Impaired regulation of emotion: Neural correlates of reappraisal and distraction in bipolar disorder and unaffected relatives
Deficient emotion regulation has been proposed as a crucial pathological mechanism in bipolar disorder (BD). We therefore investigated emotion regulation impairments in BD, the related neural underpinnings and their etiological relevance for the disorder. Twenty-two euthymic patients with bipolar-I disorder and 17 unaffected first-degree relatives of BD-I patients, as well as two groups of healthy gender-, age- and education-matched controls (N=22/17, respectively) were included. Participants underwent functional magnetic resonance imaging while applying two different emotion regulation techniques, reappraisal and distraction, when presented with emotional images. BD patients and relatives showed impaired downregulation of amygdala activity during reappraisal, but not during distraction, when compared with controls. This deficit was correlated with the habitual use of reappraisal. The negative connectivity of amygdala and orbitofrontal cortex (OFC) observed during reappraisal in controls was reversed in BD patients and relatives. There were no significant differences between BD patients and relatives. As being observed in BD patients and unaffected relatives, deficits in emotion regulation through reappraisal may represent heritable neurobiological abnormalities underlying BD. The neural mechanisms include impaired control of amygdala reactivity to emotional stimuli and dysfunctional connectivity of the amygdala to regulatory control regions in the OFC. These are, thus, important aspects of the neurobiological basis of increased vulnerability for BD
Affective Response to a Loved One's Pain: Insula Activity as a Function of Individual Differences
Individual variability in emotion processing may be associated with genetic variation as well as with psychological predispositions such as dispositional affect styles. Our previous fMRI study demonstrated that amygdala reactivity was independently predicted by affective-cognitive styles (phobic prone or eating disorders prone) and genotype of the serotonin transporter in a discrimination task of fearful facial expressions. Since the insula is associated with the subjective evaluation of bodily states and is involved in human feelings, we explored whether its activity could also vary in function of individual differences. In the present fMRI study, the association between dispositional affects and insula reactivity has been examined in two groups of healthy participants categorized according to affective-cognitive styles (phobic prone or eating disorders prone). Images of the faces of partners and strangers, in both painful and neutral situations, were used as visual stimuli. Interaction analyses indicate significantly different activations in the two groups in reaction to a loved one's pain: the phobic prone group exhibited greater activation in the left posterior insula. These results demonstrate that affective-cognitive style is associated with insula activity in pain empathy processing, suggesting a greater involvement of the insula in feelings for a certain cohort of people. In the mapping of individual differences, these results shed new light on variability in neural networks of emotion

The influence of self-reported ethnic origin and mood on elicited emotion and brain reactivity to happy and sad social films
This thesis was submitted for the degree of Docter of Philosophy and awarded by Brunel University.In recent years Social Neuroscience has started to investigate how mood and culture influence social and emotional situations. In the present study differences in elicited emotion and neural activation were investigated when participants viewed films depicting social interactions. Film clips are preferred stimuli for elicitation of emotion in laboratory studies, but given the lack of standardised film sets in the literature, two behavioural studies were conducted prior to imaging. The first study (147 females, 30 males; 98.8% 18 to 24 years) identified a set of clips that elicited emotion profiles in which the target emotion (happy, sad) was strongest, as well as neutral clips, and demonstrated an effect of participants’ stable mood. The second study (143 females, 19 males; mean age 19.2 years) optimised the stimulus set and demonstrated effects of self-reported ethnic origin, mood and interest on profiles of elicited emotion. In the fMRI investigation 33 female and 8 male participants (mean age 19.2 years) viewed film clips in a block design experiment with loose and tight t-contrasts and retrospective ratings of elicited emotion. Across all-participants, social interaction depicting sadness activated key emotion-related structures such as left amygdala and insula, and medial frontal cortex that were not significantly activated with social interaction depicting happiness. However, greater activation was observed for Europeans than for non-Europeans in orbitofrontal cortex, anterior and posterior cingulate for happy social interaction and in hippocampus, precuneus and retrosplenial cortex for sad social interaction. Individual differences in trait emotions and stable mood were measured with PANAS-X. For high fatigue participants greater activation was observed in the left amgydala for happy social interaction. For participants with high positive stable mood greater activation was observed in the insula for happy and sad social interaction. The research described here indicates that self-reported ethnic origin and mood are potentially significant influences on elicited emotion and brain reactivity to positive and negative social and emotional situations
Neural Correlates of Promotion and Prevention Goal Activation: An fMRI Study using an Idiographic Approach
Regulatory focus theory [Higgins, E. T. Beyond pleasure and pain. American Psychologist, 52, 1280-1300, 1997] postulates two social-cognitive motivational systems, the promotion and prevention systems, for self-regulation of goal pursuit. However, the neural substrates of promotion and prevention goal activation remain unclear. Drawing on several literatures, we hypothesized that priming promotion versus prevention goals would activate areas in the left versus right prefrontal cortex (PFC), respectively, and that activation in these areas would be correlated with individual differences in chronic regulatory focus. Sixteen participants underwent functional magnetic resonance imaging while engaged in a depth-of-processing task, during which they were exposed incidentally to their own promotion and prevention goals. Task-related cortical activation was consistent with previous studies. At the same time, incidental priming of promotion goals was associated with left orbital PFC activation, and activation in this area was stronger for individuals with a chronic promotion focus. Findings regarding prevention goal priming were not consistent with predictions. The data illustrate the centrality of self-regulation and personal goal pursuit within the multilayered process of social cognition

Separable Neural Components in the Processing of Black and White Faces
In a study of the neural components of automatic and controlled social evaluation, White participants viewed Black and White faces during event-related functional magnetic resonance imaging. When the faces were presented for 30 ms, activation in the amygdala—a brain region associated with emotion—was greater for Black than for White faces. When the faces were presented for 525 ms, this difference was significantly reduced, and regions of frontal cortex associated with control and regulation showed greater activation for Black than White faces. Furthermore, greater race bias on an indirect behavioral measure was correlated with greater difference in amygdala activation between Black and White faces, and frontal activity predicted a reduction in Black-White differences in amygdala activity from the 30-ms to the 525-ms condition. These results provide evidence for neural distinctions between automatic and more controlled processing of social groups, and suggest that controlled processes may modulate automatic evaluation.Psycholog
Down-Regulation of Negative Emotional Processing by Transcranial Direct Current Stimulation: Effects of Personality Characteristics
Evidence from neuroimaging and electrophysiological studies indicates that the left dorsolateral prefrontal cortex (DLPFC) is a core region in emotional processing, particularly during down-regulation of negative emotional conditions. However, emotional regulation is a process subject to major inter-individual differences, some of which may be explained by personality traits. In the present study we used transcranial direct current stimulation (tDCS) over the left DLPFC to investigate whether transiently increasing the activity of this region resulted in changes in the ratings of positive, neutral and negative emotional pictures. Results revealed that anodal, but not cathodal, tDCS reduced the perceived degree of emotional valence for negative stimuli, possibly due to an enhancement of cognitive control of emotional expression. We also aimed to determine whether personality traits (extraversion and neuroticism) might condition the impact of tDCS. We found that individuals with higher scores on the introversion personality dimension were more permeable than extraverts to the modulatory effects of the stimulation. The present study underlines the role of the left DLPFC in emotional regulation, and stresses the importance of considering individual personality characteristics as a relevant variable, although replication is needed given the limited sample size of our study
Neural mechanisms of affective instability and cognitive control in substance use
Objective: We explored the impact of affect on cognitive control as this relates to individual differences in affective instability and substance use. Toward this end, we examined how different dimensions of affective instability interact to predict substance misuse and the effect of this on two event-related potential components, the reward positivity and the late positive potential, which are said to reflect the neural mechanisms of reward and emotion processing, respectively.
Methods: We recorded the ongoing electroencephalogram from undergraduate students as they navigated two T-maze tasks in search of rewards. One of the tasks included neutral, pleasant, and unpleasant pictures from the International Affective Picture System. Participants also completed several questionnaires pertaining to substance use and personality.
Results: A principal components analysis revealed a factor related to affective instability, which we named reactivity. This factor significantly predicted increased substance use. Individuals reporting higher levels of affective reactivity also displayed a larger reward positivity following stimuli with emotional content.
Conclusion: The current study uncovered a group of high-risk substance users who were characterized by greater levels of affective reactivity and context-specific increased sensitivity to rewards.
Significance: These results help to elucidate the complex factors underlying substance use and may facilitate the creation of individually-tailored treatment programs for those struggling with substance use disorders
