which is frequently the case in applications. We can only achieve this by endowing the group with an affine connection structure that is generally not Riemannian. Utilizing it, we derive generalizations of several well-known dissimilarity measures between data distributions that can be used for various tasks, including hypothesis testing. Invariance under data translations is proven, and a connection to continuous distributions is given for one measure. A further central contribution of this thesis is that it shows use cases for all notions in real-world applications, particularly in problems from shape analysis in medical imaging and archaeology. We can replicate or further quantify several known findings for shape changes of the femur and the right hippocampus under osteoarthritis and Alzheimer's, respectively. Furthermore, in an archaeological application, we obtain new insights into the construction principles of ancient sundials. Last but not least, we use the geometric structure underlying human brain connectomes to predict cognitive scores. Utilizing a sample selection procedure, we obtain state-of-the-art results

    A Survey on Causal Discovery: Theory and Practice

    Understanding the laws that govern a phenomenon is the core of scientific progress. This is especially true when the goal is to model the interplay between different aspects in a causal fashion. Indeed, causal inference itself is specifically designed to quantify the underlying relationships that connect a cause to its effect. Causal discovery is a branch of the broader field of causality in which causal graphs is recovered from data (whenever possible), enabling the identification and estimation of causal effects. In this paper, we explore recent advancements in a unified manner, provide a consistent overview of existing algorithms developed under different settings, report useful tools and data, present real-world applications to understand why and how these methods can be fruitfully exploited

    Impact of Mild Traumatic Brain Injury and Orbitofrontal Cortex Lesion on Affective and Cognitive Brain Functions

    Lievää aivovammaa kutsutaan joskus näkymättömäksi vammaksi. Lievän aivovamman saaneet toipuvat yleensä hyvin, mutta yllättävän suuri osa kärsii pitkittyneistä oireista, vaikka tavanomaiset kuvantamistutkimukset ja neuropsykologiset testitulokset näyttäytyvät normaaleina. Myös orbitofrontaalialueen vammaan liittyy ristiriita koettujen kognitiivisten ja tunne- elämän oireiden sekä objektiivisten neuropsykologisten löydösten välillä, vaikka vamma itsessään näkyy selkeästi jo tietokonetomografiakuvissa. Orbitofrontaalinen aivokuori integroi tunnepitoista tietoa toimintaamme ja alueen vaurio heikentää tätä prosessia. Oireet ja haasteet tunteiden ohjaamassa käytöksessä ja tunnesäätelyssä sekä tarkkaavuudessa ja toiminnanohjauksessa ovat yleisiä lievän aivovamman ja orbitofrontaalialueen vaurion jälkeen, mutta niille altistavat hermostolliset mekanismit tunnetaan huonosti. Tämän väitöskirjan tavoitteena oli ymmärtää paremmin hermostollisia mekanismeja, jotka altistavat lievän aivovamman ja orbitofrontaalialueen vaurion saaneet kognitiivisille ja tunne-elämän oireille. Tavoitteena oli myös valottaa orbitofrontaalialueen roolia tunteiden ja tarkkaavuuden vuorovaikutuksessa ja aivovamman vaikutusta tähän. Tämän lisäksi pyrittiin kehittämään aivosähkökäyrään perustuva biomarkkeri lievälle aivovammalle. Tutkimme lievän aivovamman saaneita (n = 27), orbitofrontaalialueen vaurion saaneita (n = 16) sekä terveitä verrokkeja. Vamman aiheuttamia muutoksia tarkasteltiin aivojen fysiologisten ilmiöiden, herätevasteiden (ERP) ja frontaalisen alfa-asymmetrian (FAA) avulla. Aivosähkökäyrää (EEG) mitattiin kognitiivisen tehtävän (Executive RT Test) suorituksen aikana. Tehtävässä piti joko reagoida (Go) tai olla reagoimatta (NoGo) ärsykkeisiin sääntöjen mukaan ja samanaikaisesti esitettiin uhkaavia tai neutraaleja ärsykkeitä, jotka olivat joko tehtävän kannalta oleellisia ärsykkeitä tai epäoleellisia häiriöärsykkeitä. Tarkkaavuutta ja kognitiivista kontrollia kuvaavia herätevasteita (N2, P3 ja N2P3) sekä tehtäväsuoritusta vertailtiin ryhmien välillä. Tutkimme myös heijastaisiko FAA ja kehittämämme uusi indeksi, uhkaärsykkeen muovaama FAA (eFAA), lievän aivovamman jälkeisiä oireita. Lievän aivovamman saaneet raportoivat enemmän tunne-elämän oireita kuin verrokit. He myös reagoivat nopeammin uhkaärsykkeeseen sen ollessa vastaamisen kannalta oleellinen Go-tilanteessa ja tässä yhteydessä nähtiin suuremmat N2P3- amplitudit. NoGo-tilanteessa suuremmat N2P3-amplitudit nähtiin uhkaärsykkeen ollessa sekä tehtävän kannalta oleellinen ärsyke että epäoleellinen häiriöärsyke. Lievän aivovamman saaneilla havaittiin oikeavoittoinen FAA, joka heijastaa suhteellisesti suurempaa aktiviteettia oikealla otsalohkoalueella verrattuna vasempaan, ja tällainen aktivaation epäsuhta on liitetty masennusalttiuteen. eFAA kykeni erottamaan lievän aivovamman saaneista ne, joilla oli aivovamman jälkeisiä oireita ja korreloi subjektiivisten vamman jälkioireiden ja masennusoireiden kanssa. Orbitofrontaalialueen vaurion saaneilla oli suuremmat N2P3-amplitudit tehtävän kannalta oleellisen uhkaärsykkeen yhteydessä. Uhkaärsykkeen ollessa epäoleellinen häiriöärsyke, orbitofrontaalisen vaurion saaneet eivät kyenneet suuntaamaan tarkkaavuusresursseja siihen nopeasti mutta tunneärsykkeen prosessointiin liittyvät myöhäiset herätevasteaallot olivat suurentuneet. Orbitofrontaalisen vaurion saaneilla oli enemmän vaikeuksia kognitiivisessa tehtävässä, jossa tunneärsyke oli aina epäoleellinen häiriöärsyke, mutta he jopa paransivat suoritustaan uhkaärsykkeen ollessa tehtävän kannalta oleellinen. He raportoivat myös verrokkeja enemmän toiminnanohjauksen vaikeuksia sekä aivovamman jälkeisiä oireita. Tässä väitöskirjassa havaittiin muutoksia tunteiden, tarkkaavuuden ja toiminnanohjauksen vuorovaikutuksessa orbitofrontaalivaurioon ja lievään aivovammaan liittyen. Lievän aivovamman saaneet suuntasivat verrokkeja enemmän tarkkaavuutta sekä oleellisiin että epäoleellisiin uhkaärsykkeisiin kun taas orbitofrontaalisen vaurion saaneet kohdistivat verrokkeja enemmän tarkkaavuutta vain tehtävän kannalta oleellisiin uhkaärsykkeisiin. Ylikorostunut tarkkaavuuden suuntaaminen uhkaan voi altistaa tunne-elämän oireille ja masennukselle. On mahdollista, että havaitut muutokset heijastavat vaurioita etuotsalohkojen ja sen alaisten alueiden välisissä verkostoissa, jotka ovat tärkeitä tunnesäätelyssä. While normally subjects with MTBI recover well, remarkably many suffer from prolonged symptoms despite intact imaging and neuropsychological test results. A similar discrepancy between subjective cognitive and affective complaints and challenges in daily life, but intact neuropsychological test results are frequently encountered in patients with orbitofrontal cortex (OFC) lesion. The OFC has an important role in integrating emotional information into appropriate actions, with lesion to the OFC causing deficits in these processes. Symptoms and challenges related to alterations in emotion guided behaviors, emotion regulation, attention and executive functions (EFs) are common after MTBI and OFC injury, however, the underlying neural mechanisms predisposing to them remain unclear. In this thesis, we aimed to better understand the neural mechanisms underlying affective and cognitive symptoms in MTBI and OFC injury. We also aimed at unraveling the role of intact OFC in the interplay between emotion and attention and the impact of OFC lesion and MTBI on it. In addition, we aimed to develop a novel electroencephalography (EEG) based biomarker of MTBI. Subjects with MTBI (n = 27) and OFC lesion (n = 16) were included in the studies along with healthy control subjects. We used event-related potentials (ERPs) and frontal alpha asymmetry (FAA) to study injury-related alterations in brain physiology underlying emotion-attention interaction and cognitive control. EEG was recorded during a cognitive task (Executive RT Test), where threatening and neutral stimuli (task-relevant or irrelevant) were embedded into a Go-NoGo task. We assessed whether attention and cognitive control -related ERP components (N2, P3, N2P3) and task-performance differed between subjects with MTBI or OFC lesion and healthy controls. We further evaluated whether task-induced FAA and a novel index, emotional modulation of FAA (eFAA), reflect post-concussion symptoms (PCS) after MTBI. Self-report questionnaires of PCS, depression symptoms and EFs in daily life were collected to study correlations between symptom scores and objective physiological measures. The MTBI group reported more emotional symptoms than control subjects. They also had larger N2P3 ERP amplitudes in context of task-relevant threat in the Go-condition coupled with faster reaction times. In the NoGo-condition, larger N2P3 amplitudes were detected both in context of task-relevant and task-irrelevant threat in the MTBI group, and more errors were committed with task-irrelevant threat. Further, rightward FAA i.e., relatively more activity on the right compared to the left frontal regions, a pattern associated with vulnerability to depression was observed in subjects with MTBI. eFAA distinguished subjects with MTBI and prolonged PCS from those without symptoms and from controls. Moreover, the eFAA obtained during the Executive RT Test was negatively correlated with subjective reports of depressive and post-concussion symptoms. The OFC group exhibited larger N2P3 amplitudes in context with task-relevant threat in both Go- and NoGo-conditions. In contrast, when confronted with threat stimuli that were presented only as task-irrelevant distractors, they didn’t display an initial N2P3 increase but demonstrated pronounced late positive waves, indicative of emotional processing. The OFC group had generally worse task-performance when the task included emotional stimuli only as task-irrelevant distractors, whereas their task-performance seemed to slightly benefit from task-relevant threat. The OFC group also reported more challenges in EFs and more PCS than controls. To summarize, we detected alterations in interactions of affective, attentional and EF processing after MTBI and OFC lesion. Subjects with MTBI allocated more attention to both task-irrelevant and task-relevant threat than controls while subjects with OFC lesion allocated more attention particularly to task-relevant threat. Alterations in attention to threat may contribute to emotional symptoms and vulnerability to depression in MTBI. We suggest the observed alterations reflect disruption of frontal-subcortical circuits subserving emotion-cognition interactions. The OFC is important in balancing voluntary and involuntary attention, especially in gearing attention to task-irrelevant emotion. Lesion to the OFC leads to challenges in swift allocation of attention to task-irrelevant but emotionally relevant stimuli. This may underlie some of the daily challenges experienced after OFC lesion, where timely evaluation of emotional stimuli and its appropriate integration into actions is crucial. The results of this thesis shed new light on the role of OFC in attention to emotion and on the possible mechanisms of prolonged symptoms after MTBI. Finally, we introduce a novel biomarker for MTBI, known as eFAA, reflecting alterations in affective and cognitive brain circuits and functions due to brain injury

    Variation in How Cognitive Control Modulates Sentence Processing

    Prior research suggests that cognitive control can assist the comprehension of sentences that create conflict between interpretations, at least under some circumstances. However, the mixed pattern of results suggests that cognitive control may not always be necessary for accurate comprehension. We tested whether cognitive control recruitment for language processing is systematically variable, depending on the type of sentential ambiguity or conflict, individual differences in cognitive control, and task demands. Participants completed two sessions in a web-based experiment. The first session tested conflict modulation using interleaved Stroop and sentence comprehension trials. Critical sentences contained syntax-semantics or phrase-attachment conflict. In the second session, participants completed three cognitive control and three working memory tasks. Exploratory factor analysis was used to index individual differences in a cognitive control factor and a working memory factor. At the group level, there were no significant conflict modulation effects for either syntax-semantics or phrase-attachment conflict. At the individual differences level, the cognitive control factor correlated with offline comprehension accuracy but not online processing measures for both types of conflict. Together, the results suggest that the role of cognitive control in sentence processing may vary according to task demands. When overt decisions are required, individual differences in cognitive control may matter such that better cognitive control results in better language comprehension performance. The results add to the mixed evidence on conflict modulation and raise questions about the situations under which cognitive control influences online processing

    Improving passive driver fatigue, sitting health risk factors and user experience in automobiles. Conception, development and evaluation of a novel interactive seating system

    Lange Fahrten in Automobilen werden oftmals von monotoniebedingter Ermüdung durch mangelnde Stimulation sowie körperlichen Gesundheitsrisiken durch langanhaltendes Sitzen begleitet. In der Literatur werden kognitive Zusatzaufgaben als Maßnahme gegen monotoniebedingte Ermüdung vorgeschlagen. Der Stand der Technik um gesundheitliche Risiken durch langanhaltendes Sitzen im Fahrzeug zu reduzieren, beschränkt sich hauptsächlich auf Sitzsysteme, welche die Passagiere passiv mobilisieren. Mit dem Ziel die Fahrsicherheit zu erhöhen und gleichzeitig Gesundheitsrisiken zu reduzieren, wurde ein neuartiges interaktives Sitzsystem (IASS) konzipiert, entwickelt und evaluiert. Das IASS soll den Fahrer/die Fahrerin oder die Passagiere dazu motivieren mit dem Sitz zu interagieren und dabei aktive Bewegungen gegen die Sitzfläche auszuführen. Im Gegensatz zu aktuellen interaktiven Sitzsystemen erfolgt die grundlegende Interaktion nicht über ein Display. Stattdessen werden auditive Einsprachen, Luftblasen in der Lehne, Vibrationsmotoren sowie die Ambientebeleuchtung genutzt. Ein Display kommt lediglich bei der Einführung in die Nutzung des Systems oder ergänzend für die Passagiere zum Einsatz. Das ermöglicht, dass der Fahrer/die Fahrerin den Blick und damit auch die Aufmerksamkeit auf der Straße behalten kann. Dadurch soll das System, im Gegensatz zum Stand der Technik, nicht nur für die Passagiere, sondern auch für den Fahrer/die Fahrerin geeignet sein. Das wiederum eröffnet die Möglichkeit das IASS als Zusatzaufgabe neben dem Fahren zu nutzen, um einer momotoniebedingten Reduktion der Aufmerksamkeit entgegenzuwirken. Dabei ist anzumerken, dass die erhöhte Fahrsicherheit durch eine verbesserte Aufmerksamkeit nicht nur den Insassen, sondern auch den anderen Verkehrsteilnehmern dient. Gleichzeitig haben alle Passagiere die Möglichkeit, durch die physiologisch sinnvollen Bewegungen gegen den Sitz, die Gesundheitsrisiken durch langanhaltendes Sitzen zu reduzieren. Insgesamt wurden drei Probandenstudien durchgeführt, um das IASS zu entwickeln und zu evaluieren. Die erste Probandenstudie diente der Entwicklung des IASS. Um die Bewegungen des Passagiers/der Passagierin bei der Interaktion zu detektieren, wurden Drucksensoren in der Sitzlehne integriert. Zur Gewährleistung einer zuverlässigen Bewegungserkennung von Passagieren mit unterschiedlichen anthropometrischen Merkmalen, war es entscheidend, die Sensoren an optimalen Positionen in der Lehne zu platzieren. Daher war der erste Schritt im Entwicklungsprozess des IASS die Durchführung einer Probandenstudie, um diese optimalen Positionen festzulegen. Im Rahmen der Studie wurden Sitzdruckverteilungsbilder erfasst und anschließend mit einer Methode, dem Statistical Parametric Mapping ausgewertet, die für gewöhnlich bei der funktionellen Magnetresonanztomographie eingesetzt wird. In der vorliegenden Arbeit wurde diese Methode erstmalig zur Auswertung von Sitzdruckverteilungen herangezogen. Der Vorteil gegenüber herkömmlichen Methoden ist die hohe örtliche Auflösung. Damit war es möglich, die Sensoranbringungspunkte sehr präzise zu definieren. Anschließend wurde das IASS auf Basis der in der ersten Studie gewonnenen Information aufgebaut und anschließend in zwei weiteren Probandenstudien mit einem aktuellen Sitzmassagesystem (MS) hinsichtlich Wirksamkeit verglichen. In der zweiten Probandestudie sollte das Potential des IASS für die Fahrsicherheit bewertet werden. Die im Fahrsimulator durchgeführte Studie zeigte, dass das IASS die monotoniebedingte Ermüdung reduzierte, während dieser Effekt bei Nutzung des MS nicht auftrat. Das IASS wurde ebenfalls bezüglich des Nutzererlebnisses sowie der emotionalen Wahrnehmung gegenüber dem MS bevorzugt. Außerdem bewerteten die Probanden und Probandinnen, dass das IASS im Vergleich zum MS, sowohl Komfort wesentlich stärker erhöhte als auch Diskomfort stärker reduzierte. In der dritten und abschließenden Probandenstudie wurde untersucht, ob das IASS den körperlichen Gesundheitsrisiken durch langanhaltendes Sitzen besser entgegen wirkt als das MS. Hierfür wurden beide Sitzsysteme in einem Serienfahrzeug verbaut. Um die Effekte beider Sitzsysteme zu vergleichen, wurden gesundheitsrelevante Parameter erfasst. Damit fahrtbedingte Signalstörungen ausgeschlossen werden konnten, wurde die Studie im stehenden Fahrzeug durchgeführt. Das Elektrokardiogramm zeigte, dass ausschließlich das IASS die Herzfrequenz erhöhte. Elektromyographische Messungen zeigten außerdem, dass das IASS die Aktivität in den sechs erfassten Muskeln erhöhte, während beim MS lediglich ein Muskel eine Tendenz zur Aktivitätserhöhung zeigte. Entsprechend sind die gesundheitsfördernden Effekte des IASS im Vergleich zum MS als wesentlich stärker einzustufen. Nach dem Kenntnisstand des Autors, ist dies die erste wissenschaftliche Publikation welche eine motorische Zusatzaufgabe als Maßnahme gegen monotoniebedingte Ermüdung im Fahrzeug untersucht. Darüber hinaus wurde erstmalig direkt verglichen, ob ein automobiles Sitzsystem welches zu aktiven Bewegungen animiert, einem System das die Passagiere passiv bewegt, hinsichtlich der Reduktion der negativen körperlichen Effekte des Sitzens überlegen ist

    Multimodal connectivity of the human basal forebrain

    The cholinergic innervation of the cortex originates from neurons in the basal forebrain (BF) and plays a crucial role in cognitive processing. However, it is unclear how the organization of BF cholinergic neurons in the human brain is related to their functional and structural integration with the cortex. To address this, we have used high-resolution 7 Tesla diffusion and resting-state functional MRI to examine multimodal forebrain cholinergic connectivity with the neocortex in humans. Discrete parcellation analyses revealed that structural and functional parcellation broadly differentiated the anteromedial from posterolateral nuclei of BF. Next, we used gradient estimation to capture more fine-grained connectivity profile of the BF-cortical projectome and found moving from anteromedial to posterolateral BF, structural and functional gradients became progressively detethered, with the most pronounced dissimilarity localized in the nucleus basalis of Meynert (NbM). Additionally, functional but not structural connectivity with the BF grew stronger at shorter geodesic distances, with weakly myelinated transmodal cortical areas most strongly expressing this divergence. Moreover, [18F] FEOBV PET imaging was used to demonstrate that these transmodal cortical areas are also among the most densely innervated regions. This intrinsic BF cholinergic connectivity map of cortex was compared with meta-analytic connectivity map of cholinergic modulation on attention, demonstrating that patterns of brain activity evoked by directed attention are altered by pharmacological activation of acetylcholine (ACh) compared to placebo and these patterns spatially overlap with the intrinsic BF cholinergic connectivity map. Altogether, our findings provide new insights into how cholinergic signaling is organized in the human brain

Directe Elektrische Stimulatie (DES) is de techniek die momenteel wordt gezien als de gouden standaard om tijdens wakkere hersenoperaties belangrijke hersenfuncties in kaart te brengen en te bewaken. Deze techniek wordt gebruikt om de hoeveelheid tumorweefsel dat weggehaald zal worden af te wegen tegen het behouden van functioneel belangrijke gebieden. DES is ontwikkeld om tijdens een operatie vast te kunnen stellen of specifieke hersengebieden of hersenverbindingen, een belangrijke rol spelen bij hersenfuncties zoals taal en motoriek. Het bewaken van taal en motoriek levert een bewezen betere uitkomst door het voorkomen van verlammingen of taalstoornissen. De laatste jaren zijn verschillende neurochirurgische centra deze techniek ook gaan gebruiken om andere functies te monitoren, zoals executieve functies. Executieve functies zijn de regelfuncties van de hersenen die nodig zijn voor het realiseren van doelgericht en aangepast gedrag, zoals werkgeheugen, inhibitie en cognitieve flexibiliteit. Deze functies zijn van essentieel belang om alledaagse taken uit te voeren, zoals boodschappen doen, koken, autorijden, studeren en je beroep uitoefenen. De reden om executieve functies te monitoren is dat stoornissen in deze functies veel voorkomen bij patiënten met een glioom, zowel voor als na de operatie. Het is echter nog onbekend of specifieke hersenverbindingen, ook wel subcorticale banen genoemd, onmisbaar zijn voor executieve functies, waardoor de wetenschappelijke onderbouwing en klinische relevantie om deze functies te monitoren tijdens wakkere hersenoperaties voorlopig ontbreekt. Het hoofddoel van dit proefschrift was om te onderzoeken of specifieke hersenverbindingen betrokken zijn bij executieve functies en gemonitord kunnen worden met een gestandaardiseerde set van taken tijdens wakkere chirurgie bij patiënten met een glioom. Om meer inzicht te verkrijgen in de functionele rol van subcorticale banen, en ter voorbereiding op de ontwikkeling van gestandaardiseerd monitoring protocol, hebben we structurele en functionele laesie-symptoom studies verricht en de resultaten van deze studies gecombineerd met bevindingen uit de literatuur. De bevindingen toonden onder andere dat specifieke hersenbanen, zoals de Frontal Aslant Tract en de rechter en linker Superior Longitudinal Fasciculus III betrokken zijn bij executieve functies en voorzag ons van argumenten om ze tijdens wakkere hersenoperaties te monitoren. Samen met de klinische ervaring van de afdeling neurochirurgie van het ETZ Tilburg en dat van drie andere neurochirurgische centra (Lariboisière Parijs, EMC Rotterdam, UMC Utrecht), hebben we een protocol ontwikkeld om deze hersenbanen te monitoren. Dit protocol omvat een set van executieve taken die op dezelfde manier in de verschillende centra worden afgenomen. Met dit protocol werd overeenstemming bereikt voor klinisch haalbare en wetenschappelijk onderbouwde intra-operatieve cognitieve monitoring. Hiermee werd een pad gebaand voor toekomstig onderzoek dat zich richt op de mate waarin de monitoring van deze functies leidt tot een vermindering van cognitieve problemen na een hersentumoroperatie en daarmee tot een betere kwaliteit van het sociale en professionele leven

    Space, time and motion in a multisensory world

    When interacting with environmental events, humans acquire information from different senses and combine these inputs within a coherent representation of the world. The present doctoral thesis aims at investigating how humans represent space, time, and motion through auditory and visual sensory modalities. It has been widely demonstrated a predisposition of different sensory systems towards the processing of different domains of representation, with hearing that prevails in representing the time domain and vision that is the most reliable sense for processing the space domain. Given this strong link between sensory modality and domain of representation, one objective of this thesis is to deepen the knowledge of the neural organization of multisensory spatial and temporal skills in healthy adults. In addition, by using blindness as a model to unravel the role of vision in the development of spatio-temporal abilities, this thesis explores the interaction of the spatial and temporal domains in the acoustic motion perception of early blind individuals. The interplay between space and time has also been explained as the result of humans performing actions in the surrounding environment since to carry out goal-directed motor behaviors it is useful for a person to associate the spatial and temporal information of one’s target into a shared mental map. In this regard, the present project also questions how the brain processes spatio-temporal cues of external events when it comes to manually intercepting moving objects with one hand. Finally, in light of the above results, this dissertation incorporates the development of a novel portable device, named MultiTab, for the behavioral evaluation of the processing of space, time, and motor responses, through the visual and acoustic sensory modality. For the purposes of this thesis, four methodological approaches have been employed: i) electroencephalogram (EEG) technique, to explore the cortical activation associated with multisensory spatial and temporal tasks; ii) psychophysical methods, to measure the relationship between stimuli in motion and the acoustic speed perception of blind and sighted individuals; iii) motion capture techniques, to measure indices of movements during an object’s interception task; iv) design and technical-behavioral validation of a new portable device. Studies of the present dissertation indicate the following results. First, this thesis highlights an early cortical gain modulation of sensory areas that depends on the domain of representation to process, with auditory areas mainly involved in the multisensory processing of temporal inputs, and visual areas of spatial inputs. Moreover, for the spatial domain specifically, the neural modulation of visual areas is also influenced by the kind of spatial layout representing multisensory stimuli. Second, this project shows that lack of vision influences the ability to process the speed of moving sounds by altering how blind individuals make use of the sounds’ temporal features. This result suggests that visual experience in the first years of life is a crucial factor when dealing with combined spatio-temporal information. Third, data of this thesis demonstrate that typically developing individuals manually intercepting a moving object with one hand take into consideration the item’s spatio-temporal cues, by adjusting their interceptive movements according to the object’s speed. Finally, the design and validation of MultiTab show its utility in the evaluation of multisensory processing such as the manual localization of audiovisual spatialized stimuli. Overall, findings from this thesis contribute to a more in-depth picture of how the human brain represents space, time, and motion through different senses. Moreover, they provide promising implications in exploring novel technological methods for the assessment and training of these dimensions in typical and atypical populations
