    Robot docking using mixtures of Gaussians

    This paper applies the Mixture of Gaussians probabilistic model, combined with Expectation Maximization optimization to the task of summarizing three dimensionals range data for the mobile robot. This provides a flexible way of dealing with uncertainties in sensor information, and allows the introduction of prior knowledge into low-level perception modules. Problems with the basic approach were solved in several ways: the mixture of Gaussians was reparameterized to reflect the types of objects expected in the scene, and priors on model parameters were included in the optimization process. Both approaches force the optimization to find 'interesting' objects, given the sensor and object characteristics. A higher level classifier was used to interpret the results provided by the model, and to reject spurious solutions

    Visual Servoing from Deep Neural Networks

    We present a deep neural network-based method to perform high-precision, robust and real-time 6 DOF visual servoing. The paper describes how to create a dataset simulating various perturbations (occlusions and lighting conditions) from a single real-world image of the scene. A convolutional neural network is fine-tuned using this dataset to estimate the relative pose between two images of the same scene. The output of the network is then employed in a visual servoing control scheme. The method converges robustly even in difficult real-world settings with strong lighting variations and occlusions.A positioning error of less than one millimeter is obtained in experiments with a 6 DOF robot.Comment: fixed authors lis

    Where's Waldo at time t? Using spatio-temporal models for mobile robot search

    We present a novel approach to mobile robot search for non-stationary objects in partially known environments. We formulate the search as a path planning problem in an environment where the probability of object occurrences at particular locations is a function of time. We propose to explicitly model the dynamics of the object occurrences by their frequency spectra. Using this spectral model, our path planning algorithm can construct plans that reflect the likelihoods of object locations at the time the search is performed. Three datasets collected over several months containing person and object occurrences in residential and office environments were chosen to evaluate the approach. Several types of spatio-temporal models were created for each of these datasets and the efficiency of the search method was assessed by measuring the time it took to locate a particular object. The results indicate that modeling the dynamics of object occurrences reduces the search time by 25% to 65% compared to maps that neglect these dynamics

    Vesicle adhesion and fusion studied by small-angle x-ray scattering.

    We have studied the adhesion state (also denoted by docking state) of lipid vesicles as induced by the divalent ions Ca2+ or Mg2+ at well-controlled ion concentration, lipid composition, and charge density. The bilayer structure and the interbilayer distance in the docking state were analyzed by small-angle x-ray scattering. A strong adhesion state was observed for DOPC:DOPS vesicles, indicating like-charge attraction resulting from ion correlations. The observed interbilayer separations of ∼1.6 nm agree quantitatively with the predictions of electrostatics in the strong coupling regime. Although this phenomenon was observed when mixing anionic and zwitterionic (or neutral) lipids, pure anionic membranes (DOPS) with highest charge density σ resulted in a direct phase transition to a multilamellar state, which must be accompanied by rupture and fusion of vesicles.     Computational Techniques for Stochastic Reachability

    As automated control systems grow in prevalence and complexity, there is an increasing demand for verification and controller synthesis methods to ensure these systems perform safely and to desired specifications. In addition, uncertain or stochastic behaviors are often exhibited (such as wind affecting the motion of an aircraft), making probabilistic verification desirable. Stochastic reachability analysis provides a formal means of generating the set of initial states that meets a given objective (such as safety or reachability) with a desired level of probability, known as the reachable (or safe) set, depending on the objective. However, the applicability of reachability analysis is limited in the scope and size of system it can address. First, generating stochastic reachable or viable sets is computationally intensive, and most existing methods rely on an optimal control formulation that requires solving a dynamic program, and which scales exponentially in the dimension of the state space. Second, almost no results exist for extending stochastic reachability analysis to systems with incomplete information, such that the controller does not have access to the full state of the system. This thesis addresses both of the above limitations, and introduces novel computational methods for generating stochastic reachable sets for both perfectly and partially observable systems. We initially consider a linear system with additive Gaussian noise, and introduce two methods for computing stochastic reachable sets that do not require dynamic programming. The first method uses a particle approximation to formulate a deterministic mixed integer linear program that produces an estimate to reachability probabilities. The second method uses a convex chance-constrained optimization problem to generate an under-approximation to the reachable set. Using these methods we are able to generate stochastic reachable sets for a four-dimensional spacecraft docking example in far less time than it would take had we used a dynamic program. We then focus on discrete time stochastic hybrid systems, which provide a flexible modeling framework for systems that exhibit mode-dependent behavior, and whose state space has both discrete and continuous components. We incorporate a stochastic observation process into the hybrid system model, and derive both theoretical and computational results for generating stochastic reachable sets subject to an observation process. The derivation of an information state allows us to recast the problem as one of perfect information, and we prove that solving a dynamic program over the information state is equivalent to solving the original problem. We then demonstrate that the dynamic program to solve the reachability problem for a partially observable stochastic hybrid system shares the same properties as for a partially observable Markov decision process (POMDP) with an additive cost function, and so we can exploit approximation strategies designed for POMDPs to solve the reachability problem. To do so, however, we first generate approximate representations of the information state and value function as either vectors or Gaussian mixtures, through a finite state approximation to the hybrid system or using a Gaussian mixture approximation to an indicator function defined over a convex region. For a system with linear dynamics and Gaussian measurement noise, we show that it exhibits special properties that do not require an approximation of the information state, which enables much more efficient computation of the reachable set.     A multi-hypothesis approach for range-only simultaneous localization and mapping with aerial robots

    Los sistemas de Range-only SLAM (o RO-SLAM) tienen como objetivo la construcción de un mapa formado por la posición de un conjunto de sensores de distancia y la localización simultánea del robot con respecto a dicho mapa, utilizando únicamente para ello medidas de distancia. Los sensores de distancia son dispositivos capaces de medir la distancia relativa entre cada par de dispositivos. Estos sensores son especialmente interesantes para su applicación a vehículos aéreos debido a su reducido tamaño y peso. Además, estos dispositivos son capaces de operar en interiores o zonas con carencia de señal GPS y no requieren de una línea de visión directa entre cada par de dispositivos a diferencia de otros sensores como cámaras o sensores laser, permitiendo así obtener una lectura de datos continuada sin oclusiones. Sin embargo, estos sensores presentan un modelo de observación no lineal con una deficiencia de rango debido a la carencia de información de orientación relativa entre cada par de sensores. Además, cuando se incrementa la dimensionalidad del problema de 2D a 3D para su aplicación a vehículos aéreos, el número de variables ocultas del modelo aumenta haciendo el problema más costoso computacionalmente especialmente ante implementaciones multi-hipótesis. Esta tesis estudia y propone diferentes métodos que permitan la aplicación eficiente de estos sistemas RO-SLAM con vehículos terrestres o aéreos en entornos reales. Para ello se estudia la escalabilidad del sistema en relación al número de variables ocultas y el número de dispositivos a posicionar en el mapa. A diferencia de otros métodos descritos en la literatura de RO-SLAM, los algoritmos propuestos en esta tesis tienen en cuenta las correlaciones existentes entre cada par de dispositivos especialmente para la integración de medidas estÃa˛ticas entre pares de sensores del mapa. Además, esta tesis estudia el ruido y las medidas espúreas que puedan generar los sensores de distancia para mejorar la robustez de los algoritmos propuestos con técnicas de detección y filtración. También se proponen métodos de integración de medidas de otros sensores como cámaras, altímetros o GPS para refinar las estimaciones realizadas por el sistema RO-SLAM. Otros capítulos estudian y proponen técnicas para la integración de los algoritmos RO-SLAM presentados a sistemas con múltiples robots, así como el uso de técnicas de percepción activa que permitan reducir la incertidumbre del sistema ante trayectorias con carencia de trilateración entre el robot y los sensores de destancia estáticos del mapa. Todos los métodos propuestos han sido validados mediante simulaciones y experimentos con sistemas reales detallados en esta tesis. Además, todos los sistemas software implementados, así como los conjuntos de datos registrados durante la experimentación han sido publicados y documentados para su uso en la comunidad científica

    DNA-Origami-Based Fluorescence Brightness Standards for Convenient and Fast Protein Counting in Live Cells

    Fluorescence microscopy has been one of the most discovery-rich methods in biology. In the digital age, the discipline is becoming increasingly quantitative. Virtually all biological laboratories have access to fluorescence microscopes, but abilities to quantify biomolecule copy numbers are limited by the complexity and sophistication associated with current quantification methods. Here, we present DNA-origami-based fluorescence brightness standards for counting 5–300 copies of proteins in bacterial and mammalian cells, tagged with fluorescent proteins or membrane-permeable organic dyes. Compared to conventional quantification techniques, our brightness standards are robust, straightforward to use, and compatible with nearly all fluorescence imaging applications, thereby providing a practical and versatile tool to quantify biomolecules via fluorescence microscopy

    Advances in Simultaneous Localization and Mapping in Confined Underwater Environments Using Sonar and Optical Imaging.

    This thesis reports on the incorporation of surface information into a probabilistic simultaneous localization and mapping (SLAM) framework used on an autonomous underwater vehicle (AUV) designed for underwater inspection. AUVs operating in cluttered underwater environments, such as ship hulls or dams, are commonly equipped with Doppler-based sensors, which---in addition to navigation---provide a sparse representation of the environment in the form of a three-dimensional (3D) point cloud. The goal of this thesis is to develop perceptual algorithms that take full advantage of these sparse observations for correcting navigational drift and building a model of the environment. In particular, we focus on three objectives. First, we introduce a novel representation of this 3D point cloud as collections of planar features arranged in a factor graph. This factor graph representation probabalistically infers the spatial arrangement of each planar segment and can effectively model smooth surfaces (such as a ship hull). Second, we show how this technique can produce 3D models that serve as input to our pipeline that produces the first-ever 3D photomosaics using a two-dimensional (2D) imaging sonar. Finally, we propose a model-assisted bundle adjustment (BA) framework that allows for robust registration between surfaces observed from a Doppler sensor and visual features detected from optical images. Throughout this thesis, we show methods that produce 3D photomosaics using a combination of triangular meshes (derived from our SLAM framework or given a-priori), optical images, and sonar images. Overall, the contributions of this thesis greatly increase the accuracy, reliability, and utility of in-water ship hull inspection with AUVs despite the challenges they face in underwater environments. We provide results using the Hovering Autonomous Underwater Vehicle (HAUV) for autonomous ship hull inspection, which serves as the primary testbed for the algorithms presented in this thesis.     A Self-Adaptive Online Brain Machine Interface of a Humanoid Robot through a General Type-2 Fuzzy Inference System

    This paper presents a self-adaptive general type-2 fuzzy inference system (GT2 FIS) for online motor imagery (MI) decoding to build a brain-machine interface (BMI) and navigate a bi-pedal humanoid robot in a real experiment, using EEG brain recordings only. GT2 FISs are applied to BMI for the first time in this study. We also account for several constraints commonly associated with BMI in real practice: 1) maximum number of electroencephalography (EEG) channels is limited and fixed, 2) no possibility of performing repeated user training sessions, and 3) desirable use of unsupervised and low complexity features extraction methods. The novel learning method presented in this paper consists of a self-adaptive GT2 FIS that can both incrementally update its parameters and evolve (a.k.a. self-adapt) its structure via creation, fusion and scaling of the fuzzy system rules in an online BMI experiment with a real robot. The structure identification is based on an online GT2 Gath-Geva algorithm where every MI decoding class can be represented by multiple fuzzy rules (models). The effectiveness of the proposed method is demonstrated in a detailed BMI experiment where 15 untrained users were able to accurately interface with a humanoid robot, in a single thirty-minute experiment, using signals from six EEG electrodes only
